Abstract— The isolation of a mutant, strain PM-9, of Rhodopseudonionus spheroides with an abnormal complement of carotenoid pigments is described.
PM-9 accumulates phytoene, phytofluene, ζ-carotene and neurosporene. Semi-aerobic cultures form more ζ-carotene and neurosporene relative to total carotenoids than do photo-synthetic cultures.
PM-9 is killed on exposure to light and oxygen.
By making use of the effect of oxygen on the nature of the carotenoids in PM-9, we have shown that these pigments do not directly influence the in vivo spectrum of bacteriochlorophyll.
Diphenylamine inhibits the synthesis of coloured carotenoids in Rhodopseudonionos gelatinosa but does not change the bacteriochlorophyll spectrum.  相似文献

Abstract— –The presumed photoreceptor for phototaxis, the paraflagellar body, in the flagellate Euglena gracilis , was isolated still attached to the flagellum. After solubilization, fast protein liquid chromatography (FPLC) analysis yielded four major protein fractions with the chromophoric groups still attached. Fluorescence spectra showed that three fractions had excitation peaks at 380 nm and emission peaks around 450 nm indicative of pterins, while the fourth chromoprotein had a fluorescence emission at 520 nm and an excitation peak at 450 nm, indicative of a flavin. The separated proteins were analyzed by gel electrophoresis: the pterin binding proteins have apparent molecular masses between 27 000 and 31 600 and the flavin binding protein has an apparent molecular mass of 33 500.  相似文献

Abstract— The photosynthetic accessory pigments R-phycoerythrin and C-phycocyanin were extracted from a red sea-weed and a filamentous blue-green alga. Investigation of quenching of phycoerythrin fluorescence by phycocyanin at room and liquid nitrogen temperatures yielded rates for the SternVolmer quenching constants ( Kq From these values, rate constants k(D–A) were calculated for the transfer of energy from phycoerythrin to phycocyanin.
The transfer rate constants obtained were such that a collisional transfer mechanism was ruled out, and were of similar order of magnitude as the rate constants for transfer of electronic energy in mixed organic crystals.  相似文献

Abstract— The pigments in vivo of single cells of Chlorella pyrenoidosa were studied by the microspectrophotometric technique. An accessory recording the first derivative of absorption was used to obtain fine resolution and enhanced accuracy. The results suggest that there are several long-wavelength components of Chl a in vivo. In addition, there seem to be four short-wave forms of Chl a. It is also likely that Chl b exists in vivo in two different forms. The existence of all these forms was demonstrated at room temperature.  相似文献

以4,4′,4″ 三氨基三苯胺为母体,以含有4种不同取代基的萘酚AS为偶合组分合成了一系列三芳胺三偶氮类有机光导颜料.用元素分析、红外光谱、紫外光谱、质谱和X射线衍射光谱对结构进行了表征.研究结果表明,含硫杂环三偶氮类有机光导颜料在520-730nm之间都有较强吸收.  相似文献

ON THE MECHANISM OF WAVELENGTH REGULATION IN VISUAL PIGMENTS   总被引:9,自引:0,他引:9  
Abstract— The contributions of different factors that might be responsible for the 500 nm absorption maximum of bovine rhodopsin are evaluated in detail. These include: (1) electrostatic interactions between the chromophore and a charged amino acid on the apoprotein; (2) exciton interactions with aromatic amino acids; (3) twisting about single bonds which have considerable double bond character; (4) weak interactions between the Schiff base and a putative counter-ion. Analysis of these mechanisms in terms of theoretical and experimental results suggests that(2–4) are all capable of contributing to the protein induced spectral shifts. However, the "external point charge" model proposed previously, i.e. mechanism (1), appears to provide the crucial interaction. In this paper, the theoretical basis for this model is discussed in detail. The model is briefly evaluated in light of the amino-acid sequence of bovine rhodopsin and possible implications for other visual pigments are considered.  相似文献

When the synthetic estrogen and tumourogenic compound diethylstilbestrol is exposed to horseradish peroxidase (HRP) and H2O2 in the presence of the cationic surfactant hexadecyltrimethylammonium bromide (CTAB), a burst of oxygen consumption and concomitant light emission are observed. The quinone form of the product is not seen in the absorption spectrum because CTAB strongly catalyses its conversion to Z,Z-dienestrol. The emission spectrum shows several peaks. Total emission is dramatically enhanced by chlorophyll and by xanthene dyes. A key intermediate in chemiexcitation is 4-hydroxypropiophenone. The ability to promote chemiexcitation is retained through various generations of metabolites, giving origin to a cascade of excited states. Since the biological effects of diethylstilbestrol appear to be connected with its peroxidative metabolism, chemiexcitation may eventually prove to be of importance in, for example, toxicity of the drug.  相似文献

Abstract— Intact disks and inverted disks were prepared from bovine retinal rods and the distribution in the disk membrane of such enzymes as guanyl cyclase, cyclic nucleotide phosphodiesterase, GTP binding protein (GTPase), 5'-nucleotidase and rhodopsin kinase was investigated. Guanyl cyclase was not detected in the disk; the enzyme activity was high in a membranous fraction containing the cilium or axoneme and the rod outer segment plasma membrane. Cyclic nucleotide phosphodiesterase, GTP binding protein (GTPase) and rhodopsin kinase were associated on the external surface of disk in the presence of 2 m M Mg2+. The enzymes dissociated from the membrane when Mg2+ was depleted. Thus, magnesium ion seems to regulate the state of these enzymes in the outer segment. 5'-Nucleotidase activity was low in intact disks but was significantly enhanced after inversion of the disk. The catalytic site of the enzyme, therefore, must be located on the internal (intradiscal) surface. Since the disks are known to be formed by invagination of the plasma membrane, 5'-nucleotidase, by inference, would have its catalytic site exposed on the external surface of the plasma membrane. Preliminary experiments showed that the capability of light-activated rhodopsin to activate cyclic nucleotide phosphodiesterase was inhibited by phosphorylation of the pigment. This supports the idea that rhodopsin kinase, cyclic nucleotide phosphodiesterase and GTPase exist as a functional complex on rod membranes.  相似文献

Abstract— The regeneration processes of visual pigments and the dark adaptation processes of rod photoreceptor after irradiation by bleaching light were studied by spectrophotometric, electroretinographic(ERG) methods and the measurement of early receptor potentials (ERPs) in bullfrog retina. After irradiation by bleaching light, rhodopsin in the isolated retina regenerated to an extent depending on the wavelength and intensity of the bleaching light as well as pH. Intense blue light and a weak alkaline environment (pH 7.5–9.5) favoured the regeneration. The regeneration of pigment in the green rods could not be detected in these experiments on the isolated retina. The regeneration of cone pigment was studied by measuring ERPs from both isolated retinas and retinas with pigment epithelium-choroid complex separated from scleras, which are called PEC-retinas. In the PEC-retinas, cone pigment regenerated more rapidly and with better efficiency than in the isolated retinas.
Rod photoreceptors desensitized permanently by bleaching light did not demonstrate hypersensitivity at 0.1 m M [Ca2+]out, which induced hypersensitivity in non-desensitized photoreceptor, but showed the hypersensitivity when the [Ca2+]out, was lowered further by the addition of EGTA.  相似文献

The human red and green cone pigments differ at either 15 or 16 amino acids, depending upon which polymorphic variants are compared. Seven of these amino acid differences involve the introduction or removal of a hydroxyl group. One of these differences, a substitution of alanine for senne at position 80, was found previously to produce a 5 nm blue shift. To determine the role of the remaining six hydroxyl group differences in tuning the absorption spectra of the human red and green pigments, we have studied six site-directed mutants in which single amino acids from the green pigment have been substituted for the corresponding residues in the red pigment. Blue shifts of 7 and 14 nm were observed upon substitution of phenylalanine for tyrosine at position 277 and alanine for threonine at position 285, respectively. Single substitutions at positions 65, 230, 233, and 309 produced spectral shifts of 1 nm or less. These data are in good agreement with a model based upon sequence comparisons among primate pigments and with the properties of site-directed mutants of bovine rhodopsin. Nonadditive effects observed in comparing the absorption spectra of red-green hybrid pigments remain to be explained.  相似文献

Effects of Photofrin II on energy metabolism and metabolic viability were studied in a mammalian transformed cell line (BHK-21) in dark and after photo-irradiation with visible light. Cells were allowed to accumulate Photofrin by incubating for 4 h in buffer containing Photofrin (5-60 micrograms/ml). The results show that Photofrin significantly affects the cellular energy metabolism even in the absence of light; activity of cytochrome c oxidase is decreased and glucose utilization and lactate production (glycolysis) are increased. Irradiation with light resulted in a significant decrease in the activity of cytochrome c oxidase, glycolysis, ATP content, energy charge, ratios of adenine nucleotides like ATP/ADP, ATP/AMP and cell viability (dye exclusion test). Presence of inhibitors of energy metabolism, potassium cyanide (respiration) and 2-deoxyglucose (glycolysis), further enhanced the cytotoxic effects induced by hematoporphyrin derivative and light.  相似文献
