Summary 1. The fatty-acid compositions of the oil, the total phospholipids, and the main groups of phospholipids of the seed kernels of the fine-fibered cotton plant of variety 5904-I have been determined. The complete qualitative identity of the acids present in them and quantitative similarity of the acids in the oil and in the total phospholipids have been shown: in the degree of increasing saturation the phospholipids form the following sequence: phosphatidylcholines, phosphatidylethanolamines, phosphatidylinositols.2. It has been established by enzymatic hydrolysis with phospholipase A2 of kufi venom that the unsaturated fatty acids are present in position of the PCh, PE, and PI molecules — to the extent of 90.6%, 96.5%, and 85.4%, respectively.3. The free, bound, and strongly bound lipids of the seed kernels of the cotton plant of variety 108-F have been studied. It has been shown that the free and bound lipids include 97.3% and 47.6% of neutral lipids, respectively. The polar lipids consist mainly of phospholipids and glycolipids. From two-day etiolated shoots of the cotton plant of variety 108-F we have isolated the total histone and purified it on a column of CM-cellulose. It has been shown by gel electrophoresis and amino acid analysis that the total histone of the cotton plant, like the total histone of wheat, consists of five main fractions, the amount of the lysine-rich fraction A1 being greater in the cotton plant than in wheat (23% and 19%, respectively), and the amount of arginine-rich fraction A4 being less (19.2 and 24.3%, respectively). The total histone of the cotton plant is less basic (ratio of basic amino acids to acidic amino acids in the cotton-plant histone 1.24, in wheat histone 1.9, and in calf thymus histone 1.78) but richer in lysine as compared with arginine than the histones of wheat and calf thymus.V. Summary The diglyceride and monoacetyldiglyceride derivatives of the phosphatidylcholines of the seed kernels of the cotton plant of variety S-6029 have been separated into individual subfractions in a thin layer of silica gel impregnated with silver nitrate.The total fatty-acid compositions of the individual subfractions and the position distributions of the fatty acids in the glyceride moiety of the molecules have been determined, and on this basis the molecular compositions of the phosphatidylcholines have been established: for the DGs 46 species and for the MADGs 43 species. The distribution of phosphorus with the secretion of phospholipids (PLs) in six stages of development of the cotton plant has been studied. It has been shown that additional extraction with methanol leads to the isolation of further PLs. Both the seeds and cotton-plant bushes during growth contain phosphatidylcholine, phosphatidylinositol, phosphatidylethanolamine, X1, and X2, and also unidentified PLs, one of which, Y2, is phosphatidic acid according to qualitative reactions, chromatographic mobility, and literature information.  相似文献

An N-arylphosphatidylethanolamine has been isolated from the total phospholipids of the cotton plant of variety S-6029. The composition and the position distribution of the fatty-acid radicals has been studied with the aid of enzymatic hydrolysis with phospholipase A1 (from the fungusRhisophus microsporus UzLT-1) and by alkaline and acid hydrolyses. From the results obtained, the possible molecular composition of the N-acylphosphatidylethanolamine has been calculated by Coleman's method. Summary 1. The qualitative and quantitative compositions of the PLs of an industrial medium-fiber cotton plant of variety 159-F (1972 and 1974 harvests) have been considered. It has been shown that the total PL amounted to 1.76 and 1.85% and consisted of 60–65% PC, 16–18% PI, 6–8% PE, and 10–14% of combined minor components, respectively.2. The distribution of the lipid phosphorus has been studied during the isolation of PLs by selective solvents. The lipid phosphorus amounted to 5.5% of the total phosphorus in the seed kernels.3. The qualitative and quantitative compositions of the fatty acids of the individual lipid fractions and of the individual PCs, PIs, and PEs have been determined. The kinetics of some outer-sphere electron-transfer reactions with the participation of the complexes [Pt(NH3)nX6–n]2 (n=6–0, X=Cl, Br) and dipyridyl complexes of Os(II), Ru(II), Ir(III), and Cr(II) have been investigated by means of luminescence-quenching measurements and flash photolysis. The diaphorase activities of the proteins of different varieties of two species of cotton plant —Gossypium hirsutum L. andG. barbadense L. — have been studied. It has been shown that the proteins of the seeds of the cotton plantG. barbadense possess a low diaphorase activity in comparison with the proteins of the seeds of aG. hirsutum plant. On an electrophoretogram of the proteins, diaphorase activity was localized in two zones, with Rf 0.45 and 0.70. The diaphorase with Rf 0.45 has been isolated by electrophoresis in polyacrylamide gel (PAAG) and some of its properties have been studied. The diaphorase isolated oxidizes NADH and NADPH in the presence of various artificial electron Acceptors, and has two pH optima (at 7.20 and 8.70) and is characterized by relative thermal stability (at 80°C). In the case of the total extract, brief boiling does not lead to inactivation of the enzyme, which shows the presence in cotton seeds of a factor stabilizing this diaphorase. The molecular weight of the proteins isolated, according to gel filtration on Sephadex G-150, is 59,000, and from the results of SDS-PAAG electrophoresis it is 13,600, which shows a tetrameric structure of the enzyme.Institute of Experimental Plant Biology, Academy of Sciences of the Uzbek SSR, Tashkent. The influence of the composition on the AC carrier transport of the composite films containing ferromagnetic CoFeZr nanoparticles in amorphous aluminium oxide matrix has been investigated. The films 3–5 μm in thicknesses and with variable composition 30 at.% < X < 60 at.% were sputtered on a single substrate from the compound target in the chamber with argon–oxygen gas mixture. TEM and SEM measurements and Mössbauer spectroscopy data have shown that all the studied films of (Co0.45Fe0.45Zr0.10)X(Al2O3)1 − X with 30 at.% < X < 65 at.% have revealed the structure with crystalline granular metallic alloy (with particles of a few nanometers in size) and amorphous alumina. AC conductance measurements were performed over the frequency range 102–106 Hz at temperatures from 80 to 330 K. DC conductance measurements have been carried out for this temperature region also. The presence of two critical regions for the metallic fraction (X1 = 33–40% and X2 = 50–55%), where diagram “electric property–composition” exhibited pronounced peculiarities, has been confirmed. A qualitative structural model of nanocomposite was offered to explain this behavior. In accordance with the model, the first critical region at X1 is associated with a shift of percolation threshold due to the formation of oxide layer on metallic nanoparticles, owing to the presence of oxygen in gas ambient during the sputtering process. The second critical region of the composition at X2 is ascribed to the formation of percolation net of magnetic metallic nanoparticles in the dielectric amorphous alumina matrix.  相似文献

The individuality of the 5S rRNA from cotton seeds has been determined by the production of a single narrow band on gel electrophoresis and by the characteristic UV spectrum with values of the ratios E260/E230 and E260/E280 of 2.03–2.1 and 2.08–2.14, respectively. The melting curve of the 5S rRNA has been obtained and a total hyperchromic effect of 25% has been calculated. The nucleotide composition of the 5S rRNA has been established by determining the amount of nucleotides in an alkaline hydrolysate (22.6% of AMP; 30.2% of GMP; 22.6% of UMP; and 24.6% of CMP). The nucleotides were identified from the position of their appearance in the elution profile, from their UV spectra in 0.1 N HC1 and 0.1 KOH, and also by one- and two-dimensional chromatography of the nucleotides obtained, in the presence of markers.Institute of the Chemistry of Plant Substances of the Academy of Sciences of the Uzbek SSR, Tashkent. The surface lipids of the coat and the lipids of the kernel of the seeds of a wild form of the medium-fibered cotton plantGossypium mexicanum var.nervosum have been studied in comparison with cultured species. Using the methods of CC, TLC, and GLC, and UV and mass spectrometry, in the cuticular lipids ofG. mexicanum three classes of neutral lipids have been detected, and in the kernel lipids 10 classes of neutral lipids, 2 classes of glycolipids, and 7 classes of phospholipids. The structures of the epoxyacyl-, hydroxyacyl-, and diacylglycerols of the kernel have been established. The greatest differences between the seeds of the wild form of the cotton plant and the cultured species are the greater weight of the seed coat, the presence in it of a considerable amount of the 18:1 and of medium-molecular-weight fatty acids, and a higher level in the kernel of gossypol pigments, polar lipids, and acylglycerols containing the 18:2 acid.Institute of the Chemistry of Plant Substances, Academy of Sciences of the Uzbek SSR, Tashkent.
