Diamino-2-quinoxalinols are reacted with salicylaldehyde derivatives to produce 2-quinoxalinol imines regioselectively as one isomer in good yield. Regioselectivity has been determined through the use of isotopic 15N labeling experiments. The 2-quinoxalinol imines may then be reacted without further purification with additional salicylaldehyde derivatives to yield asymmetrically substituted 2-quinoxalinol salens.  相似文献

Metal complexes of 2-quinoxalinol salen (salqu) ligands can be prepared in a one-pot metal templated synthesis resulting in significantly enhanced yields than if the ligand were prepared and isolated prior to introducing the metal for complexation. Using this method, 12 salqu metal complexes have been prepared and characterized from +2 metal ions.  相似文献

A fluorous isatoic anhydride and isocyanate are synthesized and used as scavengers for amines in solution-phase parallel synthesis of urea, thiourea, and β-hydroxyamine analogs. The resulting fluorous derivatives are readily separated from the reaction mixture by solid-phase extractions (SPE) over FluoroFlash™ cartridges to give products with good purity. A commercially available Argonaut VacMaster-96 plate-to-plate solid-phase extraction (SPE) station equipped with 24 FluoroFlash cartridges is employed for parallel purification of fluorous reaction mixtures. Each cartridge charged with 3 g of fluorous silica gel has the capability to produce up to 100 mg of purified small molecules. The 24-well receiving plate has a standard footprint that can be directly concentrated in a Genevac vacuum centrifuge. Important issues such as sample loading, product cross-contamination, cartridge reuse, and reproducibility are investigated. The SPE system has been demonstrated in the purification of three small libraries that were produced involving amine scavenging reactions with fluorous isatoic anhydride, amide coupling reactions with 2-chloro-4,6-bis[(perfluorohexyl)propyloxy]-1,3,5-triazine (fluorous CDMT), and amide coupling reactions with a newly developed fluorous Mukaiyama condensation reagent.  相似文献

A parallel solution-phase synthesis of 2-quinoxalinol analogues is described. The key step-simultaneous reductions of m-Ar(NO(2))(2) to m-Ar(NH(2))(2) was investigated extensively. We obtained preliminary pharmacological activity of those analogues for the inhibition of LPS-induced TNF-alpha release on mouse macrophage in vitro. Two compounds revealed inhibitory activity, with IC(50) values of 0.40 microM (7-amino-6-[(3-methoxypropyl)amino]-3-methyl-2-quinoxalinol) and 2.2 microM (7-amino-6-[(3-butoxypropyl)amino]-3-methyl-2-quinoxalinol), respectively.  相似文献

Complexes of chiral salen and its related tetradentate Schiff-base ligands adopt three different configurations, trans, cis-alpha and cis-beta. Of these complexes, trans-complexes have been widely used as catalysts for various asymmetric reactions. However, recent studies have disclosed that cis-beta metallosalen and its related complexes show unique asymmetric catalyses that cannot be achieved by trans-metallosalen complexes. The present article summarizes generation of cis-beta metallosalen and its related complexes, their structural features, and their application to asymmetric syntheses.  相似文献

A solution-phase approach for the synthesis of phosphorothioate oligonucleotides that circumvents the use of chromatographic purifications of protected phosphorothioate intermediates was developed. Implementation of a two-step sulfurization protocol in the phosphoramidite method allows efficient isolation of the intermediate phosphorothioates by extractions. The viability of this approach is demonstrated by the synthesis of a hexameric phosphorothioate oligonucleotide fragment.  相似文献

Described is an efficient synthesis of silyl-substituted salen ligands, used for the preparation of enantioselective catalysts. The salicylaldehyde precursors are synthesized from the silyl ethers of 2,6-dibromophenols via a one-pot double lithium halogen exchanges, to induce an intramolecular retro-Brook rearrangement and allow introduction of the aldehyde group. Condensation of the salicylaldehyde products with a chiral diamine affords the silyl-substituted salen ligands in high yields. The use of other electrophiles allows easy access to silyl-substituted phenolic esters, ketones, and boronic acids.  相似文献

An in-house developed automated synthesis suite was used to prepare a library of 72 tetrapeptide derivatives, the starting materials for pharmaceutically attractive pentapeptides, employing a convergent strategy. An initial set of 18 dipeptides were synthesized on a large-scale (100-1000 g) using automated synthesis workstations, and then 72 tetrapeptides were synthesized on a medium scale (5-10 g) using an automated system. Each di- or tetrapeptide was prepared in a single operating cycle using a modified methanesulfonic acid method, then a sub-library of 56 pentapeptides were synthesized in parallel, on a small-scale (100 mg-1 g) using a robotic workstation.  相似文献

Iron carbonyl-stabilized cations have been employed to develop methodology for carbon-carbon and carbon-heteroatom formation suitable for the preparation of combinatorial libraries. Different nucleophiles were added to tricarbonyl(cyclohexa-1,3-dienylcarboxylic acid 4-nitro-phenyl ester)iron hexafluorophosphate. Aminolysis, followed by decomplexation, yielded substituted cyclohexadienyl amides of high purity. Carbon, oxygen, and sulfur nucleophiles gave good results, while amine nucleophiles gave products of somewhat lower purity.  相似文献

Using a Cu(II) 2-quinoxalinol salen complex as the catalyst and tert-butyl hydroperoxide (TBHP) as the oxidant, allylic activations of olefin substrates can be converted to the corresponding enones or 1,4-enediones. Excellent yields can be achieved (up to 99%) within a very short reaction time and with great tolerance for additional functional groups. Possible mechanistic pathways have been characterized using Raman spectroscopy, cyclic voltammetry, and theoretical calculations.  相似文献

A practical, one-pot synthesis of enantiopure unsymmetrical salen ligands is described, using a 1:1:1 molar ratio of a chiral diamine and two different salicylaldehydes. Utilizing combinatorial synthesis and a preparative LC-MS automated chromatography system we have prepared and purified a library of 4-[2-(1,2,4-oxadiazolyl)]piperidines that were designed to be novel and selective dopamine D4 ligands. In one round of synthesis we identified N-4-chlorobenzyl-4-[2-(3-(2-thienyl)-1,2, 4-oxadiazolyl)]piperidine with a Kd of 5 nM for the human D4 receptor.  相似文献

A three step efficient strategy for the synthesis of substituted 5-nitropiperidones in high de, employing Michael addition of N-p-tolylsulfinyl beta-nitroamines to alpha,beta-unsaturated esters, hydrolysis of the sulfinyl group, and cyclization of the resulting free amines, has been developed. An efficient parallel synthesis of 6,7-dimethoxytetrahydroisoquinolines is reported. The key reaction step is 3,4-dimethoxyphenethylimines reacting with acid chlorides to form an N-acyliminium ion intermediate, which undergoes Pictet-Spengler condensation to give the desired products in >80% yield. Both solution-phase and solid-phase synthesis of 6,7-dimethoxytetrahydroisoquinolines are described.  相似文献
