A wide range of proteinaceous inhibitors are present in plants to protect themselves from hydrolytic enzymes. In this study, turmerin, a water-soluble peptide in turmeric rhizomes, was evaluated for its inhibitory potential against glucosidase and its antioxidant (AO) capacity. Turmerin inhibited α-amylase and α-glucosidase activities with IC?? values 31 and 192?μg?mL?1, respectively. Under the experimental conditions, those values for a standard glucosidase inhibitor, acarbose, were 81 and 296?μg?mL?1, respectively. The AO capacity of turmerin was evaluated using in vitro assay systems. Turmerin showed good DPPH (IC???=?29?μg?mL?1) and superoxide (IC???=?48?μg?mL?1) and moderate ABTS (IC???=?83?μg?mL?1) radical scavenging and Fe(II) chelation (IC???=?101?μg?mL?1) capacities. The inhibitory potential showed by turmerin against enzymes linked to type 2 diabetes, as well as its moderate AO capacity, could rationalise the traditional usage of turmeric rhizome preparations against diabetes.  相似文献

Methanolic extracts of 18 Thai plants from Doi Tung, the north of Thailand, were examined for brine shrimp lethality and antioxidative activity using 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay. In brine shrimp lethality, the highly toxic plant extracts were Mitrephora wangii (LC(50) 14.8?μg?mL(-1)) and Hydrocotyle javanica (LC(50) 13.3?μg?mL(-1)). The extracts of Sageretia cordifolia, Ulmus lancaefolia and Acer chiangdaoense exhibited highly antioxidative activities with IC(50) values of 6.7, 8.1 and 9.8?μg?mL(-1), respectively. This study was designed to examine the chemical composition and in vitro antioxidant activity of the essential oil and various extracts of Haplophyllum robustum. GC-MS analysis of the oil resulted in the identification of 30 compounds, representing 99.2% of the oil; 1,8-cineole (38.1%), myrcene (10.7%), α-pinene (8.5%), 4-terpineol (7.0%) and sabinene (6.1%) were the main components. The antioxidant potential of the oil and extracts was evaluated using three separate methods: inhibition of the free radical 2,2-diphenyl-1-picrylhydrazyl (DPPH), β-carotene-linoleic acid assay and reducing power systems. The essential oil of the plant was able to reduce the stable free radical DPPH with an IC(50) of 72.0?±?1.2?μg?mL(-1) (the IC(50) for ascorbic acid was determined as 5.8?±?0.4?μg?mL(-1)). The essential oil also showed strong reducing power. The level of total phenolics was highest in the essential oil (179.5?±?2.1?μg?mg(-1)).  相似文献

Diospyros peregrina is an edible seasonal fruit found in coastal West Bengal, India. The fruits have been reported to possess a significant antioxidant activity. In this study, the aim was to isolate the lead compound responsible for the above-mentioned activity. The aqueous extract of D. peregrina fruit was subjected to dereplication coupled with an in?vitro 1,1-diphenyl-2-picryl-hydrazyl (DPPH) radical scavenging assay. The n-butanol fraction of the aqueous extract of D. peregrina fruit exhibited significant antioxidant activity (IC(50), 131.10?μg?mL(-1)) as compared with the parent extract (IC(50), 285.15?μg?mL(-1)). The n-butanol fraction was subjected to silica gel column chromatographic separation coupled with a chemo-autographic study of column eluents, employing ethanolic DPPH as a spraying reagent. Two bioactive flavonoid glycosides, namely luteoline-4'-methyl-ether-7-O-glucoside and quercetin-3-O-(glucosyl)-glucoside, were identified to exhibit IC(50) values of 74.04 and 65.78?μg?mL(-1), respectively in the DPPH assay.  相似文献

The experimental and theoretical study for evaluation of scavenging activity of edaravone (S1) and related derivatives, such as antipyrine (S2), dipyrone (S3), and phenylbutazone (S4), was carried out against DPPH and ABTS radicals. Structure–activity relationship study was performed using quantum chemical calculations at the DFT/B3LYP level of theory along with the 6-31G* basis sets. S1 and S4 are more effective scavengers against DPPH and ABTS. We observed little effects of S2 and S3 at several concentrations against these two free radicals. The calculations of HOMO, ionization potential, and bond dissociation energy confirmed that a hydrogen transfer is more preferential than an electron transfer. The radical stability of these compounds is related with spin densities. In accordance with experimental and theoretical results, edaravone is more active than phenylbutazone as scavenging drug.  相似文献

In this study, we evaluate the antioxidant properties of the various extracts of Protaetia brevitarsis Lewis (Coleoptera: Scarabaedia) at different growth stages. The antioxidant activities of six different extracts from larvae, pupae and imago were measured by 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulphonic acid) (ABTS) and singlet oxygen (1O? ). The larval methanol extracts (LME) and imago methanol extracts (IME) displayed the greatest effect in DPPH and ABTS radical scavenging assay, but the activity of water extracts was weaker in the all tested assays. However, LME and IME could be compared to ascorbic acid in 1O? quenching ability (the effective concentrations of 50% 1O? quenching: EC?? 0.174, 0.149 and 0.177?mg?mL?1, respectively). The antioxidant ability of the extracts to scavenge free radicals could significantly change the contents of gallic acid equivalent, an important factor based on the value of R2. The results suggest that our study may contribute to the development of new bioactive products with potential applications to reduce oxidative stress as well as play a vital role in protecting insect organisms against oxidative damage caused by undesirable conditions.  相似文献

In the traditional Sardinian system of medicine, Rubia peregrina L. (Rubiaceae) is reported as an aphrodisiac herb. Since the aphrodisiacs may also have antioxidant and dopaminergic activities, the aim of this study was to study the effect of ethanolic extract of aerial parts of R. peregrina for the scavenging of free DPPH radicals and the inhibition of haloperidol-induced catalepsy in mice and reserpine-induced orofacial dyskinesia in rats. The extract exhibited significant antioxidant activity in a free radical DPPH assay with IC(50)?=?55.6?μg?mL(-1), which was very close to IC(50) of ascorbic acid. This study showed for the first time the antioxidant and hypoglycaemic properties of the methanol, n-hexane and ethyl acetate extracts from Senecio stabianus Lacaita, a plant that belongs to the Asteraceae family. The antioxidant activities were carried out using two different in vitro assays, namely 2,2-diphenyl-1-picrylhydrazyl (DPPH) test and 2,2'-azinobis(3-ethylbenzthiazoline-6-sulphonate) (ABTS) test. The ethyl acetate extract showed the highest activity with inhibitory concentration 50% (IC(50)) values of 35.5 and 32.7?μg?mL(-1) on DPPH test and ABTS test, respectively. This activity may be related to a good total phenol and flavonoid content. All extracts were also tested for their potential inhibitory activity of α-amylase and α-glucosidase digestive enzymes. The n-hexane extract exhibited the highest α-amylase inhibition with an IC(50) value of 0.21?mg?mL(-1). Through bioassay-guided fractionation processes seven fractions (A-G) were obtained and tested. Based on the phytochemical analysis, the activity of n-hexane extract may be related to the presence of monoterpenes and sesquiterpenes.  相似文献

Abstract

Thirty-three compounds (133) were isolated from the rhizomes of Osmunda japonica. Their structures were elucidated on the basis of spectral analysis and identified as ent-kaurene terpenoids (1–3), anthraquinones (4–8), flavonoids (9–12), steroids (13–15), and other compounds (1633). Compound 114, 1927 were isolated from the genus osmunda for the first time. Compound 28–29 were isolated from the plant for the first time. Cytotoxicities of all compounds against Hela, HepG2 and A549 cell lines were measured by MTT assay, and their antioxidant activities were determined by DPPH and ABTS radical scavenging assays. Compound 2 exhibited potent cytotoxicity against Hela, HepG2 cell lines with IC50 values of 9.31 and 9.66?μM, respectively. Compound 9 showed good antioxidant activity. The Trolox-equivalent antioxidant capacity (TEAC) value was 0.95?mM in ABTS assay, and the IC50 value was 18.63?μM in DPPH assay.  相似文献

A new compound, 3,5-dihydroxy-N′-(5-bro-2-hydroxybenzylidene)benzohydrazide hydrate, has been synthesized and characterized by single-crystal X-ray diffraction. The title compound crystallizes in a monoclinic system, space group P21/c with a = 14.267(6), b = 8.079(4), c = 13.157(6) , β = 108.114(4), Z = 4, V = 1441.4(11) 3, Dc = 1.701 g/cm3, F(000) = 744.0, μ = 2.878 and S = 1.021. The final R = 0.0457 and wR = 0.1149 for 3500 observed reflections with I > 2 (I). The ability with which the compound scavenges free radicals of ABTS and DPPH, IC50, is 34.13 and 101.6 μM, respectively.  相似文献

The 70% alcohol extract of Oxytropis myriophylla (PALL.) DC. (Leguminosae) exhibited high radical scavenging activity (IC(50) value: 88.0?μg?mL(-1) and 86.7?μg?mL(-1)) on 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulphonic acid) (ABTS) radical scavenging assays. Further chemical investigation led the isolation of one new lignan, namely myriophylloside G (1), together with three known compounds. Their structural elucidations of all the compounds were based on extensive spectroscopic methods, including HRESIMS and 2D NMR experiments (HSQC, HMBC and (1)H-(1)HCOSY) and by comparison with reference values.  相似文献

Helichrysum gymnocephalum essential oil (EO) was prepared by hydrodistillation of its leaves and characterized by GC-MS and quantified by GC-FID. Twenty three compounds were identified. 1,8-Cineole (47.4%), bicyclosesquiphellandrene (5.6%), γ-curcumene (5.6%), α-amorphene (5.1%) and bicyclogermacrene (5%) were the main components. Our results confirmed the important chemical variability of H. gymnocephalum. The essential oil was tested in vitro for cytotoxic (on human breast cancer cells MCF-7), antimalarial (Plasmodium falciparum: FcB1-Columbia strain, chloroquine-resistant) and antioxidant (ABTS and DPPH assays) activities. H. gymnocephalum EO was found to be active against MCF-7 cells, with an IC(50) of 16 ± 2 mg/L. The essential oil was active against P. falciparum (IC(50) = 25 ± 1 mg/L). However, the essential oil exhibited a poor antioxidant activity in the DPPH (IC(50) value > 1,000 mg/L) and ABTS (IC(50) value = 1,487.67 ± 47.70 mg/L) assays. We have reviewed the existing results on the anticancer activity of essential oils on MCF-7 cell line and on their antiplasmodial activity against the P. falciparum. The aim was to establish correlations between the identified compounds and their biological activities (antiplasmodial and anticancer). β-Selinene (R2 = 0.76), α-terpinolene (R2 = 0.88) and aromadendrene (R2 = 0.90) presented a higher relationship with the anti-cancer activity. However, only calamenene (R2 = 0.70) showed a significant correlation for the antiplasmodial activity.  相似文献

The antioxidant and anticancer properties of a medicinal plant, Romulea tempskyana (R. tempskyana) (Iridaceae) were investigated. The fresh corm water extract of R. tempskyana significantly increased cell viability against H(2)O(2) cytotoxicity(.) The extract also showed high inhibition of the lipid peroxidation activity (78.20%), reducing power (IC(50) 64.99?μg?mL(-1)) activity and hydroxyl (IC(50) 38.66?μg?mL(-1)), superoxide (IC(50) 25.99?μg?ml(-1)) and DPPH (IC(50) 19.88?μg?mL(-1)) radical scavenging activity. On the other hand, treatment of the cells with higher extract concentration showed the anticancer activity inducing cytotoxicity. The extract significantly affected Hepatoma G2 and H1299 cell proliferation (IC(50); 103.79 and 88.15?μg?mL(-1)). The amount of MDA (2-fold and 2.5-fold) and activities of several cellular antioxidant enzymes, including Se-GSPx (30%, 15%), non-Se-GSH-Px (11%, 16%) and GST (17%, 23%) increased in Hepatoma G2 and H1299 cells treated with IC(50) concentrations of extract, respectively. These findings suggest that R. tempskyana extract exhibits antioxidant and carcinogenesis-reducing potential.  相似文献

Two new flavone glucosides, nitensosides A and B (1, 2), together with four known compounds, sorbifolin (3), sorbifolin 6-O-beta-glucopyranoside (4), pedalitin (5), and pedalitin 6-O-beta-glucopyranoside (6) were isolated from Pterogyne nitens. Their structures were elucidated from 1D and 2D NMR analysis, as well as by high resolution mass spectrometry. All the isolated flavones were evaluated for their myeloperoxidase (MPO) inhibitory activity. The most active compound, pedalitin, exhibited IC50 value of 3.75 nM on MPO. Additionally, the radical-scavenging capacity of flavones 1-6 was evaluated towards ABTS and DPPH radicals and compared to standard compounds quercetin and Trolox.  相似文献

In this work, an evaluation of the biological activities of Globularia alypum L. extracts and their global chemical composition was realized. Extracts from G. alypum were obtained by two extraction methods. The composition of polyphenols (8.5-139.95 g gallic acid equivalent/Kg of dry mass), tannins (1.39-18.65 g catechin equivalent/Kg of dry mass), anthocyanins (8.17-70.69 mg cyanidin equivalent/Kg of dry mass) and flavonoids (0.31-19.28 g quercetin equivalent/Kg of dry mass) was evaluated. The samples were subjected to a screening for their antioxidant activities using the DPPH* and ABTS*+ assays. For the first time, the anti-tuberculosis activity (H(37)Rv) for G. alypum was tested against Mycobacterium tuberculosis. The strongest antioxidant activity was obtained for the methanol extract (IC(50 ) = 15.58 ± 0.168 mg/L) and the best anti-tuberculosis activity was obtained for the petroleum ether extract (IC(50 )= 77 mg/L). We have found a positive correlation between the total phenolics content and the antioxidant activity R2 = 0.88 (DPPH*) and R2 = 0.97 (ABTS*+). We have found also a positive correlation between the flavonoid content and the antioxidant activity R2 = 0.91 (DPPH*) and R2 = 0.91 (ABTS*+).  相似文献

The occurrence of three known benzophenones, namely annulatophenonoside, acetylannulatophenonoside and annulatophenone as well as a flavonol O-glycoside guajaverin in the aerial parts of Hypericum maculatum Crantz was established. In addition, hyperoside, isoquercitrin and miquelianin were isolated from this plant, as well. Radical scavenging and antioxidant activities of the isolated compounds were examined using 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) diammonium salt (ABTS) free radicals, ferric reducing antioxidant power (FRAP) assay and inhibition of lipid peroxidation in linoleic acid system by the ferric thiocyanate method. Isoquercitrin demonstrates the highest DPPH radical scavenging (96.6?±?0.3%), FRAP (23.8?±?0.2 Trolox equivalent, TE?mol?1) and antioxidant activity in linoleic acid system. Guajaverin and acetylannulatophenonoside show significantly strong ABTS radical scavenging activity (93.9?±?0.4% and 93.4?±?0.6%, respectively), which is comparable to that of ascorbic acid (96.2?±?0.4%).  相似文献

Erythrina abyssinica Lam. is an important medicinal plant growing in Sudan; its seeds were investigated for the first time for their alkaloidal constituents and biological activity. The in vitro cytotoxicity of the crude alkaloidal fraction (CAF) against the cell lines HeLa, Hep-G2, HEP-2, HCT116, MCF-7 and HFB4 showed promising activity, with IC?? values of 13.8, 10.1, 8.16, 13.9, 11.4 and 12.2?μg?mL?1, respectively. Doxorubicin (positive control) showed in vitro cytotoxic activity with IC?? values 3.64, 4.57, 4.89, 3.74, 2.97 and 3.96?μg?mL?1, respectively. Bioassay-guided fractionation and isolation of the CAF led to the isolation of five Erythrina alkaloids, identified as erythraline, erysodine, erysotrine, 8-oxoerythraline and 11-methoxyerysodine. These were evaluated for their in vitro cytotoxic activity against Hep-G2 which resulted in IC?? values 17.60, 11.80, 15.80, 3.89 and 11.40?μg?mL?1, respectively. Furthermore, in vitro cytotoxic activity against HEP-2 was evaluated, which resulted in IC?? values 15.90, 19.90, 21.60, 18.50 and 11.50?μg?mL?1, respectively. The CAF caused a reduction in the viability of mock-infected MT-4 cells with a CC?? of 53?μM and a 50% protection of MT-4 cells against HIV-1 induced cytopathogeneticy with a EC?? of >53?μM, compared with EFV as a positive control, which had a CC?? of 45?μM and an EC?? of 0.003?μM. Melastoma malabathricum (MM) is a well-known plant in Malaysian traditional medicine, locally known as senduduk. Its ethanol and aqueous extracts have been used in the present investigation to study the immunomodulatory role on human peripheral blood mononuclear cell (PBMC), and the DPPH, ABTS and FRAP free radical scavenging activities were also measured. Total flavonoids and total phenolic contents were assayed and the antibacterial effect was tested against four species of bacteria; two Gram-positive (Staphylococcus aureus and Streptococcus agalactiae) and two Gram-negative (Escherichia coli and Klebsilla pneumonia). The tests were carried out using the disc diffusion, minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) methods. Moreover, the acute toxicity was evaluated in vivo on the ethanol extract of MM to establish its safety when administered orally. In our results, both extracts of MM showed abilities to scavenge DPPH and ABTS free radicals, IC(50) values: (11.599 ± 0.84, 10.573 ± 0.58 μmol/L) and (62.657 ± 0.78, 63.939 ± 0.48 μmol/L) for ethanol and aqueous extracts respectively. Indeed the ethanol extract evidenced high phenolic content (384.33 ± 0.005 mg/g), flavonoids contents (85.8 ± 0.009 mg/g) and ferric reducing antioxidant power (33,590 ± 0.038 mmol/g), with high activity against S. aureus and S. agalactiae (11 ± 0.3 and 12 ± 0.6 mm inhibition zones). Likewise, the percentage of peripheral blood mononuclear cells (PBMC) viability was increased in response to MM, IC(50) values (1.781 ± 1.2 and 6.545 ± 0.93 μg/mL) for ethanol and aqueous extracts, respectively. In addition, our results showed that the MM extract is safe even at a high dose of 5,000 mg/kg and has no oral toxicity. These findings suggest the excellent medicinal bioactivity of MM and explain the popularity of this plant in the folk medicine as a remedy for different illnesses.  相似文献

A series of novel 1,3,4-oxadiazole derivatives based on benzisoselenazolone has been prepared and tested for antiproliferative activity in vitro against the cells of human cancer cell lines: SSMC-7721 (human liver cancer cell), MCF-7 (human breast cancer cell) and A549 (human lung cancer cell). All the compounds obtained exhibited antiproliferative activity and showed selective cytotoxicity against different cancer cells. Compounds 7d and 7i showed significant antiproliferative activities against MCF-7 cells, with IC50 values of 1.07 and 1.76?μM respectively. Compound 7d were found to be the most potent compound against SSMC-7721 cells, with IC50 values 4.46?μM.  相似文献

This study was designed to investigate antioxidant and anticholinesterase potential of Iris germanica var; florentina. Acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) inhibitory potential of plant samples were investigated by Ellman’s assay. Antioxidant activity was performed using DPPH, H2O2 and ABTS free radical scavenging assays. Total phenolics and flavonoids contents were expressed in mg GAE/g dry weight and mg RTE/g, respectively. In AChE inhibition assay, Ig.Fl, Ig.Sp and Ig.Cf fractions exhibited highest activity with IC50 values of < 0.1, 5.64 and 19 μg/mL, respectively. In BChE inhibitory assay, Ig.Fl, Ig.Sp, Ig.Cf and Ig.Cr were most active with IC50 of < 0.1, < 0.1, 31 and 78 μg/mL, respectively. In DPPH assay, Ig.Fl and Ig.Cf exhibited highest inhibition of free radicals, 80.52% (IC50 = 9 μg/mL) and 78.30% (IC50 = 8 μg/mL), respectively. In ABTS assay Ig.Cr, Ig.Cf, Ig.Fl and Ig.Sp exhibited IC50 values of < 0.1, 2, 2 and 3 μg/mL, respectively.  相似文献
