Summary 2-Benzoylamino-6,7-dimethoxy-4H-3,1-benzoxazin-4-one was prepared by thermal treatment of 2-(3-benzoylthioureido)-4,5-dimethoxybenzoic acid and by benzoylation of 2-amino-6,7-dimethoxy-4H-3,1-benzoxazin-4-one. The inactivation of chymotrypsin and human leukozyte elastase by the title compound and 2-benzoylamino-4H-3,1-benzoxazin-4-one is reported.

Zusammenfassung 2-Benzoylamino-6,7-dimethoxy-4H-3,1-benzoxazin-4-on wurde durch thermische Behandlung von 2-(3-Benzoylthioureido)-4,5-dimethoxybenzoesäure und durch Benzoylierung von 2-Amino-6,7-dimethoxy-4H-3,1-benzoxazin-4-on hergestellt. Über die Inaktivierung von Chymotrypsin und humaner Leukozyten-Elastase durch die Titelverbindung und 2-Benzoylamino-4H-3,1-benzoxazin-4-on wird berichtet.

The reaction of 2-ethoxycarbonylmethyl-4H-3,1-benzoxazin-4-one with malononitrile in dry pyridine leads to 1-hydroxy-3,6-dioxo-4,6-dihydro-3H-pyrimido[1,2-a]quinoline-5-carbonitrile. 1,4-二氢-2H-3,1-苯并噁嗪-2-酮作为一种重要的母体骨架广泛存在于生物活性化合物中。此外,在有机合成中它可作为经受热脱羧生成氮杂-邻二亚甲基苯的有效工具。文献报道的合成1,4-二氢-2H-3,1-苯并噁嗪-2-酮的方法有:2-氨基苄醇与光气或其替代物反应,钯或硫催化的2-氨基苄醇与 CO的羰基化反应,钯或硒催化的2-硝基苄醇与 CO的羰基化反应,钯催化的2-叠氮基苄醇直接羰基化反应或2-叠氮基苄醇的氮杂-维悌希(aza-Wittig)/杂累积多烯调节的环合反应,苯并呋喃酮的胺解-霍夫曼重排反应,硼氢化锂还原1,2-二氢-3,1-苯并噁嗪-2,4-二酮,以及2-羟甲基苯基氨基甲酸酯的分子内亲核取代反应。上述合成方法存在原料毒性高或成本高且来源不便、原子经济性低、有腐蚀性废物或 CO2排放、CO利用率低、催化剂昂贵且难以循环使用、反应步骤较多等缺陷,因此发展绿色、高效、经济的合成新途径具有重要意义。本文采用廉价易得的非金属硒作催化剂,用 CO作羰基化试剂, O2作氧化剂,通过硒催化2-氨基苄醇的氧化羰基化反应直接合成了目标产物1,4-二氢-2H-3,1-苯并噁嗪-2-酮。通过考察反应时间、反应温度、催化剂硒的用量、助催化剂种类及用量、CO和 O2的比例及溶剂种类等影响因素,得到了优化的反应条件,目标产物收率最高可达87%。实验证实,该 Se/CO催化体系具有相转移催化功能。反应前硒以粉末形式存在于反应体系中,为多相体系;反应开始后,硒粉参与羰基化反应形成可溶活性化合物,从而成为均相体系;反应完成后硒粉经氧化可重新从反应介质中沉淀析出,又变为多相体系。因此,该体系既实现了高效的均相催化反应,又便于催化剂分离回收,且回收的硒可重复使用,其催化活性基本保持不变。结合相关文献,我们提出了该反应的机理:在助催化剂三乙胺存在下,硒首先与 CO反应原位生成羰基硒,然后羰基硒先后接受2-氨基苄醇中氨基和羟基的亲核进攻生成目标产物,同时释放出硒化氢,硒化氢再被 O2氧化为硒,从而进入下一轮催化循环反应。总之,我们成功开发出一条绿色、高效、经济的1,4-二氢-2H-3,1-苯并噁嗪-2-酮合成新途径。用廉价易得且能循环使用的硒替代贵金属钯作催化剂,用 CO替代剧毒光气或其衍生物作羰基化试剂, O2作氧化剂,硒催化的2-氨基苄醇氧化羰基化反应可顺利进行,以87%的良好收率得到目标产物,具有成本低、原子经济性高、CO利用率高、步骤简短、无腐蚀性废物或温室气体 CO2排放、无光气使用及环境相对友好等优点。  相似文献

New coordination compounds, (bmmpaH)[Fe(pydc)2] · (EtOH)0.8(H2O)0.2 (1), (8QH)[Fe(pydc)2] · H2O (2), (2ampyH)2[Mn(pydc)2] · H2O (3), (2ampyH)[Cr(pydc)2](2ampy)0.5 · H2O (4), [Co(H2O)5-μ-(pydc)Co(pydc)] · 2H2O (5), [Ni(pydcH)2] · H2O (6), and [Cu(pydcH)2] (7), where bmmpa, 8Q, 2ampy, pydcH2 are 5-bromo-6-methyl-2-morpholinepyrimidine-4-amine, 8-hydroxyquinoline, 2-amino-6-methylpyridine, and pyridine-2,6-dicarboxylic acid, respectively, have been synthesized and structurally characterized by elemental analyses, infrared, UV spectroscopic methods, and X-ray crystallography. Metal ions of 1 and 5 are six-coordinate with distorted octahedral geometries. Compound 1 is an anionic mononuclear complex and 5 is a binuclear compound constructed from cationic and anionic parts. The crystal data of 5 reveal that the cationic part is formed by five terminal waters and one μ-carboxylate oxygen O2 from the anionic portion and the anionic complex is built from two deprotonated (pydc)2? moieties. In the compounds, pydcH2 is tridentate by one nitrogen of pyridine ring and two oxygens of carboxylate.  相似文献

In this study, a novel 1,3,4-thiadiazole derivative containing 3-mercaptobutan-2-one and quinazolin-4-one moieties (Compound 3) is synthesized by the coupling of 2-amino-1,3,4-thiadiazole-5-(3-mercaptobutan-2-one) (Compound 1) with 2-Phenyl-4H-3,1-benzoxazin-4-one (Compound 2) in one molecule moiety. Compound 3 is found to exist as two types of intra-molecular hydrogen bonding with keto-enol tautomerism characters, which is further confirmed using FTIR, 1H-NMR, 13C-NMR, mass spectrometer, and UV-Visible spectra. The 1H-NMR and UV-Visible spectra of Compound 3 are investigated in different solvents such as methanol, chloroform, and DMSO. Compound 3 exhibits keto-enol tautomeric forms in solvents with different percentage ratios depending on the solvent polarity. The 1H-NMR and UV-Visible spectral results show that Compound 3 favors the keto over the enol form in polar aprotic solvents such as DMSO and the enol over the keto form in non-polar solvents such as chloroform. The 13C-NMR spectrum gives two singles at δ 204.5 ppm, due to ketonic carbon, and δ 155.5 ppm, due to enolic carbon, confirming the keto-enol tautomerism of Compound 3. Furthermore, the molecular ion at m/z 43 and m/z 407 in the mass spectrum of Compound 3 and fragmentation mechanisms proposed reveal the existence of the keto and enol forms, respectively.  相似文献

Summary. Novel N-alkyl-4-imino-1,4-dihydro-2H-3,1-benzoxazin-2-ones were synthesized in a single step by BaeyerVilliger oxidation of N-alkyl-3-imino-2-indolinone derivatives in high yields. The structures of the products were determined by spectral data and by X-ray diffraction. Besides their novel structures, these compounds may have important biological activities and industrial applications.  相似文献

The reaction of 2-amino-4-methyl-6-(2-pyridyl)-7,8-dihydroindazolo[4,5-d]thiazole, obtained by treating 3-methyl-4-oxo-1-(2-pyridyl)-4,5,6,7-tetrahydroindazole with pyridinium bromide perbromide and then with thiourea, and 2-amino-4-methyl-6-phenyl-7,8-dihydroindazolo[4,5-d]thiazole with 4-bromo-, 4-fluoro-, 4-dimethylamino-, 4-methoxy-, 3,4-dimethoxy-, and 3,4-methylenedioxybenzaldehydes, furfural, pyridinecarbaldehyde, and thiophenecarbaldehyde gave the corresponding Schiff bases. The products of the condensation of these aminothiazoles with cinnamaldehyde, 1-(2-pyridyl)- and 4-chloro-1-(2,4-difluorophenyl)-5-formyl-3-methyl-6,7-dihydroindazoles, 2-formyl-dimedone, and 2-formyl-1,3-indanedione were also obtained.  相似文献

Summary The reaction of 3-aroyl-6-aryl-4-hydroxy-2H-pyran-2-ones (Ar=p-tolyl, 1,1-biphenyl-4-yl or thienyl) with aniline and substitutedo-phenylenediamine (R=H, CH3 or Cl) yields a series of new Schiff bases2a–f in 51–72% yield. Bromination of1a gave the 5-bromo derivative1c, while the compounds1a,1b,2b,2e, and2f were converted into 2,6-diaryl-4H-pyran-4-ones3a–c. All products have been fully characterized.

Summary N-Acylanthranilamides react with dibromotriphenylphosphorane in the presence of triethylamine as HBr captor to give 4-imino-4H-3,1-benzoxazines in good yields. If the reaction is carried out without acid acceptor, N-acetylanthranilamides yield 2-methyl-4-quinazolones, whereas N-benzoylanthranilamides give 2-phenyl-4-imino-4H-3,1-benzoxazines. It has also been found that 2-methyl-4-imino-4H-3,1-benzoxazines rearrange under the influence of HCl or HBr into the respective 2-methyl-4-quinazolones; 2-phenyl-4-imino-4H-3,1-benzoxazines, however, do not undergo such a rearrangement.
Synthese und Umlagerung von 4-Imino-4H-3,1-benzoxazinen
Zusammenfassung Die Umsetzung von N-Acyl-anthranilsäure-amiden mit Triphenyldibromphosphoran in Gegenwart von Triethylamin als HBr-Akzeptor führt mit guten Ausbeuten zu 4-Imino-4H-3,1-benzoxazinen. Wird die Reaktion ohne säurebindendes Mittel durchgeführt, dann entstehen aus N-Acetyl-anthranilsäure-amiden 2-Methylchinazolone-4, jedoch erhält man aus N-Benzoylanthranilsäure-amiden 2-Phenyl-4-imino-4H-3,1-benzoxazine. 2-Methyl-4-imino-4H-3,1-benzoxazine erleiden unter dem Einfluß von HBr oder HCl eine Umlagerung in entsprechende 2-Methylchinazolone-4, während 2-Phenyl-4-imino-4H-3,1-benzoxazine zu einer solchen Umlagerung nicht befähigt sind.

A synthesis of novel derivatives of 6-methyluracil, 6-methyl-2-thioxo-, and 2-imino-6-methyl-2,3-dihydro-1H-pyrimidin-4-one containing a 2-(phenoxy)ethyl substituent at position 5 of the pyrimidine ring has been carried out. It was found that 5-[2-(phenoxy)ethyl] derivatives of 6-methyl-2-thioxo- and 2-imino-6-methyl-2,3-dihydro-1H-pyrimidin-4-one are obtained by the condensation of the corresponding ethyl 3-oxo-2-(2-phenoxyethyl)butanoates with thiourea or guanidine. 6-Methyl-5-[2-(phenoxy)ethyl]uracils can be prepared by treating 6-methyl-5-[2-(phenoxy)ethyl]-2-thioxo-2,3-dihydro-1H-pyrimidin-4-ones with an excess of aqueous monochloroacetic acid solution. __________ Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 8, pp. 1213–1217, August, 2005.  相似文献

4-Methoxy-1,1,1-trifluoropent-3-en-2-one reacts with cyanothioacetamide to give 3-cyano-4-methyl-6-trifluoromethylpyridine-2(1H)-thione, which was transformed into 3-amino-4-methyl-6-trifluoromethylthieno[2,3-b]pyridine-2-carboxanilide by the reaction with chloroacetanilide. A sequential one-pot, two-step reaction for an efficient preparation of 2-(4-hydroxy-6-methyl-2-oxo-2H-pyran-3-carbonyl)-6,6-dimethyl-3-phenyl-3,5,6,7-tetrahydro-2H-benzofuran-4-one derivatives has been described. One-pot reaction of in situ–formed benzopyran-substituted pyridinium ylides with aromatic aldehydes and dimedone gives corresponding 2,3-dihydrofurans in good yields. The structures of all the newly synthesized compounds were confirmed from their analytical and spectral data.  相似文献
