A capillary electrophoresis (CE) procedure with contactless conductivity detection (C(4)D) has been developed for monitoring of neutral mono- and disaccharides in drinks and foodstuffs. The separation of a mixture of seven neutral saccharides (glucose, fructose, galactose, mannose, ribose, sucrose and lactose) employed a quartz capillary, 5 μm i.d., with an effective length of 18.3 cm, and 75 mM NaOH (pH 12.8) as the background electrolyte (BGE). The limit of detection (LOD) values obtained lied within a range from 0.4 μmol L(-1) for lactose to 0.9 μmol L(-1) for ribose, with a separation time shorter than 140 s. The procedure was successfully applied to determinations of saccharides in fruit juices, Coca-Cola, milk, red and white wines, yoghurts, honey and a foodstuff additive.  相似文献

建立了毛细管区带电泳-间接紫外检测快速测定食品中乳糖、蔗糖、葡萄糖和果糖的方法。以水或5 mmol/L醋酸为样品提取液,未涂层熔融石英毛细管(30.2 cm(有效长度20 cm)×50 μm)为分离柱,4 mmol/L山梨酸钾+10 mmol/L磷酸钠+30 mmol/L NaOH(pH 12.56)+0.5 mmol/L十六烷基三甲基溴化铵(CTAB)为分离缓冲液,在-8 kV下分离,于254 nm波长下检测,10 min内实现了食品中上述4种糖的同时分离与测定。乳糖、蔗糖、葡萄糖和果糖的检出限(S/N=3)分别为50、75、25和25 mg/L,定量限(S/N=10)分别为150、225、75和75 mg/L,回收率在87.0%~107.0%之间,相对标准偏差在1.2%~4.7%之间。整个实验过程未使用有机溶剂。用该法测定了9种食品样品及1个质控样品,结果表明该法简单、快速、准确,适用于食品中乳糖、蔗糖、葡萄糖和果糖的日常测定。  相似文献

In the present work, an evaporative light scattering detector was used as a high-temperature liquid chromatography detector for the determination of carbohydrates. The compounds studied were glucose, fructose, galactose, sucrose, maltose, and lactose. The effect of column temperature on the retention times and detectability of these compounds was investigated. Column heating temperatures ranged from 25 to 175°C. The optimum temperature in terms of peak resolution and detectability with pure water as mobile phase and a liquid flow rate of 1 mL/min was 150°C as it allowed the separation of glucose and the three disaccharides here considered in less than 3 min. These conditions were employed for lactose determination in milk samples. Limits of quantification were between 2 and 4.7 mg/L. On the other hand, a temperature gradient was developed for the simultaneous determination of glucose, fructose, and sucrose in orange juices, due to coelution of monosaccharides at temperatures higher than 70°C, being limits of quantifications between 8.5 and 12 mg/L. The proposed hyphenation was successfully applied to different types of milk and different varieties of oranges and mandarins. Recoveries for spiked samples were close to 100% for all the studied analytes.  相似文献

建立了一种离子色谱-柱切换-安培检测技术同步测定饮料中三氯蔗糖、葡萄糖、果糖和蔗糖的方法。以CarboPac PA10阴离子交换保护柱和分析柱进行分离,用水和250 mmol/L NaOH梯度淋洗,脉冲安培检测。弱保留糖类在25 mmol/L NaOH流动相淋洗下分离检测,通过柱切换将保护柱切换至分析柱后,同时切换至250 mmol/L NaOH淋洗,三氯蔗糖仅通过较短的保护柱分离,4种糖类能够得到同步检测。4种糖类在0.01~20 mg/L范围内有良好的线性关系和较低的检出限,重复性好,样品测定的回收率分别为90.38%~102.88%(三氯蔗糖)、95.56%~102.75%(葡萄糖)、101.66%~114.33%(果糖)和105.03%~106.49%(蔗糖)。该方法可广泛应用于食品中强保留物质的测定。  相似文献

以对甲氧基苯胺为衍生试剂,采用毛细管电泳法分析了藏红花植物细胞多糖中的单糖组成。对衍生条件进行了优化,并对毛细管分离条件进行了系统的研究。衍生反应在醋酸含量9.5%(v/v)、80 ℃下反应2 h的衍生产率最大,衍生产物紫外检测波长234 nm。在优化的毛细管电泳分离条件(未涂层熔融石英毛细管柱(60 cm(有效长度50 cm)×50 μm),柱温25 ℃,电压20 kV,使用350 mmol/L硼酸电解液(pH 10.21),压力(3.4475 kPa)进样5 s)下,基线分离了11种结构相近的醛糖(来苏糖、木糖、核糖、葡萄糖、甘露糖、半乳糖、鼠李糖、纤维二糖、麦芽糖、乳糖)、酮糖(果糖)的衍生产物。应用该方法定量检测了藏红花植物细胞多糖水解物中糖的成分,各糖的回收率为94.3%~105.4%,相对标准偏差为3.3%~4.6%。  相似文献

A method for the determination of large amounts of carbohydrates (glucose, lactose, maltose, mannose, sucrose, and fructose) and sweeteners (xylitol and sorbitol) by reversed-phase liquid chromatography with refractive index detection without derivatization has been developed. The limits of determination for glucose, fructose, and sucrose in liquid samples were 0.1 g/L, and for xylite, lactose, maltose, mannose, and sorbite, 1 g/L. In solid samples the limits of determination for glucose, fructose, and sucrose were 0.1%, and for xylite, lactose, maltose, mannose, and sorbite, 0.6%. The method is applicable to the analysis of samples of wine, juice, honey, cookies, dairy products, and biologically active additives. The developed method for the determination of carbohydrates and sweeteners in foods and biologically active additives was certified in the Mendeleev Institute for Metrology (St. Petersburg).  相似文献

This study reports the separation of fructose, galactose, glucose, lactose and sucrose on glass microchip electrophoresis (ME) devices using a microfluidic platform adapted with external reservoirs for controlling the electrolysis phenomenon. The connections between external reservoirs and microfluidic platform were performed by saline bridges created using silicone tubing filled with BGE. The separation conditions were optimized and the best results were achieved using a BGE containing 75 mmol/L NaOH and 15 mmol/L trisodium phosphate. Electrophoretic separations were monitored using a capacitively coupled contactless conductivity detection system. The controlled electrolysis has successfully allowed the application of a higher voltage on the separation channel promoting the baseline separation of five carbohydrates within 180 s with great run‐to‐run repeatability (RSD < 1%). The achieved efficiencies ranged from 45 000 ± 6000 to 70 000 ± 3000 plates/m demonstrating a performance better than ME devices without controlled electrolysis. The proposed system offered good linearity from 1 to 10 mmol/L and LODs between 150 and 740 μmol/L. The use of external tubes for controlling the electrolysis phenomenon on ME devices has solved common problems associated to run‐to‐run repeatability and analytical reliability required for routine and quantitative analysis.  相似文献

Saccharides are the most common carbon source for Streptococcus thermophilus, which is a widely used bacterium in the production of fermented dairy products. The performance of the strain is influenced by the consumption of different saccharides during fermentation. Therefore, a precise measurement of the concentrations of saccharides in the fermentation media is essential. An 18-min long method with limits of quantitation in the range of 0.159–0.704 mg/L and with 13C labelled internal standards employing hydrophilic interaction chromatography coupled to mass spectrometric detection-(HILIC-LC-MS) allowed for simultaneous quantification of five saccharides: fructose, glucose, galactose, sucrose, and lactose in the fermentation samples. The method included a four-step sample preparation protocol, which could be easily applied to high-throughput analysis. The developed method was validated and applied to the fermentation samples produced by Streptococcus thermophilus.  相似文献

A simple and effective method using ion chromatography was developed for the simultaneous determination of five monosaccharides (arabinose, glucose, fructose, xylose, and ribose) and two disaccharides (sucrose and lactose) in raw sugar samples. The separation was performed on a CarboPac PA 10 column using the gradient elution of sodium hydroxide and water as the mobile phase. Monosaccharides and disaccharides were detected by an integrated pulsed amperometric detection (IPAD) using gold working electrode. Acid hydrolysis was used for sample preparation before the analysis of glucose and fructose. All the studied sugars showed good linear ranges within 0.5–100 µg mL−1 with the correlation coefficients higher than 0.997. The limits of detection were all less than 0.5 µg mL−1. The RSDs of the method were less than 10 %. The recoveries of the sugars that spiked in raw sugar samples ranged from 96.1 to 102.4 %. The method was successfully applied for the analysis of sugars in raw sugar samples. Sucrose is the major constituent found in the samples at 97 %.

利用循环伏安法将金纳米粒子和钼氧化物共同电沉积在玻碳电极表面,制备了金纳米粒子和钼氧化物复合膜修饰电极,利用SEM和XPS研究了MoOx/AuNPs复合膜的表面形态,并研究其修饰电极对葡萄糖的电催化氧化过程. 首次提出了阳极扫描极化反向催化伏安法,即在反向扫描过程中纯的催化氧化电流通过扣减背景电流的方法被提取出来. 显著提高电流测量灵敏度改善了信噪比. 制备的MoOx/AuNPs复合膜修饰电极在0.01-4.0 mmol/L对葡萄糖具有线性响应,电流灵敏度为2.35 mA·L/(mmol·cm2),检测限为9.01 μmol/L(信噪比为3).  相似文献

用高效阴离子交换色谱-脉冲安培检测法(HPAEC-PAD)测定了烤烟中的水溶性葡萄糖、果糖和蔗糖。采用水浸取及膜过滤法处理烤烟样品,以Dionex CarboPac PA-1阴离子交换柱为色谱柱,0.2 mol/L NaOH水溶液为淋洗液进行分离测定。葡萄糖、果糖和蔗糖的含量与其峰面积的线性关系良好,回收率均在97%以上。方法简便易行,灵敏度高,重现性良好,可以实现对烟草中单糖的快速分离和测定。  相似文献

Abstract

The photopolymerization of vinyl monomers by metal salt-saccharide system was investigated in various solvents. The rate of polymerization in the presence of a iron(III) salt in aqueous media was remarkably accelerated by the addition of saccharides. The acceleration caused by a series of saccharides was found to be in the following order: glucose > fructose > lactose maltose. α-Methyl-d-glucoside and sucrose exhibited very little effect. The overall activation energy for the photopolymerization of acrylonitrile in the presence of glucose was found to be 2.7 kcal/mole, about half of the value obtained in the absence of glucose.  相似文献

建立了梯度洗脱优化-离子色谱-脉冲安培检测技术分析婴幼儿配方乳粉中麦芽糖醇、半乳糖、葡萄糖、蔗糖、果糖、乳糖6种与代谢疾病紧密相关的糖和糖醇的方法。该方法对样品的前处理、色谱柱的选择、淋洗液的梯度等色谱条件进行了优化,重点研究了淋洗液梯度对各组分分离效果的影响。将梯度洗脱程序分解为弱保留组分分离、乳糖分离、基质消除、系统平衡4个子程序,并分别对其进行了条件优化,实现了6种组分的完全分离,并有效解决了弱保留组分色谱峰易重叠、乳糖色谱峰易拖尾等现象。6种组分的线性范围为0.5~100 mg/L,相关系数(r)>0.99,方法的检出限(S/N=3)为0.06~0.40 mg/L。该方法具有分离度好、灵敏度高等优点,可满足例行分析的要求,同时也为淋洗液的梯度优化提供了可借鉴的方法。  相似文献

 发展了在 195nm波长下直接检测葡萄糖及其衍生物的毛细管区带电泳方法。在未涂渍石英毛细管中 ,以 5 0mmol/LNa2 HPO4 5 0mmol/LNaH2 PO4 为缓冲液体系 (接近生理条件 pH 7 4) ,分离了葡萄糖及其衍生物 (葡萄糖胺、N 乙酰葡萄糖胺和葡萄糖酸钠 )。在各自相应的浓度范围内 ,峰面积与样品浓度之间呈现良好的线性关系。方法简单、快速、重复性好 ,为研究葡萄糖及其衍生物与凝集素的相互作用奠定了分离检测的基础。  相似文献

A gas chromatography-mass spectrometric method was developed for profiling of tobacco leaves. The differentiation among tobacco leaves planted in two different regions was investigated. Prior to analysis, the extraction solvent formulation was optimized and a combination of water, methanol and acetonitrile with a volume ratio of 3:1:1 was found to be optimal. The reproducibility of the method was satisfactory. Kendall tau-b rank correlation coefficients were equal to 1 (p<0.05) for 82% of the resolved peaks (up to 95% of the overall peak areas), indicating the good response correlation. Forty-four compounds including 9 saccharides, 9 alcohols, 9 amino acids, 16 organic acids and phosphoric acid were identified based on standard compounds. The method was successfully applied for profiling of tobacco leaves from Zimbabwe and Yunnan of China. Our result revealed that levels of saccharides and their derivatives including xylose, ribose, fructose, glucose, turanose, xylitol and glyceric acid were more abundant while sucrose, glucitol and D-gluconic acid were less abundant in tobacco leaves from Yunnan as compared to those from Zimbabwe. Amino acids such as L-alanine, L-tyrosine and L-threonine were found to be richer in Zimbabwe tobacco than in Yunnan tobacco.  相似文献

This paper focuses on the optimization with a design of experiments of a new CE method for the simultaneous separation of four carbohydrates of interest (fructose, glucose, lactose, and sucrose) and five potentially interfering carbohydrates (ribose, xylose, maltose, mannose, and galactose) with a highly alkaline separation electrolyte for subsequent applications to food, beverage, forensic, or pharmaceutical samples. First, the factors that potentially affect the carbohydrate migration were identified: NaOH concentration in the separation electrolyte, separation temperature, and separation electrolyte conductivity. A central composite design was then carried out to determine and model the effects of these three factors on normalized migration times and separation efficiency. From the model, an optimization of the separation was carried out using a desirability analysis based on resolutions between adjacent peaks and analysis time. The optimum conditions obtained were a separation electrolyte composed of 98 mM NaOH and 120 mM NaCl to adjust the conductivity at 4.29 S/m and a separation temperature fixed at 26.5°C. Finally, these conditions were experimentally confirmed and the robustness of the obtained separation was checked.  相似文献

基于高效液相色谱-质谱联用分析技术(HPLC-MS),在温和的NH3·H2O条件下,采用1-苯基-3-甲基-5-吡唑啉酮(PMP)衍生化试剂成功地标记了果糖及多种醛单糖,同时提出了PMP标记果糖的衍生化机理。采用Kromasil-C18色谱柱(100 mm×4.6 mm,3.5 μm)分离,梯度洗脱,选择离子模式检测,建立了8种常见单糖标记物的结构确认及含量测定的分析方法。该方法在一定质量浓度范围内具有良好的线性(相关系数> 0.9947),检出限为0.003~0.05 mg/L,定量限为0.01~0.15 mg/L。平均回收率为65.1%~116.2%,相对标准偏差≤10.2%(n=5)。自制4个产地枸杞多糖,对其单糖组成的分析结果表明,它们均由甘露糖、果糖、鼠李糖、半乳糖、葡萄糖、木糖、阿拉伯糖和脱氧核糖8种单糖组成,但各种单糖含量的分布有较大的差异。该法简便,灵敏度高,重复性好,可用于水热法水解枸杞多糖水解液中单糖标记物结构的确认及测定,对规范植物多糖的质量控制具有重要意义。  相似文献

Molecularly imprinted polymers (MIPs) represent a new class of materials possessing high selectivity and affinity for the target molecule. The main goal of this study was to prepare a galactose imprinted polymer and its potential application for the recognition of different saccharides. The selectivity of galactose imprinted polymer for several saccharides; glucose, mannose, fructose, maltose, lactose, sucrose and raffinose was investigated. Macroporous polymer was prepared utilizing ethyleneglycoldimethacrylate as a crosslinking agent, in the presence of galactose as a template molecule with acrylamide as a functional monomer. After the synthesis of polymer, galactose was removed by methanol:acetic acid washing. The selectivity of galactose imprinted polymer for other saccharides was utilized by batch rebinding assay. The arrangement of functional groups within cavities versus shape selectivity is discussed. The results showed that, the orientation of the functional groups was the dominating factor for the selectivity of galactose imprinted polymer. The dissociation constants of polymer were determined by Scatchard analysis.  相似文献
