A former spectroscopic study of Cu(II) coordination by the 13-membered ring cyclic tetrapeptide c(Lys-dHis-βAla-His) (DK13), revealed the presence, at alkaline pH, of a stable peptide/Cu(III) complex formed in solution by atmospheric dioxygen oxidation. To understand the nature of this coordination compound and to investigate the role of the His residues in the Cu(III) species formation, Cu K-edge XANES, and EXAFS spectra have been collected for DK13 and two other 13-membered cyclo-peptides: the diastereoisomer c(Lys-His-βAla-His) (LK13), and c(Gly-βAla-Gly-Lys) (GK13), devoid of His residues. Comparison of pre-edge peak features with those of Cu model compounds, allowed us to get information on copper oxidation state in two of the three peptides, DK13 and GK13: DK13 contains only Cu(III) ions in the experimental conditions, while GK13 binds only with Cu(II). For LK13/Cu complex, EXAFS spectrum suggested and UV-vis analysis confirmed the presence of a mixture of Cu(II) and Cu(III) coordinated species. Theoretical XANES spectra have been calculated by means of the MXAN code. The good agreement between theoretical and experimental XANES data collected for DK13, suggests that the refined structure, at least in the first coordination shell around Cu, is a good approximation of the DK13/Cu(III) coordination species present at strongly alkaline pH. All the data are consistent with a slightly distorted pyramidal CuN(4) unit, coming from the peptide bonds. Surprisingly, the His side-chains seemed not involved in the final, stable, Cu(III) scaffold.  相似文献

In electron-transfer reactions, the change in the oxidation states of the reactants is generally accompanied by structural changes, which influence the electron-transfer kinetics. Previous studies on the systems of Cu(II)/(I) complexes involving cyclic tetrathiaether ligands indicated that inversion of coordinated donor atoms is a major geometric change during the overall electron-transfer process. Complex formation and isomerization studies on complexes with the 1,4,8,11-tetraazacyclotetradecane ligand have demonstrated that a necessary condition for conformational change is deprotonation followed by inversion of coordinated N atoms. When one or more nitrogen donor atoms in a ligand are replaced with sulfur, there is a choice of N or S inversion. It has been hypothesized that donor atom inversion (N or S donors) is a major factor that can lead to conformationally limited electron-transfer kinetics of copper systems. In the current study, the thermodynamic properties, electron-transfer kinetics and conformational changes in copper(II)[1,4,8-trithia-11-azacyclotetradecane], copper(II)[1,8-dithia-4,11-diazacyclotetradecane] and copper(II)[1,11,-dithia-4,8-diazacyclotetradecane] were determined in order to determine the effect of inversion of coordinated N atoms on electron-transfer rates as a function of low concentrations of water in an aprotic solvent (acetonitrile). By using controlled amounts of water as a hydrogen ion acceptor, deprotonation of amine nitrogen and nitrogen donor inversion was followed by comparing self-exchange rate constants for reduction and oxidation of the copper complexes. Data on thermodynamic properties and electron-transfer kinetics are presented. Possible conformational changes and kinetic pathways for complexes with ligands having mixed N and S donor sets are presented.  相似文献

The reduction of the title complexes was studied by cyclic voltammetry in aqueous media. It proceeds through a one-electron process generating intermediate copper(I) dipeptide complexes. The copper(I) dipeptide complexes are found to be short-lived and undergo transformations eventually generating Cu0 at the mercury electrode. The unchanged fraction of the copper(I) species is re-oxidised to the copper(II) complexes. The Cu0 generated undergoes a two-electron oxidation at a more anodic potential than the copper(I) complexes. pH-dependence of the title complexes is also investigated by cyclic voltammetry.  相似文献

We here describe the first metal complex system in which electronic signals can be repeatedly extracted by converting bistable states related to an intramolecular ligand rotational motion, which is fueled by visible light. The molecular structure for relating an electron transfer and a motion consists of a copper center and a coordinated unsymmetrically substituted pyrimidine derivative, whose rotational isomerization causes an electrochemical potential shift. To harness light energy effectively through metal-to-ligand charge transfer (MLCT) excitation, we prepared a simple copper(I) complex coordinated by a 4-methyl-2-(6'-methyl-2'-pyridyl)pyrimidine and a bulky diimine. The thermodynamic and kinetic parameters of redox and rotational reactions were analyzed by cyclic voltammograms at variable temperatures, by considering four stable isomers related to copper(II)/(I) states and rotational isomeric states. The key feature of this compound is that the rotation is frozen in the copper(I) state (rate constant for the rotation, k(Ii→o) = 10(-4) s(-1)) but is active in the copper(II) state (k(IIi→o) = 10(-1) s(-1)) at 203 K. The compound makes a bypass route to the isomeric metastable copper(I) state, via a tentative copper(II) state formed by photoelectron transfer (PET) in the presence of a redox mediator, decamethylferrocenium ion (DMFc(+)), or upon a partial oxidation of the complex. Light- and heat-driven rotation in the copper(I) state with a potential shift (ΔE°' = 0.14 V) was analyzed by electrochemical measurements of the complex in the solution state. The rotor could be reset to the initial state by heating, thereby completing the cycle and enabling repeated operation fueled by light energy. A significant redox potential shift associated with the copper(II)/(I) transition accompanied the rotation, thereby providing a new type of molecular signaling system.  相似文献

The reaction of an O-confused porphyrin with a pendant pyrrole 4 and copper(II) acetate yields an organocopper(III) diamagnetic complex 4-Cu(III) substituted at the C(3) position by the pyrrole and H. The transformation of 4-Cu(III), performed in aerobic conditions, gave a rare copper(II) organometallic compound 6-Cu(II). In the course of this process, the tetrahedral-trigonal rearrangement originated at the C(3) atom but effects the whole structure. The electron paramagnetic resonance spectroscopic features correspond to a copper(II) oxidation state. A crystallographic analysis of 6-Cu(II) confirmed the formation of a direct metal-C bond [Cu(II)-C 1.939(4) A]. It was found that the Cu(II) complex of O-confused oxaporphyrin is sensitive to oxidative conditions. The degradation of 6-Cu(II) to yield copper(II) tripyrrinone complexes has been observed, which was considered as a peculiar case of dioxygen activation in a porphyrin-like environment. This process is accompanied by regioselective oxygenation at the inner C to form the 2-oxa-3-(2'-pyrrolyl)-21-hydroxycarbaporphyrinatocopper(II) complex ((pyrr)OCPO)CuII (8). The reaction of 6-Cu(II) with hydrogen peroxide, performed under heterophasic conditions, resulted in quantitative regioselective hydroxylation centered at the internal C(21) atom, also producing 8. Treatment of 8 with acid results in demetalation to form the nonaromatic 21-hydroxy O-confused porphyrin derivative ((pyrr)OCPOH)H (9).  相似文献

An important function of many copper-containing proteins is activation of O2 and subsequent substrate oxidation. The Cu (III) oxidation state is generally considered to be less accessible because of the highly positive Cu (III)/Cu (II) redox potentials with typical amino acid ligands. Here, we employ density functional (DFT) calculations to explore to what extent copper (III) may be accessed in a biologically-relevant coordination environment around a mononuclear copper center, by breaking the oxygen-oxygen bond in a copper-(hydro) peroxide complex. In agreement with previous findings by Solomon and co-workers on copper models with related coordination patterns, the formally high-valent copper complex produced by O-O bond cleavage appears to harbor both oxidizing equivalents on the ligands. The potential energy surface for such a reaction reveals that with the three-histidine binding motif at the copper, O-O bond cleavage is not impossible, but rather disfavored thermodynamically.   相似文献

The oxidation of cyanide with hexacyanoferrate(III) is a thermodynamically possible but kinetically slow reaction, which is catalysed by copper(II). The catalysed reaction has a second-order dependence on hexacyanoferrate(III) concentration, and the pseudo second-order rate constant increases linearly with the copper concentration, at least in the range from 10(-7) to 10(-3)M.  相似文献

A very sensitive catalytic spectrophotometric method for the determination of copper(II) concentrations as low as 0.06 ng/ml is described. This method is based on the oxidation of 3-hydroxy-2-naphthoic acid by hydrogen peroxide in ammoniacal medium, catalysed by copper(II). The results of an exhaustive study of interferences are given. The new method is applied to determination of copper in atmospheric aerosol samples. A qualitative study of the mechanism by means of electron paramagnetic resonance spectroscopy is also included.  相似文献

In this study, new chalcone compound 1 , new phthalonitrile derivatives 2 and 3, new copper(II), manganese(III) phthalocyanines bearing chalcone groups at peripheral or nonperipheral positions were synthesized. Electrochemistry of tetra-(4-{(2 E )-3-[2-fluoro-4-(trifluoromethyl)phenyl]prop-2-enoyl}phenoxy) substituted Co(II)Pc and Mn(III)Pcs were studied with cyclic voltammetry (CV) to determine the redox properties of the phthalocyanines. According to the results, while the CuPcs 2a and 3a showed two Pc based reduction reactions and one Pc based oxidation reaction, MnPcs 2b and 3b gave two metal-based reduction reactions. All the redox processes are shifted toward positive potentials as a result of the increased electron-withdrawing ability of the trifluoromethyl substituents.  相似文献

Metal-catalyzed oxidation (MCO) of proteins is mainly a site-specific process in which one or a few amino acids at metal-binding sites on the protein are preferentially oxidized. The oxidation of proteins by MCO can lead to oxidation of amino acid residue side chains, cleavage of the peptide bonds and formation of covalent protein-protein cross-linked derivatives. In an attempt to elucidate the products of the copper(II)-catalyzed oxidation of the 29-56, M29-D30-56 and Ac-M29-D30-56 fragments of alpha-synuclein, high performance liquid chromatography (HPLC) and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) methods and Cu(II)/hydrogen peroxide as a model oxidizing system were employed. The peptide solution (0.50 mM) was incubated at 37 degrees C for 24 h with metal : peptide : hydrogen peroxide 1 : 1 : 4 molar ratio in phosphate buffer, pH 7.4. Oxidation targets for all studied peptides are the histidine residues coordinated to the metal ions. For the M29-D30-56 and Ac-M29-D30-56 peptides the oxidation of the methionine residue to methionine sulfoxide and sulfone is observed. The cleavage of the peptide bond M29-D30 for the M29-D30-56 peptide was detected as metal binding residues. The fragmentations of the M29-D30-56 peptide near the Lys residues were observed supporting the participation of this (Lys) residue in the coordination of the copper(II) ions.  相似文献

A new series of binuclear copper(II) complexes were synthesised and studied by magnetic, spectral, ESR and cyclic voltammetry methods. The μeff values per copper atom correspond to the values observed for mononuclear copper(II) complexes. ESR spectral data in solution indicate weak interactions resulting from the electron delocalisation through the ligand system. Two nearly reversible red-ox couples are identified at +?0.50 V and +?0.75 V vs SCE. They correspond to Cu(II)αCu(III) red-ox processes, successively occurring at the two copper sites in the binuclear complexes.  相似文献

A series of copper(II) and nickel(II) 1,3-bis(thiosemicarbazonato) complexes have been synthesised by the reaction of the metal acetates with pyrazoline proligands. In each case the complexes have an overall neutral charge with a dianionic ligand. The copper 1,3-bis(4-methyl-3-thiosemicarbazonato complex has been characterised by X-ray crystallography, which shows the copper is in an essentially square-planar symmetric N(2)S(2) environment. The nickel 1,3-bis(4-methyl-3-thiosemicarbazonato) and nickel 1,3-bis(4-phenyl-3-thiosemicarbazonato) complexes have been characterised by X-ray crystallography and show that in these cases the nickel is in a distorted square-planar environment, but the bonding mode of the ligands is unusual; the nickel binds to one of the aza-methinic nitrogen atoms and one hydrazinic nitrogen, creating one five-membered N-N-C-S-Ni chelate ring and one four-membered N-C-S-Ni chelate ring. Interestingly, the X-ray structure of the ethyl analogue [1,3-bis(4-ethyl-3-thiosemicarbazonato)nickel(II)] shows that in this case the nickel is symmetrically coordinated in the usual manner. The nickel complexes are diamagnetic and the different coordination modes are confirmed in solution by NMR spectroscopy. The complexes are susceptible to oxidation in air and a nickel complex, in which the central methylene carbon has been oxidised, has been characterised by X-ray crystallography and NMR spectroscopy. Electrochemical measurements show that the copper complexes undergo a reversible one-electron reduction at biologically accessible potentials.  相似文献

Self-decomposition of the nickel(III) doubly deprotonated peptide complex of Gly2HisGly occurs by base-assisted oxidation of the peptide. At < or =p[H+] 7.0, the major pathway is a four-electron oxidation (via 4 Ni(III) complexes) at the alpha carbon of the N-terminal glycyl residue. The product of this oxidation is oxamylglycylhistidylglycine, which hydrolyzes to yield ammonia and oxalylglycylhistidylglycine. Both of these peptide products decompose to give isocyanatoacetylhistidylglycine. A small amount (2%) of oxidative decarboxylation also is observed. In another major pathway above p[H+] 7.0, two Ni(III)-peptide complexes coordinate via an oxo bridge in the axial positions to form a reactive dimer species. This dimer generates two Ni(II)-peptide radical intermediates that cross-link at the alpha carbons of the N-terminal glycyl residues. In 0.13 mM Ni(III)-peptide at p[H+] 10.3, this pathway accounts for 60% of the reaction. The cross-linked peptide is subject to oxidation via atmospheric O2, where the 2,3-diaminobutanedioic acid is converted to a 2,3-diaminobutenedioic acid. The products observed at 相似文献

It is based on the differential reaction kinetics of iron(II) and iron(III) with Tiron in a double-injection FI system. The proposed method employs the accelerating action of copper(II) for the oxidation of iron(II) in the presence of Tiron. Copper(I) complexes of the ligand cis-1,3,5-tris(cinnamylideneamino)cyclohexane (L) have been prepared from a versatile precursor complex, [Cu(I)(L)NCMe]BF4, which incorporates a labile acetonitrile ligand that can be exchanged to give a range of new Cu(L)X complexes (where X = Cl, Br, NO2, SPh). 1H NMR spectra and X-ray structures of the Cl, Br and NO2 complexes show L coordinated in a symmetric fashion about the copper centre. The complexes have been further characterised using UV/Visible spectroscopy and cyclic voltammetry. CuLCl shows an electrochemically reversible Cu(I/II) redox couple at 0.51 V (vs. Ag/AgCl) while the CuLNO2 complex shows an analogous quasi-reversible wave at 0.41 V (vs. Ag/AgCl).  相似文献
