The reactions of 2-(1-hydropolyfluoro-1-alkenyl)-4H-3,1-benzoxin-4-ones (2) with hydrazine hydrate and phenyl hydrazine were investigated. The reaction of 2 with hydrazine hydrate in ethanol under reflux condition readily gave 2-fluoroalkyl-4H-pyrazolo[5,1-b]quinazolin-9-ones (3) in high yields. The reaction of 2 with phenyl hydrazine, however, resulted in the formation of 2-(2-phenyl-5-fluoroalkyl-2H-pyrazol-3-yl) benzoic acids (7). Further treatment of 7 with PPA gave 1-phenyl-4,9-dihydro-3-fluoroalkyl-1H-pyrozolo[3,4-b]quinolin-4-ones (4) in 65-80% overall yields.  相似文献

A method for the preparation of 3-fluoroalkyl substituted 2-aza-butadienes 6 by aza-Wittig reaction of 3-fluoroalkyl-N-vinylic phosphazenes 4 and aldehydes 5 is reported. [4+2] Cycloaddition reaction of these heterodienes 6 with enamines 9 gives fluoroalkyl substituted pyridine 15, 16, 24-27 and isoquinoline 12-14, 20 derivatives.  相似文献

N-Arylmethyl-7-azabicyclo[2.2.1]heptane (I) derivatives have been synthesized by deprotection of N-protected, N-(arylmethyl)cyclohex-3-enamines, bromination of the resulting secondary cyclohex-3-enamines, followed by base-promoted cyclization (route a), or by bromination of N-protected, N-(arylmethyl)cyclohex-3-enamines followed by deprotection and base-mediated cyclization (route b). In these protocols we have observed that the bromination of the key intermediates (12, 13, and 19) is stereoselective leading to major trans-3-cis-4-dibromides (14, 17, and 20), whose mild base-mediated heterocyclization (on compound 14), or the two-step acid hydrolysis plus base-promoted cyclization (on compounds 17 and 20), gave products 6 and 7 in good yield. A mechanistic investigation using DFT has been carried out to explain the results observed in this work.  相似文献

Ytterbium-catalyzed tandem carboalkoxylation/Friedel-Crafts reaction of arylidenecyclopropanes 1 with acetals 2 afforded the corresponding indene derivatives 3 in good to high yields. For example, in the presence of 10 mol % of Yb(OTf)3 the reaction of 1-phenylbenzylidenecyclopropane 1a with the dimethyl acetals of benzaldehyde 2a, p-tolualdehyde 2b, and p-anisaldehyde 2c gave 1,3-diphenyl-2-(2-methoxyethyl)indene 3a, 2-(2-methoxyethyl)-3-phenyl-1-(p-tolyl)indene 3b, and 1-(p-anisyl)-2-(2-methoxyethyl)-3-phenylindene 3c in 82%, 80%, and 80% yields, respectively.  相似文献

New catecholate Sb(V) complexes triphenyl(3,6-di-tert-butylcatecholato)antimony(V) Ph3Sb(3,6-DBCat) (1) and triphenyl(perchloroxanthrenecatecholato)antimony(V) Ph3Sb(OXCatCl) (2) were synthesized by the oxidative addition reaction of corresponding o-quinones (3,6-di-tert-butyl-o-benzoquinone and perchloroxanthrenequinone-2,3) with triphenylantimony. Catecholates 1 and 2 can alternatively be synthesized by reacting the appropriate thallium catecholate with triphenylantimony dichloride. The oxidative addition reaction of an equimolar ratio of 4,4′-di-(3-methyl-6-tert-butyl-o-benzoquinone) and triphenylantimony yielded 4-(2-methyl-5-tert-butyl-cyclohexadien-1,5-dion-3,4-yl)-(3-methyl-6-tert-butyl-catecholato)triphenylantimony(V) Ph3Sb(Cat-Q) (3); in the case of a 1:2 molar ratio, complex 4,4′-di-[(3-methyl-6-tert-butyl-catecholato)triphenylantimony(V)] Ph3Sb(Cat-Cat)SbPh3 (4) resulted. Complexes 1-4 were characterized by IR- and 1H NMR spectroscopy. Molecular structures of 1, 2 and 4 were determined by X-ray crystallography to be a distorted tetragonal-pyramidal.  相似文献

2-Alkoxyiminoimidazolidines 2-3 react with acetylene dicarboxylates and ethyl phenylpropiolate to give 8-alkoxy-imidazo[1,2-a]pyrimidin-5(3H)-ones C, which subsequently undergo a sterically induced multihetero-retro-ene fragmentation to give imidazo[1,2-a]pyrimidin-5(1H)-ones 4-7 together with formaldehyde or benzaldehyde. On the other hand, a similar reaction of 2-3 with ethyl propiolate gives corresponding 8-alkoxy-imidazo[1,2-a]pyrimidin-5(3H)-ones 8-10. The unsubstituted imidazo[1,2-a]pyrimidin-5(1H)-one 11 can be prepared by retro-ene reaction of 9 upon prolonged heating in refluxing ethanol. A direct synthetic approach to 1-formyl-7-phenyl-imidazo[1,2-a]pyrimidine-5(1H)-one 14 is reported using DMF/sulfonyl chloride as a new Vilsmeier-type N-formylating reagent.  相似文献

The first synthesis of a series of nitro-substituted 2,2-diphenyl-2H-1-benzopyrans is reported. Our synthetic approach is based on a linear synthesis in two steps from appropriate brominated 2,2-diphenyl-2H-1-benzopyrans 12-17, which requires the preliminary preparation of bromophenols 7-11. These latter were easily obtained by the reaction of phenols 1-5 with a mild and selective brominating agent tetrabutylammonium tribromide (TBA·Br3). The key intermediates 12-17 were efficiently elaborated through an univocal classic chromenization between the commercially available 1,1-diphenyl-2-yn-1-ol and the brominated phenols 6-11. The compounds 12-17 so obtained were converted into arylboronic acids 18-23 by a metalation/boronylation sequence, followed by acid hydrolysis. From advanced building blocks 18-23, the introduction of nitro group, which constitutes the ultimate step of our strategy, was achieved by an ipso-nitration reaction using the Crivello's reagent. This highly selective method provides only the ipso-nitrated products 24-29 in moderate to high yield.  相似文献

Two new types of pyrido-fused tris-heterocycles (1a,b and 2a,b) have been prepared from 3-aminopyridine in five/six steps. A synthetic strategy for the preparation of the novel pyrido[3,4-b]thieno[2,3- and 3,2-d]pyrroles (1a,b) and pyrido[4,3-e]thieno[2,3- and 3,2-c]pyridazines (2a,b) has been studied. The Suzuki cross coupling of the appropriate 2- and 3-thienoboronic acids (3,4) and 4-bromo-3-pyridylpivaloylamide (9) afforded the biaryl coupling products (10,11) in high yields (85%). Diazotization of the hydrolysed (2-thienyl)-coupling product (12) and azide substitution gave the 3-azido-4-(2-thienyl)pyridine intermediate (72%, 14). 3-Azido-4-(3-thienyl)pyridine (15) was prepared by exchanging the previous order of reactions. The desired β-carboline thiophene analogues (1a,b) were obtained via the nitrene by thermal decomposition of the azido precursors (14,15). By optimising conditions for intramolecular diazocoupling, the corresponding pyridazine products (72-83%, 2a,b) were afforded.  相似文献

Novel 6-chloropyrazolo[3,4-b]pyridine-5-carbaldehydes 5 have been synthesized from the 4,5-dihydropyrazolo[3,4-b]pyridine-6-ones 4 via Vilsmeier-Haack reaction. Further treatment of carbaldehydes 5 with acetophenones 6 and hydrazine hydrate afforded chalcone analogues 7 and dipyrazolo[3,4-b:4′,3-e]pyridines 8, respectively.  相似文献

tert-Butyldimethylsililoxy-2-aza-1,3-butadienes react with 2H-azirine 3 leading to Diels-Alder cycloadducts in moderate yields. The reactions are endo- and regioselective with the azirine being added by its less hindered face. There is only one product in the case of 1b, 4b. There are two isomers (4 and 5) from 1a, 1c and 1d. A different result was obtained with the diene 1e. Diene 1e formed products 4e and 8. Some of compounds 4 and 5 have been hydrolysed leading to functionalised aziridines 7. Thermolysis of substituted methyl 1-methyleneamino-4,5-dioxo-4,5-dihydro-1H-pyrrole-2-carboxylates 2a,b led to substituted dimethyl 3,9-dioxo-1,5,7,11-tetrahydro-1H,7H-dipyrazolo[1,2-a;1′,2′-d][1,2,4,5]tetrazine-1,7-dicarboxylates 4a,b and methyl 2,5-dihydro-5-oxo-1H-pyrazole-3-carboxylates 5a,b as minor products. The structure of compound 4a was determined by X-ray crystallography. The proposed mechanism of this conversion includes generation of (N-methyleneamino)imidoylketenes 6a,b and its intramolecular transformation to azomethine imines—5-oxo-2,5-dihydropyrazole-1-methylium-2-ides 7a,b, which undergo dimerization in head-to-tail manner yielding products 4a,b and partially hydrolyse to compounds 5a,b.  相似文献

Substituent-controlled regioselective synthesis of highly functionalized 1,2-teraryls 3a-k has been achieved through ring transformation of 6-aryl-4-(pyrrolidin-1-yl/piperidin-1-yl)-2H-pyran-2-one-3-carbonitriles 1a-g by aryl acetones 2a-c in the presence of powdered KOH in DMF in very good yield. Under similar reaction conditions, 6-aryl-4-methylsulfanyl-2H-pyran-2-ones 5a-f afforded 1,7-diaryl-2-methyl-4H,5H-pyrano[3,4-c]pyran-4,5-diones 6a-j as major products and 3,4-diaryl-2-methyl-6-methylsulfanylbenzonitriles as minor constituents 7a-j.  相似文献

An acid-promoted intramolecular Friedel-Crafts reaction of the Morita-Baylis-Hillman adducts 3 derived from 2-arylthioquinolin-3-carbaldehydes 2 was investigated. Interestingly, promoted by sulfuric acid, the substrates with electron-donating groups on the aromatic ring gave six-membered fused-ring 12H-thiochromeno[2,3-b]quinolines 4 with good yields, while those with electron-withdrawing groups afforded eight-membered fused-ring 5H-benzo[7,8]thiocino[2,3-b]quinolines 5 in moderate yields. Using triflic acid instead of sulfuric acid, only products 4 could be obtained under the similar condition.  相似文献

A novel and efficient regioselective synthesis of various arylated highly congested 7-aryl-5-methylsulfanylindan-4-carbonitriles (3a-f), methyl 7-aryl-5-methylsulfanylindan-4-carboxylates (10a-e) and 7-aryl-5-methylsulfanylindan-4-carboxylic acids (11a-e) through base-catalyzed reaction of 6-aryl-4-methylsulfanyl-2-oxo-2H-pyran-3-carbonitriles (1a-f) and methyl 6-aryl-4-methylsulfanyl-2-oxo-2H-pyran-3-carboxylates (9a-e) by cyclopentanone (2) has been delineated. The synthetic potential of 2-pyranone was explored further to generate molecular diversity using 6-aryl-4-sec-amino-2-oxo-2H-pyran-3-carbonitriles (7a-h), 5,6-diaryl-4-methylsulfanyl-2-oxo-2H-pyran-3-carbonitriles (5a,b) and methyl 5,6-diaryl-4-methylsulfanyl-2-oxo-2H-pyran-3-carboxylates (12a,b) as precursors for the ring transformation by cyclopentanone to assess the effects of substituents on the course of the reaction to obtain highly congested indans, 6,7-diaryl-5-methylsulfanylindan-4-carbonitriles (6a,b), 7-aryl-5-(piperidin-1-yl)indan-4-carbonitriles (8a-h) and methyl 6,7-diaryl-5-methylsulfanylindan-4-carboxylates (13a,b).  相似文献

A series of new para-substituted (E)-(N)-styrylcarbazoles, i.e., eight (E)-9-[2-(aryl)ethenyl]-9H-carbazoles (5-12) and 1,4-bis[(E)-2-(9H-carbazol-9-yl)vinyl]benzene (13), have been synthesized in high yield and stereoselectively by a sequential silylative coupling-Hiyama coupling reaction, i.e., coupling of commercially available 9-vinylcarbazole with vinyltriethoxysilane or divinyltetramethyldisiloxane in the presence of [RuHCl(CO)(PCy3)2] (I), followed by Pd (II) catalyzed cross-coupling with para-substituted iodobenzenes.The tandem procedure has facilitated the synthesis of 13. Nine 2-substituted pyrrolidin-4-ones 4a-i were obtained via a series of functional group transformation of known prolinol 5 by facile six kinds of methodologies. The target structure of 1,3-amino alcohols 2a-i was constructed in the regioselective Baeyer-Villiger lactonization of ketones 4a-i and reduction of the resulting 4-substituted tetrahydro-1,3-oxazin-6-ones 3a-i. A new and straightforward synthesis of (3S,4S)-statine (6) has been established starting from trans-(2S,4R)-4-hydroxyproline (1).  相似文献

A facile method has been developed for the synthesis of 4H-chromene-3-carboxylates 3ad by the nucleophilic substitution reaction of 2-hydroxy-2H-chromene-3-carboxylates 2ad with triethylsilane in the presence of BF3·O(C2H5)2. Cyclocondensation of 4H-chromene-3-carboxylates 3ad with benzylamines 4ad afforded a series of 2,3-dihydrochromenopyrrolones 5ap and with propargylamine afforded 2-propynyl-2,3-dihydrochromenopyrrolones 6ad. Click reaction of 6ad with benzyl azides 7ad provided a series of 1H-1,2,3-triazolylmethyl-2,3-dihydrochromenopyrrolones 8ap. Thus synthesized compounds 3ad, 5ap, 6ad, and 8ap are novel heterocyclic compounds and being reported for the first time.  相似文献

Reactions of ω-diphenylphosphinofunctionalized alkyl phenyl sulfides Ph2P(CH2)nSPh (n = 1, 1a; 2, 2a; 3, 3a), sulfoxides Ph2P(CH2)nS(O)Ph (n = 1, 1b; 2, 2b; 3, 3b) and sulfones Ph2P(CH2)nS(O)2Ph (n = 1, 1c; 2, 2c; 3, 3c) with dinuclear chlorido bridged rhodium(I) complexes [(RhL2)2(μ-Cl)2] (L2 = cycloocta-1.5-diene, cod, 4; bis(diphenylphosphino)ethane, dppe, 5) afforded mononuclear Rh(I) complexes of the type [RhCl{Ph2P(CH2)nS(O)xPh-κP}(cod)]1 (n/x = 1/0, 6a; 1/1, 6b; 1/2, 6c; 2/0, 8a; 2/1, 8b; 2/2, 8c; 3/0, 10a; 3/1, 10b; 3/2, 10c) and [RhCl{Ph2P(CH2)nS(O)xPh-κP}(dppe)] (n/x = 1/0, 7a; 1/1, 7b; 1/2, 7c; 2/0, 9a; 2/1, 9b; 2/2, 9c; 3/0, 11a; 3/1, 11b; 3/2, 11c) having the P^S(O)x ligands κP coordinated. Addition of Ag[BF4] to complexes 6-11 in CH2Cl2 led with precipitation of AgCl to cationic rhodium complexes of the type [Rh{Ph2P(CH2)nS(O)xPh-κPS/O}L2][BF4] having bound the P^S(O)x ligands bidentately in a κPS (13a-18a, 15b-18b) or a κPO (13b, 14b, 13c-18c) coordination mode. Unexpectedly, the addition of Ag[BF4] to 6a in THF afforded the trinuclear cationic rhodium(I) complex [Rh3(μ-Cl)(μ-Ph2PCH2SPh-κPS)4][BF4]2·4THF (12·4THF) with a four-membered Rh3Cl ring as basic framework. Addition of sodium bis(trimethylsilyl)amide to complexes 6-11 led to a selective deprotonation of the carbon atom neighbored to the S(O)x group (α-C) yielding three different types of organorhodium complexes: a) Organorhodium intramolecular coordination compounds of the type [Rh{CH{S(O)xPh}CH2CH2PPh2CP}L2] (22a-c, 23a-c), b) zwitterionic complexes [Rh{Ph2PCHS(O)xPh-κPS/O}L2] having κPS (21a, 21b) and κPO (20b/c, 21c) coordinated anionic [Ph2PCHS(O)xPh] ligands, and c) the dinuclear rhodium(I) complex [{Rh{μ-CH(SPh)PPh2CP}(cod)}2] (19). All complexes were fully characterized spectroscopically and complexes 15b, 15c, 12·4THF and 19·THF additionally by X-ray diffraction analysis. DFT calculations of zwitterionic complexes gave insight into the coordination mode of the [Ph2PCHS(O)Ph] ligand (κPS versus κPO).  相似文献

Reaction of 3-acyl-4-methoxy-1-methylquinolinones 2a,b with hydroxylamine has been investigated under different experimental conditions. Whereas compound 2a gives rise selectively and exclusively to the regioisomeric isoxazolo[4,5-c]- or isoxazolo[4,3-c]quinolin-4(5H)one (compound 3a or 4a, respectively), reaction of 2b always led to a mixture of the required isoxazole together with the oxazole derivative. Structural elucidation of all products has been independently achieved by multinuclear (13C and 15N) magnetic resonance spectroscopy.  相似文献
