A route to selective functionalization of polyhydroxypyrrolidines is described. The method is based on orthogonal protection/deprotection along the process of synthesis of the referred pyrrolidines, which consist in hydroxylation of the double bond of 2-azabicyclo[2.2.1]hept-5-enes followed by its oxidative cleavage and in situ reduction of the intermediate dialdehyde. The synthesis of a novel N-hydroxypyrrolidine is also described.  相似文献

InCl3-CH3CN-H2O has been found to be an efficient catalyst-solvent combination for the synthesis of Perlin aldehydes and related compounds. While acetylated glycals afforded the Perlin aldehydes directly with InCl3 and water, benzylated glycals on the other hand provided the hemiacetals under identical condition. The methodology reports a non-mercurial approach to Perlin aldehydes. Noteworthy is that this reaction is more facile as well as highly selective with glycals possessing a hydroxyl as a leaving group than with a benzyloxy group. Extension of this reaction to 2-C-hydroxymethyl glycals resulted in the formation of the corresponding hemiacetals, which were further transformed in to unsaturated azasugars with an exo-methylene group at C-2 position. Glycosidase inhibition studies reveal that these compounds display selectivity in inhibiting glucosidases rather than galactosidases.  相似文献

We present a highly stereoselective procedure for the preparation of (2S and 2R,3S,4R,5S)-5-methyl-3,4-dihydroxy-2- ethoxycarbonylmethylpyrrolidines based on conjugate addition of ammonia to unsaturated aldonic esters derived from d-ribose followed by tandem cyclization. Derivatisation of these compounds to 2-hydroxyethyl-, benzymidazolylmethyl-, biphenyl-1-aminoethyl and naphthalene-l-aminoethyl-pyrrolidines is also presented.  相似文献

A practical synthesis of Entecavir (1) has been accomplished in 10 steps with 21% overall yield. The key steps to construct the five-membered carbocyclic framework 2 are a ring-closing metathesis and a diethyl-aluminum 2,2,6,6-tetramethyl piperidide (DA-TMS) mediated epoxide isomerization. Furthermore, the guanine was introduced by modified Mitsunobu reaction.  相似文献

Synthesis of l-3-epi-isofagomine, its homo-, n-butyl derivatives and its bicyclic analogue as potent glycosidase inhibitor has been achieved from readily available d-glucose. Inhibition of some commercially available glycosidases was also carried out with the newly synthesized inhibitors which showed reasonably good inhibitions (9.4–198.2 μM). An enantioselective synthesis of the tetracyclic ABCD ring system (4) of kampanols, novel Ras farnesyltransferase inhibitors from a microorganism, was efficiently achieved for the first time starting from the known trans-decalone derivative 9. The synthetic method involves the following two key steps: (i) a conjugate addition reaction between the α-methylene ketone 6 and the Grignard reagent (7) of the ortho-disubstituted bromobenzene derivative 8 to deliver the coupling product 21 with stereoselectivity at the C9 position and (ii) a phenylselenium-mediated cyclization reaction of the phenol derivative 5 to stereoselectively construct the requisite tetracyclic intermediate 25 possessing the cis-fused connectivity of the B/C rings.  相似文献

The stereocontrolled asymmetric synthesis of alpha-hydroxy-beta-amino acids has been investigated via the Lewis acid-promoted cyanation of (5R,6S)-2-acetoxy-4-(benzyloxycarbonyl)-5,6-diphenyl-2,3,5,6-tetrahydro-4H-1,4-oxazines with trimethylsilyl cyanide. Base-catalyzed hydrolysis of the resulting cyano compounds proceeds with excellent stereoselectivity, providing access to diastereomerically pure oxazine-2-carboxylic acids which were readily converted to each enantiomer of the alpha-hydroxy-beta-amino acids isothreonine and nor-C-statine.  相似文献

The synthesis of the first examples of eight-membered iminoalditols has been achieved from 2,3,4,6-tetra-O-benzyl-d-glucopyranose by way of a ring-closing metathesis. The (2R,3R,4R,5S)-2-hydroxymethyl-azocane-3,4,5-triol (3a), which has the d-gluco configuration for C-2-C-5 and appears to exist predominantly in a boat-chair conformation, is a weak inhibitor of glycosidases.  相似文献

Lewis acid-catalyzed intramolecular reactions of oxocarbenium ions with propargylsilanes are described. A simple synthetic approach has been developed towards one pot synthesis of 2-imidazolines under mild acidic conditions from N-acetyl glucosamine via reductive amination followed by dehydrative cyclization. Synthetic studies were explored in detail with different amines and sugar derivatives. A short and practical procedure for the preparation of C-2 substituted polyhydroxypyrrolidines is described. The C-2 substituent is introduced by a stereoselective addition of a Grignard reagent to a 2,3,5-protected aldofuranose and the cyclization to the pyrrolidine ring system is performed through a bis-mesylation/double nucleophilic displacement sequence. The efficiency of the methodology was demonstrated by its application to the synthesis of HomoDMDP and DMDP.  相似文献

The development of a laboratory and practical synthesis of Suvorexant 1, using intramolecular Mitsunobu cyclization reaction of intermediate 5 as the key reaction, has been reported. Compound 5 was obtained from known chiral ester 2 in three steps, and the key cyclization proceeded smoothly to provide the core seven-membered ring compound 6, which was transformed into 1 by an additional four-step sequence. The procedure described here needs no chiral-HPLC separation, no classical resolution, and no unique enzyme reactions, and offers an alternative practical synthesis of 1.  相似文献

Protected glycals are converted in high yields and selectivities in less than 2 h at low temperatures to 2-deoxy-2-iodoglycosyl acetates using the simple, inexpensive reagent mixture of ammonium iodide, hydrogen peroxide and acetic anhydride/acetic acid in acetonitrile. The corresponding 2-deoxy-2-bromoglycosyl acetates are obtained using ammonium bromide instead of the iodide, although longer reaction times are required and selectivities are inferior.  相似文献
