New serotonine 2 (5-HT2) antagonists with a monocyclic or bicyclic 2,4(1H,3H)-pyrimidinedione have been prepared and their activities evaluated. In a series of monocyclic compounds, 1-substituted 5-phenyl-2,4(1H,3H)-pyrimidinedione 14 showed potent in vitro activity, and the corresponding 3-substituted 5-phenyl and 6-phenyl derivatives 3, 8 and 20a also showed moderate activity. In the bicyclic compounds, 3-substituted 5,6,7,8-tetrahydro-2,4(1H,3H)-quinazolinedione 33 exhibited the most potent activity among the compounds prepared in this paper. The in vivo antagonist activity of 33 was comparable to that of ketanserin, a typical peripheral 5-HT2 antagonist.  相似文献

将邻羟苯基引入1,2,3-三唑结构中, 设计合成了10个1-(4-取代苯基)-4-苯基-5-取代-1,2,3-三唑类衍生物. 首先, 以对位取代的芳胺为原料, 经重氮化、叠氮化、闭环和缩合反应制得1-(4-取代苯基)-4-苯基-5-水杨醛亚胺-1,2,3-三唑类衍生物(3a~3e), 再用硼氢化钠还原制得1-(4-取代苯基)-4-苯基-5-(2-羟基苄基)氨基-1,2,3-三唑类衍生物(4a~4e). 目标化合物的结构经核磁、IR及元素分析确认. 抑菌活性测试表明, 当质量浓度为0.1 mg/L时, 除化合物3e和4e外, 所有化合物对白色念球菌的抑菌率均达95%以上, 对大肠杆菌的抑菌率达85%以上, 具有强抑菌活性, 表明该类化合物在抗菌药物开发方面有重要应用价值.  相似文献

为了进一步研究3-取代苄基-6-三氟甲基嘧啶-2,4(1H,3H)-二酮衍生物的除草活性, 以期发现更高活性化合物, 合成了16个未见文献报道的3-取代苄基-6-三氟甲基嘧啶-2,4(1H,3H)-二酮衍生物, 其结构均经过1H NMR, IR和元素分析确证. 生测结果表明, 嘧啶环1-位取代基的变化, 不仅影响化合物的抑制活性与选择性, 可能还改变了化合物的作用方式. 定量的结构与活性关系研究表明, 当作用对象为油菜时, 化合物的活性可能主要与取代基R的摩尔分子折射常数有关; 当作用对象为稗草时, 化合物的活性可能主要与取代基R的电性参数有关. 1-位为氢时, 有利于对油菜生长的抑制; 1-位为甲基时, 有利于对稗草生长的抑制.  相似文献

在冰醋酸存在下, 4-取代苯乙酮和溴素容易进行溴代反应, 得到α-溴-4-取代苯乙酮; 将其和1,2,4-三唑在丙酮中反应, 得到α-(1,2,4-三唑-1-基)-4-取代苯乙酮2; 在冰醋酸催化下, 2和4-取代苯甲酰肼1进行缩合, 得到相应酰腙衍生物3; 化合物3在乙酸酐的作用下环化生成一系列2-[1-(1,2,4-三唑)甲基]-3-乙酰基-2,5-二取代芳基-1,3,4-噁二唑啉4. 化合物4的结构经IR, 1H NMR, MS, 元素分析和单晶衍射确证.  相似文献

Cycloadditions of 1,3-butadiene derivatives having an electron-rich group at the 1-position with 4- or 3-substituted 2(1H)-quinolones were carried out to give the richly functionalized phenanthridines under both atmospheric and high pressure conditions. Furthermore, the reactivity of 4- or 3-substituted 2(1H)-quinolones acting as a dienophile with 1-substituted dienes was examined using MO calculation.  相似文献

The current work reports an organocatalytic strategy for the asymmetric catalysis of chiral benzofuran-2(3H)-ones bearing 3-position all-carbon quaternary stereocenters. Accordingly, highly enantioselective Michael addition reactions of 3-substituted benzofuran-2(3H)-ones to nitroolefins have been developed by utilizing a bifunctional tertiary-amine thiourea catalyst. The reactions accommodate a number of nitroolefins and 3-substituted benzofuran-2(3H)-ones to give the desired chiral benzofuran-2(3H)-one products with moderate to excellent yields (up to 98%) and moderate to very good selectivities (up to 19?:?1 dr and up to 91% ee). Theoretical calculations using the DFT method on the origin of the stereoselectivity were conducted. The effect of the nitroolefin substituent position on the stereoselectivity of the Michael addition reaction was also theoretically rationalized.  相似文献

Diels-Alder reactions of 3-nitro-2(1H)-quinolones with 1,3-butadiene derivatives were carried out to give the phenanthridone derivatives under both atmospheric and high pressure conditions. Alkylation of various primary amines with crotyl bromide, followed by DMAP-promoted acylation with methyl malonyl chloride to 4 and then manganic triacetate dihydrate/cupric acetate induced radical cyclization, gave 1-substituted-4-vinyl-3-carbomethoxy-2-pyrrolidinones (5). Thiation to the thiolactams 6 and guanidine cyclization then gave a series of 2-amino-3,4-dihydro-4-oxo-5-vinyl-7-substituted pyrrolo[2,3-d]pyrimidines (7). Palladium-catalyzed C-C coupling with diethyl 4-iodobenzoylglutamate led in one step via an unexpected redox reaction to the diethyl esters 9 of a series of 7-substituted derivatives of ALIMTA (LY231514, MTA), from which the target analogues 10 were readily prepared by saponification. Attempted deprotection at position 7 was successful in only one case (9d, R = CH(2)C(6)H(3)(OMe)(2)(-3',4'), which resulted in a known pentultimate precursor (9, R = H) of ALIMTA. The 7-substituted derivatives 10 proved to be inactive in vitro as inhibitors of cell division.  相似文献

The 1H and 13C NMR spectra of several isomeric N-substituted tetrazoles have been investigated. 13C NMR is shown to be more useful for distinguishing between structural isomers of N-substituted tetrazoles except for those carrying electropositive substituents like SnBu3. Correlations of δC-5 (inverse) and 1J(C-5,H) with s?1 found for 1-substituted tetrazole allowed the identification of the N SnBu3 derivative as 1-(tri-n-butylstannyl)tetrazole. The phenyl carbon chemical shift difference ΔC′ = δC-3′-δC-2′ is insignificant for structure elucidation and conformational studies of N-substituted 5-phenyltetrazoles; ΔH′ from 1H NMR spectra seems to be more useful.  相似文献

A highly diastereo- and enantioselective Michael addition reaction with respect to prochiral 3-substituted benzofuran-2(3H)-ones and maleimides by a chiral bifunctional thiourea-tertiary amine catalyst was investigated. The corresponding adducts, containing a quaternary center at the C3-position of the benzofuran-2(3H)-one as well as a vicinal tertiary center, were generally obtained in high yields (up to 99%) with very good diastereo- (up to >20:1 dr) and enantioselectivities (up to 97% ee).  相似文献

The intramolecular 1,3-chirality transfer reaction of chiral amino alcohols 1 with 99% ee was developed to construct chiral 1-substituted tetrahydroisoquinoline 2. Bi(OTf)(3) (10 mol %)-catalyzed cyclization of 1 (R = H) afforded (S)-1-(E)-propenyl tetrahydroisoquinoline 2 (R = H) in 83% yield with a ratio of 98:2. The stereochemistry at the newly formed chiral center was produced by a syn S(N)2'-type process. In this reaction, the substituent on the benzene ring of 1 significantly affected the reactivities and selectivities. A plausible reaction mechanism was proposed.  相似文献

The influence of substituents at the C(2) carbon of N(1)-substituted thiosemicarbazones, {C(4)H(3)X-C(2)(CH(3))=N(3)-N(2)H-C(1)(=S)N(1)HR(2)} (X = O, S) on the geometry of nickel(ii) complexes has been investigated. The presence of a methyl group at the C(2) position of 2-acetylfuran-N(1)-substituted thiosemicarbazones {(C(4)H(3)O)-C(2)(CH(3))=N(3)-N(2)H-C(1)(=S)N(1)HR(2), R(2) = CH(3), HaftscN-Me; C(2)H(5), HaftscN-Et; C(6)H(5), HaftscN-Ph} induces unusual coordination by the furan ring and yielded high spin octahedral nickel(II) complexes, [Ni(κ(3)-O, N(3), S-aftscN-R(2))(2)], CH(3)1, C(2)H(5)2, and 2[Ni((κ(3)-O, N(3), S-aftscN-Ph)(2)] 3 (μ(eff) = 2.98, 1; 2.96, 2; 2.92, 3). With 2-acetylthiophene-N(1)-substituted thiosemicarbazones, {(C(4)H(3)S)-C(2)(CH(3))=N(3)-N(2)H-C(1)(=S)N(1)HR(2), R(2) = CH(3), HattscN-Me; C(2)H(5), HattscN-Et; C(6)H(5), HattscN-Ph}, N(3), S chelated low spin trans square planar complexes, {[Ni(κ(3)-O, N(3), S-attscN-R(2))(2)], R(2) = CH(3), 4; C(2)H(5), 5; C(6)H(5), 6} with pendant thiophene rings have been obtained. The bigger sized sulfur atoms of the thiophene rings form short intramolecular contacts with the deprotonated hydrazinic nitrogen atoms (SN(2)) inhibiting its lability for possible coordination to nickel(II). Complexes have one independent molecule (1) or two independent molecules (2, 3) in their respective crystal lattices. The simultaneous presence of methyl groups at the C(2) and N(1) atoms of 2-acetylthiophene-N(1)-methylthiosemicarbazone (HattscN-Me) have facilitated the binding of triphenylphosphine in three-coordinate copper(i) halide complexes, [CuX(η(1)-S-HattscN-Me)(Ph(3)P)] (X, Br, 7; Cl, 8), which represent an unusual donor set of ligands, namely, triphenylphosphine, sulfur of a thio-ligand and a halide.  相似文献

3-Amino-2-methyl-4(3H)-quinazolinone has been doubly lithiated, on nitrogen and in the 2-methyl group, with n-butyllithium. The lithium reagent thus obtained reacts with a variety of electrophiles (D(2)O, benzophenone, cyclohexanone, cyclopentanone, acetophenone, benzaldehyde, tetraisopropylthiuram disulfide (TITD)) to give the corresponding 2-substituted derivatives in very good yields. Reactions of the dilithio reagent with 2 molar equiv of methyl iodide or phenyl isocyanate give disubstituted derivatives. Double lithiation of the 2-ethyl and 2-propyl analogues have been achieved using LDA, and subsequent reactions with most electophiles are then similar. In the reaction of the dianion of the 2-ethyl compound with TITD, deamination from position 3 takes place with the formation of the 2-substituted derivative. In reactions with prochiral ketones, the dianion of the 2-ethyl compound gives very high diastereoselectivity. Lithiation and subsequent reactions of 3-(methylamino) analogues take place in a similar manner, thus providing access to a range of substituted 3-(methylamino)-2-alkyl-4(3H)-quinazolinones by a general procedure. Lithiation of 3-(dimethylamino)-2-ethyl-4(3H)-quinazolinone did not take place under similar conditions. Lithiation of 3-amino-2-unsubstituted-4(3H)-quinazolinone was also unsuccessful.  相似文献

Enantioselective synthesis of 4-substituted 4,5-dihydro-1H-[1,5]benzodiazepin-2(3H)-ones has been accomplished through chiral Lewis base-catalyzed hydrosilylation. The corresponding products were obtained in excellent yields (up to 99%) and enantioselectivities (up to 98%). The absolute configuration of product 3n has been determined as S by X-ray crystallographic analysis.  相似文献

Organolithium and Grignard reagents are added to pyrimidine-2(1H)-thione, yielding 4-substituted 3,4-dihydropyrimidine-2-(1H)-thiones. Since the synthetic procedure can be performed on a multigram scale, and since pyrimidine-2(1H)-thione as well as the majority of organometallic reagents are readily available, the process described provides an easy access and complementary to other methods to 4-substituted 3,4-dihydropyrimidine-2-(1H)-thiones.  相似文献

本文用β-三氯锗基取代丙酸为中间体合成了六个β-三芳基锗取代丙酸化合物,八个β-三(α-噻吩基)锗取代丙酸化合物, 所有化合物均经元素分析、IR、^1HNMR 证实了它们的组成和结构。另外还合成了十个未见文献报道的β-(N-芳基酰胺基)丙基锗倍半氧化物, 用元素分析和IR证实了它们的组成和结构。  相似文献
