Four different stationary phases and a variety of solvents in varying proportions were examined in this study. Daidzein, genistein, formononetin, biochanin A and coumestrol were separated within 24 min on a phenyl column with acetonitrile-water (33:67, v/v) as eluent. The proposed method showed an acceptable repeatability with a RSD of quantitation <6%. The mean recoveries of daidzein, genistein, formononetin, biochanin A and coumestrol from soybean ranged from 89 to 104%. The identity of the individual analytes was confirmed by LC-MS-MS. The four isoflavones and coumestrol were isolated from soybean by hydrolysis with acid and heat. Neutralization of the soybean samples prior to identification did not alter the concentration of daidzein and genistein in soybean.  相似文献

The urinary catecholamine metabolites, vanimandelic acid, homovanillic acid, 3,4-dihydroxyphenylacetic acid and 5-hydroxyindoleacetic acid, were extracted on a silica-bonded strong-anion-exchanger cartridge (SAX) and then injected into an high-performance liquid chromatographic (HPLC) system by column switching. Chromatography was performed on a reversed-phase analytical column with electrochemical detection. Full automation was obtained by coupling two devices: a solid-phase automatic sampler and intelligent autosampler. For each substance the recovery was greater than 95% and the coefficient of variation was ca. 3%; the analysis takes 11 min. Substance instability problems are overcome, because the samples are extracted and injected in rapid succession. The normal values and correlation with manual HPLC were established for a large number of samples.  相似文献

A simple procedure for the simultaneous determination of carbamazepine and its major metabolite, carbamazepine epoxide, in plasma and urine is described. The assay involves two extractions of the drugs and an internal marker, clonazepam, from the alkalinized sample. The extract is evaporated to dryness at 45 degrees C and the residue is redissolved in methanol (30 microliters). A 25-microliters aliquot is injected into the liquid chromatograph and eluted with acetonitrile-water (40:60, v/v) on a C18 pre-column linked to a 5-microns C8 reversed-phase column. The eluent is detected at 215 nm. The method has been used to investigate the steady-state concentrations of carbamazepine and carbamazepine epoxide in the plasma and urine of a manic-depressive patient.  相似文献

A rapid and sensitive high-performance liquid chromatographic method is described for the quantitative analysis of dipotassium clorazepate (CZP) and its major metabolite nordiazepam (ND) in fresh human and dog plasma. The method consists of two separate selective ND extractions from a plasma sample without and with conversion of all the CZP to ND. For quantitation, diazepam (DZP) is used as the internal standard. The chromatographic phase utilized in a reversed-phase Hibar EC-RT analytical column prepacked with LiChrosolv RP-18 with a solvent system consisting of acetonitrile-0.05 M sodium acetate buffer, pH 5.0 (45:55). The UV absorbance is monitored at 225 nm using a variable-wavelength detector. The mean assay coefficient of variation over a concentration range of 20-400 ng per ml of plasma is less than 3% for the within-day precision. Recoveries of ND, DZP and CZP (as ND) are essentially quantitative at all levels investigated. The calibration curves of ND are rectilinear (r2 = 0.99) from the lower limit of sensitivity (2 ng/ml) to at least 2000 ng/ml in plasma. Applicability of the method to CZP and ND disposition studies in the anaesthetized mongrel dog is illustrated. When the two separate selective nordiazepam extractions from plasma cannot be performed immediately after blood sampling, an extrapolation kinetic method is suggested for the estimation of CZP concentration. In all previous in vivo studies, CZP has been determined only with gas-liquid chromatographic methods.  相似文献

Summary Rapid HPLC methods are described for the determinations of either free N-n-propylnorapomorphine (NPA) or the free and conjugated NPA in blood plasma. The methods employ a methanol protein-precipitation step that produces relatively good absolute recoveries, while permitting a simple work-up procedure without derivatization. Only 0.25 ml of plasma is required for a single determination at a sensitivity limit of 0.4g NPA/ml plasma. Satisfactory accuracy and precision are achieved with the proposed assay.

Zusammenfassung Rasche Methoden zur Bestimmung von freiem NPA oder freiem und gebundenem NPA in Blutplasma wurden beschrieben. Nach Eiweißfällung mittels Methanol erhält man relativ gute Absolutwerte nach einfacher Aufarbeitung ohne Derivatisierung. Für eine Einzelbestimmung werden nur 0,25 ml Plasma benötigt. Die Erfassungsgrenze liegt bei 0,4g NPA/ml Plasma. Das Verfahren ist hinreichend genau.

ZLR-8 is a nitric oxide releasing derivative of diclofenac for the treatment of inflammation. In this paper, a sensitive and reliable high-performance liquid chromatography method for simultaneous determination of ZLR-8 and its active metabolite diclofenac in the plasma of beagle dogs has been developed and validated. After the addition of ketoprofen as the internal standard (IS), plasma samples were extracted with n-hexane-isopropanol (95:5, v/v) mixture solution and separated by HPLC on a reversed-phase C(18) column with a mobile phase of gradient procedure. Analytes were determined by the UV detector which was set at 280 nm. The method was proved to be sensitive and specific by testing six different plasma batches. Calibration curves of ZLR-8 and diclofenac were linear over the range 0.05-4.0 microg/mL. The within- and between-batch precisions (RSD%) were lower than 10% and accuracy ranged from 85 to 115%. The lower limit of quantification was identifiable and reproducible at 0.05 microg/mL. The proposed method has been readily implemented in preclinical pharmacokinetics studies of ZLR-8 and its active metabolite diclofeance. Representative plasma concentration vs time profiles resulting from administration of ZLR-8 to beagle dogs are presented in this communication.  相似文献

Oxycodone (14-hydroxy-7,8-dihydrocodeinone) is a potent opioid receptor agonist. In the present study, a liquid-liquid extraction-based reversed-phase HPLC method with UV detection was validated and applied for the analysis of oxycodone and its major metabolite, noroxycodone, in human plasma. The analytes were separated using a mobile phase, consisting of acetonitrile and phosphate buffer (8:92, v/v) at a flow rate of 1 mL/min, and UV detection at 205 nm. The retention times for oxycodone, noroxycodone and codein (internal standard) were 14.7, 13.8 and 10.2 min, respectively. The validated quantitation range of the method was 2-100 ng/mL for oxycodone and 10-100 ng/mL for noroxycodone. The developed procedure was applied to assess the pharmacokinetics of oxycodone and its metabolite following administration of a single 20 mg oral dose of oxycodone hydrochloride to one healthy male volunteer.  相似文献

A method for the simultaneous determination of pirprofen and its metabolite, the pyrrole derivative, in human plasma is described. The two compounds and the butyric acid analogue of the pyrrole derivative used as internal standard are extracted from plasma with chloroform, then back-extracted into an alkaline buffer. After addition of acid, the aqueous phase is assayed by high-performance liquid chromatography using a fixed-wavelength ultraviolet detector at 254 nm. The limit of quantitation is 0.1 micrograms/ml (0.396 mumol/l for pirprofen and 0.400 mumol/l for the pyrrole derivative).  相似文献

A method is described for the determination of cortisol in human plasma by high-pressure liquid chromatography. The simplified extraction procedure makes the method applicable to routine clinical assays. Partition chromatography is carried out on a Zorbax-Sil column with the eluent system dichloromethane-ethanol-water. A 78% recovery was obtained for cortisol. The detection limit is 1 mug per 100 ml in 1 ml of plasma. Cortisol values were determined in samples from a random selection of patients.  相似文献

A column liquid chromatographic method suitable for the quality control of tylosin A is described. The determination can be carried out on different C8 or C18 columns, using a mobile phase containing acetonitrile, 0.2 M tetrabutylammonium hydrogensulphate, 0.2 M phosphoric acid and water. The flow-rate is 1 ml/min and detection is performed at 280 nm. The method shows good selectivity towards the major components tylosin A, B, C and D and demycinosyltylosin. Minor degradation products, mainly observed in solutions, are also separated. The compositions of several standards are compared and results for a number of commercial samples are presented.  相似文献

An automated column-switching HPLC system is described for the simultaneous determination of midodrine, an alpha-adrenergic stimulating drug, and its active metabolite, ST-1059. Serum or plasma (850 microliters) is directly injected onto a RP18 (30 micrograms particle size) pre-column (9 x 4 mm ID) which acts as an on-line liquid-solid extractor and analyte enrichment system. The injection is followed by washing steps. The fraction containing the analytes is transferred onto an analytical RP18 column via step gradient elution where the final analysis is performed. Fluorescence detection is used (lambda ex 290 nm and lambda em 322 nm), and method detection limits of 0.8 ng/mL plasma were reached. These were sufficiently low to determine the plasma concentration-time profiles for both compounds following oral administration of 2.5 mg and 5 mg midodrine hydrochloride. The assay in serum or plasma was linear in the range of 1 to 15 ng analyte/mL, the recovery was greater than 95%, and the reproducibility was sufficient. The assay was rugged and was maintained by routinely changing the home-made, dry packed pre-column every 20th serum injection.  相似文献

A simple and sensitive high-performance liquid chromatographic method for the simultaneous assay of amiodarone and desethylamiodarone in plasma, urine and tissues has been developed. The method for plasma samples and tissue samples after homogenizing with 50% ethanol, involves deproteinization with acetonitrile containing the internal standard followed by centrifugation and direct injection of the supernatant into the liquid chromatograph. The method for urine specimens includes extraction with a diisopropyl ether-acetonitrile (95:5, v/v) mixture at pH 7.0 using disposable Clin-Elut 1003 columns, followed by evaporation of the eluate, reconstitution of the residue in methanol-acetonitrile (1:2, v/v) mixture and injection into the chromatograph. Separation was obtained using a Radial-Pak C18 column operating in combination with a radial compression separation unit and a methanol-25% ammonia (99.3:0.7, v/v) mobile phase. A wavelength of 242 nm was used to monitor amiodarone, desethylamiodarone and the internal standard. The influence of the ammonia concentration in the mobile phase on the capacity factors of amiodarone, desethylamiodarone and two other potential metabolites, monoiodoamiodarone (L6355) and desiodoamiodarone (L3937) were investigated. Endogenous substances or a variety of drugs concomitantly used in amiodarone therapy did not interfere with the assay. The limit of sensitivity of the assay was 0.025 micrograms/ml with a precision of +/- 17%. The inter- and intra-day coefficient of variation for replicate analyses of spiked plasma samples was less than 6%. This method has been demonstrated to be suitable for pharmacokinetic and metabolism studies of amiodarone in man.  相似文献

A high-performance liquid chromatographic method has been developed for the simultaneous determination of metoprolol and its alpha-hydroxylated metabolite in plasma, Metoprolol, alpha-hydroxymetoprolol and alprenolol (internal standard) are extracted from plasma at alkaline pH with diethyl ether-dichloromethane (4:1, v/v) and back-extracted with 0.01 N sulfuric acid. A 100-microliter volume of the acidic extract is injected into the chromatographic system. The compounds are eluted in about 12 min with acetonitrile-acetate buffer (75:25, v/v) on a LiChrosorb RP-8 (5 micron) column. The quantitative determinations are made fluorometrically. Concentrations down to 35 nmol/1 (10 ng/ml) of metoprolol base and 30 nmol/1 (8 ng/ml) of alpha-hydroxymetoprolol base in plasma can be determined with good precision and accuracy.  相似文献
