The present review presents the state of the art of the developments, key strategies and analytical applications of chemiluminescence detection coupled to CE (CE‐CL). Different parts considering the most common CL systems have been included, such as the tris(2,2′‐bipyridine)ruthenium(II) system, the luminol and derivatives reaction, the peroxyoxalate CL or direct oxidations. New advances in homemade configurations and applications in different fields such as clinical, pharmaceutical, environmental and food analysis have been included. The focus is on studies which appeared from 2000 to the end of 2009.  相似文献

This paper reviews the latest developments and analytical applications of chemiluminescence detection coupled to capillary electrophoresis (CE-CL). Different sections considering the most common CL systems have been included, such as the tris(2,2?-bipyridine)ruthenium(II) system, the luminol and acridinium derivative reactions, the peroxyoxalate CL or direct oxidations. Improvements in instrumental designs, new strategies for improving both resolution and sensitivity, and applications in different fields such as clinical, pharmaceutical, environmental and food analysis have been included. This paper presents an overview of flow-based methods in food and environmental analysis using chemiluminescence (CL) detection covering the period from 2005 to the present. The review discussses both automated flow methods of analysis [such as flow-injection analysis (FIA), sequential-injection analysis (SIA) and their variants] and separation techniques [liquid chromatography (LC) and capillary electrophoresis (CE) coupled to CL detection]. We provide an overview of powerful applications of chemiluminescence (CL) analysis for biomolecules, mainly in gel electrophoresis. In routine immunoassays, CL labels and detection reactions are widely used for peroxidase and alkaline-phosphatase enzymes. The sensitivity, the dynamic range and the diversity of CL assays have led to a vast range of applications, notably in protein and nucleic-acid blotting. Non-enzymatic CL detection is also being developed gradually.Direct CL detection of biomolecules in gels has emerged recently, with simple, convenient and rapid methods. It offers substantial potential to detect many proteins or nucleic acids in complex biological samples. In addition, metallic nanoparticles and catalytic nucleic acids are also potential candidates for CL detection of biomolecules in the future.  相似文献

A sensitive method has been developed for liquid chromatographic determination of short aliphatic amines in water samples. Analytes are preconcentrated and dansylated on solid sorbents (C18 solid-phase extraction cartridges). The dansyl derivatives are chromatographed and post-column mixed with peroxyoxalate (TCPO) and H2O2 in order to perform chemiluminescence detection. Optimal results have been obtained using a sample volume of 5 ml. The method has been applied to the quantification or screening of several aliphatic amines: methylamine, ethylamine, butylamine, diethylamine, pentylamine and hexylamine. The screening procedure has been developed including also polyamines (putrescine, cadaverine, spermidine and spermine). The results obtained by using chemiluminescence (CL) detection have been compared with other detection systems (fluorescence and UV). The sensitivity can increase from 3 to 75 times respect UV detection and from 2 to 10 times respect fluorescence detection depending on the amine. The detection limits achieved were between 0.15 and 0.9 microg/l.  相似文献

化学发光分析是利用化学发光反应的发光现象,对化学发光物质由激发态跃迁回基态时发出的光信号进行测量的一种分析方法。化学发光分析具有无需外来光源、灵敏度高、操作方便、分析快速以及易于实现自动化等优点,可与其它分析技术联用,在临床检验、药物分析和环境监测等领域具有广泛应用。近年来,纳米材料、生物芯片及微流控技术的引入促进了化学发光分析技术的发展。本文综述了化学发光分析与高效液相色谱、毛细管电泳、量子点、微流控芯片和微阵列、以及滚环扩增、等温指数扩增和两级等温扩增联用技术的发展,介绍了化学发光分析技术在DNA、生物小分子、生物酶、蛋白质和金属离子检测中的应用研究进展,并展望了其发展趋势。  相似文献

The possibility of direct analytical applications of ferrate(VI) solution, which was freshly electrogenerated in low-concentration NaOH electrolyte, was studied by a flow-injection chemiluminescence (FI–CL) system. It was found that some inorganic ions, organic molecule and biomolecule could enhance the chemiluminescence emission caused by ferrate(VI)–luminol reaction. V(V), Ca(II), Mg(II), phloroglucinol, and bovine hemoglobin (Hb) chosen as samples were successfully detected by this developed method. The analytical characteristics of the system for the analytes determination including linear ranges, correlation coefficients, limits of detection combined with FI analysis were studied.  相似文献

The identification of radical species is possible by the electron spin resonance technique. However, the antioxidants in complex matrices such as biological and food samples are difficult to determine. Hence, we developed novel screening systems for antioxidants, which are mainly eliminating superoxide anion radical (O(2)(-)) and hydrogen peroxide (H(2)O(2)), by HPLC with luminol-based chemiluminescence (CL) detection. When the sample contains antioxidants, inhibited peaks corresponding to each antioxidant are observed on the chromatogram. The antioxidant activities of catechins and flavones were determined with flow injection analysis by the proposed indirect CL. The scavenging activity for H(2)O(2) and O(2)(-) were different from each catechin and flavone. Furthermore, the potential was dependent upon the number and the position of OH functional group in the structure. Some applications such as the screening of antioxidants in tea products were also investigated. In spite of many peaks appeared on the chromatogram at UV detection, only the peaks corresponding to the compounds having elimination effect to O(2)(-) and/or H(2)O(2) were detected as inhibited peaks. Consequently, the proposed HPLC-CL seems to provide new screening systems for antioxidants possessing inhibition activity of O(2)(-) and H(2)O(2).  相似文献

In recent years, chemiluminescence (CL)-based detection coupled to capillary electrophoresis (CE) as separation technique has attracted much interest due to new advances in home-made configurations, sample-treatment techniques for application to real matrixes, development of a commercial instrument and use of miniaturization techniques to obtain micro total analysis systems incorporating CE separation and CL detection in microchips. In this review, the recent evolution and the state of the art of photochemical reactions coupled with chemiluminescence processes are presented. Different chemiluminescence systems have been considered together with suitable photochemical derivatization processes that can affect either the analyte of interest or even the chemiluminogenic reagent, producing some derivatives able to participate more efficiently in the CL reactions and enhancing the CL emission. The on-line integration of the photochemical reactions as well as the coupling of this resulting photoinduced chemiluminescence (PICL) method with dynamic analytical systems, such as flow injection analysis, liquid or gas chromatography and capillary electrophoresis, have been discussed. Important applications of PICL have been proposed in environmental, pharmaceutical and food analysis.  相似文献

The improvement in hyphenated analytical techniques has significantly widened their applications to the analysis of biomaterials. In this article, we discuss recent advances in applications of hyphenated chromatographic techniques including capillary electrophoresis to the analyses of biological samples. As tools of separation, gas chromatography, high-performance liquid chromatography and capillary electrophoresis are considered with special emphasis on applications utilizing the hyphenation of these methods to mass spectrometry. Moreover, applications using other detection methods such as Fourier transform infrared spectroscopy hyphenated to gas chromatography and photodiode array detector combined with high-performance liquid chromatography or capillary electrophoresis are also discussed. Owing to their high sensitivity, luminescence-based detection systems such as laser-induced fluorescence and chemiluminescence are also included in this review.  相似文献

The role of high-performance liquid chromatography (HPLC) in methods of analysis for anabolic compounds in biological samples is reviewed. Special attention is given to both the separation and detection of anabolic compounds. A distinction is made between on-line detection systems, such as ultraviolet detection and diode-array detection, and off-line detection methods with special emphasis on immunochemical detection methods using non-isotopic labels. A number of applications are given to elucidate the possibilities of HPLC in the analysis of anabolic compounds.  相似文献

以4种磺胺类药物(Sulfonamides, SAs), 即磺胺脒(Sulfaguanidine, SGD)、磺胺嘧啶(sulfadiazine, SDZ)、磺胺噻唑(sulfathiazole, STZ)和磺胺二甲嘧啶(Sulfamethazine,SMZ)为分析物,基于其在碱性介质中对Ag配合物-鲁米诺(Luminol)与Ni配合物鲁米诺两化学发光体系发光强度均具有抑制作用的性质,建立了高效液相色谱-化学发光法检测牛奶中4种磺胺类药物的方法.将化学发光体系作为高效液相色谱的新型检测器,并对两种化学发光体系的检测器性能进行了比较.4种磺胺药物经高效液相色谱分离后,分别与Ag-Luminol及Ni-Luminol化学发光体系作用.色谱条件为:反相C18分离柱(250 mm × 4.6 mm,5 μm);0.1%甲酸-甲醇为流动相(V/V);梯度洗脱;流速1 mL/min.化学发光条件:Ag、Ni-Luminol两体系中,Ag配合物浓度1.4×10.-4 mol/L(含0.12 mol/L NaOH);Ni配合物浓度1.5×10.-5 mol/L(含0.12 mol/L NaOH);Luminol浓度均为1.2×10.-7 mol/L;试剂流速均为1.0 mL/min.在最佳的分离检测条件下,Ag-Luminol体系检测4种磺胺类药物的检出限分别为0.15、0.96、1.10和1.50 μg/mL,加标回收率为81.0%~101.5%;Ni-Luminol体系检测SGD、SDZ、STZ 3种磺胺类药物的检出限分别为1.5、17.2和16.8 μg/mL,加标回收率为83.9%~110.8%.相比之下,Ag-Luminol体系作为高效液相色谱检测器更佳.应用本方法对牛奶中4种磺胺类药物残留量进行检测,结果令人满意.  相似文献

We have demonstrated an integrated platform for microfluidics and chemiluminescence (CL) detection that is capable of parallel cell culture, convenient liquid manipulation, and sensitive chemiluminescent detection. Luminol-dependent CL responses induced by three different stimuli, phytohemagglutinin (PHA), concanavalin A (ConA), and lipopolysaccharides (LPS), which can evoke a CL response in macrophages, were evaluated on this microfluidic chip. We studied the dose-dependent effect of these three stimuli on CL response in murine macrophages. PHA produced the highest CL response compared to LPS and ConA. The CL intensity produced by PHA was 6.85 and four times higher than that by LPS and ConA, respectively, at the low concentration of 100 μg/ml. We have found microfluidic based CL to be a very useful screening tool, which is less laborious and more sensitive. This microfluidic system is disposable and capable of rapid device prototyping; it may prove to be very useful in clinical and pharmaceutical applications.  相似文献

On-line high-performance liquid chromatography (HPLC) coupled with biochemical detection (BCD) has been developed to screen compounds showing antioxidant action, enzyme inhibition and receptor affinity in complex matrixes. This review summarizes HPLC methods combining different post-column detection methods, such as diode-array detection (DAD), mass spectrometry (MS), chemiluminescence (CL) and nuclear magnetic resonance, for antioxidant screening. The methods based on a single relatively stable reagent such as DPPH and ABTS•+ were the most popular. Oxygen free radical scavengers mainly depended on post-column CL detection. The on-line hyphenated HPLC–BCD systems based on post-column UV/DAD fluorescence and MS detection were also widely applied to screen enzyme- and receptor-active compounds. These strategies provide a convenient tool for quick identification and quantification of active compounds in complex matrixes.

This review discusses and compares the methods given for the determination of rare earth elements (REE) in natural water samples, including sea, river, lake, tap, ground and waste waters as well as Antarctic ice. Since REE are at very low concentrations in natural waters, numerous different preconcentration methods have been proposed to enable their measurement. These include liquid liquid extraction, dispersive liquid-liquid micro-extraction and solidified floating drop micro-extraction. In addition to liquid-liquid extraction methods, solid phase extraction using commercial resins, resins made in-house, silica-based exchange materials and other solid media is also discussed. These and other techniques such as precipitation/co-precipitation and flotation are compared in terms of speed, preconcentration factors achieved, precision, accuracy and limits of detection (LOD). Some papers have discussed the direct determination of REE in these sample types. Some have used specialised sample introduction systems such as ultrasonic nebulization whereas others have used a standard sample introduction system coupled with inductively coupled plasma mass spectrometry (ICP-MS) detection.
