An improved procedure for determination of the residual DPPH (1,1-diphenyl-2-picrylhydrazyl) free radical concentration was proposed taking into account the absorbance of both DPPH free radicals and DPPH nonradical (1,1-diphenyl-2-picrylhydrazine) stable form. The calculated residual DPPH free radical concentrations were compared with those obtained from a calibration curve and variation coefficients below 10 % were found.  相似文献

For more than half of humanity, rice is life. Rice is a grain which has shaped the history, culture, diet and economy of billions of people in Asia. In Thailand, it is the essence of life. Archaeological evidence revealed that rice had been planted in northeastern area of Thailand more than 5,500 years ago which is earlier than in China and India. The ancient rice grains were found in various archaeological sites in Thailand such as Nakhon Nayok, Suphan Buri and Prachin Buri Provinces. In this work, the ancient black rice from Nakhon Nayok Province was elementally analyzed using scanning electron microscope coupled with energy dispersive X-ray fluorescence spectroscopy, proton induced X-ray emission spectroscopy and micro-beam energy dispersive X-ray fluorescence spectroscopy. Infrared (IR) spectroscopy was also used to study the chemical composition and bio-molecular structure. The grains were oblique in shape with a rough surface. Three major elements (Si, Ca and Al) and other trace elements were detected. The IR spectra provided some information about the presence of molecular bonds.  相似文献

The nutrient value of Thai rice and bean in terms of minor and trace element content has been evaluated using INAA. Six varieties of rice and six of beans bought from three local markets in 2006–2007 were investigated under this study. Elemental compositions were determined using the comparator technique. Precision, accuracy and detection limits of each element found were also calculated. Three minor (Ca, K, Mg) and eight trace elements (Al, As, Br, Cl, Cu, Fe, Mn, Zn) were determined in these Thai foods.  相似文献

The photo-chemical behaviour of a number of mono- and polyfunctional commercial phenolic antioxidants has been examined using kinetic micro-second flash photolysis. The technique provides useful information on the relationship between antioxidant structure and the efficiency of phenoxy radical production. The kinetics of decay of the phenoxy radicals are also found to be dependent on structure. Mono-functional antioxidants give phenoxy radicals which decay by a second-order process whereas polyfunctional antioxidants give phenoxy radicals that decay by a first-order process. In the former case dimerisation to give bisphenolic coupling products is observed whereas, with the latter, this process is sterically inhibited. The value of flash photolysis as a probe for studying antioxidant activity is discussed.  相似文献

Antioxidant activity has been assessed in many ways. The limitation of many newer methods is the frequent lack of an actual substrate in the procedure. The combination of all approaches with the many test methods available explains the large variety of ways in which results of antioxidant testing are reported. The measurement of antioxidant activities, especially of antioxidants that are mixtures, multifunctional or are acting in complex multiphase systems, cannot be evaluated satisfactorily by a simple antioxidant test without due regard to the many variables influencing the results. Several test procedures may be required to evaluate such antioxidant activities. A general method of reporting antioxidant activity independent of the test procedure is proposed.  相似文献

The inhibition of the autoxidation of hydrocarbons and polypropylene by aliphatic, aromatic, sterically hindered and cyclic phosphites has been studied by means of volumetric and 31P-NMR techniques. The antioxidant activity of phosphites depends on the rate of their reactions with peroxyl radicals and on the way they react with alkoxyl radicals. Only those phosphites which react by substitution to give free aryloxyl radicals are effective as chain-breaking antioxidants.The reaction modes of various phosphites with various peroxyl and alkoxyl radicals have been studied in some model reactions and the relationship between structure, reaction mechanism and antioxidant activity has been elucidated.  相似文献

From the black colored rice bran of Oryza sativa cv. Heugjinjubyeo, a new 2-arylbenzofuran, 2-(3,4-dihydroxyphenyl)-4,6-dihydroxybenzofuran-3-carboxylic acid methyl ester, oryzafuran (1), was isolated. Its structure has been elucidated on the basis of spectral data. This compound showed strong antioxidative activity in a 1,1-diphenyl-2-picrylhydrazyl free radical scavenging assay.  相似文献

Considering a DNA to carcinogen electron transfer mechanism for the carcinogenesis process whereas carcinogens react as a electrophilic and DNA as nucleophilic species we propose that the cancer protecting compounds will compete with the DNA for the donation of the electron for the carcinogens. In this work, theoretical calculations of several compounds with well-known protecting activity are performed. The results were treated by a multivariant analysis (principal component analysis) in order to relate the calculated electronic parameters with the protecting activities.  相似文献

以3,4-二甲氧基苯甲醛(1)为起始原料,使用文献已报道的三氟乙酸法合成了3个含两个甲氧基取代基的新黄酮类化合物4a-4c,在新制碘化铝的催化下发生脱甲基反应,合成了3个含多个羟基取代基的新黄酮类化合物5a-5c,产率均高于90%.所有目标化合物均经过IR,~1H NMR,ESI-MS对其进行了结构确认.通过测定羟基自由基和DPPH自由基清除活性两个方面评价其抗氧化活性,结果发现,脱甲基后产物活性均增强.  相似文献

A series of di- and tripeptide-based ebselen analogues has been synthesized. The compounds were characterized by (1)H, (13)C, and (77)Se NMR spectroscopy and mass spectral techniques. The glutathione peroxidase (GPx)-like antioxidant activity has been studied by using H(2)O(2) , tert-butyl hydroperoxide (tBuOOH), and cumene hydroperoxide (Cum-OOH) as substrates, and glutathione (GSH) as a cosubstrate. Although all the peptide-based compounds have a selenazole ring similar to that of ebselen, the GPx activity of these compounds highly depends on the nature of the peptide moiety attached to the nitrogen atom of the selenazole ring. It was observed that the introduction of a phenylalanine (Phe) amino acid residue in the N-terminal reduces the activity in all three peroxide systems. On the other hand, the introduction of aliphatic amino acid residues such as valine (Val) significantly enhances the GPx activity of the ebselen analogues. The difference in the catalytic activity of dipeptide-based ebselen derivatives can be ascribed mainly to the change in the reactivity of these compounds toward GSH and peroxide. Although the presence of the Val-Ala-CO(2) Me moiety facilitates the formation of a catalytically active selenol species, the reaction of ebselen analogues that has a Phe-Ile-CO(2) Me residue with GSH does not generate the corresponding selenol. To understand the antioxidant activity of the peptide-based ebselen analogues in the absence of GSH, these compounds were studied for their ability to inhibit peroxynitrite (PN)-mediated nitration of bovine serum albumin (BSA) and oxidation of dihydrorhodamine 123. In contrast to the GPx activity, the PN-scavenging activity of the Phe-based peptide analogues was found to be comparable to that of the Val-based compounds. However, the introduction of an additional Phe residue to the ebselen analogue that had a Val-Ala dipeptide significantly reduced the potency of the parent compound in PN-mediated nitration.  相似文献

Two thiophenes; 5-(3-buten-1-ynyl)-2,2′-bithiophene (2) and α-tertthienyl (9), two alkaloids; echinopsine (10) and echinorine (11), three flavonoids; genkwanin (5), apigenin (6), and rutin (7), two triterpenoids; lupeol acetate (1) and lupeol linoleate (4), together with 2,6,10-trimethyldodeca-2,6,10-triene (4) and β-sitosterol glucoside (8) were isolated from the aerial parts of Echinops albicaulis. Antioxidant, antimicrobial and antiprotozoal activities were evaluated. E. albicaulis aqueous methanolic extract (50, 10, and 1 mg/mL) showed significant antioxidant activity comparable to the potent antioxidant, N-acetyl cysteine, moreover, the aqueous methanolic extract (1 mg/mL) significantly reduced intracellular reactive oxygen species in active cell cultures of human peripheral blood mononuclear cells under oxidative stress more than the reference antioxidant N-acetyl cysteine. None of the isolated compounds showed antimicrobial or antiprotozoal activities at concentration up to 20 μg/mL.  相似文献

One new megastigmane glycoside, ficalloside (1), and eleven known compounds, were isolated from methanol extract of Ficus callosa leaves by repeated column chromatography. Their structures were established on the basis of spectral and chemical evidence. The antioxidant activities of these compounds were measured using the oxygen radical absorbance capacity (ORAC) assay. Compound 8 exhibited potent antioxidant activity of 10.6 microM trolox equivalents at the concentration of 2 microM. At this concentration, compounds 4-7 and 9-12 showed significant antioxidant activity with ranging of 2.1-6.1 microM trolox equivalents.  相似文献

A convenient and highly convergent method for the synthesis of new imidazo[1,2-a]pyridine-based coelenterazine analogues is reported. The imidazo[1,2-a]pyridine core was constructed through a condensation between 2-aminopyridine analogues and arylglyoxals. Additionally, a new approach to the synthesis of benzylglyoxals was introduced. The imidazo[1,2-a]pyridines display moderate antioxidant activities at a low micromolar level in 2,2-diphenyl-1-picrylhydrazyl (DPPH).  相似文献

In this study, the chemical constituents of Campanula alliariifolia Willd. (Campanulaceae) are being investigated for the first time with the aid of this article. Five known compounds, which were quercetin-3-O-glucoside, quercetin-3-O-rutinoside, kaempferol-3-O-glucoside, lobetyolin (9-O-beta-D-glucopyranosyl-2,10-tetradecadien-4,6-diyne-8,14-diol) and lobetyol (2,10-tetradecadien-4,6-diyne-8,9,14-triol), were isolated from the methanol extract. The antioxidant activity of the methanol extract and the purified compounds of the plant was investigated with DPPH (1,1-diphenyl-picrilyhydrazyl) (free radical scavenging activity) and reducing power methods. The methanol extract has antioxidant capacity according to the mentioned methods. Lobetyol and lobetyolin showed significant antioxidant activity more than both methanol extract and other purified compounds.  相似文献

Tannic acid, a naturally occurring plant polyphenol, is composed of a central glucose molecule derivatized at its hydroxyl groups with one or more galloyl residues. In the present paper, we examines the in vitro radical scavenging and antioxidant capacity of tannic acid by using different in vitro analytical methodologies such as 1,1-diphenyl-2-picryl-hydrazyl free radical (DPPH) scavenging, 2,2′-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) (ABTS) radical scavenging activity, total antioxidant activity determination by ferric thiocyanate, total reducing ability determination using by Fe3+–Fe2+ transformation method, superoxide anion radical scavenging by riboflavin–methionine-illuminate system, hydrogen peroxide scavenging and ferrous ions (Fe2+) chelating activities. Also, butylated hydroxyanisole (BHA), butylated hydroxytoluene (BHT), α-tocopherol and trolox, a water-soluble analogue of tocopherol, were used as the reference antioxidant radical scavenger compounds.Tannic acid inhibited 97.7% lipid peroxidation of linoleic acid emulsion at 15 μg/mL concentration. On the other hand, the above mentioned standard antioxidants indicated an inhibition of 92.2%, 99.6%, 84.6% and 95.6% on peroxidation of linoleic acid emulsion at 45 μg/mL concentration, respectively. In addition, tannic acid had an effective DPPH scavenging, ABTS+ radical scavenging, superoxide anion radical scavenging, hydrogen peroxide scavenging, Fe3+ reducing power and metal chelating on ferrous ions activities. Also, those various antioxidant activities were compared to BHA, BHT, α-tocopherol and trolox as references antioxidant compounds. The present study shows that tannic acid is the effective natural antioxidant component that can be used as food preservative agents or nutraceuticals.  相似文献

We propose a novel approach for assessment of total antioxidant activity by monitoring kinetics of hydrogen peroxide (H2O2) scavenging after its injection into liquid sample under study. H2O2 is known to be the strongest oxidant, really presented in human body in contrast to the majority of the model oxidative systems used for evaluation of antioxidant activity. In addition, kinetic approach, being more informative than the commonly used determination of the final product, obviously provides better discrimination of potential antioxidants. Prussian Blue based sensor due to its high sensitivity and operational stability allowed to monitor kinetics of hydrogen peroxide consumption in turbid and colored samples. The pseudo-first order kinetic constants of hydrogen peroxide scavenging in the presence of different food additives correlated with total antioxidant activity of these samples evaluated via standard procedure based on lipid peroxidation. However, in contrast to the standard method, the proposed kinetic approach is expressed and does not require fresh biological tissues.  相似文献

The simultaneous monitoring of malondialdehyde, pentanal and hexanal, final products of lipid peroxidation is reported, using a headspace solid-phase microextraction (HS-SPME) technique with on-fibre derivatisation. The aldehydes are extracted and subjected to on-sorbent derivatisation into stable hydrazones with 2,4,6-trichlorophenylhydrazine (TCPH) and analyzed. The degree of inhibition of oxidation is performed by monitoring the chlorinated hydrazones after thermal desorption, by gas chromatography-electron capture detection. The procedure was employed to evaluate in vitro the antioxidant activity of Hypericum perforatum L. extracts and of the well-known antioxidant vitamin E following induction of oxidation of sunflower oil, as a model lipid system. Prior to the measurement of antioxidant activity, the optimal process conditions, i.e. headspace volume, temperature, agitation, extraction/derivatisation time and desorption time and temperature were properly established. Aqueous extracts of H. perforatum L. exhibited the highest antioxidative effect. The method is shown to be promising for screening purposes for antioxidant substances and natural extracts.  相似文献
