A novel flexible scanning optical probe is constructed with a finely etched optical fiber strung through a platinum coil in the lumen of a dissipative polymer. The packaged probe is 2.2 mm in diameter with a rigid length of 6mm when using a ball lens or 12 mm when scanning the fiber proximal to a gradient-index (GRIN) lens. Driven by constant high voltage (1-3 kV) at low current (< 5 microA), the probe oscillates to provide wide forward-viewing angle (13 degrees and 33 degrees with ball and GRIN lens designs, respectively) and high-frame-rate (10-140 fps) operation. Motion of the probe tip is observed with a high-speed camera and compared with theory. Optical coherence tomography (OCT) imaging with the probe is demonstrated with a wavelength-swept source laser. Images of an IR card as well as in vivo Doppler OCT images of a tadpole heart are presented. This optomechanical design offers a simple, inexpensive method to obtain a high-frame-rate forward-viewing scanning probe.  相似文献

We present the smallest reported side-viewing needle probe for optical coherence tomography (OCT). Design, fabrication, optical characterization, and initial application of a 30-gauge (outer diameter 0.31 mm) needle probe are demonstrated. Extreme miniaturization is achieved by using a simple all-fiber probe design incorporating an angle-polished and reflection-coated fiber-tip beam deflector. When inserted into biological tissue, aqueous interstitial fluids reduce the probe's inherent astigmatism ratio to 1.8, resulting in a working distance of 300 μm and a depth-of-field of 550 μm with beam diameters below 30 μm. The needle probe was interfaced with an 840 nm spectral-domain OCT system and the measured sensitivity was shown to be only 7 dB lower than that of a comparable galvo-scanning sample arm configuration. 3D OCT images of lamb lungs were acquired over a depth range of ~600 μm, showing individual alveoli and bronchioles.  相似文献

We have developed a dual-channel optical coherence tomography-Indocyanine Green dye (OCT-ICG) fluorescence system based on a previously reported ophthalmic OCT confocal imaging system. The confocal channel is tuned to the fluorescence wavelength range of the ICG, and light from the same optical source is used to generate the OCT image and to excite the ICG fluorescence. The system enables the clinician to visualize simultaneously en face OCT slices and corresponding ICG angiograms of the ocular fundus, displayed side by side. C-scan (constant depth) and B-scan (cross section) images are collected by a fast en face scan (T scan). The pixel-to-pixel correspondence between the OCT and angiography images allows the user to capture OCT B scans precisely at selected points on the ICG confocal images.  相似文献

An intracavitary probe for magnetic resonance imaging of the pelvis has been developed that takes advantage of the “inside-out” spatial characteristics of a birdcage resonator. The probe consists of an eight-leg, birdcage resonator in a low-pass configuration operating in receive-only mode. The resonator circuit is mounted on a solid rod, is encased in Teflon®, and has been used to obtain detailed images of pelvic anatomy in a male canine. The approximate cylindrical symmetry of the external sensitivity profile of this type of circuit, employed in an intracavitary application, demonstrates the potential superiority of this type of probe design over single-loop intracavitary coils. Axial, coronal, and sagittal MR images, obtained with 8 and 16 cm fields of view, are presented to illustrate the advantages of this type of intracavitary probe compared with conventional body-coil images. The prototype described in this report has been designed for clinical use in human subjects and is currently undergoing testing to determine its efficacy in the evaluation of rectal, prostate, and gynecologic pathology.  相似文献

We discuss the design and implementation of forward-imaging instruments for optical coherence tomography (OCT), which require the delivery, scanning, and collection of single-spatial-mode optical radiation. A hand-held surgical probe for use in open surgery can provide cross-sectional images of subsurface tissue before surgical incisions are made. A rigid laparoscope for minimally invasive surgical OCT imaging provides a simultaneous enface view of the area being imaged. OCT imaging is demonstrated on in vitro human specimens.  相似文献

Doppler optical coherence tomography (OCT) can image tissue structure and blood flow at micrometer-scale resolution but has limited imaging depth. We report a novel, linear-scanning, needle-based Doppler OCT system using angle-polished gradient-index or ball-lensed fibers. A prototype system with a 19-guage (diameter of approximately 0.9 mm) echogenic needle is constructed and demonstrates in vivo imaging of bidirectional blood flow in rat leg and abdominal cavity. To our knowledge, this is the first demonstration of Doppler OCT through a needle probe in interstitial applications to visualize deeply situated microcirculation.  相似文献

We report on a novel scheme for extending the depth of focus (DOF) of ultrathin (125 μm diameter) fiber probes for optical coherence tomography (OCT) using a simple phase mask consisting of graded-index (GRIN) fiber. The technique is compatible with existing all-in-fiber probe fabrication techniques, and our simulations show that it can provide a DOF gain of ~2 at a modest ~5 dB reduction of peak sensitivity. In a prototype device using commercially available GRIN fiber, a DOF gain of 1.55 is obtained, validated by beam profiling and OCT imaging.  相似文献

A high-speed single-mode fiber-based polarization-sensitive optical coherence tomography (PS OCT) system was developed. With a polarization modulator, Stokes parameters of reflected flight for four input polarization states are measured as a function of depth. A phase modulator in the reference arm of a Michelson interferometer permits independent control of the axial scan rate and carrier frequency. In vivo PS OCT images of human skin are presented, showing subsurface structures that are not discernible in conventional OCT images. A phase retardation image in tissue is calculated based on the reflected Stokes parameters of the four input polarization states.  相似文献

A fiber probe-based positioning scan approach was established, whose precision can reach as high as 0.8 μm. And a set of modified microscope system was designed utilizing this approach, in which the scanning probe microscope (SPM) combined with an optical microscope was manipulated. The optical microscope and scanning probe can conveniently be switched through a switch panel. The observation period of samples can significantly be shortened. And more reliable images can be provided using this approach. Our design can effectively solve the inherent disadvantages of SPM technology, which makes SPM scan and image more reliably, conveniently, safely and rapidly.  相似文献

We describe a novel instrument capable of acquiring, simultaneously, adaptive optics enhanced scanning laser ophthalmoscopy and optical coherence tomography (OCT) images of the human cone mosaic in vivo. The OCT system is based on transversal scanning of the sample with a line scan rate of 14 kHz, approximately 20 times faster than a previously reported instrument. We demonstrate the capability of this instrument with the measurement of the human cone spacing in perifoveal retina.  相似文献

An experimental tracking optical coherence tomography (OCT) system has been clinically tested. The prototype instrument uses a secondary sensing beam and steering mirrors to compensate for eye motion with a closed-loop bandwidth of 1 kHz and tracking accuracy, to within less than the OCT beam diameter. The retinal tracker improved image registration accuracy to <1 transverse pixel (<60 microm). Composite OCT images averaged over multiple scans and visits show a sharp fine structure limited only by transverse pixel size. As the resolution of clinical OCT systems improves, the capability to reproducibly map complex structures in the living eye at high resolution will lead to improved understanding of disease processes and improved sensitivity and specificity of diagnostic procedures.  相似文献

We experimentally and theoretically investigated the performance of a fiber-optic based Fourier-domain common-path optical coherence tomography (OCT). The fiber-optic common-path OCT operated at the 840-nm center wavelength. The resolution of the system was 8.8 μm (in air) and the working depth using a bare fiber probe was approximately 1.5 mm. The signal-to-noise ratio (SNR) of the system was analyzed. OCT images obtained by the system were also presented.  相似文献

The specific contrast of scanning electron microscope (SEM) images of ferroelectric domains observed in the pyrocurrent (pyroprobe) mode is analyzed. Calculations taking into account both the nonuniform heating of domains and the heat diffusion via the domain walls and the crystal boundaries are performed. It is established that the heat diffusion smears the domain images of small domains. Along with the probe diameter, the scan rate is shown to be an important factor determining the character of the SEM image contrast. A decrease in the scan rate may appreciably lower the resolution even in case of a fairly thin probe.  相似文献

Combined confocal scanning ophthalmoscopy/en face T-scan-based ultrahigh-resolution optical coherence tomography (OCT) of the human retina in vivo is reported for the first time to our knowledge. The system uses a superluminescent diode-based broadband source, which gives an axial resolution of 3.2 microm in the retina. We demonstrate acquisition of T-scan-based OCT B-scan and simultaneous confocal/C-scan images of the human retina of large lateral size (covering a field of up to 20 degrees ) at a frame rate of 2Hz.  相似文献

A fiber-optic-bundle-based optical coherence tomography (OCT) probe method is presented. The experimental results demonstrate this multimode optical fiber-bundle-based OCT system can achieve a lateral resolution of 12 microm and an axial resolution of 10 microm with a superluminescent diode source. This novel OCT imaging approach eliminates any moving parts in the probe and has a primary advantage for use in extremely compact and safe OCT endoscopes for imaging internal organs and great potential to be combined with confocal endoscopic microscopy.  相似文献

We demonstrate en face swept source optical coherence tomography (ss-OCT) without requiring a Fourier transformation step. The electronic optical coherence tomography (OCT) interference signal from a k-space linear Fourier domain mode-locked laser is mixed with an adjustable local oscillator, yielding the analytic reflectance signal from one image depth for each frequency sweep of the laser. Furthermore, a method for arbitrarily shaping the spectral intensity profile of the laser is presented, without requiring the step of numerical apodization. In combination, these two techniques enable sampling of the in-phase and quadrature signal with a slow analog-to-digital converter and allow for real-time display of en face projections even for highest axial scan rates. Image data generated with this technique is compared to en face images extracted from a three-dimensional OCT data set. This technique can allow for real-time visualization of arbitrarily oriented en face planes for the purpose of alignment, registration, or operator-guided survey scans while simultaneously maintaining the full capability of high-speed volumetric ss-OCT functionality.  相似文献

We report on imaging of microcirculation by calculating the speckle variance of optical coherence tomography (OCT) structural images acquired using a Fourier domain mode-locked swept-wavelength laser. The algorithm calculates interframe speckle variance in two-dimensional and three-dimensional OCT data sets and shows little dependence to the Doppler angle ranging from 75 degrees to 90 degrees . We demonstrate in vivo detection of blood flow in vessels as small as 25 microm in diameter in a dorsal skinfold window chamber model with direct comparison with intravital fluorescence confocal microscopy. This technique can visualize vessel-size-dependent vascular shutdown and transient vascular occlusion during Visudyne photodynamic therapy and may provide opportunities for studying therapeutic effects of antivascular treatments without on exogenous contrast agent.  相似文献

We present a full range handheld probe type spectral domain optical coherence tomography (SD-OCT) method. Here, the sample arm is composed of a tilted fiber-optic cantilever scanner; thus, the phase shift concurrently occurs while sample scanning. With the phase shift, we could achieve a full range complex-conjugate-free OCT image with no additional phase shifters in the reference arm. To realize this technique, a magnetically actuated probe was adopted. Full range SD-OCT images of a pearl, human fingernail, and human tooth were subsequently obtained using this suggested probe. The scanning range and acquisition speed were 3?mm and 20 frames/s?, respectively.  相似文献

A distally actuated, rotational-scanning micromotor endoscope catheter probe is demonstrated for ultrahigh-resolution in vivo endoscopic optical coherence tomography (OCT) imaging. The probe permits focus adjustment for visualization of tissue morphology at varying depths with improved transverse resolution compared with standard OCT imaging probes. The distal actuation avoids nonuniform scanning motion artifacts that are present with other probe designs and can permit a wider range of imaging speeds. Ultrahigh-resolution endoscopic imaging is demonstrated in a rabbit with <4-microm axial resolution by use of a femtosecond Cr:forsterite laser light source. The micromotor endoscope catheter probe promises to improve OCT imaging performance in future endoscopic imaging applications.  相似文献

We propose the two-dimensional scanning probe operating with a single actuator, which is thought useful as a sample probe for optical coherence tomography (OCT). The probe was designed to use a single-body lensed fiber cantilever loaded with an iron-bead and driven by a single-solenoid actuator. Elliptic spiral trace patterns were achieved using off-axis magnetic fields of the solenoid. A three-dimensional OCT image was obtained for a scanning area of 3.8 mm × 3.4 mm at an acquisition speed of 16.7 s/V. Up to 27 Hz B-scan rate, the proposed probe worked well, and 1000 A-scans were made per each B-scan.  相似文献
