Two new monobasic bidentate ligands and their Pd(II) complexes have been synthesized and characterized by analytical and spectroscopic methods. The structures of the complexes were confirmed by single-crystal X-ray diffraction. The bimolecular binding of the ligands and complexes has been carried out and described. Interestingly, both the bidentate chelating ligands replaced all the triphenyl arsine and chloride ions from the metal precursor in the formation of new complexes and were found to be approximately square planar. The interaction of the ligands and the complexes with calf thymus DNA and bovine serum albumin was studied by electronic and emission spectroscopy techniques, which suggested an intercalation mode of binding. It is well-known that the viscosity of a DNA solution increases if any compound added binds to it through intercalation because this process lengthens the DNA helix due to the increased separation of the DNA base pairs when the compound slides in between, whereas a partial, nonclassical intercalation could bend (or kink) the DNA helix, which leads to a reduction in length and thereby reducing its viscosity. By contrast, there will be no change in the viscosity when the compounds bind with DNA grooves or by partial intercalation, which was further confirmed by viscosity measurements and molecular docking studies. It has been found that the compounds cleaved supercoiled DNA into nicked DNA without any external agent. The in vitro cytotoxicity studies of the ligands and complexes against human lung (A549) and breast (MCF7) cancer cell lines showed significant activity for both species.  相似文献

Pyrazolone derivatives (Z)-4-((2-hydroxyethylimino)(p-tolyl)methyl)-3-methyl-1-phenyl-1H-pyrazol-5(4H)-one [PMP-EA] (1), (Z)-1-(3-chlorophenyl)-4-((2-hydroxyethylimino)(p-tolyl)methyl)-3-methyl-1H-pyrazol-5(4H)-one [MCPMP-EA] (2), and (Z)-4-((2-hydroxyethylimino)(p-tolyl)methyl)-3-methyl-1-p-tolyl-1H-pyrazol-5(4H)-one [PTPMP-EA] (3) have been synthesized and characterized. The molecular geometry of 2 has been determined by single-crystal X-ray study. These ligands exist in amine-one tautomeric form in the solid state. Three copper(II) complexes, [Cu(PMP-EA)(H2O)2] (4), [Cu(MCPMP-EA)(H2O)2] (5), and [Cu(PTPMP-EA)(H2O)2] (6), respectively, have been synthesized using these ligands and characterized by microanalytical data, molar conductivity, IR, UV–Visible, FAB-Mass, magnetic measurement, TG-DTA studies, and ESR spectral studies; Cu(II) is five-coordinated with [ML(H2O)2] composition. The interaction of the complexes with CT-DNA (calfthymus) was investigated using different methods. The results suggest that the copper complexes bind to DNA via intercalation and can quench the fluorescence intensity of EB bound to DNA.  相似文献

Transition metal complexes of type M(L)2(H2O)x were synthesized, where L is deprotonated Schiff base 2,4‐dihalo‐6‐(substituted thiazol‐2‐ylimino)methylphenol derived from the condensation of aminothiazole or its derivatives with 2‐hydroxy‐3‐halobenzaldehyde and M = Co2+, Ni2+, Cu2+ and Zn2+ (x = 0 for Cu2+ and Zn2+; x = 2 for Co2+ and Ni2+). The synthesized Schiff bases and their metal complexes were thoroughly characterized using infrared, 1H NMR, electronic and electron paramagnetic resonance spectroscopies, elemental analysis, molar conductance and magnetic susceptibility measurements, thermogravimetric analysis and scanning electron microscopy. The results reveal that the bidentate ligands form complexes having octahedral geometry around Co2+ and Ni2+ metal ions while the geometry around Cu2+ and Zn2+ metal ions is four‐coordinated. The geometries of newly synthesized Schiff bases and their metal complexes were fully optimized in Gaussian 09 using 6–31 + g(d,p) basis set. Fluorescence quenching data reveal that Zn(II) and Cu(II) complexes bind more strongly to bovine serum albumin in comparison to Co(II) and Ni(II) complexes. The ligands and their complexes were evaluated for in vitro antibacterial activity against Escherichia coli ATCC 25922 (Gram negative) and Staphylococcus aureus ATCC 29213 (Gram positive) and cytotoxicity against lever hepatocellular cell line HepG2.  相似文献

The DNA cleavage activities of nickel(II) ion and four closely related macrocyclic nickel(II) complexes NiL1 ∼ NiL4 in the absence of any added redox cofactors are compared and the structure of NiL3 methanol solvate has been characterized by single crystal X-ray analysis, where L1 ∼ L4 are the dianions of tetraazamacrocyclic oxamido Schiff bases. In NiL3·MeOH, the macrocyclic [N4] ligand coordinates to the central Ni(II) ion forming a distorted square–planar geometry. The adjacent mononuclear molecules are linked by O–H?O hydrogen bonds and Ni?O and Ni?L van der Waals forces into 2D supramolecular structure. Agarose gel electrophoresis studies indicate that the ability of these nickel(II) complexes to cleave DNA is highly dependent upon the ligand employed. In the absence of any added oxidizing agents, only NiL3 is a relatively good DNA cleavage agent, and the process of plasmid DNA cleavage is much sensitive to ionic strength and pH value. The NiL3-mediated DNA cleavage reaction is a typical pseudo-first-order consecutive reaction, and the rate constants of 0.148 ± 0.007 h−1 (k1) and 0.0118 ± 0.0018 h−1 (k2) for the conversion of supercoiled to nicked DNA and nicked to linear DNA are obtained in presence of 0.5 mmol L−1 NiL3. The results of DNA cleavage experiments, combining with those of circular dichroism (CD) and fluorescence spectroscopy indicate that the main binding modes between DNA and the complexes should be groove binding and electrostatic interaction.  相似文献

Ruthenium(II) complexes with 2-acetylpyridine-thiosemicarbazones (L1–L4) were synthesized and characterized by analytical and spectral (FT-IR, UV–vis, NMR [1H, 13C and 31P], and ESI-Mass) methods. Systematic biological investigations, free radical scavenging, anticancer activities, and DNA cleavage studies, were carried out for the complexes. Antioxidant studies showed that the complexes have significant antioxidant activity against DPPH, hydroxyl, nitric oxide radicals and hydrogen peroxide assay. The in vitro cytotoxicity of complexes against breast cancer (MCF-7) cell line was assayed showing high cytotoxicity with low IC50 values indicating their efficiency in destroying the cancer cells even at very low concentrations. The DNA cleavage studies showed that the complexes efficiently cleaved DNA.  相似文献

The ternary complexes of [CuII(Hist)(Tyr)]+1 and [CuII(Hist)(Trp)]+2 have been synthesized, structurally characterized and their DNA binding and cleavage abilities probed. The intrinsic binding constants (Kb) for complexes/CT-DNA were also determined (Kb = 2.7 × 102 for complex 1 and Kb = 2.2 × 102 for complex 2). These complexes exhibit their nuclease activity on plasmid DNA, which seems to depend on the nature of the aromatic moiety. The DNA hydrolytic cleavage rate constants were also determined for complexes 1 and 2, which are 0.91 and 0.79 h−1, respectively.  相似文献

Three isoxazole Schiff bases 2-((E)-(3,5-dimethylisoxazol-4-ylimino)methyl)-6-methoxyphenol (L1), 2-((E)-(3,5-dimethylisoxazol-4-ylimino)methyl)-4,6-diiodophenol (L2), 2-((E)-(3,5-dimethylisoxazol-4-ylimino)methyl)-6-bromo-4-chlorophenol (L3), and their Cu(II) complexes [Cu(L1)2] (1), [Cu(L2)2] (2) and [Cu(L3)2] (3) were synthesized. All the complexes have been characterized by elemental analysis, FT-IR, ESI mass, UV-Visible, ESR, TGA, magnetic moments, and single-crystal X-ray diffraction analysis. Based on analytical data, a square planar geometry is assigned to the Cu(II) complexes with N2O2 donors from the Schiff base ligands. The single-crystal X-ray diffraction measurements of 1 and 2 confirmed the square planar geometry. DNA binding studies from electronic absorption titrations, viscosity measurements, and fluorescence quenching studies indicated an intercalation mode of binding of Cu(II) complexes with CT-DNA. DNA cleavage experiments of Cu(II) complexes with supercoiled plasmid pBR322 DNA have also been investigated by agarose gel electrophoresis in the presence of H2O2 (oxidative cleavage) and UV light (photolytic cleavage). The synthesized compounds were screened for antibacterial (Escherichia coli, Pseudomonas putida, Klebsiella pneumoniae, Bacillus subtillis and Staphylococcus aureus) and antifungal (Candida albicans and Aspergillus niger) activities by the paper disk method. The Cu(II) complexes showed better activity than corresponding Schiff bases.  相似文献

Two mononuclear Cu(II) complexes, [Cu(L1H2)](ClO4)1.25Cl0.75·1.25H2O (1) and [Cu(L2H2)](ClO4)2 (2), of the pyridoxal Schiff base ligands N,N′-dipyridoxylethylenediimine (L1H2) and N,N′-dipyridoxyl-1,3-propanediimine (L2H2) are reported. X-ray crystal structures of both complexes are also reported. In both complexes the pyridoxal nitrogen atoms remain protonated. In the solid state, the tetradentate Schiff base ligand is virtually planar in 1, while in 2 the ligand conformation is like an inverted umbrella. In cyclic voltammetry experiments it is found that in these complexes the Cu(III) and Cu(I) states are more easily accessible than in their salen type analogs. The pyridoxal Schiff base complexes are also found to be resistant to oxidative electro-polymerization, unlike their corresponding salicyl aldehyde Schiff base complexes.  相似文献

[Cu(L)] (1) and mixed ligand copper(II) complexes [Cu(L)(A)] (2 and 3), where L is the Schiff base derived from o-vanillin and l-tryptophan and A is pyridine (2) and imidazole (3), were synthesized and characterized using conventional and spectral techniques. 2 was structurally characterized using single crystal X-ray crystallography showing that Cu(II) is coordinated through N2O2 donors in a square plane. The EPR spectra of the complexes in frozen solution support a square-based structure. Electrochemical behavior of the complexes has been studied by cyclic voltammetry. The DNA-binding properties of L and 1–3 with calf thymus DNA were investigated by spectral and kinetic methods. For all the complexes, the maximum value of binding constant (0.38 × 106) was achieved with 3 by spectroscopic titration. The ability of compounds to break pUC19 DNA was checked by gel electrophoresis. The ligand and copper complexes exert cytotoxicity against MCF-7 cell line.  相似文献

We have reported herein the synthesis of three new Cu(II) complexes of tri- and tetradentate Schiff base ligands containing N3 or N4 donor set along with terminal NNN or SCN ligands: [L1Cu(NCS)]ClO4 (1), [L2Cu(NCS)2] (2) and [L3Cu(NNN)]ClO4 (3) [L1 = NC5H4C(CH3)=N(CH2)3N=C(CH3)C5H4N, L2= Me2N–(CH2)3–N=C(CH3)C5H4N and L3 = NC5H4CH=N–(CH2)4–N=CHC5H4N]. The complexes have been systematically characterised by elemental, spectroscopic and electrochemical techniques. Antimicrobial activities of the Schiff base ligands and their metal complexes have been studied using the disc diffusion method on the strains of Candida tropicalis and Bacillus megaterium. Structures of all the complexes have been unequivocally established from single crystal X-ray diffraction analyses that show the monomeric units containing a five-coordinated copper center in highly distorted square pyramidal geometry with thiocyanate or azide anion coordinated as terminal ligand. The complexes 1 and 3 crystallise in monoclinic (P21/c) and 2 in triclinic (P-1) space group, respectively.  相似文献

Reaction of Cu(II) nitrate with a new pyrazole-based Schiff base ligand, 5-methyl-3-formylpyrazole-N-(2′-methylphenoxy)methyleneimine (MPzOA), afforded two types of Cu(II) complexes at different reaction temperatures, [Cu(MPzOA)(NO3)]2 (1) and [Cu(3,7,11,15-tetramethylporphyrin)(H2O)](NO3)2 (2), reported together with a Ni(II) complex, [Ni(MPzOA)2(H2O)2]Br2 (3). The compounds are characterized by single crystal X-ray structure analyses along with several physico-chemical and spectral parameters. Complex 1 is authenticated as a bis(μ-pyrazolato)dicopper(II), while 2 is a porphyrinogen and 3 is a distorted octahedral complex. Structural analyses of the complexes reveal that 1 crystallized in monoclinic P21/n space group while 2 and 3 crystallized in monoclinic C2/c space group. DNA-binding studies of the complexes have shown that the complexes interact with CT-DNA. DNA-cleavage studies with plasmid DNA have shown that 1 and 2 induce extensive DNA cleavage in the presence of H2O2 as an additive, whereas there is no change in degradation of super-coiled DNA by 3 in the presence of additive. The antimicrobial studies of the complexes against Escherichia coli DH5α bacteria strain indicated that all the complexes were capable of killing E. coli with different LD50 values.  相似文献

Schiff base mixed-ligand copper complexes [CuL1(phen)Cl2], [CuL1(bipy)Cl2], [Cu(L1)2Cl2], [Cu(L2)2Cl2], [CuL2(bipy)Cl2], and [CuL2(phen)Cl2] (where L1?=?4-[3,4-dimethoxy-benzylidene]-1,5-dimethyl-2-phenyl-1,2-dihydro-pyrazole-3-one; L2?=?4-[3-hydroxy-4-nitro-benzylidene]-1,5-dimethyl-2-phenyl-1,2-dihydro-pyrazole-3-one; phen?=?1,10-phenanthroline; and bipy?=?2,2′-bipyridine) have been synthesized and characterized. Their DNA-binding properties have been studied by electronic absorption spectra, viscosity, and electrochemical measurements. The absorption spectral and viscosity results suggest that the copper(II) complexes bind to DNA via partial intercalation. The addition of DNA resulting in the decrease of the peak current of the copper(II) complexes indicates their interaction. Interaction between the complexes and DNA has also been investigated by submarine gel electrophoresis. The copper complexes cleave supercoiled pUC19 DNA to nicked and linear forms through hydroxyl radical and singlet oxygen in the presence of 3-mercaptopropionic acid as the reducing agent. These copper complexes promote the photocleavage of pUC19 DNA under irradiation at 360?nm. Mechanistic study reveals that singlet oxygen is likely to be the reactive species responsible for the cleavage of plasmid DNA by the synthesized complexes. The in vitro antimicrobial study indicates that the metal chelates have higher activity against the bacterial and fungal strains than the free ligands.  相似文献

A new series of mercaptopyrimidine Ru(III) complexes were synthesized and characterized using various spectral techniques like single‐crystal X‐ray diffraction, Fourier transform infrared and NMR spectroscopies, thermogravimetric analysis and energy‐dispersive X‐ray analysis. The complexes were evaluated for their pharmacological activities like in vitro antimicrobial, anticancer, antituberculosis and antioxidant activities. The DNA binding of the complexes was investigated by absorption and emission spectral measurements which indicated that the complexes bind to DNA via intercalation, with molecular docking studies validating the results. Cu(II) complexes of three bis(pyrrol-2-yl-methyleneamine) ligands were synthesized and characterized by elemental analyses, mass spectra, and IR spectra. X-ray diffraction analysis shows that [CuL3]2 is a dinuclear complex with an extremely distorted square-planar geometry. Furthermore, the antioxidant activities of the compounds have been investigated. The electrochemical properties of the Cu(II) complexes have also been studied by cyclic voltammetry. The Cu(II) complexes show similar superoxide dismutase (SOD) activity compared with that of the native Cu, Zn-SOD.  相似文献

Ternary Cu(II) complexes [Cu(II)(saltyr)(B)] (1,2), (saltyr = salicylidene tyrosine, B = 1,10 phenanthroline (1) or 2,2′ bipyridine (2)) were synthesized and characterized by various techniques. The complexes exhibit square pyramidal (CuN3O2) geometry. CT-DNA binding studies revealed that the complexes show good binding propensity (Kb = 3.47 × 104 M−1 and 3.01 × 104 M−1 for 1 and 2, respectively). The role of these complexes in the oxidative and hydrolytic DNA cleavage was studied. The catalytic ability of 1 and 2 follows the order: 1 > 2. The rate constants for the hydrolysis of phosphodiester bond were determined as 2.80 h−1 and 2.11 h−1 for 1 and 2, respectively. It amounts to (0.58-0.77) × 108 fold rate enhancement compared to non-catalyzed DNA cleavage, which is significant.  相似文献

(E)-2-(2-hydroxybenzylideneamino)isoindoline-1,3-dione (Hbid) was prepared by condensation of N-aminophthalimide and salicylaldehyde and characterized by elemental analysis, IR, 1H-NMR, and mass spectral studies. Mononuclear complexes [(phen)CuII(μ-Hbid)2H2O] (1), [(phen)CoII(Cl)2(μ-Hbid)]6H2O (2) (phen?=?1,10-phenanthroline) and binuclear complexes [CuII(μ-Hbid)]2 (3), and [CoII(μ-Hbid)]2 (4) with Hbid were prepared and characterized by elemental analysis, IR, UV-Vis, molar conductance, and thermogravimetric (TG) techniques. DNA-binding properties of 14 were investigated by UV spectroscopy, fluorescence spectroscopy, and viscosity measurements. The results suggest that 1 and 2 bind to DNA by partial intercalation, whereas 3 and 4 find different groove-binding sites. The cleavage of these complexes with super coiled pUC19 has been studied using gel electrophoresis; all the complexes displayed chemical nuclease activity in the absence and presence of H2O2 via an oxidative mechanism. Complexes 14 inhibit the growth of both Gram-positive and Gram-negative bacteria.  相似文献

Two mixed-ligand complexes, [Cu(L)(2imi)] (1) and [Ni(L)(2imi)]·MeOH (2) [L = 2-(((5-chloro-2-oxyphenyl)imino)methyl)phenolato) and 2imi = 2-methyl imidazole], have been prepared by the reaction of appropriate metal salts with H2L and 2-methyl imidazole. Their structures were characterized by microanalysis, FT-IR, UV–vis, molar conductivity, and 1H NMR for [Ni(L)(2imi)]·MeOH. The structures were determined using single crystal X-ray diffraction. Each four-coordinate metal center, Cu(II) in 1 and Ni(II) in 2, is surrounded by donors of Schiff base (L2?) and N of 2-methyl imidazole in square planar geometries. α-Amylase activities of these compounds have also been investigated. The experimental data showed that α-amylase was inhibited by Ni(II) complex while the Cu(II) complex causes a 1.3-fold decrease in Km value. Antimicrobial results show that these compounds, especially the Cu(II) complex, have potential for antibacterial activity against Gram negative and Gram positive bacteria and antifungal activity against Aspergillus fumigatus.  相似文献

Reliable compounds with low toxicity are tempting potential chemotherapeutics. With an aim of achieving less toxic but more potent metallodrugs, four new‐generation hydrophilic Cu(II) and Zn(II) complexes with DNA‐targeting properties were synthesized and characterized using various physicochemical data. The excellent DNA binding and cleavage results confirmed the mode of binding of DNA with the complexes and their ability to denature it. The profound in vitro cytotoxicity exhibited by complex 3 against a panel of cell lines (HeLa, MCF‐7 and HepG‐2) along with NHDF (normal human dermal fibroblasts) with distinct activity towards HepG‐2 and low toxicity to NHDF prompted in vivo studies of induced hepatocellular carcinoma‐affected Swiss albino rats. On evaluating various serum hepatic, biological and histopathological parameters, complex 3 showed excellent activity in restoring the damaged liver to normal. As a means of identifying the pathway of DNA damage, flow cytometric evaluation of cell cycle analysis was performed, which revealed S phase arrest‐induced apoptosis in HepG‐2 cells by complex 3 , making it a cell cycle‐specific drug.  相似文献
