Monolayer films of phospholipids at the air-water interface have been transferred to solid substrates under conditions of continuously varying surface pressure, an approach termed COVASP. The molecular and supramolecular properties of the film constituents have been characterized with two complementary techniques. IR spectroscopy was used to monitor chain conformation as a function of transfer surface pressure. Results were compared to those from Langmuir films determined directly at the A/W interface by IR reflection-absorption spectroscopy (IRRAS). The methylene stretching frequencies for both proteated and acyl chain perdeuterated 1,2-dipalmitoylphosphatidylcholine (DPPC and DPPC-d62) in the transferred molecules indicate that the phospholipids retain at least, in part, their surface pressure-dependent chain-conformational order characteristics. The line widths of these modes are somewhat reduced, suggestive of slower rates of reorientational motion in the Langmuir-Blodgett (LB) films. Epifluorescence microscopy reveals a progressive condensation gradient, including nucleation and growth of probe-excluding condensed domains along the transfer line. DPPC condensation, observed along a single LB film, was qualitatively comparable to compression-driven condensation as observed in situ or in conventional LB films transferred at constant pressures. However, condensation along the compression isotherm in COVASP-LB films was reduced by 15-20% as compared to films equilibrated at different constant pressures, probably the result of kinetic differences in equilibration processes. As a preliminary demonstration of the utility of this new approach, the monolayer --> multilayer transition known to occur (Eur. Biophys. J. 2005, 34, 243) in a four-component model for pulmonary surfactant has been examined. IR parameters from both the lipid and the protein constituents of the film all indicate that the transition persists during the transfer process. This new approach for the study of transferred films will permit the efficient characterization of lipid-protein interactions and structural transitions occurring in pulmonary surfactant films subjected to dynamic compression.  相似文献

The respreading of a lung surfactant monolayer at the air-water interface is investigated with broad bandwidth sum frequency generation (BBSFG) spectroscopy. The lung surfactant mixture contains chain perdeuterated dipalmitoylphosphatidylcholine (DPPC-d62), palmitoyloleoylphosphatidylglycerol (POPG), palmitic acid (PA), and KL4 (a 21-residue polypeptide analogue to the surfactant protein SP-B). DPPC-d62 serves as a probe molecule for the spectroscopic investigation. The BBSFG spectra of DPPC-d62 in the lung surfactant mixture are obtained in the C-D stretching region in real-time during film compression and expansion in a Langmuir trough. The BBSFG intensity of the CD3 stretch peak from DPPC-d62 terminal methyl groups is used as a measure of the interfacial density of DPPC-d62 after careful consideration of orientation effects. For the first time, the interfacial loss of DPPC in a complex lung surfactant mixture is quantified. Spectroscopic results reveal that there is an 18% DPPC-d62 interfacial loss during film respreading. However, the surface pressure-area isotherm measurements demonstrate that there is a rather large trough area reduction (37%) during film expansion. The relatively small interfacial loss of DPPC-d62 and the rather large trough area reduction indicate that the respreading of DPPC and non-DPPC components in the lung surfactant is not uniform and a surface refinement process exists during film compression and expansion. This refinement process results in a DPPC-enriched monolayer with a significant depletion of non-DPPC components after film respreading. Implication for replacement surfactant design from this work is discussed.  相似文献

At the air-water interface, interfacial molecular structure, intermolecular interactions, film relaxation and film respreading of model lung surfactant monolayers were studied using vibrational sum frequency generation (VSFG) spectroscopy combined with a Langmuir film balance. Chain-perdeuterated dipalmitoylphosphatidylcholine (DPPC-d62), palmitoyloleoyl-phosphatidylglycerol (POPG), palmitic acid (PA) and tripalmitin were investigated. In the DPPC-d62-PA binary monolayer, PA showed a condensing effect on the DPPC chains. On the contrary, in the DPPC-d62-POPG binary monolayer, POPG showed a fluidizing effect on the DPPC chains. In the ternary monolayer system of DPPC-d62-POPG-PA, the balance between the fluidizing and the condensing effect was also observed. In addition, the film relaxation behavior of DPPC-d62 and the enhanced film stability of DPPC-d62 caused by the addition of tripalmitin were observed. Real-time VSFG was also employed to study the respreading properties of a complex lung surfactant mixture containing DPPC-d62, POPG, PA and KL4 (a mimic of SP-B) peptide, which revealed DPPC enrichment after film compression.  相似文献

对不同链长的2-烷基-苯并咪唑衍生物(BzCn,烷基链长从C5到C15)在硝酸银亚相上的成膜行为及形成的LB膜的结构进行了研究.表面压-面积曲线的结果表明,短链(C5~C9)的2-烷基-苯并咪唑可在银离子亚相上形成稳定的单分子膜,而长链(C13和C15)衍生物则形成多层膜.利用LB技术可将上述Langmuir膜转移到固体基板上形成LB膜,其吸收光谱的结果说明了苯并咪唑和银离子配位.利用AFM、XRD及FT-IR等技术研究了烷基链长对LB膜结构的影响.实验结果表明,除了BzC15,其余的衍生物都可形成规整的层状结构.短链衍生物的单层LB膜具有均一、平整的形貌;而对于BzC15,观察到多层结构.  相似文献

Phosphatidylglycerols are components of biological membranes. The phase behavior of these phospholipids was extensively investigated. However, there is still no definite picture about the dependence of the ionization state and monolayer structure on subphase composition. The major problem of previous investigations is that none of the methods used allow obtaining the ionization degree directly. In the present work we apply techniques developed in the past decades for Langmuir monolayers: infrared reflection absorption spectroscopy (IRRAS) as well as X-ray diffraction and reflectivity techniques, which provide straightforward information about structure and ionization state of a L-1,2-dipalmitoylphosphatidylglycerol (DPPG) monolayer. The Gouy-Chapman model is applied to evaluate the intrinsic pKa. Therewith, the ionization degree can be determined even at low pH values. The experimental titration curves are in good agreement with theoretical curves based on the Gouy-Chapman model. The obtained instrinic pKa amounts to 1. The ionization degree of a DPPG monolayer is independent of the monovalent cation size. In contrast, the structure of a DPPG monolayer is strongly affected by the type of divalent cations.  相似文献

The action mechanism of surfactant protein C (SP-C) in the lung surfactant monolayers is studied. On the basis of the SP-C molecular structure, a detailed interaction model is developed to describe the interaction of phospholipids/SP-C in the lung surfactant monolayers. It is supposed that: (1) in an alveolus monolayer, SP-C molecules are surrounded by phosphatidylglycerol (PG). When the monolayer is compressed, SP-C molecules can promote PG molecules to be squeezed out; (2) during compressing of the monolayer, unsaturated-PG molecules form a collapse pit firstly when liquid-expanded state (LE) components achieve the collapse pressure. Then, SP-C's alpha-helix is attracted by the collapse pit and both alpha-helix and PG molecules are squeezed out speedily. Finally, the squeezed-out matters can form a lipid-protein aggregation in the subphase. The lipid-protein aggregation, in the centre of which, there is the hydrophobic alpha-helix section surrounded by PG molecules; (3) during the monolayer expanding, because of the increasing of the monolayer's surface tension, the structure of the lipid-protein aggregation is disturbed and reinserts into the surface of the monolayer rapidly. On the basis of analyzing the energies change of the squeeze-out process, a mathematical model is obtained to calculate the squeezed-out number of DPPG molecules when a SP-C molecule squeezes out in a monolayer. According to the model, it is concluded that SP-C has the capability to promote the squeeze-out and the reinsertion of most of PG component in an alveolus monolayer, the prediction data agree well with the experimental data.  相似文献

An investigation has been made of the interaction between chlorpromazine (CPZ) and monolayers of 1,2-dipalmitoyl-sn-3-glycerophosphatidylcholine (DPPC) and 1,2-dipalmitoyl-sn-3-glycero[phospho-rac-(1-glycerol)] (DPPG), both at the air/water interface and in transferred Langmuir-Blodgett films. The Gibbs free energy, DeltaG, and the compressibility modulus (C(S)(-1)), obtained from the surface pressure isotherms, indicated changes in the in-plane interactions of CPZ/DPPG mixed monolayers, with positive values of DeltaG. The arrangement of CPZ in the zwitterionic DPPC monolayers causes a weaker interaction in CPZ/DPPC mixed monolayers, with the DeltaG fluctuating around zero. IR measurements in transferred monolayers showed that CPZ did not affect the conformational order of the acyl chains, its effects being limited to the bands corresponding to the headgroups. Furthermore, since no shift was observed for the acyl chain bands, the phase transition induced by CPZ is not a liquid expanded (LE) to liquid condensed (LC) transition, as the latter is associated with chain ordering. Taken together, the IR and compressibility results demonstrate that the effect from CPZ cannot be correlated with temperature changes in the subphase for pure monolayers, in contrast to models proposed by other authors.  相似文献

Pulmonary lung surfactant is a mixture of surfactants that reduces surface tension during respiration. Perfluorinated surfactants have potential applications for artificial lung surfactant formulations, but the interactions that exist between these compounds and phospholipids in surfactant monolayer mixtures are poorly understood. We report here, for the first time, a detailed thermodynamic and structural characterization of a minimal pulmonary lung surfactant model system that is based on a ternary phospholipid-perfluorocarbon mixture. Langmuir and Langmuir-Blodgett monolayers of binary and ternary mixtures of the surfactants 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC), 1,2-dipalmitoyl-sn-glycero-3-phosphoglycerol (DPPG) and perfluorooctadecanoic acid (C18F) have been studied in terms of miscibility, elasticity and film structure. The extent of surfactant miscibility and elasticity has been evaluated via Gibbs excess free energies of mixing and isothermal compressibilities. Film structure has been studied by a combination of atomic force microscopy and fluorescence microscopy. Combined thermodynamic and microscopy data indicate that the ternary monolayer films were fully miscible, with the mixed films being more stable than their pure individual components alone, and that film compressibility is minimally improved by the addition of perfluorocarbons to the phospholipids. The importance of these results is discussed in context of these mixtures' potential applications in pulmonary lung surfactant formulations.  相似文献

Composite monolayers of cadmium stearate and polyaniline processed with camphor sulphonic acid have been obtained at the air–water interface and subsequently transferred onto substrates as uniform Langmuir–Blodgett multilayers. Pressure–area (Π-A) isotherm studies indicated that polyaniline molecules are not squeezed out of the cadmium stearate matrix during compression or in the compressed state. Monolayer stability is seen to decrease when the polymer content is increased beyond 60% (in weight) which is probably associated with the formation of microaggregates that also affected the monolayer surface potential at large areas per molecule. With increasing amount of polyaniline in the mixture, a transition from Y-type to Z-type deposition has been observed. Transferred multilayer LB films were characterized by UV-vis, FTIR, XRD, surface potential, and dc electrical conductivity measurements. UV-vis results indicated that though the polyaniline was in the doped state in the spreading solution, the transferred films were in the emeraldine base state. FTIR studies revealed that the transferred films mainly contained cadmium stearate salt in addition to polyaniline. XRD results indicated that the stacking order is dependent on the polyaniline content in the composite films, the order was found to decrease upon increasing the polymer amount. The uniformity of transferred films was confirmed by surface potential studies. A possible packing arrangement in these composite Langmuir and Langmuir–Blodgett films has been proposed.  相似文献

Lipid rafts being rich in cholesterol and sphingolipids are considered to provide ordered lipid environment in the neuronal membranes, where it is hypothesized that the cleavage of amyloid precursor protein (APP) to Aβ (1-40) and Aβ (1-42) takes place. It is highly likely that the interaction of lipid raft components like cholesterol, sphingomylein or GM1 leads to nucleation of Aβ and results in aggregation or accumulation of amyloid plaques. One has investigated surface pressure-area isotherms of the lipid raft and Aβ (1-40) Langmuir monolayer. The compression-decompression cycles and the stability of the lipid raft Langmuir monolayer are crucial parameters for the investigation of interaction of Aβ (1-40) with the lipid raft Langmuir monolayer. It was revealed that GM1 provides instability to the lipid raft Langmuir monolayer. Adsorption of Aβ (1-40) onto the lipid raft Langmuir monolayer containing neutral (POPC) or negatively charged phospholipid (DPPG) was examined. The adsorption isotherms revealed that the concentration of cholesterol was important for adsorption of Aβ (1-40) onto the lipid raft Langmuir monolayer containing POPC whereas for the lipid raft Langmuir monolayer containing DPPG:cholesterol or GM1 did not play any role. In situ UV-vis absorption spectroscopy supported the interpretation of results for the adsorption isotherms.  相似文献

4-amino-2-phenyl, 6(p-fluor-phenyl)-5-carbonitrile-pyrimidine (APCP) is a new derivative of pyrimidine with low solubility in water and anti-inflammatory properties. We compared the interfacial behaviors of spread films of poly(ethylene glycol)-grafted phospholipid (DSPE-PEG2000), 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC), and APCP and a mixture of these molecules. The surface pressure–area (Π–A) isotherm showed that APCP and DSPE-PEG2000 molecules were stable at the air/water interface and could be evenly inserted into a DPPC floating monolayer. The introduction of APCP into the DPPC/(DSPE-PEG2000) binary monolayer generally causes an overall increase in surface potential. Analyses of distance variation between the grafted sites are associated with a change of mushroom to brush conformation and this behavior is observed for the DPPC/(DSPE-PEG2000) and DPPC/(DSPE-PEG2000)/APCP monolayers. Langmuir–Blodgett (LB) films of molecules of biological interest were transferred onto mica in order to investigate their interaction. AFM images do not show any regular shape or size and are randomly distributed.  相似文献

Langmuir monolayer and Langmuir-Blodgett (LB) films of cyclopalladated ferrocenylimine 1 were prepared and characterized. The surface pressure (π)-area (A) isotherm of complex 1 indicated the formation of highly condensed monolayer on the subphase. Ultraviolet-visible (UV-vis) and Fourier transform infrared (FTIR) spectroscopy showed that complex 1 monolayer was transferred successfully onto solid supports. Atomic force microscopy (AFM) image suggested that LB films transferred on the solid substrate were well-ordered, homogeneous. Cyclic voltammograms of LB films deposited on glassy carbon electrodes showed quasi-reversible oxidation/reduction waves of ferrocene moiety. From the average thickness of monolayer, the hydrocarbon chain could be fairly directed perpendicular to the substrates. Finally, LB films of complex 1 presented a largely improved catalytic efficiency for Suzuki reaction with respect to its cast films and homogeneous reactions under the same conditions. The results might have an implication on the catalytic mechanism of this reaction.  相似文献

A trisilanol polyhedral oligomeric silsesquioxane (POSS), trisilanolcyclohexyl-POSS (TCyP), has recently been reported to undergo a series of phase transitions from traditional Langmuir monolayers to unique rodlike hydrophobic aggregates in multilayer films that are different from "collapsed" morphologies seen in other systems at the air/water interface. This paper focuses on the phase transitions and morphology of films varying in average thickness from monolayers to trilayers and the corresponding viscoelastic properties of trisilanolcyclohexyl-POSS molecules at the air/water interface by means of surface pressure-area per molecule (Pi-A) isotherms, Brewster angle microscopy (BAM), and interfacial stress rheometry (ISR) measurements. The morphology studies by BAM reveal that the TCyP monolayer can collapse into different 3D structures by homogeneous or heterogeneous nucleation mechanisms. For homogeneous nucleation, analysis by Vollhardt et al.'s nucleation and growth model reveals that TCyP collapse is consistent with instantaneous nucleation with hemispherical edge growth at Pi = 3.7 mN.m(-1). Both surface storage (Gs') and loss (Gs") moduli obtained by ISR reveal three different non-Newtonian flow regimes that correlate with phase transitions in the Pi-A isotherms: (A) A viscous liquidlike "monolayer"; (B) a "biphasic regime"between a liquidlike viscous monolayer and a more rigid trilayer; and (C) an elastic solidlike "trilayer". These observations provide interesting insights into collapse mechanisms and structures in Langmuir films.  相似文献

Polyhedral oligomeric silesquioxanes (POSS) with eight polyether substituents were mixed with the liquid crystal (LC) 4-octyloxy-4′-cyanobiphenyl and spread at the air/water interface. The surface pressure-area and surface potential-area isotherms were recorded for different weight ratios of both components. The obtained results showed that POSS molecules had beneficial influence on LC monolayer improving its stability and rigidity. Moreover, it was found that some LC–POSS mixtures collapse reversibly and form multilayer films on the top of LC monolayer. On the other hand, interfacial dilatational and shear rheology indicated decrease of elasticity of the films after mixing. Brewster angle microscopy revealed multilayer structure of the condensed film and formation of net-like structures in the expanded film. These films were successfully transferred on solid substrates using the Langmuir–Blodgett technique. The scanning electron microscopy images confirmed the film deposition and formation of networks by POSS–LC mixtures. These findings may be useful in the fabrication of electronic devices based on LCs.  相似文献

"?Langmuir monolayers and LB films of 4-((s)-2-methylbutoxy)phenyl-(4'-(10-undecen-1-oyloxy)phenyl) methylenimine (MPUOPM) were investigated by ultraviolet-visible, polarized infrared spectroscopy. ?-A isotherms showed well-defined Langmuir monolayers were formed at an air/water interface for the MPUOPM and their mixture with SA. An inflection point at 13 mN/m appeared on the isotherm, which was due to the transition from the monolayer to multilayer. The polarized IR spectra of LB films of MPUOPM had provided new insight into the molecular orientation and structure. In LB films, the tilted angle between the alkyl chain and the normal line of the substrate was 48ffi, the tilted angle between the dipole moment of C=N and the normal line of the substrate was 51ffi. The alkyl chains assumed a trans-zigzag conformation but it included a few gauche conformers. The C=N groups were almost in one plane in the LB films. "  相似文献

The Langmuir and Langmuir-Schaefer (LS) films of two coumarin derivatives, 4-octadecyloxylcoumarin (4-CUMC18) and 7-octadecyloxylcoumarin (7-CUMC18), were newly synthesized, and their interfacial assemblies were investigated. Owing to the different substituent position of the long octadecyloxy chain in the coumarin parent, the two compounds showed completely different behaviors in the interfacial assemblies. When they were spread at the air/water interface, 7-CUMC18 formed a monolayer while 4-CUMC18 formed a multilayer film on the water surface. The spreading films on the water surface were transferred onto solid substrates by a Langmuir-Schaefer method, and the transferred films were characterized by UV-vis, Fourier transfer infrared, X-ray diffraction, circular dichroism, and atomic force microscopy spectra. Different packing of the molecules in the multilayer films was observed. While coumarin groups stacked in a face-to-face way in 7-CUMC18 film, they stacked in a head-to-tail manner in 4-CUMC18 film. Furthermore, distinct properties of the multilayer films were observed. It is revealed that a reversible [2+2] photodimerization and photocleavage could be induced in the LS film of 7-CUMC18 under photoirradiation with UV light of 365 and 254 nm, respectively. No photodimerization occurred in the 4-CUMC18 film. However, the film of 4-CUMC18 showed a supramolecular chirality although the compound itself is achiral.  相似文献

The Langmuir monolayer is a special class of lyotropic liquid crystalline system wherein phase transition essentially depends on surface density, temperature and ion-content in the aqueous medium. The variety of surface phases can be transferred onto devices by the Langmuir–Blodgett (LB) technique. The Langmuir monolayer of pristine single-walled carbon nanotubes (SWCNTs) exhibited gas and liquid-like phases. The LB film of SWCNTs shows target surface pressure dependent interesting morphologies. The methane gas sensing using parallel alignment of SWCNTs was found to be better than that of randomly oriented SWCNTs. The SWCNTs can be functionalised chemically to enhance the ease of film processability and affinity towards analytes. These are essential parameters for the development of a sensor. In this article, we present our work on Langmuir monolayer and LB films of octadecylamine functionalised SWCNTs (ODACNTs) and its sensing application towards bio-analytes, e.g. L-aspartic acid and bovine serum albumin. The sensing performance of LB film of ODACNTs was compared with that of spin-coated films of ODACNTs. The sensing performance of LB films of ODACNTs indicated a potential platform for bio-sensing application.  相似文献
