A rapid and efficient one-pot three-component protocol for the synthesis of 2H-indazolo[2,1-b]phthalazine-1,6,11-triones 4 and 1H-pyrazolo[1,2-b]phthalazine-5,10-diones 6 has been developed by domino coupling of phthalhydrazide, 1,3-diketones, and aldehydes under solvent-free conditions at 80 °C as well as under solvent-free ultrasound irradiation at room temperature promoted by (S)-camphorsulfonic acid.  相似文献

An efficient, inexpensive and environmentally friendly synthesis of novel 3-amino-2-benzoyl-1-aryl-1H-pyrazolo[1,2-b]phthalazine-5,10-dione and 3-amino-2-benzoyl-1-aryl-1H-pyrazolo[1,2-a]pyridazine-5,8-dione derivatives has been developed via one-pot three-component reaction of phthalhydrazide or maleic hydrazide, aldehydes and arylacetonitrile in the presence of catalytic amount of InCl3 as a Lewis acid catalyst under solvent-free conditions. The most important features of the present protocol are mild reaction conditions, short reaction times, high yields, and a wide range of functional group tolerance.  相似文献

Protonation of the highly reactive 1:1 intermediate produced in the reaction between alkyl isocyanides and electron-deficient acetylenic esters with phthalhydrazide, leads to a vinylisonitrilium cation, which undergoes an addition reaction with the conjugate base of the phthalhydrazide to produce dialkyl 3-(alkylamino)-5,10-dioxo-5,10-dihydro-1H-pyrazolo[1,2-b]phthalazine-1,2-dicarboxylates in fairly good yields at room temperature.  相似文献

A new, environmentally benign two-step synthesis of 5H[1,3]-thiazolo[2,3-b]quinazoline-3,5-(2H)-dione and 5H-thiazolo[2,3-b]quinazolin-5-one derivatives has been accomplished stepwise. The substituted 2-aminobenzoic acid upon condensation with thiourea in 1-butyl-3-methylimidazolium bromide at moderate temperature under nitrogen atmosphere yielded 2-thioxo-1H-4-quinazolinones. The resulting intermediate 2-thioxo-1H-4-quinazolinones, when reacted with 2-chloroethanoic acid/2-chloropropanal underwent cyclization to yield the desired product in excellent yields.  相似文献

The synthesis of 1H-pyrazolo[1,2-b]phthalazine-5,10-dione derivatives by NiCl2-catalyzed novel one-pot four-component condensation reaction of phthalimide, hydrazine, malononitrile (or ethyl cyanoacetate), and aromatic aldehydes was reported. CuI nanoparticles as an efficient catalyst have been used for the preparation of 1H-pyrazolo[1,2-b]phthalazine-5,10-diones by the four-component condensation reaction of phthalic anhydride, hydrazine monohydrate, aromatic aldehydes and malononitrile or ethyl cyanoacetate under solventfree conditions in good to excellent yields, short reaction times and environmentally benign, milder reaction conditions.  相似文献

A new, convenient, environmentally benign two-step synthesis of 4(1H)-quinolones, 5H-thiazolo[3,2-a]pyrimidin-5-one and 4H-pyrimido[2,1-b]benzothiazol-4-ones have been developed by first condensing substituted arylamine/2-aminothiazole/2-aminobenzenethiazole with Meldrum’s acid and trimethylorthoformate in 1-butyl-3-methylimidazolium bromide at a moderate temperature to afford 5-{(substituted aryl/4-methylthiazolyl/substituted benzothiazolyl)methylene}-2,2-dimethyl-1,3-dioxane-4,6-dione. The resulting compounds upon cyclization in 1-butyl-3-methyl tetrafluoroborate/triflate at a moderate temperature gave the title compounds in excellent yields.  相似文献

Efficient syntheses of novel 10-aryl-5a-(arylamino)-9-hydroxy-5a,6,7,8-tetrahydroindeno[1,2-b]chromen-11(10H)-one derivatives has been reported by [4+2] cycloaddition reactions of electron-deficient 2-(arylmethylene)-1H-indene-1,3(2H)-dione heterodienes with electron-rich enaminones in [bmim]BF4 at 80?°C and in acetic acid at 80?°C. Dimedone/cyclohexane-1,3-dione enaminones have been used as dienophiles in Inverse Electron Demand hetero-Diels-Alder reactions. A simple and efficient synthesis of 4-aryl-4,5-dihydro-1H-indeno[1,2-b]pyridine derivatives has been achieved via a four-component cyclocondensation of 1,3-indanedione, aromatic aldehydes, β-ketoesters and ammonium acetate in one-pot in the absence of catalyst and solvent at room temperature on grinding. For the first time, a synthesis of 1-hydroxy-3,4-dihydro-1H-benz[g]isochromene-5,10-dione (3), which is claimed to be a bioactive compound isolated from Psychotria camponutans, was achieved with a phthalide annulation reaction using 3-cyano-1(3H)-isobenzofuranone (5) and 5,6-dihydropyran-2-one (6) and subsequent reduction of the lactone moiety in the key steps. However, full spectral characterization of the synthesized target compound revealed that the isolated compound is not 1-hydroxy-3,4-dihydro-1H-benz[g]isochromene-5,10-dione (3). Structure revision shows the previously isolated compound to be the known psychorubrin (2).  相似文献

A simple, efficient and green approach to the synthesis of 1H-pyrazolo[1,2-b]phthalazine-5,10-diones has been developed via one-pot three-component reaction of aromatic aldehyde, malononitrile and phthalhydrazide catalyzed by zinc–proline complex (Zn[L-proline]2) using H2O: PEG400?=?6: 4 as solvent. Atom economy, good to excellent yield, operational simplicity and easy workup are important features of this method.

An efficient ionic liquid system was developed for the preparation of various heterocycle-fused pyrimidine-2, 4(1H,3H)-diones in moderate to excellent yields (52–95%). It was found that [HDBN+][TFE?], a simple and easily prepared ionic liquid, could act as both the solvent and reaction promoter, and that the reactions could be efficiently carried out at atmospheric pressures of CO2.  相似文献

Various trisubstituted [1]benzopyrano[4,3-b]pyrrol-4(1H)-one derivatives have been synthesized from 4-hydroxycoumarin with a 30-40% yield over six steps. The key step of the synthesis is a base-promoted intramolecular cyclization of enamines 5, followed by dehydration to generate the fused pyrrole ring.  相似文献

A new and efficient strategy for the synthesis of 3-substituted 1-hydroxybenz[g]isoquinoline-5,10-diones by reaction of 2-methoxycarbonyl-1,4-naphthoquinone with different pyridinium salts under Kröhnke conditions is disclosed. Treatment of N-substituted 2-methyl-1H-indoles 1 with S2Cl2 and DABCO in chloroform gave the corresponding [1,2]dithiolo[4,3-b]indole-3(4H)-thiones 5 by the addition of triethylamine in high yield. 1,2-Dithiole-3-thiones 5 underwent cycloaddition with one or two DMAD equivalents to afford either 2-(3-thioxo-1,3-dihydro-2H-indol-2-ylidene)-1,3-dithioles 10 or fused 4,5-dihydrothiopyrano[3,2-b]indoles 9.  相似文献

A convenient and efficient procedure has been developed for the synthesis of a new ring system, pyrrolo[1,2-b]pyrazolone, via two-component coupling reaction followed by base mediated intramolecular cyclization. Single-pot synthesis replacing the two step process has also been successfully carried out. A series of polycyclic pyrrolo[1,2-b]pyrazolone derivatives have been obtained by employing the procedure along with some fused pyrrol-1-ylamine system. The products were formed very rapidly in catalyst free condition in a good yield (up to 75%) and also it had tolerance to a wide scope of substrates.  相似文献
