Ochratoxin A (OTA) is a mycotoxin (potentially carcinogenic secondary metabolite derived from fungal contamination), produced by some Aspergillus and Penicillium strains. Although present and legislated in different food sources in the human diet, the regulation for wine intake is still under discussion. The Office International de la Vigne et du Vin (OIV) recommended maximum levels in wine of 2 μg l−1. Some reports refer to OTA contamination in wines up to 15 μg l−1 and a special incidence in red wines from the southern regions of Europe and the north of Africa, but the majority of the data available are below 1 μg l−1. When working at such low concentrations, the problem of the uncertainty of the results becomes decisive towards the implementation of legal limits. In order to assess the global uncertainty associated with OTA determination in wines and widen the data set and knowledge of the situation in Portugal, 340 wines were analysed (189 Port Wine, 85 Vinho Verde and 66 wines from other regions in the country) by a high performance liquid chromatography (HPLC)-fluorescence detection (FD) method using immunoaffinity columns for clean up. OTA was detected in 69 wines by the method used, but in concentrations below 0.5 μg l−1, except for two which showed levels up to a maximum of 2.1 μg l−1. However, the global uncertainty for OTA is close to 37% for concentrations above 0.5 μg l−1, and therefore, such value can be below or exceed the OIV limit. In the vicinity of the limit of detection, 0.084 μg l−1, the global uncertainty rises exponentially to a maximum of about 70%. This can be an obstacle when discussing safety intake limits. Ethanol and glucose content did not interfere in the clean up of OTA by immunoaffinity columns.  相似文献

Competitive electrochemical enzyme-linked immunosorbent assays based on disposable screen-printed electrodes have been developed for quantitative determination of ochratoxin A (OTA). The assays were carried out using monoclonal antibodies in the direct and indirect format. OTA working range, I50 and detection limits were 0.05-2.5 and 0.1-7.5 μg L−1, 0.35 (±0.04) μg L−1 and 0.9 (±0.1) μg L−1, 60 and 100 μg L−1 in the direct and indirect assay format, respectively. The immunosensor in the direct format was selected for the determination of OTA in wheat. Samples were extracted with aqueous acetonitrile and the extract analyzed directly by the assay without clean-up. The I50 in real samples was 0.2 μg L−1 corresponding to 1.6 μg/kg in the wheat sample with a detection limit of 0.4 μg/kg (calculated as blank signal −3σ). Within- and between-assay variability were less than 5 and 10%, respectively. A good correlation (r = 0.9992) was found by comparative analysis of naturally contaminated wheat samples using this assay and an HPLC/immunoaffinity clean-up method based on the AOAC Official Method 2000.03 for the determination of OTA in barley.  相似文献

Ochratoxin A (OTA) exhibits potent nephrotoxic, carcinogenic and teratogenic effects and its maximum level in wines has been set to 2 μg L−1 by regulation. Consequently, the analytical procedures for OTA determination in wines have to be both very sensitive and reliable. In this paper, we compared two quantification methods: the stable isotope dilution assay (SIDA) and the diastereomeric dilution assay (DIDA). For this purpose, non-natural analogues of OTA were synthesized: the labeled OTA (OTA-d4) as a diastereomeric mixture for the SIDA and one non-natural OTA’s diastereomer (OTA-dia) for the DIDA. To quantify OTA in red grapes, musts or wines, the sample preparation was optimized using immunoaffinity column extraction and the analysis was performed by LC–MS/MS in Multiple Reaction Monitoring mode. A validation procedure in agreement with the International Organization of Vine and Wine recommendations was conducted. It appeared that SIDA quantification exhibited excellent sensitivity (LOD < 1 ng L−1), accuracy (recovery = 98%), repeatability (RSD < 3%) and intermediate reproducibility (RSD < 4%) compared to quantification by DIDA. Indeed, DIDA method did not provide satisfactory results demonstrating that immunoaffinity extraction is exclusively selective for the natural OTA and not for its diastereomer, which therefore cannot be considered as a good internal standard for this particular method.  相似文献

A flow injection hydride manifold was coupled to a 150 W tungsten coil electrothermal atomizer for in situ hydride collection followed by selenium and arsenic determination by ET AAS. Rhodium (200 μg), thermally reduced over the double layer tungsten atomizer, was very efficient at collecting selenium or arsenic hydrides. Prior to analysis, biological samples were digested in closed-vessels microwave digestion system. Prior to the hydride formation, both selenium and arsenic were reduced to valence state (IV) and (III), respectively. The detection limit was 35 ng L−1 for selenium and 110 ng L−1 for arsenic. Sample throughput was 70 h−1 using 30 s of hydride trapping time. Method accuracy was evaluated by analyzing biological-certified reference materials from the National Institute of Standard and Technology (SRM-1577a and SRM-1577b “bovine liver” and RM-8414 “bovine muscle powder”) and from the International Agency for Energy Atomic (A-13 “animal blood”) and one water-certified reference material from the National Institute of Standard and Technology (SRM-1640 trace elements in natural water). By applying a t-test, there was no significant difference at the 95% probability level between the results obtained with the proposed method and those certified values.  相似文献

A spectrophotometric flow injection procedure involving N,N-dimethyl-p-phenylenediamine (DMPD) is applied to the sulfide monitoring of a sugar fermentation by Saccharomyces cerevisiae under laboratory conditions. The gaseous chemical species evolving from the fermentative process, mainly CO2, are trapped allowing a cleaned sample aliquot to be collected and introduced into the flow injection analyzer. Measurement rate, signal repeatability, detection limit and reagent consumption per measurement were estimated as 150 h−1, 0.36% (n = 20), 0.014 mg L−1 S and 120 μg DMPD, respectively. The main characteristics of the monitoring record are discussed. The strategy is worthwhile for selecting yeast strain, increasing the industrial ethanol production and improving the quality of wines.  相似文献

A sensitive reagent-injection flow analysis method for the spectrophotometric determination of nitrate in marine, estuarine and fresh water samples is described. The method is based on the reduction of nitrate in a micro column containing zinc granules at pH 6.5. The nitrite formed is reacted with sulfanilamide and N-(1-naphthyl)ethylene diamine (Griess reagent), and the resulting azo compound is quantified spectrophotometrically at 520 nm. Water samples in the range of 3-700 μg L−1 NO3-N can be processed with a throughput of up to 40 samples per hour, a detection limit of 1.3 μg L−1 and reproducibility of 1.2% RSD (50 μg L−1 NO3-N, n = 10). The proposed method was successfully applied for the determination of nitrate in estuarine waters and the reliability was assessed by the analyses of certified reference materials and recovery experiments. The method is suitable for waters with a wide range of salinities, and was successfully used for more than 3200 underway nitrate measurements aboard SV Pelican1 in the “Two Bays” cruise in January 2010.  相似文献

A pyrimethanil-imprinted polymer (P1) was prepared by iniferter-mediated photografting a mixture of methacrylic acid and ethylene dimethacrylate onto homemade near-monodispersed chloromethylated polydivinylbenzene beads. The chromatographic behaviour of a column packed with these imprinted beads was compared with another column packed with irregular particles obtained by grinding a bulk pyrimethanil-imprinted polymer (P2). The comparison was made using the kinetic model of non-linear chromatography, studying the elution of the template and of two related substances, cyprodinil and mepanipyrim. Extension of the region of linearity, capacity factors for the template and the related substances, column selectivity, binding site heterogeneity, apparent affinity constant (K) and lumped kinetic association (ka) and dissociation rate constant (kd) were studied during a large interval of solute concentration, ranging between 1 and 2000 μg/ml. From the experimental results obtained, in the linearity region of solute concentration column selectivity and binding site heterogeneity remained essentially the same for the two columns, while column capacity (at 20 μg/ml, P1 = 23.1, P2 = 11.5), K (at 20 μg/ml, P1 = 8.3 × 106 M−1, P2 = 2.5 × 106 M−1) and ka (at 20 μg/ml, P1 = 3.5 μM−1 s−1, P2 = 0.47 μM−1 s−1) significantly increased and kd (at 20 μg/ml, P1 = 0.42 s−1, P2 = 0.67 s−1) decreased for the column packed with the imprinted beads. These results are consistent with an influence of the polymerisation method on the morphology of the resulting polymer and not on the molecular recognition properties due to the molecular imprinting process.  相似文献

Liquid–liquid equilibria of systems water (A) + CiEj surfactant (B) + n-alkane (C) have been modeled by a mass-action law model previously developed and so far successfully applied to a series of binary water + CiEj systems and to the ternary system water + C4E1 + n-dodecane. These calculations provide the basis for the presented modeling. The aqueous systems give information about the association constants and the χAB-parameter of the Flory–Huggins theory and the ternary C4E1-system provides universal temperature functions for the χAC- and the χBC-parameter. The three-phase equilibrium for seven ternary CiEj systems (i = 6–12, j = 3–6) has been calculated by fitting one additional parameter for each of both temperature functions to the characteristic “fish-tail” point. The agreement with the experimental data is reasonably well. For systems with very small three-phase areas the results can considerably be improved by individual temperature functions that incorporate the experimental temperature maximum of the “fish” into the parameter fit. Based on the parameters of the system water + C8E4 + n-C8H18 the “fish-shaped” phase diagram of the system water + C8E4 + n-C14H30 was predicted reasonably well.  相似文献

We report the synthesis of a novel bistriazene, 4,4′-bis(3-(4-phenylthiazol-2-yl)triazenyl)biphenyl (BPTTBP), and its highly sensitive color reaction with Hg2+. The new reagent was synthesized in good yield by coupling 2-amino-4-phenylthiazole with 4,4′-biphenyldiamine bisdiazonium salt. Using a blend of surfactants N-cetylpyridinium chloride (CPC) and polyethylene glycol n-octanoic phenyl ether (OP) as a micelle sensitizer, the red colored reagent assembles with Hg2+ in pH 9.8 borate buffer according to a 1:1 stoichiometry, forming a blue oligomeric/polymeric chelating complex with a high apparent stability constant (1.1 × 108 M−1). Whereas the maximum absorption of reagent occurs at 510 nm with an extinct coefficient of 1.35 × 104 M−1 cm−1, the complex absorbs at 611 nm, with an apparent extinct coefficient of 1.04 × 105 M−1 cm−1. Beer's law is obeyed in the range of 0-15 μg/25 mL Hg2+, and Sandell's sensitivity is 1.92 × 10−3 μg/cm2. In the presence of thiourea and Na4P2O7 as masking agents, the method was found free from interferences of foreign ions commonly occurring with mercury. The optimized protocol has been successfully applied to spectrophotometric determination of mercury in waste water samples. The features of the new reagent associated with its special structure were discussed, and an unprecedented “domino effect” was proposed to account for its unique chelating stoichiometry with Hg2+.  相似文献

Present study described a simple, sensitive, and viable method for the determination of bisphenol A, 4-n-nonylphenol and 4-tert-octylphenol in water samples using temperature-controlled ionic liquid dispersive liquid-phase microextraction coupled to high performance liquid chromatography-fluorescence detector. In this experiment, 1-octyl-3-methylimidazolium hexafluorophosphate ([C8MIM][PF6]) was used as the extraction solvent, and bisphenol A, 4-n-nonylphenol and 4-tert-octylphenol were selected as the model analytes. Parameters affecting the extraction efficiency such as the volume of [C8MIM][PF6], dissolving temperature, extraction time, sample pH, centrifuging time and salting-out effect have been investigated in detail. Under the optimized conditions, good linear relationship was found in the concentration range of 1.0-100 μg L−1 for BPA, 1.5-150 μg L−1 for 4-NP, and 3-300 μg L−1 for 4-OP, respectively. Limits of detection (LOD, S/N = 3) were in the range of 0.23-0.48 μg L−1. Intra day and inter day precisions (RSDs, n = 6) were in the range of 4.6-5.5% and 8.5-13.3%, respectively. This method has been also successfully applied to analyze the real water samples at two different spiked concentrations and excellent results were obtained.  相似文献

A trace element preconcentration procedure is described utilizing a minicolumn of yeast (Yamadazyma spartinae) immobilized TiO2 nanoparticles for determination of Cr, Cu, Fe, Mn, Ni and Zn from water samples by inductively coupled plasma atomic emission spectrometry. The elements were quantitatively retained on the column between pH 6 and 8. Elution was made with 5% (v/v) HNO3 solution. Recoveries ranged from 98 ± 2 (Cr) to 100 ± 4 (Zn) for preconcentration of 50 mL multielement solution (50 μg L−1). The column made up of 100 mg sorbent (yeast immobilized TiO2 NP) offers a capacity to preconcentrate up to 500 mL of sample solution to achieve an enrichment factor of 250 with 2 mL of 5% (v/v) HNO3 eluent. The detection limits obtained from preconcentration of 50 mL blank solutions (5%, v/v, HNO3, n = 11) were 0.17, 0.45, 0.25, 0.15, 0.33 and 0.10 μg L−1 for Cr, Cu, Fe, Mn, Ni and Zn, respectively. Relative standard deviation (RSD) for five replicate analyses was better than 5%. The retention of the elements was not affected from up to 500 μg L−1 Na+ and K+ (as chlorides), 100 μg L−1 Ca2+ (as nitrate) and 50 μg L−1 Mg2+ (as sulfate). The method was validated by analysis of freshwater standard reference material (SRM 1643e) and applied to the determination of the elements from tap water and lake water samples.  相似文献

A method based on solid-phase microextraction (SPME) and gas chromatography with mass spectrometry (GC/MS) for the determination of 18 organophosphorus pesticides (OPPs) in textiles is described. Commercially available SPME fibers, 100 μm PDMS and 85 μm PA, were compared and 85 μm PA exhibited better performance to the OPPs. Various parameters affecting SPME, including extraction and desorption time, extraction temperature, salinity and pH, were studied. The optimized conditions were: 35 min extraction at 25 °C, 5% NaSO4 content, pH 7.0, and 3.5 min desorption in GC injector port at 250 °C. The linear ranges of the SPME-GC/MS method were 0.1-500 μg L−1 for most of the OPPs. The limits of detection (LODs) ranged from 0.01 μg L−1 (for bromophos-ethyl) to 55 μg L−1 (for azinphos-methyl) and the RSDs were between 0.66% and 9.22%. The optimized method was then used to analyze 18 OPPs in textile sample, and the determined recoveries were ranged from 76.7% to 126.8%. Moreover, the distribution coefficients of the OPPs between 85 μm PA fiber and simulative sweat solution (Kpa/s) were determined. The determined Kpa/s of the OPPs correlated well with their octanol-water partition coefficients (r = 0.764 and 0.678) and water solubility (= −0.892 and −0.863).  相似文献

A highly selective sequential injection (SI) method for the automated determination of weak-acid-dissociable cyanides is reported. The analytical procedure is based on the on-line reaction of the analyte with ninhydrin in carbonate medium to form a coloured product (λmax = 510 nm). Cyanides are removed from sample matrix by acidification through a gas-diffusion step incorporated in the SI manifold. The effect of instrumental and chemical variables was studied. By adopting an on-line standard addition protocol, the sensitivity of the proposed method was enhanced drastically, without affecting the determination range. The assay was validated in terms of linearity (up to 200 μg L−1), limit of detection (cL = 2.5 μg L−1), limit of quantitation (cQ = 7.5 μg L−1), precision (sr < 2.5% at 100 μg L−1) and selectivity. High tolerance against critical species such as sulfides and thiocyanates was achieved. The applicability of the method was demonstrated by analyzing tap and mineral water samples at levels below the limits established by international E.U. and U.S. organizations. The percent recoveries were satisfactory in all cases, ranging between 94.2 and 103.6%.  相似文献

An analytical methodology based on headspace solid phase microextraction (HS-SPME) combined with comprehensive two-dimensional gas chromatography—time-of-flight mass spectrometry (GC × GC–ToFMS) was developed for the identification and quantification of the toxic contaminant ethyl carbamate (EC) directly in fortified wines. The method performance was assessed for dry/medium dry and sweet/medium sweet model wines, and for quantification purposes, calibration plots were performed for both matrices using the ion extraction chromatography (IEC) mode (m/z 62). Good linearity was obtained with a regression coefficient (r2) higher than 0.981. A good precision was attained (R.S.D. <20%) and low detection limits (LOD) were achieved for dry (4.31 μg/L) and sweet (2.75 μg/L) model wines. The quantification limits (LOQ) and recovery for dry wines were 14.38 μg/L and 88.6%, whereas for sweet wines were 9.16 μg/L and 99.4%, respectively. The higher performance was attainted with sweet model wine, as increasing of glucose content improves the volatile compound in headspace, and a better linearity, recovery and precision were achieved. The analytical methodology was applied to analyse 20 fortified Madeira wines including different types of wine (dry, medium dry, sweet, and medium sweet) obtained from several harvests in Madeira Island (Portugal). The EC levels ranged from 54.1 μg/L (medium dry) to 162.5 μg/L (medium sweet).  相似文献

A colloidal suspension of nanostructured poly(N-butyl benzimidazole)-graphene sheets (PBBIns-Gs) was used to modify a gold electrode to form a three-dimensional PBBIns-Gs/Au electrode that was sensitive to hydrogen peroxide (H2O2) in the presence of acetic acid (AcOH). The positively charged nanostructured poly(N-butyl benzimidazole) (PBBIns) separated the graphene sheets (Gs) and kept them suspended in an aqueous solution. Additionally, graphene sheets (Gs) formed “diaphragms” that intercalated Gs, which separated PBBIns to prevent tight packing and enhanced the surface area. The PBBIns-Gs/Au electrode exhibited superior sensitivity toward H2O2 relative to the PBBIns-modified Au (PBBIns/Au) electrode. Furthermore, a high yield of glucose oxidase (GOD) on the PBBIns-Gs of 52.3 mg GOD per 1 mg PBBIns-Gs was obtained from the electrostatic attraction between the positively charged PBBIns-Gs and negatively charged GOD. The non-destructive immobilization of GOD on the surface of the PBBIns-Gs (GOD-PBBIns-Gs) retained 91.5% and 39.2% of bioactivity, respectively, relative to free GOD for the colloidal suspension of the GOD-PBBIns-Gs and its modified Au (GOD-PBBIns-Gs/Au) electrode. Based on advantages including a negative working potential, high sensitivity toward H2O2, and non-destructive immobilization, the proposed glucose biosensor based on an GOD-PBBIns-Gs/Au electrode exhibited a fast response time (5.6 s), broad detection range (10 μM to 10 mM), high sensitivity (143.5 μA mM−1 cm−2) and selectivity, and excellent stability. Finally, a choline biosensor was developed by dipping a PBBIns-Gs/Au electrode into a choline oxidase (ChOx) solution for enzyme loading. The choline biosensor had a linear range of 0.1 μM to 0.83 mM, sensitivity of 494.9 μA mM−1 cm−2, and detection limit of 0.02 μM. The results of glucose and choline measurement indicate that the PBBIns-Gs/Au electrode provides a useful platform for the development of oxidase-based biosensors.  相似文献

A new sample preparation procedure, termed pH-controlled dispersive liquid–liquid microextraction (pH-DLLME), has been developed for the analysis of ionisable compounds in highly complex matrices. This DLLME mode, intended to improve the selectivity and to expand the application range of DLLME, is based on two successive DLLMEs conducted at opposite pH values. pH-DLLME was applied to determination of ochratoxin A (OTA) in cereals. The hydrophobic matrix interferences in the raw methanol extract (disperser, 1 mL) were removed by a first DLLME (I DLLME) performed at pH 8 to reduce the solubility of OTA in the extractant (CCl4, 400 μL). The pH of the aqueous phase was then adjusted to 2, and the analyte was extracted and concentrated by a second DLLME (extractant, 150 μL C2H4Br2). The main factors influencing the efficiency of pH-DLLME including type and volume of I DLLME extractant, as well as the parameters affecting the OTA extraction by II DLLME, were studied in detail. Under optimum conditions, the method has detection and quantification limits of 0.019 and 0.062 μg kg−1, respectively, with OTA recoveries in the range of 81.2–90.1% (n = 3). The accuracy of the analytical procedure, evaluated with a reference material (cereal naturally contaminated with OTA), is acceptable (accuracy of 85.6% ± 1.7, n = 5).  相似文献

A new automated spectrophotometric method for the determination of total sulfite in white and red wines is reported. The assay is based on the reaction of o-phthalaldehyde (OPA) and ammonium chloride with the analyte in basic medium under SI conditions. Upon on-line alkalization with NaOH, a blue product is formed having an absorption maximum at 630 nm. The parameters affecting the reaction - temperature, pH, ionic strength, amount concentration and volume of OPA, amount concentration of ammonium chloride, flow rate and reaction coil length - and the gas-diffusion process - sample and HCl volumes, length of mixing coil, donor flow rate - were studied. The proposed method was validated in terms of linearity (1-40 mg L−1, r = 0.9997), limit of detection (cL = 0.3 mg L−1) and quantitation (cQ = 1.0 mg L−1), precision (sr = 2.2% at 20 mg L−1 sulfite, n = 12) and selectivity. The applicability of the analytical procedure was evaluated by analyzing white and red wine samples, while the accuracy as expressed by recovery experiments ranged between 96% and 106%.  相似文献

A facile, sensitive and universal method was established for analysis of biogenic amines using micellar electrokinetic chromatography coupled with chemiluminescent (CL) detection. It was found that diperiodatocuprate (III) (K5[Cu(HIO6)2], DPC), a transition metal chelate at unstable high oxidation state, could effectively enhance the reaction between luminol-type compound and hydrogen peroxide, to produce very strong CL signal. In addition, triethylamine was found to be able to effectively improve the yield of the derivatization reaction between biogenic amines and a luminol-type derivatization reagent, N-(4-aminobutyl)-N-ethylisoluminol (ABEI). Based on these facts, three biogenic amines were pre-column derivatized with ABEI, and post-column detected using high sensitive luminol-hydrogen peroxide-DPC CL system. Since the background was quite low, and the signal was quite strong, a considerable improved sensitivity was obtained. The presented method had been successfully applied to simultaneously analyze glycine, proline and phenylalanine with the detection limits (S/N = 3) of 0.030 μmol L−1, 0.23 μmol L−1 and 0.21 μmol L−1, respectively. To evaluate its potential application value, glycine in saliva and urine samples was detected using this method, and satisfied results were obtained. This approach can be further extended to detection of many other compounds such as peptides and drugs by using luminol-type derivatization reagent.  相似文献

The effect of thickness on the glass transition dynamics in ultra-thin polystyrene (PS) films (4 nm < L < 60 nm) was studied by thin film ac-calorimetry, dielectric spectroscopy (DRS) and capacitive dilatometry (CD). In all PS-films, a prominent α-process was found in both the ac-calorimetric and dielectric response, indicating the existence of cooperative bulk dynamics even in films as thin as 4 nm. Glass transition temperatures (Tg) were obtained from ac-calorimetric data at 40 Hz and from capacitive dilatometry, and reveal a surprising, marginal thickness dependence Tg(L). These results, which confirm recent data by Efremov et al. [Phys. Rev. Lett. 91 (2003)] but oppose many previous observations, is rationalized by differences in film annealing conditions together with the fact that our techniques probe exclusively cooperative dynamics (ac-calorimetry) or allow the effective separation of surface and “bulk”-type mobility (CD). Two other observations, a significant reduction in cp towards lower film thickness and the decrease in the contrast of the dilatometric glass transition, support the idea of a layer-like mobility profile consisting of both cooperative “bulk” dynamics and non-cooperative surface mobility.  相似文献

We report a new device for the estimation of the content of chlorophyll a pigment in water samples as an indicator of water quality. The extraction of the pigment from water was also optimized. 10 mL of water was filtered through a nylon filter (45 μm pore size and 13 mm of diameter), after the chlorophylls were dissolved by immersing the filter in 1 mL of a low non-hazardous solvent as ethanol. An in-valve in-tube SPME device coupled to capillary liquid chromatography with diode array detection was designed. A capillary column of 70 cm in length (0.32 mm i.d. coated with 5% diphenyl-95% polydimethylsiloxane, 3 μm coating thickness) was used as the loop of the injection valve for preconcentration and a Zorbax SB C18 (SiO2-based) 150 mm × 0.5 mm i.d., 5 μm column (Agilent) was used as analytical column. The achieved detection limit was 0.05 μg L−1 and the working range of concentrations was 0.1-1 μg L−1. % RSD values between 2 and 11 were obtained. Chlorophyll a in several water matrices was determined with good results in presence of other pigments such as chlorophyll b, pheophytin a and pheophytin b.  相似文献
