Previously reported theories for cyclical electrical field flow fractionation (CyElFFF) are severely limited in that they do not account for diffusion, steric, or electric double layer effects. Experiments have shown that these theories overpredict the retention of particles in CyElFFF. In this work, we present a model for prediction of steric, diffusion, and electrical effects. The electrical double layer effects are treated using a lumped electrical circuit model that accounts for the field shielding by the electrical double layer formed at the electrode-carrier interface. The electrical effects are shown to dominate retention times and outweigh the contributions of diffusion and particle size. Detailed results from the simulations are presented in this work, and a comparison between the theoretical and experimental results obtained from the retentions of polystyrene particle standards is presented in this paper. The models are shown to correctly predict the retention of the polystyrene standards in CyElFFF with a reasonable error, while existing models are shown to have significant failings.  相似文献

We recently built a magnetic separation system to extend the applications of split-flow thin (SPLITT) fractionation to magnetically susceptible particles. Here, we characterize the magnetic SPLITT system using magnetically susceptible particles and ion-labeled particles. The flow axis of separation channel was orientated parallel and perpendicular to gravitational forces to exclude and include, respectively, gravitational effects on separation. Both operating modes were used to test the theory experimentally, with emphasis on the parallel mode. The magnetic susceptibilities of carrier and ion-labeled particles were varied, and various ion-labeled and unlabeled particles were studied experimentally, resulting in successful separation of labeled particles, yeasts, and cells from unlabeled ones. The minimal difference in magnetic susceptibility (delta(chi)) required for complete particle separation was about 1.75 x 10(-5) [cgs], corresponding to about 10(9) labeling ions per particle in this study. The throughput was around 7.2 x 10(8) particles/h using the present setup. Magnetic SPLITT fractionation shows good potential for use in obtaining particles magnetic susceptibilities from a simple theoretical treatment.  相似文献

Split-flow thin (SPLITT) fractionation devices have been widely used to separate macromolecules, colloids, cells and particles. Recently, the quadrupole magnetic flow sorter (QMS) has been reported in the literature as another family of SPLITT fractionation device. However, the separation performance observed in the experimental measurements is generally found to deviate from the ideal behaviour. Possible causes such as hydrodynamic lift force, high particle concentration and imperfect geometries have been extensively examined. However, the effects of flow development regions and fringing magnetic force field at the separation channel inlet and outlet, which are ignored by the theory, have not been investigated. The error introduced by ignoring these effects need to be rigorously studied so that the theory can be used to optimise operation flow rates with confidence. Indeed, we find in this paper that these ignored effects are responsible to the discrepancy between the experimental data and the theoretical predictions. A new theory has been proposed for optimisation of device operation.  相似文献

Reinjection of one ore more collected fractions of eluted samples is recognized as a useful procedure in analytical separation techniques, among which field-flow fractionation (FFF), to improve the actual separation of complex samples. Hollow-fiber flow FFF (HF5) is a micro-channel subset of flow FFF (F4), which has recently reached a performance comparable to that of standard, flat-channel F4. To further improve HF5 of complex protein samples, we present a new device and method for in-line, reinjection HF5 that we call tandem HF5 (HF5/HF5). HF5 is ideally suited for tandem operation because (1) small channel volume and low operation flow rates allow reducing dilution and volume of the collected fractions, and (2) the relaxation/focusing step that takes place between the 1st and 2nd run (refocusing) allows reestablishing the volume and concentration of the sample plug before the 2nd elution. HF5/HF5 proves particularly effective in the case of oligomeric proteins since it allows collecting and reinjecting the bands that correspond to each separated oligomeric form. This provides information on the dynamic equilibria between the different oligomers. For HF5/HF5 operations, a modified, prototype HF5 instrumentation is presented which includes a "trap" constituted of a four-port, two-way valve positioned downstream the UV detector and a collection loop. The effect of refocusing conditions on HF5/HF5 performance is investigated by varying refocusing time. With a complex protein samples such as blood serum, HF5/HF5 can improve detectability of the low abundance components since overloading effects due to high-abundance components are reduced. This is shown for serum lipoproteins: while after the 1st run high density lipoproteins (HDLs) are not separated from high-abundance serum proteins, after the 2nd run it is shown possible to separate the HDL subclasses.  相似文献

The combined employment of the SPLITT (split-flow thin) cell--a relatively new system for fast, continuous binary separation--and of gravitational field-flow fractionation (GrFFF)--a fractionation technique suitable for micron particle size distribution determination--was investigated for starch separation and characterization. Emphasis is placed on the main advantages of both techniques: operating under gentle earth gravity field, low cost and ease of maintenance. The reproducibility of GrFFF is demonstrated. Both the SPLITT separation and GrFFF fractionation results were checked by optical microscopy. Application examples of typical starch fractionation experiments are reported and discussed.  相似文献

Pinched inlet gravitational split-flow thin fractionation (PI-GSF) has been applied to the continuous size fractionation of marine sediments in order to study the difference in sediment size distribution and the concentration of PCDD/Fs contained in different particle sizes. A PI-GSF channel, known to improve the separation efficiency by reducing the sample inlet thickness, was utilized to fractionate sediments collected from three different bay areas (Geoje, Ulsan, and Pohang) in Korea into 5 different sub-populations (<2.0, 2.0-5.0, 5.0-10, 10-20, 20-63 microm in diameter). The sorted sediment fractions from PI-GSF were examined using electron microscopy to obtain size distribution and the results showed a variation in particle size distribution between bay areas. When the collected particle fractions were examined for size dependent levels of PCDD/Fs, the concentrations of total PCDD/Fs were shown to be much greater for samples collected close to heavy industry complexes than sediments from bay areas without major industry.  相似文献

The gravitational split-flow lateral transport thin fractionation is known to be a fast, simple, theoretically tractable and tunable tool for the binary separation of molecular or particulate samples into different dimensional fractions. This fractionation is performed in a so-called SPLITT cell and is due to the combined effect of the gravitational force field and the flow rates inside the separation channel. It is known that separation performance is strongly dependent on the flow rate conditions and feed flow concentration, however, to date, few studies have been conducted to investigate the effect non-specific crossover has on separation. The aim of this work is to establish whether diffusive processes stemming from hydrodynamic effects contribute in any way to the quality of separation. A silica sample of known granule size distribution was chosen for this study which has environmental applications.  相似文献

Cyclical electrical field flow fractionation (CyElFFF) is a variation on electrical field flow fractionation (ElFFF) where cyclical electrical fields are used instead of steady DC fields to increase the effective field experienced by particles in the flow channel. Even though the effective field increases more than 20-fold compared to normal ElFFF, the retention and resolution in CyElFFF has not been shown to be better than in ElFFF. In this paper we report how one can optimize operational parameters in CyElFFF to obtain good retention and resolution in CyElFFF. The effects of offset voltage, frequency, flowrate, concentration of particles and sample size on retention, resolution and retained peak/void peak ratio have been observed. The results obtained from these experiments were analyzed and suggestions have been made to improve both retention and resolution. A 4-fold improvement in retention without a significant increase in band broadening is reported.  相似文献

Cyclical electrical field flow fractionation (Cy/ElFFF) is demonstrated in a standard electrical field flow fractionation (ElFFF) channel for the first time. Motivation for the use of alternating current (AC) fields in a traditionally direct current (DC) technique are discussed. The function of the system over a wide range of operating conditions is explored and challenges associated with various operating conditions reported. Retention of polystyrene nanoparticle standards is accomplished and the effect of varying parameters of the applied field, such as voltage and frequency, are explored. The first separations using this technique are demonstrated. The experimental results are compared to analytical models previously reported in the literature. The general trend of the experimental results is similar to those predicted in theoretical models and possible reasons for discrepancies are elucidated. Suggestions are made for improving the separation and analysis method, and possible applications explored.  相似文献

The purpose of this study was to investigate the influence of ionic strength of the carrier liquid, cross flow rate, focus flow rate, and sample load on the retention behavior of liposomes in asymmetric flow field‐flow fractionation (AF4). Two differently prepared samples of large unilamellar vesicles (LUV) were used. Experiments were performed varying the factors systematically and evaluating their effect on both retention behavior of the liposomes and on particle size as obtained from online coupled multi‐angle light scattering (MALS) analysis. The results showed that the focus flow rate had the least influence on the elution of liposomes. Elution of LUV is mainly governed by the chosen cross flow condition and ionic strength of the carrier liquid as well as its sample load. Optimal fractionation and size analysis were achieved using a sample load of about 10 μg, a cross flow gradient from 1.0 to 0.1 mL/min over 35 min and a carrier solution of NaNO3 with a concentration of 10 mM.  相似文献

非对称流场流分离技术对于蛋白质等生物大分子的分析具有温和、分离范围广的特点。然而,在场流分离通道中,受载液组成的影响而产生的蛋白质与通道膜的相互作用和蛋白质在通道内的聚集行为,会影响分析物的回收率和尺寸形态,这些现象一定程度上限制了场流分离仪器的进一步应用。该文研究了载液组成对于卵白蛋白在非对称流场流分离中膜吸附和聚集行为的影响。考察了不同pH (6.2、7.4、8.2)、阳离子种类(Na+、K+、Mg2+)及多种离子强度(0~0.1 mol/L)等条件对卵白蛋白洗脱过程的影响。结果表明a)载液的离子强度越大,卵白蛋白的吸附和聚集行为越严重;b) pH和蛋白质的等电点pI的相对大小决定了蛋白质的表面电荷,从而影响蛋白质的吸附聚集行为;c)二价阳离子Mg2+更易引发通道中蛋白质的吸附和聚集。这些结果有助于今后使用非对称流场流分离技术分析蛋白质样品时,改善载液组成以获得更高的回收率,降低蛋白质聚集作用,对AF4更广泛地应用于蛋白质生化分析中有较好的参考价值。  相似文献

Summary Several types of membrane have been tested for use in organic solvent flow field-flow fractionation in an asymmetric channel. The practical problems most commonly encountered were leakage of air and solvent through the support layer on which the membranes are cast, and unequal swelling of the membrane and the support layer in the organic solvent, leading to ridging of the membrane in the channel. Three types of membrane were found suitable for the separation of polystyrene standards with tetrahydrofuran as solvent. The best results were obtained with a fluoropolymer membrane. Fair agreement was found between theory and practice for the dependence of retention times on the relative molecular mass of the standards and on the flow regime. Use of scanning electron microscopy revealed that for a number of the membrane materials some pores were much larger than expected on the basis of the indicated molecular weight cut-off. Whereas these materials could not be used for the fractionation of soluble polymers, they could be applied with some success to the separation of solid latex and silica particles. A PTFE membrane could be used for the separation of latexes and silica particles suspended in acetonitrile as carrier liquid. In general, however, the retention times of these particles were shorter than theoretically predicted.  相似文献

In electrokinetically driven microfluidic systems, the driving voltage applied during operation tends to induce a Joule heating effect in the buffer solution. This heat source alters the solution's characteristics and changes both the electrical potential field and the velocity field during the transport process. This study performs a series of numerical simulations to investigate the Joule heating effect and analyzes its influence on the electrokinetic focusing performance. The results indicate that the Joule heating effect causes the diffusion coefficient of the sample to increase, the potential distribution to change, and the flow velocity field to adopt a nonuniform profile. These variations are particularly pronounced under tighter focusing conditions and at higher applied electrical intensities. In numerical investigations, it is found that the focused bandwidth broadens because thermal diffusion effect is enhanced by Joule heating. The variation in the potential distribution induces a nonuniform flow field and causes the focused bandwidth to tighten and broaden alternately as a result of the convex and concave velocity flow profiles, respectively. The present results confirm that the Joule heating effect exerts a considerable influence on the electrokinetic focusing ratio.  相似文献

A new system design and setup are proposed for the combined use of asymmetrical flow field-flow fractionation (AF4) and hollow-fiber flow field-flow fractionation (HF5) within the same instrumentation. To this purpose, three innovations are presented: (a) a new flow control scheme where focusing flow rates are measured in real time allowing to adjust the flow rate ratio as desired; (b) a new HF5 channel design consisting of two sets of ferrule, gasket and cap nut used to mount the fiber inside a tube. This design provides a mechanism for effective and straightforward sealing of the fiber; (c) a new AF4 channel design with only two fluid connections on the upper plate. Only one pump is needed to deliver the necessary flow rates. In the focusing/relaxation step the two parts of the focusing flow and a bypass flow flushing the detectors are created with two splits of the flow from the pump. In the elution mode the cross-flow is measured and controlled with a flow controller device. This leads to reduced pressure pulsations in the channel and improves signal to noise ratio in the detectors. Experimental results of the separation of bovine serum albumin (BSA) and of a mix of four proteins demonstrate a significant improvement in the HF5 separation performance, in terms of efficiency, resolution, and run-to-run reproducibility compared to what has been reported in the literature. Separation performance in HF5 mode is shown to be comparable to the performance in AF4 mode using a channel with two connections in the upper plate.  相似文献

A new concept of isopycnic focusing exploiting the simultaneous action of coupled electric and gravitational fields is described. Experimental implementation of this concept is demonstrated under conditions of focusing field-flow fractionation and of newly proposed thin layer isopycnic focusing. Both methods are aimed for particle separations. Although the demonstration was performed by using model cross-linked polymer and latex particles, the most important field of potential applications is intended for analytical and preparative separations of biological particles.  相似文献

We describe the development and testing of a setup that allows for DEP field‐flow fractionation (DEP‐FFF) of irreversibly electroporated, reversibly electroporated, and nonelectroporated cells based on their different polarizabilities. We first optimized the channel and electrode dimensions, flow rate, and electric field parameters for efficient DEP‐FFF separation of moderately heat‐treated CHO cells (50°C for 15 min) from untreated ones, with the former used as a uniform and stable model of electroporated cells. We then used CHO cells exposed to electric field pulses with amplitudes from 1200 to 2800 V/cm, yielding six groups containing various fractions of nonporated, reversibly porated, and irreversibly porated cells, testing their fractionation in the chamber. DEP‐FFF at 65 kHz resulted in distinctive flow rates for nonporated and each of the porated cell groups. At lower frequencies, the efficiency of fractionation deteriorated, while at higher frequencies the separation of individual elution profiles was further improved, but at the cost of cell flow rate slowdown in all the cell groups, implying undesired transition from negative into positive DEP, where the cells are pulled toward the electrodes. Our results demonstrate that fractionation of irreversibly electroporated, reversibly electroporated, and nonelectroporated cells is feasible at a properly selected frequency.  相似文献

Various operational parameters affecting the formation of the density gradient generated by the electric field action on a binary pseudo-continuous carrier liquid composed of charged colloidal silica particles suspended in water and the isopycnic focusing of sample particles were investigated under conditions of static thin layer focusing and dynamic focusing field-flow fractionation. The properties and the behavior of the density gradient forming carrier liquid were studied. The experimental results are compared with theoretical predictions and discussed with respect to potential applications of the proposed concept not only for separation purposes but also for studies of interparticle interactions.  相似文献

The flow field-flow fractionation (FIFFF) technique is a promising method for separating and analysing particles and large size macromolecules from a few nanometers to approximately 50 μm. A new fractionation channel is described featuring well defined flow conditions even for low channel heights with convenient assembling and operations features. The application of the new flow field-flow fractionation channel is proved by the analysis of pigments and other small particles of technical interest in the submicrometer range. The prediction of optimal times of separation as a function of the applied electrical field and cation valence have been studied for the case of field flow fractionation [Martin M., Giddings J. C., J. Phys. Chem. 1981, 85, 727] with charged solutes. These predictions can be very useful to a priori design or identify optimal operating conditions for a Couette-based device for field flow fractionation when the orthogonal field is an electrical field. Mathematically friendly relationships are obtained by applying the method of spatial averaging to the solute species continuity equation; this is accomplished after the role of the capillary geometrical dimensions on the applied electrical field equations has been assessed [Oyanader M. A., Arce P., Electrophoresis 2005; 26, 2857]. Moreover, explicit analytical expressions are derived for the effective parameters, i.e. diffusivity and convective velocity as functions of the applied (orthogonal) electrical field. These effective transport parameters are used to study the effect of the cation valence of the solutes and of the magnitude of the applied orthogonal electrical field on the values of the optimal time of separation. These parameters play a significant role in controlling the optimal separation time, leading to a family of minimum values, for particular magnitudes of the applied orthogonal electrical field.  相似文献

Recent and earlier models of electrical field flow fractionation (ELFFF) have assumed that the electric field within the fluid domain is governed by Laplace's equation. This assumption results in a linear potential and a spatially constant field across the channel and is generally true for very dilute systems and relatively high effective potentials. Experimental studies show, however, that the effective potential within the channel may be less than 1% of the applied potential; this is apparently due to double layer formation and charge buildup at the poles. In such cases, local analyte concentrations can, nonetheless, be orders of magnitude higher than the bulk mean and the local potential small, both of which can lead to a nonlinear spatial distribution of the field strength. In such cases Poisson's equation must be used rather than Laplace's equation. Steady-state ELFFF simulations were performed using a Poisson's equation-based model. The domain in which Laplace's equation is valid was identified and the effects of concentration and effective field strength on device performance were explored.  相似文献
