The Hpn-like protein (Hpnl), a histidine- and glutamine-rich protein, is critical for Helicobacter pylori colonization in human gastric muscosa. In this study, the thermodynamic properties of Ni(II), Cu(II), Co(II), and Zn(II) toward Hpnl were studied by isothermal titration calorimetry (ITC). We found that Hpnl exhibits two independent binding sites for Ni(II) as opposed to one site for Cu(II), Co(II), and Zn(II). Protease digestion and chemical denaturation analysis further revealed that Ni(II) confers a higher stability upon Hpnl than other divalent metal ions. The potential Ni(II) binding sites are localized in the His-rich domain of Hpnl as confirmed by mutagenesis in combination with modification of histidine residues of the protein. We also demonstrated that the single mutants (H29A and H31A) and tetrameric mutant (H29-32A) cut nearly half of the binding capacity of Hpnl towards nickel ions, whereas other histidine residues (His30, 32, 38, 39, 40, and 41) are nonessential for nickel coordination. Escherichia coli cells that harbored H29A, H31A, and H29-32A mutant genes exhibited less tolerance toward high concentrations of extracellular nickel ions than those with the wild-type gene. Our combined data indicated that the conserved histidine residues, His29 and His31 in the His-rich domain of Hpnl, are critical for nickel binding, and such a binding is important for Hpnl protein to fulfill its biological functions.  相似文献

Pyrrolidinedithiocarbamate (PDTC) chelates of Zn(II), Cu(II), Ni(II), Co(III), Fe(III), Mn(II), Cr(III), and VO(II) were analysed by capillary GC on a DB-1701 column (30 m x 0.25 mm id) with flame ionisation detection (FID). Linear calibrations were attained within "1-30 microg/mL" for Ni(II), Fe(III), Mn(II), Cr(III), Cu(II), and VO(II), and within "2-50 microg/mL" for Co(III) and Zn(II). The limits of detection were in the "150-500 ng/mL" range, corresponding to 15-50 pg amounts reaching the FID system. The optimised method was applied to the determination of Cu(II) and Ni(II) in coins, and that of Zn(II), Cu(II), Ni(II), Fe(III), Mn(II), Cr(III), and VO(II) in pharmaceutical preparations with relative standard deviations within 1.1-5.2%. The results obtained are in good agreement with sewage water samples and the declared values for the pharmaceutical formulations, or with the results of AAS of metal contents in coins, pharmaceutical preparations, and sewage water samples.  相似文献

Immobilized metal affinity chromatography (IMAC) was investigated as a method of recovery for green fluorescent protein (GFPuv). It was found that in the absence of genetic modification to enhance metal affinity, GFPuv displayed strong metal affinity to Cu(II) and Ni(II), and weak or negligible affinity to Zn(II) and Co(II). Changes in the mobile phase NaCl concentration during Ni(II)-IMAC strongly affected purity and yield of GFPuv, with fine resolution under higher NaCl concentrations. Finally, IMAC via Cu(II) and Zn(II) with intervening diafiltration was used to recover GFPuv with high yield and purity.  相似文献

A phenanthroline-based macrocycle 1 has been newly developed which has two chemically equivalent metal chelating sites within the spatially restricted cavity for dinuclear metal arrangement. The macrocycle 1 reacts with Zn(CF(3)CO(2))(2) or ZnCl(2) to form homodinuclear Zn(II)-complexes. A single-crystal X-ray structural analysis of the resulting Zn(2)1(CF(3)CO(2))(4) determined the complex structure in which two Zn(II) ions are bound by two phenanthroline sites and two CF(3)CO(2)(-) ions bind to each Zn(II) ion in a tetrahedral geometry. Similarly, a homodinuclear Cu(I)-macrocycle was formed from 1 and Cu(CH(3)CN)(4)BF(4). Notably, from 1 and an equimolar mixture of Cu(CH(3)CN)(4)BF(4) and Zn(CF(3)CO(2))(2), a heterodinuclear Cu(I)-Zn(II)-macrocycle was exclusively formed in high yield (>90%) because of the relatively low stability of the dinuclear Cu(I)-macrocycle. A heterodinuclear Ag(I)-Zn(II)-macrocycle was similarly formed with fairly high selectivity from a mixture of Ag(I) and Zn(II) ions. Such selective heterodinuclear metal arrangement was not observed with other combinations of M-Zn(II) (M = Li(I), Mg(II), Pd(II), Hg(II), La(III), and Tb(III)).  相似文献

Salicylaldoxime-immobilized silica gel was characterized and used as a potential sorbent for heavy metal ions, viz. Cu(II), Ni(II), Co(II), and Zn(II). The experimental conditions were optimized both in batch and column processes to achieve the maximum efficiency. Kinetic and thermodynamic parameters as well as isotherm constants were evaluated to test the feasibility of the process. The role of various metal ions and different anions were tested in order to monitor the process in case of real samples. The alkali metal, alkaline earth metal, and ammonium salts do not have any effect on the said process. This differential behavior can be effectively used for the decontamination of alkali metal, alkaline earth metal, and ammonium salts from Cu(II), Ni(II), Co(II), and Zn(II) ions via solid phase extraction following AAS measurement. The purification of the salts was confirmed by voltammetric experiment.  相似文献

The linear (absorption and emission) and nonlinear optical (NLO) properties of a series of D(3) [(Fe(II), Ru(II), Ni(II), Cu(II), Zn(II)] octupolar metal complexes featuring the 4,4'-bis[(dibutylamino)styryl]-2,2'-bipyridine ligand are reported. Zinc(II), nickel(II), and copper(II) complexes exhibit similar absorption spectra in the visible region (lambda(ILCT) = 474-476 nm) which are assigned to intraligand charge-transfer (ILCT) bands. The quadratic and cubic NLO properties are strongly influenced by the nature of the metallic center. Harmonic light scattering studies at lambda = 1.91 microm reveal that these chromophores display large first hyperpolarizabilities beta(1.91) in the range of (211-340) x 10(-30) esu; replacing the Zn(II) metal ion by Ni(II) or Cu(II) results in a decrease of the static beta(0) coefficient by a factor of 1.5-1.6. Z-scan measurements at 765 and 965 nm reveal relatively large two-photon absorption cross-sections [650 < sigma(2) < 2200 GM], showing that both beta and sigma(2) values can be tuned by simple modification of the metal ion.  相似文献

Several mixed ligand Ni(II), Cu(II) and Zn(II) complexes of 2-amino-3-hydroxypyridine (AHP) and imidazoles viz., imidazole (him), benzimidazole (bim), histamine (hist) and L-histidine (his) have been synthesized and characterized by elemental and spectral (vibrational, electronic, 1H NMR and EPR) data as well as by magnetic moment values. On the basis of elemental analysis and molar conductance values, all the complexes can be formulated as [MAB]Cl except histidine complexes as MAB. Thermogravimetric studies reveal the presence of coordinated water molecules in most of the complexes. From the magnetic measurements and electronic spectral data, octahedral structure was proposed for Ni(II) and Cu(II)-AHP-his, tetrahedral for Cu(II)-AHP-him/bim/hist, but square planar for the Cu(II)-AHP complex. The g∥/A∥ calculated supports tetrahedral environment around the Cu(II) in Cu(II)-AHP-him/bim/hist and distorted octahedral for Cu(II)-AHP-his complexes. The morphology of the reported metal complexes was investigated by scanning electron micrographs (SEM). The potentiometric study has been performed in aqueous solution at 37 °C and I=0.15 mol dm(-3) NaClO4. MABH, MAB and MAB2 species has been identified in the present systems. Proton dissociation constants of AHP and stability constants of metal complexes were determined using MINIQUAD-75. The most probable structure of the mixed ligand species is discussed based upon their stability constants. The in vitro biological activity of the complexes was tested against the Gram positive and Gram negative bacteria, fungus and yeast. The oxidative DNA cleavage studies of the complexes were performed using gel electrophoresis method. Cu(II) complexes have been found to promote DNA cleavage in presence of biological reductant such as ascorbate and oxidant like hydrogen peroxide.  相似文献

Mn(II), Co(II), Ni(II), Cu(II) and Zn(II) complexes of salicylidene-N-cyano-acetohydrazone H2L1 and 2-hydroxy-l-naphthylidene-N-cyanoacetohydrazone H2L2 have been prepared in ethanolic solution and characterized by analytical, spectral, magnetic susceptibility, molar conductivity and TGA measurements. The analytical data show that all the complexes derived from H2L1 and H2L2 are formed in molar ratios 1M:2L, except the complexes of Mn(II), Co(II) and Cu(II) acetates of H2L2 and the complexes of Mn(II), Co(II) and Ni(II) acetates and CuCl2 of H2L1 are formed in 1:1 molar ratios. The conductance data show that all metal complexes are non-electrolytes. Electronic absorption spectra and magnetic susceptibility measurements proved that the prepared complexes have octahedral configuration except [Co(HL2)OAc] which has tetrahedral structure. The ligand field parameters were calculated for the Co(II) and Ni(II) complexes and the data show that the covalent character of the metal ligand sigma-bond is low. The ESR parameters of the Cu(II) complexes at room temperature were calculated. Thermal TGA for some solid complexes are reported.  相似文献

The interaction of Cu(II), Ni(II), Zn(II), Mn(II), Co(II), Mg and Ca ions with cytidine and the biologically important ligands histidine, histamine, glycine and oxalic acid in a 1:1:1 ratio have been investigated by potentiometric equilibrium measurements at 35°C and 0.10 M (KNO3) ionic strength. These investigations were undertaken to assess the influence of the secondary ligands on the structure and stability of the 1:1 metal—cytidine system. The stability of the binary and ternary complexes has been compared. The enhanced stability of the ternary complexes was measured in terms of Δ log K, the difference between the ternary and binary complexes. The involvement of various donor sites of histidine in metal binding was specially discussed. A general conclusion drawn from this investigation is that aromatic ligands formed more stable complexes than aliphatic ligands. This is attributed to the stacking phenomenon.  相似文献

Sorption properties of ammonium humates with respect to Fe(III), Cu(II), Al(III), Ni(II), Pb(II), and Zn(II) are studied. The effect of the metal ion concentration on the ammonium humate consumption is examined and the corresponding dependences are analyzed using regression equations. Co(II), Ni(II), Cu(II), Zn(II) and Cd(II) 2,5-dichlorobenzoates were prepared and their compositions and solubilities in water at 295 K were determined. The IR spectra and X-ray diffractograms of the obtained complexes were recorded. The complexes of Co(II), Ni(II), Cu(II), Zn(II) and Cd(II) were obtained as solids with a 1:2 molar ratio of metal to organic ligand and different degrees of hydration. When heated at a heating rate of 10 K min-1, the hydrated complexes lose some (Co, Zn) or all (Ni, Cu, Cd) of the crystallization water molecules and then decompose to oxide MO (Co, Ni) or gaseous products (Cu, Zn, Cd). When heated at a heating rate of 5 K min-1, the complexes of Ni(II) and Cu(II) lose some (Ni) or all (Cu) of the crystallization water molecules and then decompose directly to MO. The determination of metal ions by capillary isotachophoresis and the complexation equilibria between metal ions and polyaminopolycarboxylic acids has been investigated. A seven-component mixture of metal ions can be separated in 45% v/v acetone-water medium when EDTA or DCTA is used as the terminating ion. Linear calibration graphs are obtained for a standard mixture of Mn(+), Cu(2+), Zn(2+), Cd(2+), Pb(2+) and Fe(3+) in the range 0.5-5.0 nmole, with relative standard deviations of 1.0% or better. The effective mobilities of the Mn(II), Co(II), Ni(II), Cu(II) and Zn(II) complexes increase in parallel with the stability constants, except for the Cu(II) complexes. It is concluded that the abnormal behaviour of the Cu(II) complexes may be attributed to a difference in steric configuration.  相似文献

The effects of metal ions on the reduction of nitric oxide (NO) with a designed heme copper center in myoglobin (F43H/L29H sperm whale Mb, CuBMb) were investigated under reducing anaerobic conditions using UV-vis and EPR spectroscopic techniques as well as GC/MS. In the presence of Cu(I), catalytic reduction of NO to N2O by CuBMb was observed with turnover number of 2 mol NO.mol CuBMb-1.min-1, close to 3 mol NO.mol enzyme-1.min-1 reported for the ba3 oxidases from T. thermophilus. Formation of a His-heme-NO species was detected by UV-vis and EPR spectroscopy. In comparison to the EPR spectra of ferrous-CuBMb-NO in the absence of metal ions, the EPR spectra of ferrous-CuBMb-NO in the presence of Cu(I) showed less-resolved hyperfine splitting from the proximal histidine, probably due to weakening of the proximal His-heme bond. In the presence of Zn(II), formation of a five-coordinate ferrous-CuBMb-NO species, resulting from cleavage of the proximal heme Fe-His bond, was shown by UV-vis and EPR spectroscopic studies. The reduction of NO to N2O was not observed in the presence of Zn(II). Control experiments using wild-type myoglobin indicated no reduction of NO in the presence of either Cu(I) or Zn(II). These results suggest that both the identity and the oxidation state of the metal ion in the CuB center are important for NO reduction. A redox-active metal ion is required to deliver electrons, and a higher oxidation state is preferred to weaken the heme iron-proximal histidine toward a five-coordinate key intermediate in NO reduction.  相似文献

Thermodynamic stability constants of complexes of Mn(II), Ni(II), Zn(II) and Cu(II) with five closely related N-arylhydroxamic acids have been determined at 25 +/- 0.1 degrees in 50% v v aqueous dioxan medium. The stabilities of the complexes mostly follow the order of the basicity of the ligands and the electron affinities of the metal ions as measured by their second ionization potential.  相似文献

A metallic silver electrode provides a potentiometric response to Ag(I), Hg(II), Fe(III), Cu(II), Zn(II), Co(II) and Ni(II) ions, and large changes in the electrode potential of up to 400 mV are observed when these ions are injected into a 0.29 mM soulution of cyanide ion. Injection 10-μl aliquots into a reagent stream with a cyanide concentration of 0.1 mM is shown to give the expected sigmoidal-shaped response curve for peak height as a function of metal ion concentration. Sub-nanomole quantities of metal ions are detectable with a peak height of ca. 20 mV, depending on the cyanide concentration.  相似文献

Four silica gel-immobilized new metal chelate Schiff(,)s bases were synthesized (I-IV). Silica gel chemically bonded diethylenetriamine mono-naphthaldehyde and mono-salicyaldehyde Schiff's bases (phases I and III) were produced via the interaction of silica gel-modified diethylenetriamine with naphthaldehyde and salicylaldehyde, respectively. However, phases II and IV arose through the interaction of bis-naphthaldehyde and bis-salicylaldehyde Schiff(,)s bases of diethylenetriamine with 3-chloropropyltrimethoxysilane modified silica gel. The characterization of such new phases, their capabilities towards selective extraction or separation of Fe(III), Ni(II), Cu(II), Zn(II), Cd(II) and Pb(II) ions were studied and evaluated by both batch and column techniques as a function of pH and time of contact. Phases III and I showed high performance towards Cu(II) extraction, where their Cu(II) sorption determined to be 0.957 and 0.940 mmol g(-1), respectively. However, for phases IV and II, the great affinity was devoted to Fe(III) extraction followed by Cu(II) ions. The reactivity of metal ion sorption was discussed in the light of effects of bulkiness as well as orientation of immobilized chelate on sorbent reactivity. Donor sites of phases III and I (diethylenetriamine and azomethene nitrogens along with phenolic hydroxyl group oxygen) are fully active, whereas phases IV and II are partially active with only participation of oxygen and azomethene nitrogen.
