The aim of this work is the development of new radiopharmaceuticals for imaging infection and inflammation in human. Gatifloxacin (fluoroquinolone derivative) and cefepime (cephalosporine derivative) are antibiotics used to treat bacterial infections were investigated to label with one of the most important radioactive isotopes (technetium-99m). The reaction parameters that affect the labeling yield such as substrate concentration, stannous chloride dihydrate concentration, pH of the reaction mixture, and reaction time were studied to optimize the labeling conditions. Maximum radiochemical yield of 99mTc–gatifloxacin (90  ± 1.8%) complex was obtained by using 50 μg SnCl2·2H2O and 2.5 mg gatifloxacin at pH 10 while 99mTc–cefepime was prepared at pH 8 with a maximum radiochemical yield of 98  ± 1.4% by adding 99mTc to 5 mg cefepime in the presence of 50 μg SnCl2·2H2O. Biological distribution of 99mTc–gatifloxacin and 99mTc–cefepime was carried out in experimentally induced infection rats, in the left thigh, using Escherichia coli. Both thighs of the rats were dissected and counted and the ratio of bacterial infected thigh/contralateral thigh was then evaluated. T/NT for both 99mTc–gatifloxacin and 99mTc–cefepime was found to be 4.5  ± 0.3 and 8.4  ± 0.1, respectively, which was higher than that of the commercially available 99mTc–ciprofloxacin. The abscess to normal muscle ratio indicated that 99mTc–cefepime could be used for infection imaging. Besides, in vitro studies showed that 99mTc–cefepime can differentiate between bacterial infection and sterile inflammation.  相似文献

The aim of this study is to prepare radiolabeled guanine with 99mTc(CO)3+ core. For this purpose, guanine has been radiolabeled with 99mTc(CO)3+ core. Quality control study of radiolabeled guanine molecule with 99mTc(CO)3+ core was performed by thin layer radio chromatography (TLRC) and high performance liquid radio chromatography (HPLRC). The results showed that the radiolabeling yield was quite high (94 ± 3%). Beside that 99mTc(CO)3–Gua complex has showed good in vitro stability during the 24 h period. Radiopharmaceutical potential of this complex was evaluated in Wistar Albino Rats. It was concluded that 99mTc(CO)3–Gua could be used as a nucleotide radiopharmaceutical for in vivo applications.  相似文献

The labeling of garenoxacin (GXN) with technetium-99m (99mTc) using different concentrations of GXN, sodium pertechnetate (Na99mTcO4), stannous chloride dihydrate (SnCl2·2H2O) at different pH was investigated and evaluated in terms of in-vitro stability in saline, serum, binding with multi-resistant Staphylococcus aureus (MDRSA) and penicillin-resistant Streptococci (PRSC) and its biodistribution in artificially MDRSA and PRSC infected rats. 99mTc–GXN complex with 97.45 ± 0.18% radiochemical stability was prepared by mixing 3 mg of GXN with 3 mCi of Na99mTcO4 in the presence of 150 μL of SnCl2·2H2O (1 μg/μL in 0.01 N HCl) at a pH 5.6. The radiochemical stability of the complex was evaluated in normal saline up to 240 min of reconstitution. It was observed that the complex showed maximum RCP values after 30 min of the reconstitution and remained more than 90% up to 240 min. The complex showed radiochemical stability in normal saline at 37 °C up to 16 h with a 17.80% de-tagging. The complex showed saturated in-vitro binding with living MDRSA and PRSC as compared to the insignificant binding with heat killed MDRSA and PRSC. Biodistribution behavior of the complex was assessed in artificially infected with living and heat killed MDRSA and PRSC rats. It was observed that the accumulation of the complex in the infected (live MDRSA and PRSC) tissue of the rats was almost five fold than in the inflamed and normal tissue. The high radiochemical stability in normal saline at room temperature, promising in-vitro stability in serum at 37 °C, saturated in-vitro binding with living MDRSA and PRSC, specific biodistribution behavior and high infected (target) to normal (non-target) tissue and low inflamed (non-target) to normal (non-target) tissue ratios we recommend 99mTc–GXN complex for in-vivo localization of infection caused by MDRSA and PRSC effective stains.  相似文献

This work reports the synthesis, radiolabeling and preliminary biodistribution results in tumor-bearing mice of the 99mTc(CO)3–AOPA colchicine conjugate. The novel ligand was successfully synthesized by conjugation of N-(acetyloxy)-2-picolylamino (AOPA) to deacetylcolchicine via a short carbonyl-methylene linker. Radiolabeling was performed in high yield with [99mTc(CO)3]+ core. 99mTc(CO)3–AOPA colchicine conjugate was hydrophilic and was stable at room temperature. Biodistribution studies in tumor-bearing mice showed that 99mTc(CO)3–AOPA colchicine conjugate accumulated in the tumor with good uptake and retention. However, its clearance from normal organs was not so fast, resulting in poor T/NT ratios. Further modification on the linker or/and 99mTc-chelate to improve the tumor targeting efficacy and in vivo kinetic profiles is currently in progress.  相似文献

In this paper, cheap liquorice residue was used to prepare activated carbon (AC), thioacetamide (TAA) was used to modify the AC, and the adsorption experiments were conducted in the simulated acid radioactive wastewater with low uranium concentration to study the adsorption behavior and mechanism for uranium by TAA modified AC (TAA–AC). The removal efficiency by TAA–AC was 92.1–98.2% from the 1 mg L?1 uranium solution at pH 2–6. The adsorption equilibrium data were well fitted by Dubinin–Radushkevich model, and the maximum adsorption capacity was estimated to be 340 mg g?1. TAA–AC showed an enhanced selectivity for uranium in the presence of competitive ions. Furthermore, the adsorption experiments were conducted in the actual acid radioactive wastewater with low uranium concentration from an in situ leach uranium mine. The high adsorption rate (98.3%) and selectivity (Kd?=?3.78×104 mL g?1) for uranium were observed in the actual acid radioactive wastewater, and the adsorption rate was found to maintain 96.2% over six cycles of adsorption–desorption.  相似文献

This work reports the synthesis, radiolabeling and preliminary biodistribution results in tumor-bearing mice of [99mTc(CO)3(IDA–PEG3–CB)]. The novel chlorambucil derivative was successfully synthesized by conjugation of iminodiacetic acid (IDA) to chlorambucil via a pegylated linker. The ligand could be labeled by [99mTc(CO)3]+ core in high yield to get [99mTc(CO)3(IDA–PEG3–CB)], which was very hydrophilic and was stable at room temperature. Biodistribution studies in tumor-bearing mice showed that [99mTc(CO)3(IDA–PEG3–CB)] accumulated in the tumor with favorable uptake and retention. The good accumulation in tumor tissue with high tumor/muscle ratios warrants further research to improve tumor targeting efficacy and pharmacokinetic profile of radiolabeled chlorambucil derivative by structural modification.  相似文献

This paper describes the preparation of samples for an experiment to measure the cross-section for 235U(n,n′)235mU in a fast fission spectrum of neutrons provided by a fast pulsed reactor/critical assembly. Samples of 235mU have been prepared for the calibration of the internal conversion electron detector that is used for the 235mU measurement. Two methods are described for the preparation of 235mU. The first method used a U-Pu chemical separation based on anion-exchange chromatography and the second method used an alpha recoil collection method. Thin, uniform samples of 235mU+235U were prepared for the experiment using electrodeposition.  相似文献

A new method for the possible incorporation of nuclear wastes has been attempted here by using ceramic matrix of TiO2 as a host precursor for confinement. Hafnium is used as a simulant for actinide high-level waste. After incorporating 181Hf tracer into TiO2 matrix, the leaching property of the resulting matrix was studied in water, sodium chloride and humic acid solutions. The leaching was measured in each of the case by following the radioactivity of 181Hf. TiO2 matrix has also been exposed to γ-radiation in order to simulate the radiation field for nuclear waste. It has been investigated with a nuclear technique called time differential perturbed Angular Correlation (TDPAC) that the lattice structure of titania remains undisturbed even under a strong radiation field. The leaching of 181Hf has also been studied after irradiating the TiO2 matrix with γ-radiation and the leaching behavior was observed not to change from that before irradiation.  相似文献

A rapid method for the preparation of 87Y/87mSr radionuclide generator from a rubidium chloride target irradiated with 35 MeV α-particles is described. A simple two-step procedure is used to obtain a carrier-free 87mSr isotope with a high enough radiochemical yield and high purity in the final aqueous fraction.  相似文献

A detailed theoretical study is carried out at the B3LYP/6-311G(d,p) and CCSD(T)/6-311++G(3df,2pd) (single-point) levels as an attempt to investigate the mechanism of the little understand ion–molecule reaction between HCN+ and NH3. Various possible reaction pathways are considered. It is shown that six dissociation products P 1 (NH3 + + HCN), P 2 (NH4 + + CN), P 3 (NH3 + + HNC), P 9 (HCNH+ + NH2) P 10 (NCNH3 + + H), and P 12 (HNCNH2 + + H) are both thermodynamically and kinetically feasible. Among these products, P 1 is the most competitive product with predominant abundance. P 3 and P 9 may be the second feasible products with comparable yields. P 12 may be the least possible product followed by the almost negligible P 2 and P 10 . Because the isomers and transition states involved in the HCN+ + NH3 reaction all lie below the reactant, the title reaction is expected to be rapid, which is consistent with the measured large rate constant in experiment. The title reaction may have a potential relevance in Titan’s atmosphere, where the temperature is very low. Furthermore, our calculated results are compared with the previous experimental findings.  相似文献

Type 304 stainless steel specimens artificially contaminated with CsCl solution were treated with KOH solution and KNO3 solution, respectively. Cs+ ion removal tests by a Q-switched Nd:YAG laser at 1064 nm at a given fluence of 57.3 J/cm2 were performed. The surface morphology and the relative atomic mole ratio of the specimen surface were investigated by SEM and EPMA. The order of Cs+ ion removal efficiency of laser was no-treatment < KOH < KNO3 during the 42 shots. From the investigation of XPS peaks around 532.7 and 292.9 eV, KNO3 on a surface of specimen was found to be fully decomposed during the laser irradiation. It was suggested that Cs2O particulates formed by the reaction between the reactive oxygen generated from the nitrate ion and Cs+ ion on the metal surface could be easily suspended. For the KOH system, FeOOH was formed during the laser irradiation and it changed into Fe2O3. It was also suggested that Cs2O particulates were formed by the reaction between the reactive oxygen generated from the decomposition of K2O and Cs+ ion on the metal surface..  相似文献

Summary.  The diagram of the ternary system Mg2+/Cl, SO4 2−–H2O was established at 15°C by means of analytical and conductimetric measurements. Three compounds were found in this diagram, which are MgSO4·6H2O, MgSO4·7H2O, and MgCl2·6H2O. The solubility field of MgSO4·7H2O is important whereas those of MgSO4·6H2O and MgCl2·6H2O are small. The compositions (mass-%) of the two invariant points determined by the two methods are: MgSO4:MgCl2=2.73:33.80 and MgSO4: MgCl2=3.38:28.91. Both the measured and the calculated isotherm at 15°C have been used for modelling of the diagram Mg2+/Cl, SO4 2−–H2O between 0 and 35°C. The polythermal invariant point was approximately located between 15 and 10°C.  Corresponding author. The radiolabeling of trovafloxacin dithiocarbamate (TVND) with technetium-99m using [99mTc-N]2+ core was investigated and biologically assessed as prospective infection imaging agent. The achievability of the 99mTcN-TVND complex as a future MRSA infection radiotracer was investigated in artificially methicillin-resistant Staphylococcus aureus (MRSA) infected male Sprague–Dawley rats (MSDR). Radiochemically the 99mTcN-TVND complex was characterized in terms of radiochemical purity (RCP) in saline, in vitro permanence in serum, in vitro binding with MRSA and biodistribution in living and heat killed MRSA infected MSDR. Radiochemically the complex showed stability in saline with a 97.90 ± 0.22% yield and serum at 37 °C up to 4 h. The 99mTcN-TVND complex showed saturated in vitro binding with MRSA. Normal in vivo uptake in the MRSA infected MDRS was observed with a five fold uptake in the infected muscle as compared to inflamed and normal muscles. The high RCP values, in vitro permanence in serum, better in vitro binding with MRSA, biodistribution behavior and the target to non-target (infected to inflamed muscle) ratios posed the 99mTcN-TVND complex as a promising MRSA infection radiotracer.  相似文献

Supramolecular pillared oxides were prepared through the intercalation of M2+ cations into a MnO2 host matrix by the method of ion exchange between the precursor δ-K x MnO2 and the corresponding guest. The materials M-MnO2 crystallize in the hexagonal system, the same structure as the precursor, with a larger interlamellar spacing. In the case of ZrO-MnO2, extended X-ray absorption fine structure (EXAFS) determination indicates that the Zr atom locates between the MnO2 layers forming a stable structure. Compared with the precursor, the cycling property of M-MnO2 was improved distinctly, while the capacity decreased to some degree due to the strong interaction between pillars and the host matrix. Among these pillared materials, ZrO-MnO2 has an advanced reversible capacity of 161.5 mAh·g−1 and improved cycling behavior compared with the precursor.  相似文献

The partial pressures of the components in the saturated vapor of the Se–S system were determined and presented as the temperature–concentration dependences. Based on these data, the boundaries of the melt–vapor phase transition at atmospheric pressure and in vacuum (1350, 100, and 10 Pa) were calculated. A complete phase diagram was constructed, which included the vapor–liquid equilibrium fields at atmospheric and low pressures, whose boundaries allowed us to determine the behavior of sulfur and selenium during distillation separation.  相似文献

Protactinium and thorium activities were measured in eight surface sediment taken in 2004 to determine effectiveness scavenging of 231Pa at Sabah–Sarawak coastal waters. The result found that activity ratios of 231Paex/230Thex were ranged from 0.07 to 0.13 at all sampling stations. The high 231Paex/230Thex activity ratio than the production ratio of 0.093 in seawater at station SR 01, SR 02, SR 04, SB 02 and SB 05, revealed that 231Pa is effectively removed from the water column into the sediment in comparison with 230Th at those stations. Low percentage of 230Thex (90–95%) in comparison with 231Paex at all stations can be attributed to less efficiently scavenged of 230Th onto particles prior deposited at the marine sediment bed.  相似文献

The first results of activity measurement of short-lived radium isotopes dissolved in water in the Anadyr River–Bering Sea transit system are presented. The trends of the radionuclide behavior after passage through high-gradient river–coastal sea water mixing zone were identified and the exchange rate of the horizontal mixing was determined. The 224Ra/223Ra activity ratio was used to construct the “radium age” model, which served for estimating the residence time of brackish water in the Anadyr River estuary. Four new mononuclear Pd(II) complexes of the type [PdX2(tdmPz)] {X = Cl (1); Br (2); I (3); SCN (4); tdmPz = 1-thiocarbamoyl-3,5-dimethylpyrazole} have been synthesized and characterized by elemental analysis, IR spectroscopy, 1H and 13C{1H}-NMR experiments. The thermal behavior of the complexes 14 has been investigated by means of thermogravimetry (TG) and differential thermal analysis (DTA). From the initial decomposition temperatures, the thermal stability of the complexes can be ordered in the sequence: 3 < 4  2 < 1. The final products of the thermal decompositions were characterized as metallic palladium by X-ray powder diffraction.  相似文献

Garenoxacin (GXN) was modified to its dithiocarbamate followed by radiolabeling with technetium-99m (99mTc) through [99mTc-N]2+ core. The suitability of the 99mTcN–Garenoxacin dithiocarbamate (GXND) complex as a potential multiresistant Staphylococcus aureus (MDRSA) and penicillin-resistant Streptococci (PRSC) infection radiotracer was assessed in artificially infected rats (AFRT). The radiolabeled complex was investigated for its radiochemical purity (RCP), permanence in serum using HPLC and TLC methods. In vitro binding with MDRSA and PRSC was performed at 37 °C. The 99mTcN–GXND showed maximum RCP of 98.00 ± 0.22% and remained more than 90% stable up to 4 h. The 99mTcN–GXND showed saturated in vitro binding with living MDRSA and PRSC, respectively. The complex showed normal biodistribution in healthy rats (HRT), however in AFRT, seven fold uptakes was observed in infected muscle as compared to inflamed and normal muscles. Based on the high RCP, stability in serum, better in vitro binding with bacteria, biodistribution behavior and the target to non-target (infected to inflamed muscle) ratio, we recommend the 99mTcN–GXND complex for in vivo investigation of MDRSA and PRSC infection in human.  相似文献

Abstract  The title compound, labeled with 13C in the ethyl groups was synthesized from K13CN and low-molecular-weight components. The synthetic relay compound was 31(32)[13C]-xanthobilirubinic acid methyl ester in a synthetic route that leads to a label in the ethyl β-substituent of a dipyrrinone model for bilirubin. This labeled dipyrrinone was oxidatively coupled to the dimethyl ester of mesobiliverdin-XIIIα, thereby providing a route to a 13C-labeled mesobiliverdin and mesobilirubin, with one carbon of each ethyl being 98% 13C-enriched.
