Three novel copper(II) complexes, [Cu(Gly‐l ‐Val)(HPBM)(H2O)]·ClO4·H2O ( 1 ), [Cu(Gly‐l ‐Val)(TBZ)(H2O)]·ClO4 ( 2 ) and [Cu(Gly‐l ‐Val)(PBO)(H2O)]·ClO4 ( 3 ) (Gly‐l ‐Val = glycyl‐l ‐valine anion, HPBM = 5‐methyl‐2‐(2′‐pyridyl)benzimidazole, TBZ = 2‐(4′‐thiazolyl)benzimidazole, PBO = 2‐(2′‐pyridyl)benzoxazole), have been prepared and characterized with elemental analyses, conductivity measurements as well as various spectroscopic techniques. The interactions of these copper complexes with calf thymus DNA were explored using UV–visible, fluorescence, circular dichroism, thermal denaturation, viscosity and docking analyses methods. The experimental results showed that all three complexes could bind to DNA via an intercalative mode. Moreover, the cytotoxic effects were evaluated using the MTT method, and the antimicrobial activity of these complexes was tested against Bacillus subtilis, Staphylococcus aureus, Escherichia coli and Pseudomonas aeruginosa. The results showed that the activities are consistent with their DNA binding abilities, following the order of 1 > 2 > 3 .  相似文献

The compounds [(η6p‐cymene)RuCl2(4‐nitroaniline)] and [(η6p‐cymene)RuCl2(2‐halogen‐4‐nitroaniline)] were synthesized and characterized by various means. The [(η6p‐cymene)RuCl2(4‐nitroaniline)] and [(η6p‐cymene)RuCl2(2‐fluoro‐4‐nitroaniline)] compounds were determined by X‐ray diffraction, appearing in a distorted piano‐stool type of arrangement with similar bond lengths and angles around the ruthenium. The compounds exhibited moderate to strong in vitro cytotoxicity against A549 and MCF‐7 human cancer cells. Substitution of heavy halogen atom on the ortho position of para‐nitroaniline weakened the cytotoxicity against both of MCF‐7 and A549, except the cases of fluorine substitution for hydrogen atom regarding A549 and bromine substitution for chlorine atom regarding MCF‐7, which showed minor deviation.  相似文献

Two novel cyclotriphosphazene ligands ( 2 and 3 ) bearing 3‐oxypyridine groups and their corresponding Ru(II) complexes ( 4 and 5 ) were synthesized and their structures were characterized using Fourier transform infrared, 1H NMR and 31P NMR spectroscopic data and elemental analysis. The Ru(II) complexes were used as catalysts for catalytic transfer hydrogenation of p‐substituted acetophenone derivatives in the presence of KOH. Additionally, the cytotoxic activities of compounds 2 , 3 , 4 , 5 were evaluated against PC3 (human prostate cancer), DLD‐1 (human colorectal cancer), HeLa (human cervical cancer) and PNT1A (normal human prostate) cell lines. Finally the antimicrobial activities of compounds 2 , 3 , 4 , 5 were evaluated against a panel of Gram‐positive and Gram‐negative bacteria and yeast cultures. The complexes showed efficient catalytic activity towards transfer hydrogenation of acetophenone derivatives, especially those bearing electron‐withdrawing substituents on the para‐position of the aryl ring. The compounds were found to have moderate to high cytotoxic and antimicrobial activities, and Ru(II) complexation enhanced both cytotoxic and antimicrobial activities in comparison with the parent compounds. Eight novel Pt(II), Pd(II), Cu(II) and Zn(II) complexes with 4’‐substituted terpyridine were synthesized and characterized by elemental analysis, UV, IR, NMR, electron paramagnetic resonance, high‐resolution mass spectrometry and molar conductivity measurements. The cytotoxicity of these complexes against HL‐60, BGC‐823, KB and Bel‐7402 cell lines was evaluated by MTT assay. All the complexes displayed cytotoxicity with low IC50 values (<20 μm ) and showed selectivity. Complexes 3 , 5 , 7 and 8 exerted 9‐, 5‐, 12‐ and 7‐fold higher cytotoxicity than cisplatin against Bel‐7402 cell line. The cytotoxicity of complexes 3 , 5 , 6 , 7 and 8 was higher than that of cisplatin against BGC‐823 cell line. Complexes 3 , 7 and 8 showed similar cytotoxicity to cisplatin against KB cell line. Complex 7 exhibited higher cytotoxicity than cisplatin against HL‐60 cell line. Among these complexes, complex 7 demonstrated the highest in vitro cytotoxicity, with IC50 values of 1.62, 3.59, 2.28 and 0.63 μm against HL‐60, BGC‐823, Bel‐7402 and KB cells lines, respectively. The results suggest that the cytotoxicity of these complexes is related to the nature of the terminal group of the ligand, the metal center and the leaving groups. Interaction of chiral Ru(II) salen complexes (S)-1 and (R)-1 with Calf Thymus DNA (CT-DNA) was studied by absorption spectroscopy, competitive binding study, viscosity measurements, CD measurements, thermal denaturation study and cleavage studies by agarose gel electrophoresis. The DNA binding affinity of (S)-1 (6.25 × 103 M−1) was found to be greater than (R)-1 (3.0 × 103 M−1). The antimicrobial studies of these complexes on five different gram (+)/(−) bacteria and three different fungal organisms showed selective inhibition of the growth of gram (+) bacteria and were not affective against gram (−) and fungal organisms. Further, the (S)-1 enantiomer inhibited the growth of organisms to a greater extent as compared to (R)-1 enantiomer.  相似文献

A series of half‐sandwich Ru(II)–arene complexes [Ru(η6‐benzene)(diimine)Cl](PF6) ( 1 – 4 ), where diimine is 1,10‐phenanthroline ( 1 ), 5,6‐dimethyl‐1,10‐phenanthroline ( 2 ), dipyrido[3,2‐a:2′,3′‐c]phenazine ( 3 ) or 11,12‐dimethyldipyrido[3,2‐a:2′,3′‐c]phenazine ( 4 ), have been isolated and characterized using analytical and spectral methods. Complex 2 possesses a familiar pseudo‐octahedral ‘piano‐stool’ structure. The intrinsic DNA binding affinity of the complexes depends upon the diimine ligand: 3 (dppz) > 4 (11,12‐dmdppz) > 2 (5,6‐dmp) > 1 (phen). The π‐stacking interaction of extended planar ring of coordinated dppz ( 3 ) in between the DNA base pairs is more intimate than that of phen ( 1 ), and the incorporation of methyl groups on the dppz ring ( 4 ) discourages the stacking interaction leading to a lower DNA binding affinity for 4 than 3 . Docking studies show that all the complexes bind in the major groove of DNA. Interestingly, 3 shows an ability to convert supercoiled DNA into nicked circular DNA even at 20 μM concentration beyond which complete oxidative DNA degradation is observed. The protein binding affinity of the complexes decreases in the order 4 > 3 > 2 > 1 , and the higher protein binding affinity of 4 illustrates the strong involvement of methyl groups on dppz ring in hydrophobic interaction with protein. Also, 4 cleaves protein more efficiently than the other complexes in the presence of H2O2. It is notable that 2 , 3 and 4 display cytotoxicity against human cervical cancer cell lines (SiHa) with potency higher than the currently used drug cisplatin. Acridine orange/ethidium bromide staining studies reveal that 3 induces apoptosis in cancer cells much more efficiently than 4 .  相似文献

A new cobalt(II) complex ( 1 ) of 5‐chloro‐8‐hydroxyquinoline was prepared and structurally characterized using infrared spectroscopy, electrospray ionization mass spectrometry, elemental analysis, single‐crystal X‐ray diffraction as well as powder X‐ray diffraction. Its biological activities including DNA binding and anticancer activity were investigated. The DNA binding study of complex 1 suggested that it interacted with calf thymus DNA mainly via an intercalative binding mode. The in vitro anticancer activity of complex 1 was screened against a series of tumor cell lines as well as the normal liver cell line HL‐7702 using MTT assay. complex 1 showed much higher cytotoxicity than corresponding metal salt and ligand towards the five tested tumor cell lines, in which T‐24 was the most sensitive tumor cell line towards 1, with IC50 value of 7.04 ± 0.06 μM. complex 1 was found to greatly induce cell cycle arrest in T‐24 cells at S phase, and consequently to induce cell apoptosis in a dose‐dependent mode suggested by cell apoptosis analysis via Hoechst 33258 and acridine orange/ethidium bromide staining assays. The cell apoptosis mechanism of 1 was studied targeting the mitochondrion‐mediated pathway, since the apoptotic mechanism in the T‐24 cells treated by 1 was investigated by reactive oxygen species (ROS) detection, intracellular calcium concentration measurement and caspase‐9/3 activity assay. The results suggested that the cell apoptosis induced by 1 was closely related to the loss of mitochondrial membrane potential, ROS production and enhancement of intracellular [Ca2+], which would trigger the caspase‐9/3 activation via a mitochondrial dysfunction pathway. A series of new complexes of oxovanadium(IV) [VO(L)(B)] and ruthenium(II) [Ru(CO)(PPh3)2(L)] ( 1.1- 1.3,  2.1–2.3 ) (H2L = dehydroacetic acid Schiff base of S‐methyldithiocarbazate, H2smdha ( 1 ) or S‐benzyldithiocarbazate, H2sbdha ( 2 ); B = 2,2′‐bipyridine (bpy) or 1,10‐phenanthroline (phen)) have been synthesized. The structure of these complexes was authenticated using elemental analyses and spectroscopic techniques, and their magnetic properties and electrochemical behaviour were studied. The molecular structures of oxovanadium(IV) complexes [VO(smdha)(bpy)]?CH2Cl2 ( 1.1 ) and [VO(sbdha)(phen)]?2H2O ( 2.2 ) were confirmed using single‐crystal X‐ray crystallography. Analytical data showed that the ligands 1 and 2 are chelated to the metal centres in a bi‐negative tridentate fashion through azomethine N, thiol S and deprotonated hydroxyl group. The antioxidant activity of the synthesized compounds was tested against 2,2‐diphenyl‐1‐picrylhydrazyl) radical, which showed that the complexes demonstrate a better scavenging activity than their corresponding ligands. The cupric ion reducing antioxidant capacity method was also employed and the total equivalent antioxidant capacity values were found to be higher for the oxovandium(IV) complexes. DNA binding affinity of the compounds was determined using UV–visible and fluorescence spectra, revealing an intercalation binding mode. Higher cytotoxicity for the complexes compared to their ligands was found against human liver hepatocellular carcinoma (HepG2) and breast adenocarcinoma (MCF7) cell lines using MTT assay.  相似文献

New palladium(II) complexes, [Pd(PPh3)L] ( 2 ) and [Pd(AsPh3)L] ( 3 ), were synthesized using 4‐hydroxybenzoic acid (3‐ethoxy‐2‐hydroxybenzylidene)hydrazide ( 1 ) ligand (H2L), and characterized using various physicochemical techniques. The molecular structures of 2 and 3 were determined using single‐crystal X‐ray diffraction, which reveals a square planar geometry around the palladium(II) metal ion. In vitro DNA binding studies were conducted using UV–visible absorption spectroscopy, emission spectroscopy, cyclic voltammetry and viscosity measurements, which suggest that the metal complexes act as efficient DNA binders. The interaction of ligand H2L and complexes 2 and 3 with bovine serum albumin (BSA) was investigated using UV–visible and fluorescence spectroscopies. Absorption and emission spectral studies indicate that complexes 2 and 3 interact with BSA protein more strongly than the parent ligand. The free radical scavenging potential of all the synthesised compounds ( 1 – 3 ) was also investigated under in vitro conditions. In addition, the in vitro cytotoxicity of the complexes to tumour cells lines (HeLa and MCF‐7) was examined using the MTT assay method.  相似文献

Two transition metal complexes, [Cu(FH)3]⋅2Cl⋅2H2O and [Ni(FH)3]⋅2Cl⋅2H2O, were synthesized from the reactions of furan‐2‐carboxylic acid hydrazide with CuCl2⋅2H2O and NiCl2⋅6H2O. The synthesized complexes were characterized using analytical and various spectral techniques. The structures of the complexes were determined using single‐crystal X‐ray diffraction. The interactions of the complexes with calf thymus DNA (CT‐DNA) were studied using absorption, fluorescence, cyclic voltammetric and viscosity measurements. The experimental results showed that the complexes could interact with CT‐DNA through intercalation. A gel electrophoresis assay demonstrated the ability of the complexes to cleave pBR322 DNA. The binding interaction of the complexes with bovine serum albumin was investigated using a fluorescence spectroscopic method. The radical scavenging ability, assessed using a series of antioxidant assays involving 2,2‐diphenyl‐2‐picrylhydrazyl radical, hydroxyl radical and nitric oxide radical, showed that the complexes possess significant radical scavenging properties. Further, the in vitro cytotoxic effect of the complexes examined on cancerous cell lines, such as human cervical cancer cells (HeLa) and human breast cancer cell line (MCF‐7), showed that the complexes exhibit significant anticancer activity.  相似文献

Three water‐soluble complexes, [Cu2L2Cl2] ( 1 ), [CoL2(im)2] ( 2 ) and [ZnLClH2O] ( 3 ) (HL = 8‐hydroxyquinoline‐5‐sulphonic acid; im = N ‐methylimidazole), were prepared and characterized using various spectral techniques. The DNA binding behaviour of complexes 1 – 3 was studied using UV–visible and circular dichroism (CD) spectra and cyclic voltammetry. All three complexes exhibit hypochromism but complexes 1 and 3 alone give a red shift of 4 nm with a significant binding constant of K b = 2.1 × 104 and 1.0 × 104 M−1, respectively, but complex 2 shows no red shift with lower K b of 4.1 × 103 M−1. The voltammetric E 1/2 of complex 1 on interaction with herring sperm DNA shifts to a more positive potential, as expected, than complex 2 due to higher DNA affinity. Additionally, analysis of electrochemical data yields a value of K +/K 2+ greater than one suggesting that complex 1 binds to DNA through intercalation in the M(I) state. Evidently in CD spectral analysis, complex 1 exhibits a decrease in molar ellipticity with a red shift of 10 nm and a significant decrease in intensity compared to complexes 2 and 3 . This clearly indicates that complex 1 induces the B → A transition to a greater extent than 2 and 3 . Oxidative cleavage using circular plasmid pUC18 DNA with complex 1 was investigated using gel electrophoresis. Interestingly, complex 1 displays a strong DNA binding affinity and is efficient in cleaving DNA in the presence of H2O2 at pH = 8.0 at 37 °C.  相似文献

DNA and protein binding affinities of the synthesized compounds were examined using UV–visible and fluorescence titration method. Platinum(II) complexes with various selenones (L) having the general formula [PtL2Cl2] were prepared and characterized by elemental analysis and, IR and NMR (1H, 13C, and 77Se) spectroscopies. A decrease in the IR frequency of the >C=Se mode and an upfield shift in 13C NMR for the >C=Se resonance of selenones are consistent with their selenium coordination to platinum(II). The NMR data show that the complexes are stable in solution and do not undergo equilibration at 297 K. The geometrical structures of the complexes were predicted theoretically (with DFT method) using Gaussian09 program. DFT calculations predicted that the trans configurations were up to 1.7 kcal/mol more stable than the cis forms in gas phase, while in solution form the cis isomers were predicted to be more stable. The UV–vis spectra of the two complexes, 6 and 7 were also recorded at room temperature for 24 h and it was observed that the complexes were stable and did not undergo decomposition. The in vitro antitumor properties of the complexes as well as of cisplatin were evaluated on two human cancer cell lines, HeLa (cervical cancer cells) and MCF7 (breast cancer cells) using MTT assay. The results indicated that the prepared complexes exerted significant inhibition on the selected cancer cells.  相似文献

A complex of formula [Ni(pobb)2](pic)2, (pobb = 1,3‐bis(1‐propylbenzimidazol‐2‐yl)‐2‐oxapropane, pic = 2,4,6‐trinitrophenol), has been synthesized and structurally characterized by physico‐chemical and spectroscopic methods. The crystals crystallize in the monoclinic system, space group C2/c, a = 25.766(11) Å, b = 14.943(7) Å, c = 19.543(14) Å, α = 90°, β = 129.722(4)°, γ = 90°, Z = 4. The coordination environment around nickel(II) atom can be described as a distorted octahedral geometry. The interactions of the ligand pobb and the nickel (II) complex with calf thymus DNA (CT‐DNA) are investigated by using electronic absorption titration, ethidium bromide‐DNA displacement experiments and viscosity measurements. The experimental evidence indicated the compounds interact with calf thymus DNA through intercalation.  相似文献

Reliable compounds with low toxicity are tempting potential chemotherapeutics. With an aim of achieving less toxic but more potent metallodrugs, four new‐generation hydrophilic Cu(II) and Zn(II) complexes with DNA‐targeting properties were synthesized and characterized using various physicochemical data. The excellent DNA binding and cleavage results confirmed the mode of binding of DNA with the complexes and their ability to denature it. The profound in vitro cytotoxicity exhibited by complex 3 against a panel of cell lines (HeLa, MCF‐7 and HepG‐2) along with NHDF (normal human dermal fibroblasts) with distinct activity towards HepG‐2 and low toxicity to NHDF prompted in vivo studies of induced hepatocellular carcinoma‐affected Swiss albino rats. On evaluating various serum hepatic, biological and histopathological parameters, complex 3 showed excellent activity in restoring the damaged liver to normal. As a means of identifying the pathway of DNA damage, flow cytometric evaluation of cell cycle analysis was performed, which revealed S phase arrest‐induced apoptosis in HepG‐2 cells by complex 3 , making it a cell cycle‐specific drug.  相似文献

Two isomeric Zinc (II) complexes constructed by 3,5‐bis(1‐imidazoly) pyridine has been synthesized and characterized by single crystal X‐ray diffraction, elemental analyses and infrared spectroscopy. The binding mode and ability of complex 1–2 with CT‐DNA were studied by UV and fluorescence spectra. The intrinsic binding constant Kb (Kb1 = 2.305 × 104 M?1, Kb2 = 3.095 × 104 M?1) and the observed association constant Kobs (Kobs1 = 1.523*106 M?1, Kobs2 = 2.057*106 M?1) indicated that the insertion ability of complex 2 with CT‐DNA is stronger than complex 1. Gel electrophoresis showed that complexes have a good ability to hydrolyze cleavage pBR322 plasmid DNA. The cytotoxicity and apoptosis studies showed that complexes exhibited excellent cytotoxic activity against HeLa cells, especially complex 2 had better growth inhibition than Cisplatin. Molecular docking study simulated the binding model of complexes with DNA (PDB:4av1), showing an imidazole plane of complex 2 can be inserted into a DNA base pair in relative parallel. Both complexes can be used as potential anticancer agents.  相似文献

Binuclear polypyridine ruthenium compounds have been shown to slowly intercalate into DNA, following a fast initial binding on the DNA surface. For these compounds, intercalation requires threading of a bulky substituent, containing one RuII, through the DNA base‐pair stack, and the accompanying DNA duplex distortions are much more severe than with intercalation of mononuclear compounds. Structural understanding of the process of intercalation may greatly gain from a characterisation of the initial interactions between binuclear RuII compounds and DNA. We report a structural NMR study on the binuclear RuII intercalator Λ,Λ‐B (Λ,Λ‐[μ‐bidppz(bipy)4Ru2]4+; bidppz=11,11′‐bis(dipyrido[3,2‐a:2′,3′‐c]phenazinyl, bipy = 2,2′‐bipyridine) mixed with the palindromic DNA [d(CGCGAATTCGCG)]2. Threading of Λ,Λ‐B depends on the presence and length of AT stretches in the DNA. Therefore, the latter was selected to promote initial binding, but due to the short stretch of AT base pairs, final intercalation is prevented. Structural calculations provide a model for the interaction: Λ,Λ‐B is trapped in a well‐defined surface‐bound state consisting of an eccentric minor‐groove binding. Most of the interaction enthalpy originates from electrostatic and van der Waals contacts, whereas intermolecular hydrogen bonds may help to define a unique position of Λ,Λ‐B. Molecular dynamics simulations show that this minor‐groove binding mode is stable on a nanosecond scale. To the best of our knowledge, this is the first structural study by NMR spectroscopy on a binuclear Ru compound bound to DNA. In the calculated structure, one of the positively charged Ru2+ moieties is near the central AATT region; this is favourable in view of potential intercalation as observed by optical methods for DNA with longer AT stretches. Circular dichroism (CD) spectroscopy suggests that a similar binding geometry is formed in mixtures of Λ,Λ‐B with natural calf thymus DNA. The present minor‐groove binding mode is proposed to represent the initial surface interactions of binuclear RuII compounds prior to intercalation into AT‐rich DNA.  相似文献

The synthesis and characterization of Ru (II) terpyridine complexes derived from 4′ functionalized 2,2′:6′,2″‐terpyridine (tpy) ligands are reported. The heteroleptic complexes comprise the synthesized ligands 4′‐(2‐thienyl)‐ 2,2′:6′,2″‐terpyridine) or (4′‐(3,4‐dimethoxyphenyl)‐2,2′:6′,2″‐terpyridine and (dimethyl 5‐(pyrimidin‐5‐yl)isophthalate). The new complexes [Ru(4′‐(2‐thienyl)‐2,2′:6′,2″‐terpyridine)(5‐(pyrimidin‐5‐yl)‐isophthalic acid)Cl2] ( 9 ), [Ru(4′‐(3,4‐dimethoxyphenyl)‐2,2′:6′,2″‐terpyridine)(5‐(pyrimidin‐5‐yl)‐isophthalic acid)Cl2] ( 10 ), and [Ru(4′‐(2‐thienyl)‐2,2′:6′,2″‐terpyridine)(5‐(pyrimidin‐5‐yl)‐isophthalic acid)(NCS)2] ( 11 ) were characterized by 1H‐ and 13C‐NMR spectroscopy, C, H, N, and S elemental analysis, UPLC‐ESI‐MS, TGA, FT‐IR, and UV‐Vis spectroscopy. The biological activities of the synthesized ligands and their Ru (II) complexes as anti‐inflammatory, antimicrobial, and anticancer agents were evaluated. Furthermore, the toxicity of the synthesized compounds was studied and compared with the standard drugs, namely, diclofenac potassium and ibuprofen, using hemolysis assay. The results indicated that the ligands and the complex 9 possess superior anti‐inflammatory activities inhibiting albumin denaturation (89.88–100%) compared with the standard drugs (51.5–88.37%) at a concentration of 500 μg g?1. These activities were related to the presence of the chelating N‐atoms in the ligands and the exchangeable chloro‐ groups in the complex. Moreover, the chloro‐ and thiophene groups in complex 9 produce a higher anticancer activity compared with its isothiocyanate derivative in the complex 11 and the 3,4‐dimethoxyphenyl moiety in complex 10 . Considering the toxicity results, the synthesized ligands are nontoxic or far less toxic compared with the standard drugs and the metal complexes. Therefore, these newly synthesized compounds are promising anti‐inflammatory agents in addition to their moderate unique broad antimicrobial activity.  相似文献

A water soluble chloro bridged binuclear copper(II) complex (3) and mononuclear complex (4) have been synthesized from chloro substituted 2‐oxo‐1,2‐dihydroquinolin‐3‐yl‐methylene‐2 hydroxybenzohydrazide 1 and 2 and CuCl2·2H2O. The structures of the complexes have been determined by single crystal X‐ray diffraction. The binding interactions of the ligands and complexes with CT‐DNA and protein have been evaluated by absorption and emission spectroscopic method. CT‐DNA and ethidium bromide (EB) competitive studies revealed that the compounds could interact with CT‐DNA through intercalation binding mode. Interactions of the compounds with BSA were also studied by UV−visible, fluorescence and synchronous fluorescence spectroscopic methods which showed that the compounds had a strong binding affinity with BSA through static quenching process. The synthesis, spectral characterization, and biological studies of ruthenium(II) hydrazone complexes [RuCl(CO)(PPh3)2L] (where L = hydrazone ligands) have been carried out. The hydrazones are monobasic bidentate ligands with O and N as the donors and are preferably found in the enol form in all the complexes. The molecular structure of the ligands HL1, HL2, and HL3 were determined by single-crystal X-ray diffraction. The DNA binding studies of the ligands and complexes were carried out by absorption spectroscopic and viscosity measurements. The results revealed that the ligands and complexes bind to DNA via intercalation. The DNA cleavage activity of the complexes, evaluated by gel electrophoresis assay, revealed that the complexes are good DNA cleaving agents. The antioxidant properties of the complexes were evaluated against DPPH, OH, and NO radicals, which showed that the complexes have strong radical-scavenging. Further, the in vitro cytotoxic effect of the complexes examined on HeLa and MCF-7 cancer cell lines showed that the complexes exhibited significant anticancer activity.  相似文献
