We report on a combination of magnetic solid-phase extraction and spectrophotometric determination of bromate. Cetyltrimethylammonium ion was adsorbed on the surface of phenyl-functionalized silica-coated Fe3O4 nanoparticles (Ph-SiO2@Fe3O4), and these materials served as the sorbent. The effects of surfactant and amount of sorbent, the composition of the desorption solution, the extraction time and temperature were optimized. Under optimized conditions, an enrichment factor of 12 was achieved, and the relative standard deviation is 2.9 % (for n?=?5). The calibration plot covers the 1–50 ng mL?1 range with reasonable linearity (r 2?>?0.998); and the limit of detection is 0.5 ng mL?1. The method is not interfered by ionic compounds commonly found in environmental water samples. It was successfully applied to the determination of bromate in spiked water samples.

Methylisothiocyanate (MITC) is the main degradation product of metam sodium, a soil disinfectant widely used in agriculture, and is responsible for its disinfectant properties. Because MITC is highly toxic and volatile, metam sodium has to be applied in a manner that tries to reduce atmospheric emissions but still maintains adequate concentration of MITC in soil to ensure its disinfectant effect. Thus, monitoring of MITC concentrations in soil is required, and to this end sensitive, fast, and reliable analytical methods must be developed. In this work, a headspace solid-phase microextraction (HS-SPME) method was developed for MITC determination in water and soil samples using gas chromatography-tandem mass spectrometry (GC–MS–MS) with a triple-quadrupole analyzer. Two MS–MS transitions were acquired to ensure the reliable quantification and confirmation of the analyte. The method had linear behavior in the range tested (0.026–2.6 ng mL?1 in water, 1–100 ng g?1 in soil) with r 2 over 0.999. Detection limits were 0.017 ng mL?1 and 0.1 ng g?1 in water and soil, respectively. Recoveries for five replicates were in the range 76–92 %, and RSD was below 7 % at the two spiking levels tested for each matrix (0.1 and 1 ng mL?1 for water, 4 and 40 ng g?1 for soil). The potential of using multiple HS-SPME for analyzing soil samples was also investigated, and its feasibility for quantification of MITC evaluated. A functionalized gold-nanoparticle bio-barcode assay, based on real-time immuno-PCR (IPCR), was designed for the determination of 3,4,3',4'-tetrachlorobiphenyl (PCB77). 15 nm gold nanoparticles were synthesized, and modified with thiol-capped DNA and goat anti-rabbit IgG. The nanoparticle probes were used to replace antibody–DNA conjugate in the IPCR, and were fixed on the PCR tube wall via the immune reaction. Real-time PCR was performed to quantify the DNA signal directly. Under optimized conditions, the new method was used to detect PCB77 with a linearity range from 5 pg L?1 to 10 ng L?1, and the limit of detection (LOD) was 1.72 pg L?1. Real samples of Larimichthys polyactis, collected from the East China Sea, were analyzed. Recovery was from 82 % to 112 %, and the coefficient of variation (CV) was acceptable. The results were compared with GC–ECD, revealing that the method would be acceptable for providing rapid, semi-quantitative, and reliable test results for making environmental decisions.

A selective and low organic-solvent-consuming method of sample preparation combined with high-performance liquid chromatography with diode-array detection is introduced for analysis of phthalic acid esters in edible oils. Sample treatment involves initial liquid–liquid partitioning with acetonitrile, then QuEChERS cleanup by dispersive solid-phase extraction with primary secondary amine as sorbent. Preconcentration of the analytes is performed by ionic-liquid-based dispersive liquid–liquid microextraction, with the cleaned-up extract as disperser solvent and 1-hexyl-3-methylimidazolium hexafluorophosphate as extraction solvent. Under the optimized conditions, correlation coefficients (r) were 0.998–0.999 and standard errors (S y/x ) were 2.67–3.37?×?103 for calibration curves in the range 50–1000 ng g?1. Detection limits, at a signal-to-noise ratio of 3, ranged from 6 to 9 ng g?1. Intra-day and inter-day repeatability, expressed as relative standard deviation, were in the ranges 1.0–6.9 % and 2.4–9.4 %, respectively. Recovery varied between 84 % and 106 %. The developed method was successfully used for analysis of the analytes in 28 edible oils. The dibutyl phthalate content of four of the 28 samples (14 %) exceeded the specific migration limit established by domestic and international regulations.

Biomass burning is becoming an increasing contributor to atmospheric particulate matter, and concern is increasing over the detrimental health effects of inhaling such particles. Levoglucosan and related monosaccharide anhydrides (MAs) can be used as tracers of the contribution of wood burning to total particulate matter. An improved gas chromatography–mass spectrometry method to quantify atmospheric levels of MAs has been developed and, for the first-time, fully validated. The method uses an optimised, low-volume methanol extraction, derivitisation by trimethylsilylation and analysis with high-throughput gas chromatography–mass spectrometry (GC–MS). Recovery of approximately 90 % for levoglucosan, and 70 % for the isomers galactosan and mannosan, was achieved using spiked blank filters estimates. The method was extensively validated to ensure that the precision of the method over five experimental replicates on five repeat experimental occasions was within 15 % for low, mid and high concentrations and accuracy between 85 and 115 %. The lower limit of quantification (LLOQ) was 0.21 and 1.05 ng m?3 for levoglucosan and galactosan/mannosan, respectively, where the assay satisfied precisions of ≤20 % and accuracies 80–120 %. The limit of detection (LOD) for all analytes was 0.105 ng m?3. The stability of the MAs, once deposited on aerosol filters, was high over the short term (4 weeks) at room temperature and over longer periods (3 months) when stored at ?20 °C. The method was applied to determine atmospheric levels of MAs at an urban background site in Leicester (UK) for a month. We report on a method for the extraction of organophosphorus pesticides (OPPs) from water samples using mixed hemimicelles and magnetic titanium dioxide nanoparticles (Fe3O4@TiO2) modified by cetyltrimethylammonium. Fe3O4@TiO2 nanoparticles were synthesized by a hydrothermal process and then characterized by scanning electron microscopy and Fourier transform IR spectrometry. The effects of the quantity of surfactant, extraction time, desorption solvent, pH value, extraction volume and reuse of the sorbent were optimized with respect to the extraction of OPPs including chlorpyrifos, dimethoate, and trichlorfon. The extraction method was applied to analyze OPPs in environmental water using HPLC along with UV detection. The method has a wide linear range (100–15,000 ng L?1), good linearity (r?>?0.999), and low detection limits (26–30 ng L?1). The enrichment factor is ~1,000. The recoveries (at spiked levels of 100, 1,000 and 10,000 ng L?1) are in the range of 88.5–96.7 %, and the relative standard deviations range from 2.4 % to 8.7 %.

Graphene-based magnetic nanoparticles (G-Fe3O4) were prepared and used as an effective adsorbent for the solid-phase extraction of trace quantities of cadmium from water and vegetable samples. The method avoids some of the time-consuming steps associated with traditional solid phase extraction. The excellent sorption property of the G-Fe3O4 system is attributed to π - π stacking interaction and hydrophobic interactions between graphene and the Cd-PAN complex. The effects of pH, the amount of G–Fe3O4, extraction time, type and volume of eluent, desorption time and interfering ions on the extraction efficiency were optimized. The preconcentration factor is 200. Cd(II) was then quantified by flame atomic absorption spectrometry with a detection limit of 0.32 ng mL?1. The relative standard deviation (at 50 ng mL?1; for n?=?10) is 2.45 %. The method has a linear analytical range from 1.1 to 150 ng mL?1, and the recoveries in case of real samples are in the range between 93.1 % and 102.3 %.

We describe a novel magnetic metal-organic framework (MOF) prepared from dithizone-modified Fe3O4 nanoparticles and a copper-(benzene-1,3,5-tricarboxylate) MOF and its use in the preconcentration of Cd(II), Pb(II), Ni(II), and Zn(II) ions. The parameters affecting preconcentration were optimized by a Box-Behnken design through response surface methodology. Three variables (extraction time, amount of the magnetic sorbent, and pH value) were selected as the main factors affecting adsorption, while four variables (type, volume and concentration of the eluent; desorption time) were selected for desorption in the optimization study. Following preconcentration and elution, the ions were quantified by FAAS. The limits of detection are 0.12, 0.39, 0.98, and 1.2 ng mL?1 for Cd(II), Zn(II), Ni(II), and Pb(II) ions, respectively. The relative standard deviations were <4.5 % for five separate batch determinations of 50 ng mL?1 of Cd(II), Zn(II), Ni(II), and Pb(II) ions. The adsorption capacities (in mg g?1) of this new MOF are 188 for Cd(II), 104 for Pb(II), 98 Ni(II), and 206 for Zn(II). The magnetic MOF nanocomposite has a higher capacity than the Fe3O4/dithizone conjugate. This magnetic MOF nanocomposite was successfully applied to the rapid extraction of trace quantities of heavy metal ions in fish, sediment, soil, and water samples.

A novel non-instrumental bioanalysis based on colloidal-gold immunochromatography in a modified glass capillary was developed and named capillary immunochromatographic assay (CICA). In this report, glass capillary was proposed as a support in immunochromatographic assay because of its excellent characteristics. Goat anti-rabbit IgG and parvalbumin (PV) were immobilized on the inner wall of the glass capillary as control zone and test zone, respectively. The CICA was constructed, and main variables for the performance were optimized. Using an important allergen of fish products (parvalbumin, PV) as the target, the analytical efficiency of the developed technique was investigated and the visual detection limit (VDL) and semi-quantitative limit of detection (LOD) were estimated to be 70 ng mL?1 and 40 ng mL?1, respectively. The coefficient of variation (CV) for the intra-assay and inter-assay was calculated for the PV concentration of 50 ng mL?1, and the entire operation, including sample preparation, was consistently performed in 30 min. The developed technique was implemented and validated with different foodstuffs, including Scophthalmus maximus (Linnaeus), surimi products, and livestock, confirming sufficient accuracy and precision of results and verifying the method to be efficacious. These results enabled us to propose CICA as a new and promising technique for simple, rapid, and on-site screening of PV in biological samples. Graphical abstract

An ion imprinted polymer coated onto magnetite (Fe3O4) nanoparticles is shown to be a useful magnetic sorbent for the fairly selective preconcentration of vanadium. The sorbent was prepared by radical copolymerization of 3-(triethoxysilyl)propyl methacrylate (the monomer), ethylene glycol dimethacrylate (the cross-linker), and the vanadium(IV) complex of 1-(2-pyridylazo-2-naphthol) in the presence of magnetite nanoparticles. The material was characterized by IR spectroscopy, scanning electron microscopy, and thermal analysis. The vanadium(IV) ions were removed from the imprint by a solution containing thiourea and HCl, and the eluent was submitted to AAS. The analytical efficiency and relative standard deviation are 99.4 and ±2.3 %, respectively, under optimum conditions, and the limit of detection is 20 ng mL?1. The method was successfully applied to the preconcentration and determination of vanadium(IV) ions in crude oil. Figure

A sensitive, simple and rapid LC–MS–MS method has been developed and validated for the simultaneous determination of l-dopa and l-dopa n-pentyl ester hydrochloride in rat plasma in the present study. The analytes were separated on a C18 column (5 μm, 2.1 × 150 mm) with a security guard C18 column (5 μm, 4 × 20 mm) and a triple-quadrupole mass spectrometer was applied for detection. The method was linear over the concentration ranges of 25–5,000 ng mL?1 for l-dopa and 12.5–2,500 ng mL?1 for l-dopa n-pentyl ester hydrochloride. A glassy carbon electrode (GCE) was modified with nickel(II) hydroxide nanoparticles and a film of molybdenum sulfide. The nanocomposite was prepared by two-step electrodeposition. Scanning electron microscopy reveals that the nanoparticles are uniformly deposited on the film. Cyclic voltammetry and chronoamperometry indicate that this modified GCE displays a remarkable electrocatalytic activity towards nonenzymatic oxidation of glucose. Response is linear in the 10–1,300 μM concentration range (R 2 ?=?0.9987), the detection limit is very low (5.8 μM), response is rapid (< 2 s), and selectivity over ascorbic acid, dopamine, uric acid, fructose and galactose is very good.

An efficient nonenzymatic glucose sensor based on Ni(OH)2/MoSx nanocomposite modified glassy carbon electrode has been fabricated via a two-step electrodeposition approach. Hexafluoroisopropanol (HFIP)-induced coacervation in aqueous mixed systems of catanionic surfactants of dodecyltrimethylammonium bromide (DTAB) and sodium dodecyl sulfate (SDS) was described in detail, and its application in the extraction of strongly polar sulfonamides (SAs) was investigated. With 10 % (v/v) HFIP inclusion, coacervation formation and two-phase separation occur in a wide range of SDS/DTAB mole ratios (88:12~0:100 mol/mol) and total surfactant concentrations (10~200 mmol/L). The interactions between HFIP and DTAB play an important role in coacervation formation. The HFIP-induced SDS–DTAB coacervation extraction proves to be an efficient method for the extraction and preconcentration of SAs. Both hydrophobic interaction and polar interactions (hydrogen–bond, electrostatic, and π-cation) contribute to the distribution of SAs into coacervate phase. The proposed HFIP-induced SDS–DTAB coacervation extraction combined with HPLC–UV was employed for the extraction and quantitative determination of SAs in environmental water samples. Limits of detection were 1.4~2.5 ng mL?1. Excellent linearity with correlation coefficients from 0.9990 to 0.9995 was obtained in the concentration of 0.01~10 μg mL?1. Relative recoveries were in the range of 93.4~105.9 % for analysis of the lake, underground, and tap water samples spiked with SAs at 0.01, 1.0, and 10 μg/mL, respectively. Relative standard deviations were 0.7~3.2 % for intraday precision and 1.3~4.6 % for interday precision (n?=?3). Concentration factors were 17~49 for three water samples spiked with 0.01 μg/mL SAs. The results demonstrate that the proposed extraction method is feasible for the preconcentration and determination of trace SAs in real water samples. Graphical abstract

A sensitive fluorescence liquid chromatographic analytical method was developed for the simultaneous determination of carnosine enantiomers in rat plasma. The method was applied to pharmacokinetic studies. Chiral separation of carnosine enantiomers was achieved by pre-column derivatization with o-phthaldialdehyde and the thiol N-acety-l-cysteine as derivating reagents. They were separated on an ODS column and detected by fluorescence detection (λex = 350 nm, λem = 450 nm). γ-Aminobutyric acid was used as internal standard. The method was linear up to 6,000 ng mL?1 for l-carnosine, 4,000 ng mL?1 for d-carnosine. Low limit of quantitation (LLOQ) was 40 ng mL?1 for each isomer. The relative standard deviations obtained for intra- and inter-day precision were lower than 12% and the recoveries were higher than 75% for both enantiomers. The method was applied to a stereoselective study on the pharmacokinetics of carnosine after oral administration with a single dose (carnosine, 75 mg kg?1 for each isomer) to a rat. The initial data indicated that l-carnosine had a larger value of the highest plasma concentration than d-carnosine (C max 5,344 vs. 1,914 ng mL?1), and that of l-carnosine had a lower value of AUC(0?∞) and t 1/2(h) (AUC(0?∞) 5,306 vs. 6,321 ng h mL?1, t 1/2 1.43 vs. 3.37 h). Our results indicated that the pharmacokinetic of l-carnosine and d-carnosine revealed enantioselective properties significantly.  相似文献

Xanthine oxidase (XOD) catalyzes the metabolism of hypoxanthine and xanthine to uric acid, the overproduction of which could cause hyperuricemia, a risk factor for gout. Inhibition of XOD is a major treatment for gout, and biflavonoids have been found to act as XOD-inhibitory compounds. In this study, ultrafiltration liquid chromatography with photodiode-array detection coupled to electrospray-ionization tandem mass spectrometry (UF-LC-PDA–ESI-MS) was used to screen and identify XOD inhibitors from S. tamariscina. High-performance counter-current chromatography (HPCCC) was used to separate and isolate the active constituents of these XOD inhibitors. Furthermore, ultrahigh-performance liquid chromatography (UPLC) and triple-quadrupole mass spectrometry (TQ-MS) was used to determine the XOD-inhibitory activity of the obtained XOD inhibitors, and enzyme kinetics was performed with Lineweaver–Burk (LB) plots using xanthine as the substrate. As a result, two compounds in S. tamariscina were screened as XOD inhibitors: 65.31 mg amentoflavone and 0.76 mg robustaflavone were isolated from approximately 2.5 g?S. tamariscina by use of HPCCC. The purities of the two compounds obtained were over 98 % and 95 %, respectively, as determined by high-performance liquid chromatography (HPLC). Lineweaver–Burk plot analysis indicated that amentoflavone and robustaflavone were non-competitive inhibitors of XOD, and the IC 50 values of amentoflavone and robustaflavone for XOD inhibition were 16.26 μg mL?1 (30.22 μmol L?1) and 11.98 μg mL?1 (22.27 μmol L?1), respectively. The IC 50 value of allopurinol, used as the standard, was 7.49 μg mL?1 (46.23 μmol L?1). The results reveal that the method for systematic screening, identification, and isolation of bioactive components in S. tamariscina and for detecting their inhibitory activity using ultrafiltration LC–ESI-MS, HPCCC, and UPLC–TQ-MS is feasible and efficient, and could be expected to extend to screening and separation of other enzyme inhibitors. We describe a simple, environmentally friendly and selective technique for the determination of ochratoxin A (OTA) in urine. It involves (a) the use of a molecularly imprinted polymer as a sorbent in micro-solid-phase extraction in which the sorbent is contained in a propylene membrane envelope, and (b) separation and detection by capillary electrophoresis (CE). Under optimized conditions, response is linear in the range between 50 and 300 ng mL?1 (with a correlation coefficient of 0.9989), relative standard deviations range from 4 to 8 %, the detection limit for OTA in urine is 11.2 ng mL?1 (with a quantification limits of 32.5 ng mL?1) which is lower than those of previously reported methods for solid-phase extraction combined with CE. The recoveries of OTA from urine spiked at levels of 50, 150 and 300 ng mL?1 ranged from 93 to 97 %.

Ion pair solid phase extraction was applied to the simultaneous preconcentration of iron and antimony. The ion pairs consisting of FeCl4 ? or SbCl4 ? anions and the benzyldimethyltetradecyl ammonium cation were formed on the surface of multi-walled carbon nanotubes, then eluted with nitric acid, and the elements finally quantified by ETAAS. The adsorption capacities of the impregnated MWCNTs are 9.2 mg g?1 for iron and 27.5 mg g?1 for antimony. The following analytical figures of merit were determined for iron and antimony, respectively: Enrichment factors of 210 and 230, assay precisions of ±5.3 % and ±4.8 %, linear calibration plots from 0.7 to 9.4 and 13.0 to 190 ng L?1, and detection limits of 0.17 and 3.5 ng L?1. The method was applied to the determination of iron and antimony in human hair, synthetic sample, and to the certified reference materials gold ore (MA-1b) and trace elements in water (SRM 1643d).

We describe a new method for differential-pulse anodic stripping voltammetric determination of thallium(I) using a carbon paste electrode modified with dicyclohexyl-18-crown-6. The effect of supporting electrolyte (type and pH), accumulation and reduction potential, and of time and amount of modifier were investigated by differential pulse anodic stripping voltammetry. A method was then worked out for the determination of thallium at low levels. Under optimized conditions, the response to Tl(I) is linear in the range from 3.0 to 250 ng mL?1. The detection limit is 0.86 ng mL?1. The sensor displays good repeatability (with a relative standard deviation of ±2.70 % for n?=?7) and was applied to the determination of Tl(I) in water, hair samples, and certified reference materials.

Water-soluble cadmium telluride quantum dots (CdTe QDs) capped with glutathione (GSH) display chemiluminescence (CL) emission on reaction with hydrogen peroxide (H2O2) in strongly alkaline medium. It is found that the CL is strongly enhanced on addition of formaldehyde in aqueous solution. A flow injection system was developed, and it is shown that there is good linearity between CL intensity and the concentration of formaldehyde in the 0.06–3.0 μg L?1 range. The limit of detection is as low as 10 ng L?1. The method was successfully applied to the determination of formaldehyde in indoor air after adsorption into an aqueous phase. The recoveries for the real samples range from 97 % to 102.5 %, and the relative standard deviation is <3.8 % for intra- and inter-assay precision.

An electrochemical glucose biosensor was developed by immobilizing glucose oxidase (GOx) on a glass carbon electrode that was modified with molybdenum disulfide (MoS2) nanosheets that were decorated with gold nanoparticles (AuNPs). The electrochemical performance of the modified electrode was investigated by cyclic voltammetry, and it is found that use of the AuNPs-decorated MoS2 nanocomposite accelerates the electron transfer from electrode to the immobilized enzyme. This enables the direct electrochemistry of GOx without any electron mediator. The synergistic effect the MoS2 nanosheets and the AuNPs result in excellent electrocatalytic activity. Glucose can be detected in the concentration range from 10 to 300 μM, and down to levels as low as 2.8 μM. The biosensor also displays good reproducibility and long-term stability, suggesting that it represents a promising tool for biological assays. Figure
