Self-assembled aggregates of a synthetic zinc chlorin in an aqueous suspension with either α-lecithin or Triton X-100 exhibit unique structural and functional properties. Absorption, circular dichroism, fluorescence, and resonance Raman spectra indicate that the supramolecular structure in an aqueous microheterogeneous medium is very similar to that of the bacteriochlorophyll c aggregates in non-polar organic solvents and in chlorosomes, the main light-harvesting antennae of green photosynthetic bacteria. The nature of the aggregates is controlled by structure and/or concentration of the added surfactants. When a small amount of metal-free bacteriochlorin is present it acts as an efficient energy acceptor from the aggregated zinc chlorins. Thus, self-assembly of synthetic zinc chlorins, doped with appropriate energy acceptors and surrounded with surfactants, affords an artificial supramolecular light-harvesting device in aqueous environment.  相似文献

Abstract— Light-harvesting bacteriochlorophylls are believed to he aggregated in oligomcric forms in chlorosomes of green photosynthetic bacteria. Zn and Mg methyl bacteriopheophorhides d (MBPd) were synthesized from chlorophyll a and studied as model compounds for bacteriochlorophyll d . Monomeric Zn and Mg MBPd in methanol have Q y absorption maxima at 650 nm and 657 nm and fluorescence decay lifetimes of 5.1 ns and 5.4 ns, respectively, comparcd to 5.6 ns for bactcriochlorophyll d . Zn and Mg MBPd both form oligomers in nonpolar solvents and exhibit Q, absorption maxima at 728 nm and 731 nm and fluorescence decay lifetimes of 14 ps and 19 ps, respectively, compared to 730 nm and 9 ps for similar bacteriochlorophyll d aggregates. One of the diastereomers at the 3l position, R-Mg MBPd, forms intermediate-sized aggregated species that are equivalent to the dimer and a highly fluorescent species formed by bacteriochlorophylls c and d . The similarities of quantitative propcrtics between the model compounds, and the antenna pigments bacteriochlorophyll c and d indicate that Mg and Zn MBPd are good models for studying pigment interactions in chlorosomes and that the long hydrocarbon tail in the natural pigment is not required for oligomer formation. The dimer and the highly fluorescent species do not appear to be the building blocks of the oligomcr.  相似文献

Spectral forms of bacteriochlorophyll (Bchl) in chlorosomes were analyzed by linear dichroism, circular dichroism (CD), and deconvolution of these spectra. Isolated chlorosomes were embedded in polyacrylamide gels and compressed unidirectionally (along the x-axis) while allowing the gel to stretch in another direction (along the z-axis). The chlorosomes were aligned three-dimensionally due to their flat oblong shape; the longest axis was presumed to parallel the z-axis, its shortest axis was presumed to parallel the x-axis, and the intermediate-length axis was presumed to parallel the y-axis. Degrees of polarization (AI? A1)/(AI+ A1) of Bchl c and a measured from the y-axis with linearly polarized light were significantly different from those measured from the x-axis. Deconvolution of spectra into components revealed the presence of two major forms of Bchl c with peaks at 744 nm and 727 nm. The degrees of polarization of the 744 and 727 nm spectral forms were 0.76 and 0.59 from the y-axis and 0.48 and 0.39 from the x-axis, respectively. The degrees of polarization of Bchl a794 were –0.21 from the y-axis and 0.12 from the x-axis. These values indicate that the direction of the Qy transition moment of Bchl c744 is almost completely parallel to the longest axis of chlorosomes and that of Bchl c727 is also nearly, but slightly less so, parallel to the longest axis of the chlorosomes. The Qy transition moment of the baseplate Bchl a peak at 794 nm is nearly perpendicular to the longest axis and parallel to the shortest axis: that is, it is perpendicular to the associated membrane plane in the cell. These alignments of Bchl transition moments in chlorosomes were lost after suspending the chlorosomes in a solution saturated with 1-hexanol accompanying a shift in the peak position from 742 nm to 670 nm. The alignment recovered after the hexanol concentration was decreased. The presence of two major spectral forms of Bchl c was supported by the deconvolution of CD spectra and absorption spectra.  相似文献

Chlorosomes are one of the most unique natural light‐harvesting antennas and their supramolecular nanostructures are still under debate. Chlorosomes contain bacteriochlorophyll (BChl)‐c, d and e molecules and these pigments self‐aggregate under a hydrophobic environment inside a chlorosome. The self‐aggregates are mainly constructed by the following three interactions: hydrogen bonding, coordination bonding and π–π stacking. Supramolecular nanostructures of self‐aggregated BChls have been widely investigated by spectroscopic and microscopic techniques. Model compounds of such chlorosomal BChl molecules have been synthesized and the effects of esterified long alkyl chains at the 17‐propionate residue for their self‐aggregation have been studied. Structurally simple zinc chlorophyll derivatives possessing an oligomethylene chain as the esterifying group at the 17‐propionate residue were prepared as chlorosomal BChl models. The synthetic zinc BChls self‐aggregated in nonpolar organic solvents to give precipitates. The resulting insoluble self‐aggregated solids were investigated on a variety of substrates, including hydrophobic, neutral and hydrophilic substrates, by visible absorption, circular dichroism and polarized light absorption spectroscopies, as well as atomic force, transmission electron and scanning electron microscopies. The self‐aggregates of synthetic Zn‐BChls formed rods with an approximately 5 nm diameter and wires with further elongated growth of the rods (aspect ratio >200). The diameter size was consistent with that estimated for natural chlorosomal rods in a filamentous anoxygenic phototroph, Chloroflexus aurantiacus. The supramolecular formation and stability of the rod on the examined substrates depended on the length of an oligomethylene chain at the 17‐propionate residue as well as on the surface properties. Especially, the number of the 5 nm rods on the substrates increased with an elongation of the chain.  相似文献

The aggregation behavior of two homologs of bacteriochlorophyll c (BChl c) in various media was investigated for the effects of pH and salt, and the corresponding structures were analyzed by Fourier transform (FT)-IR spectroscopy. R-[P, E] BChl cF (31-R-form of BChl c with a propyl group at the C-8 position and an ethyl group at the C-12 position) and R-[E, E] BChl cF (31-R-form of BChl c with two ethyl groups at positions C-8 and C-12) were isolated from the green sulfur bacterium Chloro-bium limicola. Aggregates of each homolog showed a pH-dependent shift of the absorption maximum; at low pH, the peak moved to the red. This tendency was also revealed by circular dichroic spectra. A similar red shift of the peak was also induced by a high concentration of salt (NaCl) or buffer for both homologs. The FT-IR spectrum indicates that at low pH, both homologs formed a rather amorphous aggregate. On the other hand, a regular structure of R-[P, E] BChl cF was indicated in an acetone-water mixture. This structure was stabilized by a triangular interaction among three pigment molecules through the Mg-OH (3>) O = C (131) linkage. This structure was not found for R-[E, E] BChl cF. These results indicate that the replacement of the side chain at the C-8 position on the macrocycle induces a change in aggregation behavior. A possible heterogeneity of the in vivo rod structure of chlorosomes in green sulfur bacteria is discussed based on the above results.  相似文献

Near-infrared excited Fourier-transform Raman spectra have been measured for whole living Chlorobium limicola f. thiosulfatophilum to explore in situ structure of bacteriochlorophyll c (BChl-c). The spectra, whose Raman bands are preresonance enhanced via a Qy band of BChl-c, did not contain contributions from the major components of bacteria such as proteins and lipids. Therefore, the spectra provide selective structural information about BChl-c in the chlorosomes in a totally nondestructive manner. A marker band, appearing at 1605 cm-1 for the coordination number of the Mg atom, shows that BChl-c in the chlorosomes is five coordinate. The Raman spectrum of living bacteria closely resembles that of BChl-c in water-saturated carbon tetrachloride (w-std CC14) where it is comprised of dimer, tetramer and polymer spectra. However, a band assigned to a C=O stretching mode of the 131-keto group is identified only at 1641 cm-1. A band arising from the free keto carbonyl group, which appears in the spectrum of BChl-c in w-std CC14, is not observed in the spectra of bacteria. These observations suggest that BChl-c in the chlorosomes forms mostly coordinate polymeric species whose structure is very similar to that of BChl-c in w-std CC14.  相似文献

Chlorosomes are light-harvesting antennae of photosynthetic bacteria containing large numbers of self-aggregated bacteriochlorophyll (BChl) molecules. They have developed unique photophysical properties that enable them to absorb light and transfer the excitation energy with very high efficiency. However, the molecular-level organization, that produces the photophysical properties of BChl molecules in the aggregates, is still not fully understood. One of the reasons is heterogeneity in the chlorosome structure which gives rise to a hierarchy of structural and energy disorder. In this report, we for the first time directly measure absorption linear dichroism (LD) on individual, isolated chlorosomes. Together with fluorescence-detected three-dimensional LD, these experiments reveal a large amount of disorder on the single-chlorosome level in the form of distributions of LD observables in chlorosomes from wild-type bacterium Chlorobaculum tepidum . Fluorescence spectral parameters, such as peak wavelength and bandwidth, are measures of the aggregate excitonic properties. These parameters obtained on individual chlorosomes are uncorrelated with the observed LD distributions and indicate that the observed disorder is due to inner structural disorder along the chlorosome long axis. The excitonic disorder that is also present is not manifested in the LD distributions. Limiting values of the LD parameter distributions, which are relatively free of the effect of structural disorder, define a range of angles at which the excitonic dipole moment is oriented with respect to the surface of the two-dimensional aggregate of BChl molecules. Experiments on chlorosomes of a triple mutant of Chlorobaculum tepidum show that the mutant chlorosomes have significantly less inner structural disorder and higher symmetry, compatible with a model of well-ordered concentric cylinders. Different values of the transition dipole moment orientations are consistent with a different molecular level organization of BChl's in the mutant and wild-type chlorosomes.  相似文献

Linear dichroism (LD) spectroscopy is a widely used technique for studying the mutual orientation of the transition-dipole moments of the electronically excited states of molecular aggregates. Often the method is applied to aggregates where detailed information about the geometrical arrangement of the monomers is lacking. However, for complex molecular assemblies where the monomers are assembled hierarchically in tiers of supramolecular structural elements, the method cannot extract well-founded information about the monomer arrangement. Here we discuss this difficulty on the example of chlorosomes, which are the light-harvesting aggregates of photosynthetic green-(non) sulfur bacteria. Chlorosomes consist of hundreds of thousands of bacteriochlorophyll molecules that self-assemble into secondary structural elements of curved lamellar or cylindrical morphology. We exploit data from polarization-resolved fluorescence-excitation spectroscopy performed on single chlorosomes for reconstructing the corresponding LD spectra. This reveals that LD spectroscopy is not suited for benchmarking structural models in particular for complex hierarchically organized molecular supramolecular assemblies.  相似文献

Esterifying chains of chlorophyllous pigments play important roles in the formation of photosynthetic supramolecules, but their effects have not thoroughly been unraveled yet. Substitution of the esterifying chains in these pigments will be one possible strategy to elucidate this enigma. Recently, unnatural bacteriochlorophylls (BChls) c possessing a hydroxy group at the terminus of the esterifying chains were successfully biosynthesised in the green sulfur bacterium Chlorobaculum (Cba.) tepidum grown by supplementation of α,ω-diols. In this paper, in vitro assembling behaviours of unnatural BChls c isolated from Cba. tepidum grown with 1,8-octanediol, 1,12-dodecanediol and 1,16-hexadecanediol were characterised in aqueous Triton X-100 micelles to investigate the effects of the terminal hydroxy group in the esterifying chains of BChls c on self-aggregates such as chlorosomes, major antenna complexes in green photosynthetic bacteria. The bacteriochlorophyll (BChl) c derivatives monoesterified with α,ω-diols formed chlorosomal self-aggregates, but their formations were much slower than those of natural BChl c. The Qy absorption bands of the residual monomers of these BChl c derivatives were larger than those of natural BChl c. These suggest that the esterifying α,ω-diols in these unnatural BChls c somewhat interfered with formation of chlorosome-like aggregates compared with the natural esterifying farnesol.  相似文献

We have studied the effect of the absence of carotenoids on the organization of bacteriochlorophylls (BChls) in chlorosomes of Chlorobium (Chl.) phaeobacteroides strain CL1401. Carotenoid-depleted chlorosomes were obtained by means of 2-hydroxybiphenyl-supplemented cultures. In the presence of the inhibitor, isorenieratene (Isr) and beta-Isr biosynthesis were inhibited to more than 95%, leading to an accumulation of the colorless precursor phytoene inside the chlorosomes. In addition, there was a 30-40% decrease in the baseplate BChl a content. The absorption spectrum of the carotenoid-depleted chlorosomes showed a 10 nm blue shift in the BChl e Qy absorption peak. Under reducing conditions, a decrease in the BChl a/BChl e fluorescence emission ratio was observed in carotenoid-depleted chlorosomes relative to that in control chlorosomes, caused mainly by the decrease in the BChl a content. The steady-state fluorescence emission anisotropy in the BChl e region dropped from approximately 0.24 for native chlorosomes to approximately 0.14 for carotenoid-depleted ones, indicating reorganization of BChl e. The circular dichroism (CD) signal of the carotenoid-depleted chlorosomes was increased two times in the BChl e Qy region. A simple model based on the structure proposed was used to explain the observed effects. Carotenoids might affect the angle between the direction of the BChl e Qy transition and the axis of the rod. The orientation of BChl a in the baseplate remains unchanged in carotenoid-depleted chlorosomes, although there is a partial loss of BChl a as a consequence of a decrease in the baseplate size. The carotenoids are most likely rather close to the BChls and appear to be important for the aggregate structure in Chl. phaeobacteroides.  相似文献

Abstract— Zinc chlorins possessing 31-hydroxyl and 131-carbonyl groups self-assemble in nonpolar solvents, such as hexane, in a manner similar to bacteriochlorophyll c in the chlorosomes of green photosynthetic bacteria. Visible absorption and steady-state fluorescence measurements of zinc chlorin aggregates containing a small amount of the bacteriochlorin-zinc chlorin dyad molecules showed that singlet excitation energy transfer from the zinc chlorin aggregate to the bacteriochlorin moiety of the coaggre-gated dyad occurs in the homogeneous solution. In the coaggregated dyad, the bacteriochlorin moiety plays the role of an efficient energy trap and the chlorin moiety the role of an anchor to the donor aggregate. The artificial assembly thus mimics the structure and function of natural chlorosomes and can be considered as the first in vitro supramolecular light-harvesting antenna.  相似文献

Abstract— Two different protochlorophyllides (PChlide), PChlide 629/433 (absorption data in methanol) and PChlide 630/441 (the monovinyl (MV) and divinyl (DV) forms) were isolated from the pigment mutant C-2A'of Scenedesmus obliquus. Their spectroscopic behaviour in several organic solvents and their aggregation in toluene was investigated. In polar solvents such as ether, acetonitril or acetone, absorption maxima similar to those in methanol were observed, while in solvents such as tetrahydrofuran and pyridine a bathochromic shift of the blue absorption band compared to the spectra in methanol occurred. The absorption maxima of MV-PChlide shifted from 629 nm and 433 nm in methanol (monomeric form), to 631 nm and 443 nm in toluene (aggregated form). The absorption maxima of DV-PChlide shifted from 630 nm and 441 nm in methanol to 655 nm and 483 nm in toluene (aggregated form). The fluorescence excitation and emission spectra of the two protochlorophyllides yielded the according results. The aggregation process was faster for DV-PChlide than for MV-PChlide and was reversible upon addition of small amounts of polar solvents. The similarity of the spectral characteristics of the aggregated forms of the different protochlorophyllides after toluene treatment with those reported for “active”-PChlide in vivo are discussed.  相似文献

New “reference” circular dichroism spectra of α helix, β-structure (both parallel and antiparallel), β-bends, and the unordered form are obtained from circular dichroism spectra and x-ray data for six proteins (myoglobin, lysozyme, lactate dehydrogenase, papain, ribonuclease, and subtilisin BPN′). Circular dichroism spectra for α-helix and antiparallel β-structure are similar to those for poly(Llysine). The circular dichroism spectrum of the parallel β-structure is qualitatively similar to that theoretically calculated by Madison and Schellman. The circular dichroism spectrum of β-bends is qualitatively similar to that theoretically calculated by Woody. The spectrum of the unordered form is close to that of the denaturated proteins. These “reference” circular dichroism spectra used for the analysis of the secondary structure of ten globular proteins (besides the six reference proteins D-glyceraldehyde 3-phosphate dehydrogenase, concanavalin A, cytochrome c, and insulin).  相似文献

Systematically synthetic zinc 3-hydroxymethyl-131-oxo-chlorins esterified by different linear alcohols (methanol, 1-propanol, 1-hexanol, 1-dodecanol and 1-octadecanol) at the 17-propionate were self-assembled in the presence of cetyltrimethylammonium bromide in an aqueous solution. These zinc chlorins exhibited red-shifted Q y absorption bands and circular dichroism (CD) signals in the corresponding Q y region after incubation for 17 h, indicating that the zinc chlorins formed self-aggregates like those in natural chlorosomes of green photosynthetic bacteria. Visible absorption and CD spectra of self-aggregates of the zinc chlorins depended on the length of their esterifying alcohols. Zinc chlorins esterified by shorter alcohols gave larger changes in their visible absorption and CD spectra after incubation above 40°C, whereas zinc chlorins esterified by longer alcohols afforded smaller changes. These results indicate that hydrophobic interaction among esterifying chains of chlorin molecules as well as that between the esterifying chains and peripheral surfactants or lipids play an important role in the stability of chlorosomal self-aggregates.  相似文献

Methyl 131‐(di)cyanomethylene‐pyropheophorbides were synthesized by Knoevenagel reactions of the corresponding 131‐oxo‐chlorins prepared from modifying chlorophyll‐a with malononitrile or cyanoacetic acid. Alternatively, methyl 131‐cyanomethylene‐pyropheophorbides were produced by Wittig reactions of 131‐oxo‐chlorins with Ph3P=CHCN. Self‐aggregation of zinc complexes of the semi‐synthetic chlorophyll derivatives possessing a hydroxy or methoxy group at the 31‐position was examined in 1%(v/v) tetrahydrofuran or dichloromethane and hexane by electronic absorption and circular dichroism spectroscopy. Although intermolecular hydrogen‐bonding between the 31‐hydroxy and 131‐oxo groups of bacteriochlorophylls‐c/d/e/f was essential for their self‐aggregation in natural light‐harvesting antenna systems (=chlorosomes), zinc 31‐hydroxy‐131‐di/monocyanomethylene‐chlorins self‐aggregated in the less/lesser polar organic solvents to form chlorosome‐like large oligomers in spite of lacking the 131‐oxo moiety as the hydrogen‐bonding acceptor. Zinc 31‐methoxy‐131‐dicyanomethylene‐chlorin gave similar self‐aggregates regardless of lack of both the 31‐hydroxy and 131‐oxo groups. The present self‐aggregation was ascribable to stronger coordination of the 31‐oxygen atom to the central zinc than the conventional systems, where the electron‐withdrawing cyano group(s) increased the coordinative ability of the central zinc through the chlorin π‐system.  相似文献

Abstract— We have investigated changes in the organization of bacteriochlorophyll c (BChl c ) in chlorosomes isolated from the green sulfur bacterium Chlorobium tepidum during the formation of bacteriopheophytin and bacteriopheo-phorbide in acidic media. The reaction was much slower than that observed with BChl dissolved in methanol, suggesting that the aggregation of BChl or the presence of the chlorosome envelope constitutes a barrier to the reaction with protons in the aqueous phase. In most cases the first two-thirds of the reaction showed time courses that were close to linear. Simultaneously with the pheo-phytinization process we observed a red-shift of the Qy band of the remaining aggregated BChl c reaching a maximum extent of 9 nm. Analysis of the spectral changes provided evidence at least for two spectrally distinct pools of aggregated BChl c with different rates of reaction with acid. An HPLC analysis showed that there were no changes in the distribution of the four major homologs of BChl c remaining in chlorosomes during the course of acid treatment, at least up to the time when two-thirds had been converted to pheophytin. This suggests that the homologs of BChl c are uniformly distributed within the chlorosome.  相似文献

Self-aggregation of naturally occurring bacteriochlorophyll (BChl)-e in nonpolar organic solvents was investigated by visible absorption, fluorescence emission and circular dichroism spectra. Cultured brown-colored photosynthetic bacteria have several BChl-e as light-harvesting antenna pigments. Three major BChl-e homologs were separated from the extracts of the culture by reverse-phase high-performance liquid chromatography (HPLC) and characterized by 1H-NMR and fast-atom bombardment mass spectroscopy: 8-ethyl-12-ethyl ([E,E])-, 8-propyl-12-ethyl- and 8-isobutyl-12-ethyl-BChl-e farnesyl esters. All the homologs consisted of a mixture of the 3(1)-epimers, and epimerically pure BChl-e were also given by HPLC separation. All the separated BChl-e epimers, the epimeric mixtures and the homologous mixtures formed self-aggregates in 2% dichloromethane/hexane, giving visible absorption spectra similar to that of the whole cells, which showed two peaks (or shoulders) around 430-450 and 520 nm at the Soret region as well as a red-shifted Qy band relative to the monomeric. The spectral properties of the Soret band were basically unchanged among the epimers or epimeric/homologous mixtures. In contrast, the Qy band of aggregates of epimeric mixtures (except [E,E]) and homologous mixtures red-shifted and broadened compared with the epimerically pure. The red-shift and broadening of the Qy band are advantageous for efficient energy transfer from BChl-e aggregates to BChl-a in a baseplate in chlorosomes because their spectral overlap increases.  相似文献

Time-resolved, laser-induced changes in absorbance, delta A(lambda; t), have been recorded with a view to probing pigment-pigment interactions in chlorosomes (control as well as carotenoid-depleted) and artificial aggregates of bacteriochlorophyll e (BChle). Control chlorosomes were isolated from Chlorobium phaeobacteroides strain CL1401, whose chromophores comprise BChle, bacteriochlorophyll a (BChla) and several carotenoid (Car) pigments; Car-depleted chlorosomes, from cells grown in cultures containing 2-hydroxybiphenyl. Artificial aggregates were prepared by dispersing BChle in aqueous phase in the presence of monogalactosyl diglyceride. In chlorosomes delta A(lambda; t) shows, besides a signal attributable to triplet Car (with a half-life of about 4 microseconds), signals in the Qy regions of both BChl. The BChla signal decays at the same rate as the Car signal, which is explained by postulating that some Car are in intimate contact with some baseplate BChla pigments, and that when a ground-state Car changes into a triplet Car, the absorption spectrum of its BChla neighbors undergoes a concomitant change (termed transient environment-induced perturbation). The signal in the Qy-region of BChle behaves differently: its amplitude falls, under reducing conditions, by more than a factor of two during the first 0.5 microsecond (a period during which the Car signal suffers negligible diminution), and is much smaller under nonreducing conditions. The BChle signal is also attributed to transient environment-induced perturbation, but in this case the perturber is a BChle photoproduct (probably a triplet or a radical ion). The absence of long-lived BChle triplets in all three systems, and of long-lived BChla triplets in chlorosomes, indicates that BChle in densely packed assemblies is less vulnerable to photodamage than monomeric BChle and that, in chlorosome, BChla rather than BChle needs, and receives, photoprotection from an adjacent Car.  相似文献

Bacteriochlorophyll (BChl) e was coassembled with BChl c in Triton X‐100 micelles in aqueous solutions. The Qy absorption bands of the coaggregates were positioned between those of aggregates consisting solely of BChl c or e. The electronic absorption spectra of the coaggregates could not be reproduced by linear combinations of the spectra of the aggregates consisting solely of each pigment, but they were in line with the simulated spectra for the self‐aggregates in which both BChls were randomly distributed. These suggest that BChls c and e are not spatially separated; they are homogenously distributed over the self‐aggregates to give electronic spectra that are different from those of the aggregate consisting solely of each pigment. Deaggregation of the scrambled self‐aggregates by excess Triton X‐100 did not produce any spectral components assigned to an aggregate consisting solely of either BChl c or e. Acid‐induced decomposition of the scrambled aggregates showed different kinetics from those of the aggregates consisting solely of each pigment. These also support the homogeneous distribution of BChls c and e in the scrambled self‐aggregates. These results will be useful to investigate the major light‐harvesting antenna systems of green photosynthetic bacteria that contain two kinds of chlorosomal BChls.  相似文献
