A simple and reliable liquid chromatography–mass spectrometry (LC–MS) method was developed for simultaneous determination of saikosaponin A, saikosaponin B1, saikosaponin C, saikosaponin D and saikosaponin F in rat plasma using glycyrrhetinic acid as an internal standard (IS). The separation was operated on a Waters BEH C18 column. The mobile phases of gradient elution consisted of acetonitrile (A) and 0.1% aqueous acetic acid (B). The mass spectrometric detection was accomplished in multiple reaction monitoring mode. The five saponins displayed good linearity (r2 > 0.9996). The lower limits of quantitation of saikosaponin A, saikosaponin B1, saikosaponin C, saikosaponin D and saikosaponin F were determined to be 2.9, 2.3, 3.5, 2.9 and 3.1 ng/mL, respectively. Moreover, the intra‐ and inter‐day precisions of the five saponins showed an RSD within 2.96%, whereas the accuracy (RE) ranged from ?2.28 to 2.78%. Finally, the developed method was fully validated and applied to a comparative pharmacokinetic study of the five bioactive saponins in rats following oral administration of crude and vinegar‐processed Bupleurum scorzonerifolium.  相似文献

From the ethanolic extract of the whole plant of Hemistepta lyrate Bunge., a new lignan, named hemislin B glucoside,has been isolated and identified as α-(trans-3-methoxy-4-O-β-D-glucopyranosylbenzylidene)-β-(3,4-dimethoxybenzyl)-γ-butyrolactone. The perennial Bupleurum root is thick and woody and contains high levels of interfering compounds. Common protein extraction methods have proved refractory towards the isolation of proteins suitable for 2-DE, due to the presence of interfering compounds. A novel method for extracting proteins suitable for 2-DE was established to overcome these problems. The main characteristic of this protocol is the partitioning of the proteins into the aqueous (fraction A-2), chloroform and isoamyl alcohol phases (A-3), and the interphase (A-1). The proteins are then extracted from each of these phases. From A-1, 85% (extracted protein against total proteins) proteins could be extracted and purified. For fraction A-2, a novel phenol extraction step is employed for the extraction of proteins. Based on the well-resolved 2-DE patterns, our protein preparation is free of interfering compounds. Using these methods (A-1, A-2, and A-3-3), a total of 3662 (1526 + 1128 + 1008) spots could be separated, and a protein yield of about 1.41 mg per 1.0 g fresh root material was obtained. To our knowledge, this is the first time that a protocol for protein extraction from perennial Bupleurum root has been reported that gives reproducible results. The protocol is expected to be applicable to other recalcitrant plant tissues as well.  相似文献

The genus Mananthes (Acanthaceae) comprises about 5 species mainly distributed in south Asia and two of them occur in China. Mananthes patentiflora (Hemsl.) Bremek is a herb native to Yunnan, Hunan and Guangdong Provinces of China. No phytochemical investigation has been reported on this plant. A new lignan, 5‐hydroxyjusticidin A (= 9‐(1,3‐benzodioxol‐5‐yl)‐5‐hydroxy‐4,6,7‐trimethoxynaphtho[2,3‐c]furan‐1(3H)‐one; 1 ), and four new diphyllin‐type lignan glycosides, mananthosides C–F ( 2 – 5 ), containing glucosyl (Glc), arabinosyl (Ara), galactosyl (Gal), and/or apiosyl residues, have been isolated from Mananthes patentiflora, together with five known compounds. Three new arylnaphthalide lignans, 5‐(4‐hydroxy‐3‐methoxyphenyl)furo[3′,4′: 6,7]naphtho[2,3‐d]‐1,3‐dioxol‐6(8H)‐one ( 1 ), 10‐(4‐hydroxy‐3‐methoxyphenyl)furo[3′,4′: 6,7]naphtho[1,2‐d]‐1,3‐dioxol‐9(7H)‐one ( 2 ), and 10‐(3,4‐dimethoxyphenyl)‐6‐hydroxyfuro[3′,4′: 6,7]naphtho[1,2‐d]‐1,3‐dioxol‐9(7H)‐one ( 3 ), together with two known ones, chinensin ( 4 ) and isodiphyllin ( 5 ), were isolated from the aerial parts of Bupleurum marginatum Wall . ex DC. Supercritical fluid extraction (SFE) was used to extract saikosaponins a, c and d from the root of Bupleurum falcatum. An orthogonal array design L9(3)4 was employed as a chemometric method for the optimization of the SFE conditions. The effects of four factors including pressure (30–40 MPa), temperature (40–50°C), ethanol concentration (60–100%) and time (2.5–3.5 h) on the yields of saikosaponins were investigated by a preparative SFE system in the SFE mode. Under the optimized conditions, namely 35 MPa of pressure, 45°C of temperature, 80% of ethanol concentration and 3.0 h of time, the yields of saikosaponin c, saikosaponin a, saikosaponin d, total saikosaponins and SFE extract were 0.16, 0.12, 0.96, 1.24 and 16.48 mg/g, respectively. Determinations of the saikosaponins were performed by HPLC.  相似文献

Accelerated solvent extraction (ASE) was applied to the extraction of saikosaponin a, saikosaponin c and saikosaponin d from the roots of Bupleurum falcatum. Main extraction parameters such as the extraction solvents, extraction temperature and static extraction time were investigated and optimized. The optimized procedure employed 70% methanol as extraction solvent, 120°C of extraction temperature, 10 min of static extraction time, 60% of flush volume and the extraction recoveries of the three compounds were near to 100% with one extraction cycle. The extracted samples were analyzed by HPLC with UV detector. The HPLC conditions were as follows: Hypersil ODS2 (4.6 mm×250 mm, 5 μm) column, acetonitrile and water as mobile phase, flow rate of 1.0 mL/min, UV detection wavelength of 204 nm and injection volume of 20 μL. Compared with the traditional methods including heat‐reflux extraction and ultrasonic‐assisted extraction, the proposed ASE method was more efficient and faster to be operated. The results indicated that ASE was an alternative method for extracting saikosaponins from the roots of B. falcatum.  相似文献
