A biosensor for determination of oxalate concentration in urine has been developed by immobilisation of oxalate oxidase and peroxidase on the surface of an interdigitated gold electrode. Enzyme immobilisation was performed using BSA and glutaraldehyde. Biamperometric measurements were made in flow conditions both in aqueous oxalate solutions (tested concentration range between 50 μM and 10 mM) and in real urine samples (tested measuring range between 5 and 100 μM). Optimal working conditions were examined for flow-injection analysis, and good correlation was achieved between added oxalate quantity and the one measured by biosensor in urine matrix (R2 = 0.9983). The influence of some interferences (ascorbic acid, uric acid, paracetamol, acetylsalicylic acid) was also studied using biamperometric measurement mode.  相似文献

Zone sampling performed on a single analytical channel and associated with the stopped-flow approach was proposed and applied to the spectrophotometric kinetic determination of creatinine in urine based on Jaffé's reaction. The flow network comprised microcomputer controlled three-way solenoid valves. With multicommutation, the potentialities of zone sampling and stopped-flow were expanded, and the design of the flow set up was simplified. Influence of the main related parameters such as system configuration, reagent concentrations, temperature and timing were studied. The proposed system handled about 24 samples per hour. Baseline drift was not observed during extended operation periods (8 h). Sample pretreatment was not required. Results were reproducible (R.S.D. < 3%) and in agreement with those obtained in a batch procedure.  相似文献

A flow-injection Spectrophotometric method for the determination of adrenaline and isoprenaline, based on the reaction with metaperiodate, is described. The calibration graphs are linear up to 2 × 10−4 M. Flow injection allows the measurement of 120 samples per hour. The method was successfully applied to the determination of both catecholamines in pharmaceuticals.  相似文献

An enzymatic assay that is highly selective for l-lysine, based on flow-injection techniques combined with spectrophotometric detection, is presented. l-Lysine-α-oxidase (E.C. 1.4.3.14) from Trichoderma viride and horseradish peroxidase were used in a coupled enzyme assay. Peroxide produced in the first reaction was converted by peroxidase with phenol and 4-aminoantipyrine to a quinoneimine dye detectable at 500 nm. An analytical enzyme reactor filled with coimmobilized enzymes was incorporated in the flow-injection system. The assay has a measuring frequency of 30 samples h?1 and a response time of less than 2 min. To adapt the assay to high concentrations of l-Lysine and to minimize interferences, the injected sample volume was reduced to 2 μ-l, resulting in a linearity range of 1–16 mM l-lysine with a sensitivity of 6–7 mV 1 mmol?1, a limit of detection (3σ) of 1 mM and a reproducibility of 0.5% (repetitive injection of a 10 mM l-lysine sample). The enzyme cartridge is stable for several months and thousands of measurements.  相似文献

Immobilized mannitol dehydrogenase is used for the determination of D-fructose in a flow-injection system. The enzyme is immobilized on poly(vinyl alcohol) beads. The oxidation of NADH occurs simultaneously and the disappearance of NADH is measured fluorimetrically. The response is linearly related to fructose concentration in the range 6–600 μM; 30 samples per hour can be analysed. The immobilized enzyme retains over 80% of its initial activity after repetitive use for 2 months.  相似文献

The classical Jaffé reaction for the determination of creatinine in urine samples is tested. A comparative study of the main analytical characteristics focussed to minimize the bias error and improve the precision, for the batchwise and flow injection (FI) methods is realized. Also, the effect of the albumin concentration in the determination of creatinine has been studied. Different analytical signals were studied. Absorbance increments at different times permit to estimate the creatinine concentration free from bias error in urine by the batchwise method using the calibration graph obtained with creatinine standards and no measurement of the blank solution is needed. The lineal interval was 0.92–50 mg l−1 and seven samples can be processed per hour by an operator. No previous treatment of the urine sample is necessary. The FI method provides also good results. The lineal interval was 30–100 mg l−1 and the sample rate was around 20 samples per hour. If increased albumin levels are detected in the urine, standard addition method or the calibration graphs with standards in presence of albumin are needed in order to obtain accurate results when FI method is employed. The obtained accuracy of the both methods allows its application as diagnostic tool to establish the urinary creatinine levels.  相似文献

The determination of uric acid in urine shows clinical importance, once it can be related to human organism dysfunctions, such as gout. An analytical procedure employing a multicommuted flow system was developed for the determination of uric acid in urine samples. Cu(II) ions are reduced by uric acid to Cu(I) that can be quantified by spectrophotometry in the presence of 2,2′-biquinoline 4,4′-dicarboxylic acid (BCA). The analytical response was linear between 10 and 100 μmol L− 1 uric acid with a detection limit of 3.0 μmol L− 1 (99.7% confidence level). Coefficient of variation of 1.2% and sampling rate of 150 determinations per hour were achieved. Per determination, 32 μg of CuSO4 and 200 μg of BCA were consumed, generating 2.0 mL of waste. Recoveries from 91 to 112% were estimated and the results for 7 urine samples agreed with those obtained by the commercially available enzymatic kit for determination of uric acid. The procedure required 100-fold dilution of urine samples, minimizing sample consumption and interfering effects. In order to avoid the manual dilution step, on-line sample dilution was achieved by a simple system reconfiguration attaining a sampling rate of 95 h− 1.  相似文献

Trace (≤1 mg/l or 30 μM) and ultratrace (≤1 μg/l or 30 nM) analysis methods for phosphorus determination by flow-injection analysis are reviewed. Most of the methods cited in this review are fundamentally based on the reaction of orthophosphate with molybdate to form heteropoly acids, such as molybdenum yellow and molybdenum blue, and some of the methods are based on the formation of such secondary reactions as ion associates and their aggregates with bulky cations, such as cationic dyes and quaternary ammonium ions. The heteropoly acids themselves can be measured by spectrophotometry, and the ion associate formed with a cationic dye, Malachite Green (MG), can be measured based on the coloration of MG. Light scattering detection methods can be used for measuring the aggregates of ion associates formed with bulky cations. Highly sensitive detection of phosphorus can be accomplished by fluorophotometry; Rhodamine B (RB) and its analogues react with molybdophosphate to form ion associates, which shows fluorescence quenching of RB: LOD is about 5 nM. The detection method based on the chemiluminescence of luminal oxidized with molybdophosphoric acids is probably the most sensitive of all the detection methods reported so far: LOD of the method is as low as 1 nM. The LOD of the molybdenum blue method can be improved by using a liquid core waveguide: LOD is 0.5 nM.  相似文献

An enzymatic flow-injection procedure for the determination of -lysine was developed. A lysine oxidase reactor is combined with a fibre-optic hydrogen peroxide detector. Hydrogen peroxide detection is based on the peroxidase-catalysed luminol reaction. The chemiluminescent light is detected by a photomultiplier. -Lysine can be determined in the range 10–1000 μM. A sampling rate of up to 90 h can be achieved. The whole sensing assay works for more than 1 month. The double logarithmic graph of the peak signal height vs. the lysine concentration is linear, the slope being larger than unity (r2 = 0.991, n = 4).  相似文献

Summary A sensitive ion-exclusion chromatographic method has been developed for determination of oxalate, thiosulfate, and thiocyanate. The method is based on separation of these anions on a polymethacrylate-based, weakly acidic cation-exchange resin (TSKgel OApak-A) and detection by means of a glassy carbon (GC) electrode electrochemically modified with polyvinylpyridine (PVP), palladium, and iridium oxide (PVP/Pd/IrO2). The electrochemical behavior of oxalate, thiosulfate, and thiocyanate at this chemically modified electrode (CME) have been investigated by cyclic voltammetry. The results indicated that electrocatalytic oxidation of these anions by the electrode was efficient and that the sensitivity, stability, and lifetime of the electrode were relatively high. Combined with ion-exclusion chromatography the PVP/Pd/IrO2 electrode was used as the working electrode for amperometric detection of these anions. All linear ranges were over two orders of magnitude and detection limits, defined asS/N=3, were 9.0×10−7 mol L−1 for oxalate, 6.7×10−7 mol L−1 for thiosulfate, and 5.6×10−7 mol L−1 for thiocyanate. Correlation coefficients were all>0.998. Coupled with microdialysis sampling the method has been successfully applied to the determination of oxalate, thiosulfate, and thiocyanate in urine.  相似文献

The combination of immobilized enzymes and amperometry to build selective detection devices in flow-injection analysis and liquid chromatography is described. The pros and cons of enzyme electrodes and of immobilized enzyme reactors are discussed. The paper concentrates on the use of immobilized dehydrogenases, oxidases, peroxidases, and on electrodes on which these enzyme reactions can be selectively followed. A reliable, highly sensitive and selective method is described for the determination of the herbicide diquat in different materials. It involves the formation of a stable radical obtained by reduction of diquat with sodium dithionite. The radical is a very fluorescent species (φF=0.37), and there is a linear relationship between diquat concentration and fluorescence intensity over the range 3–900 μg l?1. The limit of detection is 0.4 μg l?1. The method can be successfully adapted as a flow system using a two-channel manifold, the peak height being proportional to the diquat concentration over the range 18–4000 μg l?1. Manual and flow-injection methods were successfully applied to the determination of diquat in commercial herbicides, waters, potatoes, flowers and soils. Both procedures were also been applied to the determination of diquat in serum and urine without prior preparation of the samples and with good results.  相似文献

Chemiluminescence analysis with flow injection was used for the determination of vitamin B12 by means of the luminol-hydrogen peroxide system. A flow-injection method for the ultraviolet spectrophotometric determination of silver, based on its reaction with nickelocyanide ion, Ni(CN)2−4, in ammoniacal buffer medium (pH 10) and subsequent measurement of the decrease in the absorption of the Ni(CN)2−4 complex at 275 nm is described. The calibration graph is linear in the range 10–400 μm silver. At a sampling rate of about 60 samples h−1 with 35 μl sample injections, precision was about 1% relative standard deviation. The proposed method was successfully applied to the determination of silver in some common silver minerals.  相似文献

A novel plant tissue-based bioelectrode obtained by incorporating sunflower (Helianthus annuus L.) leaves tissue as a source of glycolate oxidase and peroxidase into a ferrocene-mediated carbon paste electrode for the determination of glycolic acid was developed. It was coupled with the flow-injection (FI) system and used as the basis to develop a novel FI amperometric procedure for glycolic acid determination. The flow-injection amperometric measurements were performed by injecting aliquot of glycolic acid solution into the flowing stream of 0.05 mol L−1 of phosphate buffer solution having pH 8.0 with a flow rate of 0.3 mL min−1. The bioelectrode consisted of 20% (w/w) of sunflower leaves tissue and 5% (w/w) of ferrocene at 0.00 V (vs Ag/AgCl). The bioelectrode exhibited a linear response from 1.0 × 10−6 up to 2.0 × 10−3 mol L−1 glycolic acid with a detection limit (S/N = 3) and a quantitation limit (S/N = 10) of 1 × 10−6 and 3.3 × 10−6 mol L−1, respectively. The sampling rate of 12 h−1 and a relative standard deviation of 1.67% (n = 15) were achieved. The bioelectrode response decreased to 70% of the original value within 90 continuous injections. The proposed bioelectrode was satisfactorily applied to glycolic acid determination in human urine samples after appropriate sample pretreatment. Results obtained by the FI method were compared favorably with those obtained by HPLC. It offers advantages, which included rapidity, high activity, limited stability, ease of preparation and low cost.  相似文献

A flow analysis method with on-line sample digestion/minicolumn preconcentration/flame atomic absorption spectrometry is described for the determination of trace metals in urine. First, urine sample was on-line ultrasound-assisted digested exploiting the stopped-flow mode, and then the metals were preconcentrated passing the pre-treated sample through a minicolumn containing a chelating resin. A home-made minicolumn of commercially available imminodiacetic functional group resin, Chelite Che was used to preconcentrate trace metals (Cu, Fe, Mn and Ni) from urine. The proposed procedure allowed the determination of the metals with detection limits of 0.5, 1.1, 0.8 and 0.8 μg L−1, for Cu, Fe, Mn and Ni, respectively. The precision based on replicate analysis was less than ±10.0%, and the enrichment factor obtained was between 21.3 (Mn) and 44.1 (Ni), for sample volumes between 2.5 and 5.0 mL, and an eluent volume of 110 μL. This procedure was applied for determination of metals in urine of workers exposed to welding fumes and urine of unexposed persons (urine control).  相似文献
