Abstract

A liquid chromatography‐tandem triple‐quadrupole mass spectrometry assay to quantify palitaxel in rat tissue homogenates containing paclitaxel nanoliposome (PTX‐NLP) modified by PEO-PPO-PEO triblock copolymers was developed and validated. Liquid–liquid extraction with tert‐butyl methyl ether was used for preparation of tissue samples and docetaxel was used as the internal standard. Paclitaxel and docetaxel were separated on a 200 mm×4.6 mm×5 µm C18 column and quantified using a triple‐quadrupole mass spectrometer operating in positive ion electrospray selective reaction monitoring mode (ESI+‐SRM) with a total run time of 6.0 min. The peak area of the m/z 876.3→307.9 transition of paclitaxel is measured vs. that of the m/z 830.3→549.1 transition of docetaxel to generate the standard curves. The standard curves were linear over the concentration range of 0.2–2000 ng/mL for different tissues. The method had high extraction recovery (>90%) and accuracy (>90%) with the intraday and inter‐day precision <15%. Frozen stability, freeze‐thaw stability, extracted stability, and solution stability under ambient temperature were examined, which indicated the tissue samples should be extracted within 5 days and avoid being frozen and thawed repeatedly over 5 times, extracted samples after evaporation could be stored at ?20°C for 20 days without drug degradation, also, no degradation was observed after solution samples were left out at ambient temperature for 24 h. This assay was used to support an in vivo biodistribution study of paclitaxel nanoliposome modified by PEO-PPO-PEO triblock copolymers in rats.  相似文献

Docetaxel, frequently used for the treatment of breast cancer, is mainly metabolized via hepatic cytochrome P450 (CYP) 3A in humans and is also a substrate of P‐glycoprotein (P‐gp). Wogonin has been shown to be able to modulate the activities of CYPs and P‐gp, and it could serve as an adjuvant chemotherapeutic agent. However, the impacts of co‐administration of wogonin and docetaxel on their pharmacokinetics have not been studied because of a lack of an analytical method for their simultaneous measurement. In the present study, we established an HPLC–MS/MS method for simultaneous measurement of wogonin and docetaxel in rat plasma, and it was then utilized to explore the pharmacokinetics of wogonin and the herb–drug interactions between wogonin and docetaxel after their combined administration in rats with mammary tumors. The rats received 10, 20 and 40 mg/kg wogonin via oral administration, with or without docetaxel intravenously administered at 10 mg/kg, and the plasma concentrations of wogonin and docetaxel were measured using the established and validated HPLC–MS/MS method. The Cmax and AUC0–t of wogonin were proportionally increased in the dose range from 10 to 40 mg/kg, suggesting a linear pharmacokinetics of wogonin. Moreover, the Cmax and AUC0–t of docetaxel and the AUC0–t of wogonin were increased after co‐administration (p < 0.05), indicating increased in vivo exposures of both wogonin and docetaxel, which might lead to an increase in not only therapeutic but also toxic effects. Thus the alterations of pharmacokinetics should be taken into consideration when wogonin and docetaxel are co‐administered.  相似文献

Side chains of docetaxel and its isomer were obtained through Staudinger cycloaddition and catalytic hydrogenation of chlorophenyl intermediates, using chlorobenzaldehyde as starting material. Syntheses of three novel chiral azetidinone derivatives through the Staudinger cycloaddition reaction of chlorophenyl chi‐ral amine Schiff base with different substituted positions were described and their ring‐opening reaction under the catalysis of Pd/MgCO3 or Pd/C to afford side chains of docetaxel and its isomer in high yields was investigated. Finally, docetaxel and its isomer were obtained. Single crystal of (3S,4R)‐3‐hydroxy‐N‐[(S)‐(l‐phenyl)ethyl]‐4 ‐(2′‐chlorophenyl) ‐2‐azetidinone ( 4c ) was obtained, the configuration of which was determined by X‐ray diffraction. Because of the mild cyclization reaction condition and convenient asymmetric resolution operation when p‐chlorobenzaldehyde was employed instead of benzaldehyde, the yield of cyclization and hydrogenation increased dramatically and the total yield of docetaxel was higher than the result in literature. When o‐chlorobenzaldehyde was employed instead of benzaldehyde an isomer of docetaxel was obtained by the same way.  相似文献

In this study, a sensitive, simple and reliable method for the quantification of docetaxel in rat plasma was developed and validated using liquid chromatography–tandem mass spectrometry (LC‐MS/MS). The plasma samples were prepared by protein precipitation, and paclitaxel was used as an internal standard (IS). Chromatographic separation was achieved using a Gemini C18 column (2.0 × 150 mm, 5 µm) with a mobile phase consisting of 0.1% formic acid–acetonitrile (30:70, v/v). The precursor–product ion pairs used for multiple reaction monitoring were m/z 808.5 → 527.5 (docetaxel) and m/z 854.2 → 286.5 (IS, paclitaxel). A calibration curve for docetaxel was constructed over the range 1–1000 ng/mL. The developed method was specific, precise and accurate, and no matrix effect was observed. The validated method was applied in a comparative pharmacokinetic study in which two docetaxel formulations, SID530, a new parenteral formulation of docetaxel with hydroxypropyl‐β‐cyclodextrin (HP‐β‐CD), and Taxotere, were administered to rats at a dose of 5 mg/kg. For SID530 and Taxotere, the mean C0 values were 1494 and 1818 ng/mL, respectively, and the AUClast values were 837 and 755 h ng/mL, respectively. These two formulations did not show any statistical differences with regard to the pharmacokinetic parameters, thus establishing that the SID530 and Taxotere products are pharmacokinetically comparable in male rats. Abstract

A rapid, sensitive, and specific liquid chromatography–tandem mass spectrometric (HPLC-MS/MS) method has been developed for quantification of ferulic acid in rat plasma. The analyte and docetaxel (internal standard) were extracted from plasma samples with diethyl ether and analyzed on a C18 column. The chromatographic separation was achieved within 3.5 min by using acetonitrile-water as the mobile phase and the flow rate was 0.2 mL · min?1. The method was linear within the range of 0.5 ? 800 ng · mL?1. The lower limit of quantification (LLOQ) was 0.5 ng · mL?1. Finally, the method is successfully applied for the pharmacokinetic study of ferulic acid in rats following intravenous administration.  相似文献

7‐epi‐Taxane has been achieved efficiently in gram scale from natural taxane via inversion of the 7‐hydroxyl group simply using Ag2O as catalyst and DMF as solvent. The catalyst could be quantitatively recovered by filtration without loss of catalytic activity. This condition is also applicable to the direct epimerization of taxane derivatives (e.g., docetaxel and paclitaxel) to 7‐epi‐taxane derivatives (e.g., 7‐epi‐docetaxel and 7‐epi‐paclitaxel). Furthermore, 33 ester derivatives of 7‐epi‐taxane with different amino acid moieties at the position of C‐13 were successfully synthesized via esterification without protecting C‐7‐OH. Bioassay results revealed that compounds 13 and 18 have good selectivity against prostatic cancer cell line DU145, with IC50 value as low as 15.9 nmol/L for 18 .  相似文献

Combination therapy is considered a viable strategy to overcome the resistance to chemotherapeutics. Survivin as a member of the inhibitor of apoptosis protein (IAP) family, which is involved in resistance to various drugs. We investigated the role of combination therapy in downregulating survivin and increasing drug’s efficacy in MDA-MB-231 cells. MTT assay and DAPI staining were applied to study the anti-proliferative activity and apoptosis response of the agents. Real-time RT-PCR and Western blot analysis were applied to study survivin mRNA and protein. Our findings showed that combined treatment of cells with docetaxel and vinblastine reduces survivin expression and consequently decreases the IC50 value of docetaxel from 70 to 5 nM (p?p?相似文献

Prostate cancer is one of the most common cancers among men in the United States. It is also a major leading cause of cancer death among men of all races. In order to treat prostate cancer, drug combinations are often applied. Drug combinations target at different pathways of cells can potentially lead to higher efficacy and lower toxicity due to drug synergy. In this paper, we sequentially applied a two-level design and a follow-up orthogonal array composite design (OACD) to investigate combinations of five anti-cancer drugs, namely, doxorubicin, docetaxel, paclitaxel, cis-dichlorodiamine platinum and dihydroartemisinin. Our initial screening using a two-level full factorial design identified doxorubicin and docetaxel as the most significant drugs. A follow-up experiment with an OACD revealed more complicated drug interactions among these 5 anti-cancer drugs. Quadratic effects of doxorubicin and paclitaxel appeared to be significant. A further investigation on contour plots of all the two-drug pairs indicated that combination of doxorubicin and docetaxel are the most effective companion, while the combination of cis-dichlorodiamine platinum and dihydroartemisinin showed unknown antagonistic effects which diminished the individual drug anti-cancer efficacy. These observations have significant practical implications in the understanding of anti-cancer drug mechanism that can facilitate clinical practice of better drug combinations.  相似文献

Two new kinds of docetaxel compound, with a mercapto group instead of the hydroxyl on the C13 side chain (both syn and anti), via the 7,10-di-(2,2,2-trichloroethyloxycarbonyl)-10-deacetylbaccatin III route, were synthesized. A reduction-sensitive stearic acid modified-Bletilla striata polysaccharide amphiphilic copolymer is synthesized. Host–guest complexes of Docetaxel 1, an anti-cancer drug have been isolated and crystal structures are described. Docetaxel crystallized in the 1:1 molar ratio with n-butanol, dimethylformamide (DMF) and acetonitrile (ACN) during crystallization from the respective solvents. In all the three complexes (1 · n-butanol, 1 · DMF and 1 · ACN), docetaxel formed a host framework through hydrogen bonds and the guest solvent molecules occupied the channels. The host is hydrogen bonded to the guest molecules through hydroxyl moieties. Interestingly, 1 · n-butanol, 1 · DMF and a literature 1 · CH3OH · H2O (1:1:1) host–guest complexes are isomorphs. Further, 1 · ACN complex unit cell parameters are similar (same space group) to the marketed docetaxel trihydrate polymorph (form A).  相似文献

An LC–MS/MS multi-method for the simultaneous determination of the structurally different and frequently used cytostatic drugs 5-fluorouracil, gemcitabine, methotrexate, cyclophosphamide, ifosfamide, etoposide, docetaxel and paclitaxel was developed and validated. In order to perform repeated ambient monitoring in 130 German pharmacies all steps of the monitoring procedure such as sample collection, transport, storage, sample preparation and HPLC–MS/MS analysis have been adapted and optimised. Thus sensitivity and reliability as well as sample throughput were increased. The final method consists of wipe sampling from 900?cm2 surfaces and extraction of the tissues with an aqueous pH 3 solution. The limits of quantification range from 3.7 to 37?pg?cm?2. Validation showed that sampling via the individual pharmacy personnel does not affect the overall results. Recovery rates below 70% were observed on rough surfaces for the taxanes docetaxel and paclitaxel. Likewise, neither the storage nor the shipping conditions affected the results significantly.  相似文献

A novel, rapid and sensitive isocratic liquid chromatography/tandem mass spectrometry (LC/MS/MS) method was developed for quantification of docetaxel in human plasma with paclitaxel as internal standard. The high sensitivity and specificity of MS/MS detection enabled the use of a small volume of plasma (0.05 mL) and a simple liquid-liquid extraction procedure. Furthermore, a very short run-time (3 min) fulfilled the need for monitoring plasma levels of docetaxel from large-scale clinical studies. The calibration curve for docetaxel was linear over the range 5-1000 ng/mL with coefficients of correlation >0.999 using only 0.05 mL plasma. The intra- and inter-day precisions (CV) of analysis were <7%, and accuracy ranged from 96 to 110%. The applicability of the method was demonstrated in a pharmacokinetic study of a 1-h infusion of docetaxel with dosages of 75 mg/m(2). Possible conjugated metabolites of docetaxel were not detected in patients' samples.  相似文献

Twenty-seven compounds including nine triterpenoids (19), eight sterols (1017), two ribonucleotides (18, 19), four phenols (2023), three glycosides (2426), and one furan (27) were isolated from the fruiting bodies of Trametes versicolor (L.) Lloyd. This study is the first confirmation of the presence of the 11 compounds (3, 5, 6, 8, 18, 20, 21, 2325, and 27) isolated from the Polyporaceae family, with six of these (2 and 1216) from the genus Trametes. Compounds 3, 4, 10, 11, 16 and 17 were found to significantly inhibit the production of NO, TNF-α and IL-6 in a dose-dependent manner.  相似文献

The title compound, C29H36O10·2C2H6OS, forms a monoclinic crystal containing two hydrogen-bonded di­methyl sulfoxide mol­ecules per 10-de­acetyl baccatin III. The lattice is further stabilized by two additional hydrogen bonds and a dipole–dipole interaction. A comparison of 10-de­acetyl baccatin III with the structurally similar docetaxel reveals differences primarily in the benzoyl moiety, while comparisons to baccatin III identify relative differences principally in the cyclo­octane- and cyclo­hexane-ring conformations.  相似文献

Abstract

Alkylation of 4-anilino-5-phenyl-4H-1,2,4-triazole-3-thiol (1) with some halo compounds yielded the corresponding sulfides 2af. Some sulfides 2e,f were cyclized to give triazolothiadiazines 3 and 4. Triazolothiadiazoles 5 and 6 were prepared through the reaction of compound 1 with carbon disulfide or ethyl orthoformate, respectively. Treatment of compound 1 with ethyl chloroformate or phenyl isothiocyanate yielded triazolo-thiadiazole and triazole 9 and 10, respectively. Reaction of compound 1 with Lawesson's reagent gave triazolothiadiazaphosphole derivative 11. Also, compound 1 underwent cyclocondensation reactions with some bidentate reagents to give triazolothiazines 4, 12, and 13. Triazolo-thiazepines and triaziepine 1416 were synthesized via the reaction of compound 1 with β-ketoesters or ethyl cyanoacetate. Tricyclic systems 19 and 20 were prepared through the reaction of compound 4 with the appropriate reagent. Some synthesized compounds were tested for antibacterial activity.

The addition of benzenethiol to p-chlorophenylphenylacetylene results in the formation of a mixture of two pairs of diastereomeric (E)- and (Z)-1-p-chlorophenyl-2-phenyl-1-phenylthioethylenes (1 and 2) and (E)- and (Z)-1-p-chlorophenyl-2-phenyl-2-phenylthioethylenes (3 and 4). The configurations of these compounds have been established by 1 H NMR studies, by their preparation from benzyl p-chlorophenyl ketone and p-chloro-benzylphenyl ketone, and by the oxidation of the thioethylenes 1, 2, 3, and 4 to the corresponding sulphonylethylenes 5, 6, 7, and 8, respectively.  相似文献

The refluxing of 3-amino-6,8-dibromo-2-thioxo-2,3-dihydro-1H-quinazolin-4-one (5) with ethyl chloroformate and/or ethyl chloroacetate afforded compounds 6 and 7 . The reaction of 5 with ethyl bromobutyrate, chloroacetyl chloride, phenacyl chloride, and phenyl isocyanate yielded compounds 8 , 9 , 11 , and 12 . The coupling of 5 with (2,3,4,6-tetra-O-acetyl-α -D-gluopyranosyl)bromide( ABG ) in DMF at r.t. gave 3-amino-6,8-dibromo-2-(2′,3′,4′,6′-tetra-O-acetyl-β-D-glucopyranosyl)thioxo-2,3-dihydro-1H-quinazolin-4-one ( 14 ). The deblocking of 14 in sodium methoxide gave 5 . 3-Amino-6,8-dibromo-2-methylthio-3H-quinazolin-4-one ( 16 ) was prepared by stirring 5 with methyl iodide in methanol. The treatment of 16 with hydrazine hydrate afforded 4 . The condensation of 4 with aldehydes furnished 3,5-dibromo-2-arylaminobenzoic acid hydrazide ( 18a–c ). The refluxing of 18a with acetic anhydride gave 3-(benzylideneamino)-6,8-dibromo-2-methyl-3H-quinazolin-4-one ( 19 ). Hydrazones 20a–f were prepared by the condensation of 4 with pentoses and/or hexoses. The acetylation of ( 20a–f ) with acetic anhydride gave the acetyl derivatives 21a–f .  相似文献

2-Cyano-N-(tetrahydrocarbazole)acetamide (1) was utilized for the synthesis of several new arylazocarbazole derivatives (2a–e). Compound (1) reacted with phenyl isothiocyanate to yield the corresponding non-isolable intermediate (3), which gave, upon treatment with dilute hydrochloric acid, thiocarbamoyl derivative (4). Compound (3) reacted with chloroacetone, chloroacetic acid, chloroacetyl chloride, ethyl bromoacetate, and phenacyl bromide to afford thiazolone derivatives (6), (8), and (10), respectively. Compound (1) was heated in the presence of pyridine and/or hydrazine hydrate and/or isatine to give the corresponding tetrahydrocarbazole derivatives (13), (14), and (18), respectively.
