一种新的保护的2-脱氧-2-氨基葡二糖合成   总被引:1,自引:0,他引:1

以氨基葡萄糖盐酸盐为原料制得糖基给体3,4,6-三-氧-乙酰-2-脱氧-2-(2,2,2-三氯乙氧)甲酰胺基-α-D-吡喃葡萄糖基三氯乙酰亚胺酯(5)和糖基受体烯丙基-4,6-氧-亚苄基-2-脱氧-2-(2,2,2-三氯乙氧)甲酰胺基-α-D-吡喃葡萄糖苷(7).在三甲基硅基三氟甲基磺酸酯(TMSOTf)条件下,给体5与受体7反应得到一种新的二糖烯丙基-3,4,6-三-氧-乙酰-2-脱氧-2-(2,2,2-三氯乙氧)甲酰胺基-β-D-吡喃葡萄糖-(1→3)-4,6-氧-亚苄基-2-脱氧-2-(2,2,2-三氯乙氧)甲酰胺基-α-D-吡喃葡萄糖糖苷(10),收率为39.2%.当7在无水BaO和Ba(OH)2·8H2O的条件下与BnBr进行3-OH苄基化反应时,却得到了烯丙基-3-氧-苄基-4,6-氧-亚苄基-2-脱氧-2-羟甲酰胺基-α-D-吡喃葡萄糖苷(8).改用Ag2O条件下与BnBr进行苄基化,得到预期的烯丙基-3-氧-苄基-4,6-氧-亚苄基-2-脱氧-2-(2,2,2-三氯乙氧)甲酰胺基-α-D-吡喃葡萄糖苷(9),从而避免了氨基保护基三氯乙氧甲酰基的水解.标题化合物10对枯草芽孢杆菌、葡伎根霉等微生物有一定的抑制作用.  相似文献

以茴香醛保护氨基方法得到的β-构型的四乙酰基氨基葡萄糖盐酸盐(1)为原料, 采用三氯乙酰基(TCA)、三氯乙酰亚胺酯基(TCI)和乙硫基(SEt)保护体系, 经五步反应以产率40.5%得到完全β-构型保护的β-(1→3)-2-脱氧-2-氨基葡二糖7, 又经两步合成得到保护的β-(1→3)-2-脱氧-2-氨基葡二糖受体8, 共八步合成了保护的β-(1→3)-2-脱氧-2-氨基葡三糖10, 总产率为27%. 以上化合物均为未知. 同时, 还得到了用以合成β-(1→4)-保护的氨基葡四糖的受体6. 采用该保护体系可以高选择性地、较高产率地合成氨基寡糖.  相似文献

合成了新的N-乙基咔唑衍生物: 3,6-二[2-(8-羟基喹啉基)乙烯基]咔唑(4)及其锌配合物(5); 化合物4经质谱、红外光谱、核磁共振氢谱、元素分析表征其结构, 并测定了它的荧光光谱. 结果显示: 化合物4的荧光发射为蓝绿色光(500 nm), 其发射光谱随着溶剂极性的增大荧光光谱向长波方向移动(即发生红移); 同时, 考察了化合物5的荧光性质, 其荧光发光峰值为600 nm, 与2-甲基-8-羟基喹啉锌相比, 发生了明显的红移.  相似文献

在合成2-(2-氨基-3-吡啶基)-苯并咪唑的基础上,利用NMR(1H、13C、COSY、HSQC和HMBC)、MS、IR和UV进行了详细表征;通过X-ray单晶衍射仪测定了该化合物的晶体结构,实验结果表明该晶体属于三方晶系(空间群R3,a=1.833 7(3)nm,b=1.833 7(2)nm,c=1.777 7(4)nm,V=5.176 4(15)nm3,Z=18),很好地支持了波谱表征的结果。同时,结合密度泛函计算,研究了2-(2-氨基-3-吡啶基)-苯并咪唑的荧光光谱。结果表明,化合物的双荧光不是由同一种异构体发射的,而是来源于不同异构体:长波区500~600 nm的荧光由K构型发射,短波区350~450 nm的发射由异构体E1和E2共同产生,理论预测的光谱与实验一致。  相似文献

NBS滴定PFK-2的紫外光谱的结果表明,该酶可滴定Trp残基数为4个其中2个是活性必需的。 用295nm的紫外光激发果糖-6-磷酸-2-激酶(PFK-2),在335nm附近有较强的发射荧光。该荧光能较专一反映PFK-2的Trp基团的状态。底物果糖-6-磷酸对该荧光不影响,ATP引起荧光吸灭。最大吸灭幅度为30%左右。 采用荧光滴定方法测定ATP与酶的结合,得出解离常数(K_i)在低浓度ATP时为0.033mmol/L,高度时为0.23mmol/L。表明该结合具有负协同性。Mg~(2+)、无机磷均降低ATP与酶的K_i值而表现为激活因子;果糖-6-磷酸和果糖-2,6-二磷酸酸对K_i值不影响。ATP结合后引起了该酶构象上的较大的变化。 在ATP保护下,PFK-2对NBS的失活速度稍有增加。丙烯酰胺荧光滴定的结果是,在有无ATP时的Stern-Volmer常数(K_(SV))皆为4.0。  相似文献

本文综述了近年来基于大环多胺及其金属配合物与DNA相互作用的研究进展,着重介绍本课题组在有关单双核、多核以及功能化大环多胺衍生物及其金属配合物与DNA相互作用方面的研究和发现,并对其在化学核酸酶方面的应用进行了讨论。在单双核大环多胺衍生物方面,我们分别合成了以吡啶、苯环、咪唑、三氮唑为侧臂的单核大环多胺金属配合物,同时合成了以刚性桥相连的双核配合物和以柔性链相连的双核配合物。并研究这些单双核大环多胺与DNA的相互作用,发现以刚性链相连的双核大环多胺金属配合物具有很好的切割DNA的性质,可以在低浓度、短时间内切断DNA。在功能化大环多胺方面,我们合成了含有碱基、PNA单体、咪唑鎓盐、冠醚、二茂铁等功能化基团的大环多胺衍生物及金属配合物,并研究了其与DNA的相互作用。在多核大环多胺方面,我们合成了基于大环多胺的寡聚物,研究发现该类物质可与DNA形成复合物,从而有效地保护DNA免于酶解。  相似文献

Two types of complexes were prepared from a cationic cholesterol derivative, dioleoylphos-phatidylcholine and DNA. Depending on the preparation procedure complexes were either dense snarls of lipid covered DNA (type A) or multilayer liposomes with DNA between layers (type B). The transfection efficiency of the snarl-shaped complexes was low but positive. The transfection efficiency of the liposome-shaped complexes was zero, while DNA release upon their interaction with anionic liposomes was 1.7 times higher. The differences in transfection efficacy and DNA release could not be ascribed to the difference in resistance of complexes to decomposition upon interaction with anionic liposomes or intracellular environment since the lipid composition of complexes is the same. Instead the complexes in which lipoplex phase is more continuous (type A) should require more anionic lipids or more time within a cell for complete decomposition. Prolonged life time should lead to the higher probability of DNA expression.  相似文献

The zinc(II) complexes reported here have been synthesised from the ligand 4-methyl-2-N-(2-pyridylmethyl)aminophenol (Hpyramol) with chloride or acetate counterions. All the five complexes have been structurally characterised, and the crystal structures reveal that the ligand Hpyramol gradually undergoes an oxidative dehydrogenation to form the ligand 4-methyl-2-N-(2-pyridylmethylene)aminophenol (Hpyrimol), upon coordination to Zn(II). All the five complexes cleave the phiX174 phage DNA oxidatively and the complexes with fully dehydrogenated pyrimol ligands were found to be more efficient than the complexes with non-dehydrogenated Hpyramol ligands. The DNA cleavage is suggested to be ligand-based, whereas the pure ligands alone do not cleave DNA. The DNA cleavage is strongly suggested to be oxidative, possibly due to the involvement of a non-diffusible phenoxyl radical mechanism. The enzymatic religation experiments and DNA cleavage in the presence of different radical scavengers further support the oxidative DNA cleavage by the zinc(II) complexes.  相似文献

合成了香兰素缩赖氨酸希夫碱配体(HL)和15种香兰素缩赖氨酸希夫碱的稀土配合物REL(NO3)2.3H2O(RE=Y,La,Ce,Pr,Nd,Sm,Eu,Gd,Tb,Dy,Ho,Er,Tm,Yb,Lu),并通过元素分析、摩尔电导、热重分析、红外光谱、紫外光谱、核磁共振方法对配合物的组成和结构进行了表征。采用紫外光谱和荧光光谱方法初步研究了Nd,Tb,Yb,Lu这4种希夫碱配合物与DNA的相互作用,实验结果显示,随着DNA的加入,配合物在280 nm处的紫外吸收峰不断增强,同时配合物在418 nm左右的荧光发射峰随DNA浓度的增大均逐渐减小,说明配合物和DNA是以静电模式结合。  相似文献

The copper complexes of 2‐furaldehyde and furan oximes have previously demonstrated potent cytotoxicity, L1210 DNA synthesis inhibition, DNA topoisomerase II inhibition and DNA fragmentation. Currently a series of cobalt metal complexes of 2‐furaldehyde oximes were compared with copper complexes of furan oximes to determine whether the type of metal is important to the cytotoxicity and mode of action of the complexes. The cobalt complexes of furan oximes, like the copper complexes, were shown to be cytotoxic to suspended tumor cell lines, e.g. leukemias, lymphomas, acute monocytic leukemia and HeLa‐S3 uterine carcinoma. The cobalt complexes did not demonstrate dramatic cytotoxicity against the growth of tumors derived from solid human tumor lines. The cobalt complexes preferentially inhibited L1210 DNA synthesis, followed by inhibition of RNA and protein synthesis from 25 to 100 µM over 60 min. These agents, like the copper complexes of 2‐furaldehyde and furan oximes, were inhibitors of DNA polymerase α activity and de novo purine synthesis with marginal inhibition of ribonucleoside reductase and dihydrofolate reductase activities with DNA fragmentation. Unlike the copper complexes, the cobalt complexes did not inhibit L1210 DNA topoisomerase II activity but did reduce thymidylate synthetase activity. Thus, varying the type of metal within the complexes of 2‐furaldehyde and furan oximes produces differences in both cytotoxicity and mode of action. Copyright © 1999 John Wiley & Sons, Ltd.  相似文献

Two ruthenium complexes containing a new ligand phipz (phipz = 2‐(1,10‐phenanthroline)‐1H‐imidazo[4,5‐b]phenazine) were designed and synthesized. These complexes were found to inhibit the DNA supercoiled relaxation mediated by topoisomerase I (topo I), cleave DNA under irradiation and bind to calf thymus DNA through intercalative mode. Furthermore, complex 2 shows higher photocleavage activity, topo I inhibition activity and DNA affinity than complex 1 . Additionally, introduction of phenazine unit may be the reason that two complexes exhibit DNA ‘light switch’ behavior. The present work shows that two complexes might be potential as new DNA ‘light switches’, DNA photocleavers and topo I inhibitors.  相似文献

Zinc complexes with the third-generation quinolone antibacterial drugs levofloxacin and sparfloxacin have been synthesized and characterized. The deprotonated quinolones act as bidentate ligands coordinated to zinc ion through the pyridone and a carboxylato oxygen atom. The crystal structures of [bis(aqua)bis(levofloxacinato)zinc(II)], 1, and [bis(sparfloxacinato)(1,10-phenanthroline)zinc(II)], 3, have been determined by X-ray crystallography. The biological activity of the complexes has been evaluated by examining their ability to bind to calf-thymus DNA (CT DNA) by UV spectroscopy and viscosity measurements. UV studies of the interaction of the complexes with DNA have revealed that they can bind to CT DNA probably by the intercalative binding mode which has also been verified by DNA solution viscosity measurements. The DNA binding constants have been also calculated. A competitive study with ethidium bromide (EB) showed that the complexes exhibit the ability to displace the DNA-bound EB indicating that they bind to DNA in strong competition with EB for the intercalative binding site. The interaction of the complexes with human and bovine serum albumin proteins has been studied by fluorescence spectroscopy showing that the complexes exhibit good binding propensity to these proteins having relatively high binding constant values. The biological properties of the complexes have been evaluated in comparison to the previously reported Zn(II) complexes with the first- and second-generation quinolones oxolinic acid and enrofloxacin.  相似文献

A new polypyridyl ligand containing a nitro group and two new ruthenium complexes of it were synthesized. The two complexes exhibited non‐dppz DNA ‘light switch’ effects after interaction with calf thymus DNA. Introducing both electron‐withdrawing group (─ NO2) and electron‐donating group (─ CH3) may be the reason that the two complexes display DNA ‘light switch’ behaviors. Furthermore, one of the complexes showed higher photocleavage activity, topoisomerase I inhibition activity and DNA affinity than the other. The present work shows that the more active complex can be employed as a non‐dppz DNA ‘light switch’, DNA photocleaver and topoisomerase I inhibitor. In addition, the two complexes have no or weak cytotoxic activities against Eca‐109 and A549 cells.  相似文献

There is considerable research interest and vigorous debate about the DNA binding of polypyridyl complexes including the electron transfer involving DNA. In this review, based on the fluorescence quenching experiments, it was proposed that DNA might serve as a conductor. From the time-interval CD spectra, the different binding rates of A- and A-enantiomer to calf thymus DNA were observed. The factors influencing the DNA-binding of polypyridyl complexes, and the potential bio-functions of the complexes are also discussed.  相似文献
