A new procedure is developed for the extraction of polycyclic aromatic hydrocarbons (PAHs) from the particulate phase of cigarette smoke. The procedure applies solid-phase extraction using a Bond Elut CH cartridge as a sample preparation step. The efficiency of the cleanup procedure is verified using a gas chromatographic (GC)-high-resolution mass spectrometric (MS) technique, proving that no interference occurs in the PAHs' determination. The efficient cleanup allows GC detection using either high- or low-resolution MS detection. Enhanced sensitivity is obtained using GC-MS and selected ion monitoring. This new technique has several advantages over other reported techniques. The method is simple and robust and has good repeatability and accuracy. The estimated detection limit is 0.1 ng/cigarette for benzo[a]pyrene. In addition to that, the recovery from the smoke pad in which the smoke is collected is approximately 97% for all PAHs. Results for the PAH analyses for 1R5F, 1R4F, and 1R3 Kentucky reference cigarettes are reported in this study. These results provide useful evidence for clarifying the controversy about previously reported data.  相似文献

A fast and effective sample cleanup procedure for the quantification of polycyclic aromatic hydrocarbons (PAHs) in fish samples is presented. The procedure involved extraction of fish samples using acetonitrile and cleanup by an automated gel-permeation chromatography (GPC) following liquid?Cliquid partition into n-hexane. The extracted samples were analyzed by gas chromatography-mass spectrometry (GC?CMS). Electron ionization was employed in a single analysis for the determination of PAHs in the selected ion monitoring mode. Three different solvents were studied for the extraction step: acetonitrile/n-hexane, methanol/n-hexane and acetone/n-hexane. The best solvent was found to be acetonitrile/n-hexane. The cleanup technique resulted in a good separation of analytes from co-extractive matrix compounds.  相似文献

建立高效液相色谱–串联质谱法(HPLC–MS/MS)测定动物源性食品中玉米赤霉烯酮及其5种代谢产物(α-玉米赤霉烯醇、β-玉米赤霉烯醇、α-玉米赤霉醇、β-玉米赤霉醇、玉米赤霉酮)残留量。在样品中加入4种同位素内标(13C18–玉米赤霉烯酮,D7–α-玉米赤霉烯醇,D7–β-玉米赤霉烯醇,D5–α-玉米赤霉醇)后,经β-葡萄糖苷酶/硫酸酯酶酶解,用叔丁基甲基醚萃取,取上清液氮吹至近干后用三氯甲烷复溶,再用氢氧化钠溶液反向萃取,以HLB固相萃取柱净化后,用HPLC–MS/MS检测。结果表明,玉米赤霉烯酮及其代谢产物在1.0~100.0μg/L范围内线性关系良好,相关系数均大于0.996,方法的检出限为0.04~0.13μg/kg,定量限为0.11~0.43μg/kg。在1.0、4.0、10.0μg/kg三种加标浓度水平下,回收率为77.7%~105.5%,测定结果的相对标准偏差为4.8%~9.8%(n=6)。该方法准确、可靠,灵敏度高,适用于动物源性食品中玉米赤霉烯酮及其代谢产物的定量分析。  相似文献

An in-line matrix cleanup method was used for the simultaneous extraction of 15 sulfonamides and two metabolites from manure samples. The ultrasound/microwave-assisted extraction (UMAE) combined with solid–liquid–solid dispersive extraction (SLSDE) procedure provides a simple sample preparation approach for the processing of manure samples, in which the extraction and cleanup are integrated into one step. Ultrasonic irradiation power, extraction temperature, extraction time, and extraction solvent, which could influence the UMAE efficiency, were investigated. C18 was used as the adsorbent to reduce the effects of interfering components during the extraction procedure. The extracts were concentrated, and the analytes were analyzed by liquid chromatography–tandem mass spectrometry (LC–MS/MS) without any further cleanup. The isotopically labeled compounds sulfamethoxazole-d 4, sulfamethazine-d 4, sulfamonomethoxine-d 4, and sulfadimethoxine-d 6 were selected as internal standards to minimize the matrix effect in this method. The recoveries of the antibiotics tested ranged from 71 to 118 % at the three spiking levels examined (20, 200, and 500 μg?·?kg-1). The limits of detections were 1.2–3.6 μg?·?kg-1 and the limits of quantification were 4.0–12.3 μg?·?kg-1 for the sulfonamides and their metabolites. The applicability of the method was demonstrated by analyzing 30 commercial manure samples. The results indicated that UMAE–SLSDE combined with LC–MS/MS is a simple, rapid, and environmentally friendly method for the analysis of sulfonamides and their metabolites in manure, and it could provide the basis for a risk assessment of the antibiotics in agricultural environments.  相似文献

Pesticides are widely applied to protect plants from diseases, weeds, and insect damage, and they usually come into contact with soil where they may undergo a variety of transformations and provide a complex pattern of metabolites. Spreading sewage sludge on agricultural lands has been actively promoted by national authorities as an economic way of recycling. However, as a byproduct of wastewater treatment, sewage sludge may contain pesticides and other toxic substances that could be incorporated into agricultural products or be distributed in the environment. This article reviews the determination of pesticides in sewage sludge samples. Sample preparation including pretreatment, extraction, and cleanup, as well as the subsequent instrumental determination of pesticide residues, are discussed. Extraction techniques such as Soxhlet extraction, ultrasound-assisted extraction, pressurized liquid extraction, supercritical fluid extraction, and matrix solid-phase dispersion and their most recent applications to the determination of pesticides in sewage sludge samples are reviewed. Determination of pesticides, generally carried out by GC and HPLC coupled with different detectors, especially MS for the identification and quantification of residues, is summarized and discussed.  相似文献

A multiresidue method was developed for screening, quantification, and confirmation of nine natural androgen steroids and their metabolites in urine. Steroids were first extracted from urine by solid phase extraction, enzymatically deglucuronated, re-extracted using a liquid/liquid extraction for purification, and finally acetylated for GC/MS and GC/FID analysis. Each step of sample preparation, as well as analysis, was optimized: solid phase extraction, liquid/ liquid extraction, and derivatization reaction … Therefore, a rugged sample preparation procedure was developed leading to extracts of sufficient purity (recoveries >66% and few matrix compounds). The whole methodology allowed reliable detection and quantification of the nine steroids at low concentration levels. Linearity and repeatability were established and were found to be satisfactory (R2 > 0.996, RSD < 11%). Finally, the method was applied to quantify compounds of interest in real samples collected from healthy volunteers and patients treated with 4-androstenedione or dehydroepiandrosterone.  相似文献

An analytical procedure for the simultaneous determination of twelve endogenous steroids (testosterone, androstenedione, 17β-estradiol, estrone, pregnenolone, progesterone, dihydroandrostenedione, dihydrotestosterone, 11α-ketotestosterone, 17α-hydroxyprogesterone, 17α-hydroxypregnenolone, 17α,20β-dihydroxy-4-pregnen-3-one) in plasma and bile samples by solid-phase extraction (SPE) and gas chromatography/mass spectrometry (GC–MS) has been developed. After enzymatic hydrolysis for bile samples only, samples were concentrated and purified using two successive SPE (C18 and NH2) cartridges. Analytes were derivatized with a mixture of N-methyl-N-(trimethylsilyl)trifluoroacetamide (MSTFA) / mercaptoethanol / ammonium iodide (NH4I) and determined by GC–MS in selective ion monitoring mode. For most of the steroids monitored, recoveries were in the range 90–120% in plasma and in the range 60–70% in bile, and the reproducibility was below 10% for the complete procedure. Limits of detection obtained ranged from 0.1 to 0.4 ng/g in fish plasma and from 1.6 to 14 ng/g in fish bile. The developed method was successfully applied to the determination of plasma steroids in flounders (Platichthys flesus) collected from two French estuaries.  相似文献

An extraction procedure based on the Quick Easy Cheap Effective Rugged and Safe (QuEChERS) approach has been developed and used for analysis of particle-bound nitrated and oxygenated PAH derivatives (NPAH and OPAH, respectively). Several analytical conditions, for example GC injection temperature and MS detection settings, were optimized. This analytical procedure enabled simultaneous GC–NICIMS quantification of 32 NPAH and 32 OPAH (or other oxygenated compounds), including typical components of secondary organic aerosol (SOA) formed by photooxidation of PAH (e.g. 2-formyl-trans-cinnamaldehyde and 6H-dibenzo[b,d]pyran-6-one). The QuEChERS-like approach was optimized, including the nature of the extraction solvent, the sorbent used for clean-up, and extraction time. The final extraction procedure was based on brief mechanical agitation (vortex mixing for 1.5 min), with 7 mL acetonitrile as solvent. Because dispersive solid-phase extraction (d-SPE) did not provide satisfactory results, SPE using SiO2 was selected for sample purification. Identical results were obtained when the QuEChERS-like and traditional pressurised solvent extraction (PLE) procedures were compared for analysis of fortified ambient air particle samples. The procedure was validated by analysis of two aerosol standard reference materials (NIST SRM 1649b (urban dust) and SRM 2787 (fine particulate matter, <10 μm)). For numerous NPAH and OPAH, this is the first report of their quantification in both SRMs. Compared with other extraction methods, including PLE, the QuEChERS-like procedure resulted in increased productivity and reduced extraction cost. This paper shows that QuEChERS-like extraction procedures can be suitably adapted for molecular chemical characterization of aerosol samples and could be extended to other categories of compound.

The state-of-the-art of analysis for acrylamide in food is reviewed. The majority of analytical methods adopts a similar approach: addition of internal standard to the specimen, extraction with water, purification of extract using a solid-phase extraction cartridge, and then determination using either gas chromatography coupled to mass spectroscopy (GC/MS) after bromination, or direct measurement with liquid chromatography coupled to mass spectroscopy (LC/MS). The available methods generally show good agreement and are likely to be accurate. However, improvements in precision (within-laboratory) and repeatability (between-laboratory) are needed by particular data users.  相似文献

An efficient and sensitive multiresidue method has been developed for quantification and confirmation of 25 phenyl acetanilide pesticides in a wide variety of food commodities including maize, spinach, mushroom, apple, soybean, chestnut, tea, beef, cattle liver, chicken, fish, and milk. Analytes were extracted with acetone–n‐hexane (1:2, v/v) followed by cleanup using SPE. Several types of adsorbents were evaluated. Neutral aluminum and graphitized carbon black cartridge showed good cleanup efficiency. The extract was determined by GC‐MS in the selected ion monitoring mode using one target and two qualitative ions for each analyte. The limits of detection were 0.01 mg/kg for all analytes. The average recoveries ranged from 66.9 to 110.6% (mean 88.8%) and RSDs were in the range 2.0–19% (mean 10.5%) across three fortification levels. The proposed method was successfully applied to real samples in routine analysis and a satisfactory result was obtained.  相似文献

In response to recent discoveries of acrylamide in heated foods, a solid-phase extraction and cleanup protocol was developed for the determination of acrylamide in fried or baked potato samples by liquid chromatography/mass spectrometry (LC/MS). The analyte was extracted from the matrix by using 2M NaCl, and an aliquot of the initial extract was loaded onto a reversed-phase cartridge. After the analyte was eluted from the cartridge, the eluate was cleaned up on a mixed-mode cation-exchange cartridge. The eluate was then evaporated, and the residue was reconstituted in mobile phase before LC/MS analysis. Recoveries, based on the recovery of an added internal standard, ranged from 96 to 101% with relative standard deviations (RSDs) of 5-11%. The response was linear for a concentration range of 100-2000 ng/g with a coefficient of determination (R2) of 0.992 (n = 25). An interday study showed good accuracy and precision of the method over a 3-day period with a recovery of 98% and an RSD of 9.5% (n = 15). The analyses of 6 potato chip samples showed concentrations of incurred acrylamide ranging from 260 to 1500 ng/g.  相似文献

An original and highly specific method able to identify and quantify traces of five tetracycline antibiotics (TCAs) in milk and eggs is presented. This method uses a single solid-phase extraction (SPE) cartridge for simultaneous extraction and purification of TCAs in the above matrices. After diluting 5 mL of intact whole milk or 2 g egg samples with Na(2)EDTA-containing water, samples are passed through a 0.5-g Carbograph 4 extraction cartridge. After analyte elution from the SPE cartridge, an aliquot of the final extract is injected into a liquid chromatography/mass spectrometry (LC/MS) instrument equipped with an electrospray ion source and a single quadrupole. MS data acquisition is performed in the positive-ion mode and by a time-scheduled multiple-ion selected ion-monitoring program. With methanol as organic modifier, the in-source collision-induced dissociation (CID) process generated fragment ions able to pick up one methanol molecule. In several cases, these methanol-adduct fragment ions have m/z values higher than those of the protonated molecules. This event is rarely encountered in MS, thus making the analysis of TCAs by this method extremely specific. Compared with a conventional published method, the present protocol extracted larger amounts of TCAs from both milk and egg and decreased the analysis time by a factor of 3. Recovery of TCAs in milk at the 25-ppb level ranged between 81 and 96% with relative standard deviation (RSD) no larger than 9%. Recovery of TCAs in egg at the 50-ppb level ranged between 72 and 92% with RSD no larger than 7%. Estimated limits of quantification(S/N = 10) of the method were 2-9 ppb TCAs in whole milk and 2-19 ppb TCAs in eggs.  相似文献

Picroside II is one of the main active constituents of Picrorhiza kurroa, which has hepatoprotective, anticholestatic, antioxidant, and immune-modulating activity. To gain an understanding of the biotransformation of picroside II in vivo, liquid chromatography?Celectrospray ionization ion-trap mass spectrometry (LC?CESI?CIT?CMS) was used to investigate the metabolism of picroside II in rats after intravenous administration of a single dose. This method could simultaneously determine picroside II and its metabolites in rat bile. The bile samples were purified by use of a C18 solid-phase extraction (SPE) cartridge and were separated on a Hypersil ODS2 C18 analytical column. Two phase II metabolites of picroside II in rat bile were characterized, and elucidation of their structures was performed by comparing changes in molecular masses (??M), retention times, and MS2 spectral patterns of metabolites with those of the parent drug. Two metabolites identified for the first time in this research were glucuronide and sulfate conjugates.  相似文献

A method is described for the determination of 251 pesticide and degradation product residues in fruit and vegetable samples. Extraction of the sample with acetonitrile is followed by a salting-out step. Co-extractives are removed by passing a portion of the acetonitrile extract through an octadecyl (C18) solid-phase extraction cleanup cartridge and then, in a second cleanup, through a carbon cartridge coupled to an amino propyl cartridge. Determination is by gas chromatography with mass-selective detection in the selected-ion monitoring mode, and by liquid chromatography with post-column reaction and fluorescence detection for N-methyl carbamates. The method has been used for analysis of various fruits and vegetables, such as apple, banana, cabbage, carrot, cucumber, lettuce, orange, pear, pepper, and pineapple. Limits of detection range between 0.02 and 1.0 mg/kg for most compounds. Over 80% of the compounds have a limit of detection of < or = 0.04 mg/kg.  相似文献

A new procedure has been developed for the quantitation of aromatic amines in mainstream cigarette smoke. Two solid-phase extraction (SPE) cleanup steps using different retention mechanisms are required to process the samples. The first step uses a cation-exchange cartridge, followed by a second step that uses a cartridge with a hydrophobic retention character. The aromatic amines eluted from the second SPE cartridge are derivatized with heptafluorobutyric anhydride and analyzed with GC-MS selected ion monitoring in the negative chemical ionization mode. This new method has several advantages over other reported techniques, being sensitive, robust, and easily automated. The detection limits ranged from 0.02 ng/cigarette for tolidine to 1.41 ng/cigarette for aniline and the recoveries were from 79 to 109%.  相似文献

A liquid chromatographic method with comfirmation by capillary electrophoresis was used to determine 12 sulfonylurea herbicides in agricultural water. Analysis of 3 different water matrixes fortified at 2 levels gave good recoveries with adequate sensitivity at the 0.1 ppb level. A portion of the water was acidified with acetic acid and loaded onto an RP-102 solid-phase extraction (SPE) cartridge, and the extract was cleaned up on an alumina SPE cartridge. Extracts were desalted with an RP-102 SPE cartridge before instrumentation. Samples needing chemical filtration, such as pond water, required additional cleanup with a SAX SPE cartridge before the alumina cleanup step. Data were compiled for both determinative techniques and evaluated.  相似文献

This work reports a rapid, reliable and sensitive multi-residue method for the simultaneous determination of six resorcylic acid lactones in bovine milk by ultra-high-pressure liquid chromatography–tandem mass spectrometry (UHPLC–MS/MS). The resorcylic acid lactones were extracted, purified, and concentrated from milk samples in one step using a solid-phase extraction (SPE) cartridge that contained a polymeric mixed-mode anion-exchange sorbent. The analysis was performed on a Waters Acquity BEH C18 column utilizing a gradient elution profile. Each LC run was completed in 3.5 min. The analytes were detected by multiple reaction monitoring (MRM) using electrospray ionization (ESI) negative mode. Mean recoveries from fortified samples ranged from 92.6% to 112.5%, with relative standard deviations lower than 11.4%. Using 5 mL bovine milk, the limits of detection and quantification for resorcylic acid lactones were in the ranges of 0.01–0.05 and 0.05–0.2 μg/L, respectively. The application of this newly developed method was demonstrated by analyzing bovine milk samples from markets.  相似文献

A solid-phase extraction (SPE) and liquid chromatographic (LC) method was developed for the determination of selected fluoroquinolone (FQ) drugs including ofloxacin, norfloxacin, and ciprofloxacin in municipal wastewater samples. Extraction of the FQs was carried out with a weak cation exchanger SPE cartridge, the Oasis WCX. The cartridge was washed with water and methanol as a cleanup before the FQs were eluted by a mixture of methanol, acetonitrile, and formic acid. Separation of the FQs was achieved by using a Zorbax SB-C8 column under isocratic condition at a flow rate of 0.2mL/min. Recoveries of the FQs in spiked final effluent samples were between 87 and 94% with a relative standard deviation of less than 6%. Several techniques have been evaluated for the detection of FQs in sewage extracts; they included fluorescence detection and electrospray ionization (ESI) mass spectrometry using either mass-selective detection or tandem mass spectrometry (MS/MS). When they were applied to sewage influent and effluent samples, the LC-MS/MS technique operating in the multiple reaction monitoring (MRM) mode proved to be best suited for the determination of FQs in sewage samples as it provided the highest sensitivity (limit of quantification 5ng/L) and selectivity. The observation of signal suppression (matrix effect) for some FQs in ESI LC-MS and LC-MS/MS is discussed and a solution is proposed. The three FQs were detected in all the sewage samples tested in this work, with median concentrations between 34 and 251ng/L.  相似文献

A new and reliable two‐step liquid chromatography/tandem mass spectrometry (LC/MS/MS) method in combination with gas chromatography/mass spectrometry (GC/MS) for the screening and confirmation of adrafinil and its major metabolites, modafinil and modafinil acid, in human urine has been developed and validated. The method involved reversed‐phase C18 solid‐phase extraction (SPE) cartridge extraction and MS analysis by means of LC/MS/MS and GC/MS. The study illustrated that the ESI capillary temperature played a key role in the formation of the protonated molecule. The limits of detection (LODs) of the developed method for the three compounds were lower than the minimum required performance limit (MRPL) of the World Anti‐Doping Agency (WADA). The human urine samples obtained after the oral administration of modafinil and from the Beijing 2008 Olympic Games were analyzed by using the described method, which has also been successfully applied to routine analyses and the WADA Proficiency Test. This review deals with analytical methods for polycyclic aromatic hydrocarbon (PAH) determination in oils and fats. The data reported in the introduction deal with PAH dietary intake from this group of food and contamination levels recently found by some authors in different vegetable oils, stressing the importance of establishing a method suitable for routine analyses. Traditional sample preparation relies on tedious, time-consuming procedures. They generally consist of an extraction step (liquid-liquid partition, caffeine complexation, saponification) followed by one or more purification procedures (column chromatography, thin-layer chromatography, solid-phase extraction). The analytical determination is usually carried out by HPLC and spectrofluorometric detection, or through high-resolution capillary GC coupled to flame ionisation detection or mass spectrometry. LC is a valid alternative to the traditional sample preparation, and off-line LC-LC allows performing an accurate PAH analysis in less than 2 h. Also supercritical fluid extraction, allowing performing both extraction and clean-up in one combined step, is a promising technique. Hyphenated techniques such as LC-GC and LC-LC-GC seem to be very promising. A completely on-line method for alkylated PAH determination in oils or lipidic extracts contaminated with mineral oil involves a two-dimensional LC-step with intermediate eluent evaporation and GC transfer through a vaporiser/overflow interface.  相似文献
