Three novel complexes, [Zn (tib)2·(H2O)2]·(NO3)2 ( 1 ), [Co (tib)2]·2NO3 ( 2 ) and [Co2(tib)2(btc)]·H2O ( 3 ) [H4btc = 1,2,4,5‐benzenetetracarboxylic acid; H2tib = 1,3,5‐tris(1‐imidazolyl)benzene], were synthesized and characterized by single‐crystal X‐ray, IR and elemental analysis. The interaction of these complexes with FS‐DNA (fish sperm DNA) was monitored, and binding constants were determined using UV/Vis, which revealed that they have the ability to bind to FS‐DNA. DNA‐binding constants (K) for the three complexes were 2.2 × 104 m ?1, 0.7 × 104 m ?1 and 0.09 × 104 m ?1, respectively. The interaction capacity of the complexes with FS‐DNA has been investigated by fluorescence spectroscopy. Stern–Volmer quenching plot values for complexes 1 , 2 and 3 were 0.3784, 0.1028 and 0.076, respectively. The viscosity measurement suggested that complexes 1 , 2 and 3 interact with DNA in an intercalation mode. In addition, anti‐cancer activities of these complexes investigated through MTT assays in vitro indicated that the complexes showed good cytotoxic activity against cancer cell lines. Cytotoxic activity of test complexes against two different cancer cell lines (HeLa and KB cells) showed significant cancer cell inhibition rates. Flow cytometry experiments and morphological apoptosis studies showed that the complexes induced apoptosis of HeLa tumor cell lines. Finally, a further molecular docking technique was employed to confirm the binding of the complexes toward the molecular target DNA.  相似文献

Two water‐soluble 6‐(pyrazin‐2‐yl)‐1,3,5‐triazine‐2,4‐diamino (pzta)‐based Cu(II) complexes, namely [Cu(l ‐Val)(pzta)(H2O)]ClO4 ( 1 ) and [Cu(l ‐Thr)(pzta)(H2O)]ClO4 ( 2 ) (l ‐Val: l ‐valinate; l ‐Thr: l ‐threoninate), were synthesized and characterized using elemental analyses, molar conductance measurements, spectroscopic methods and single‐crystal X‐ray diffraction. The results indicated that the molecular structures of the complexes are five‐coordinated and show a distorted square‐pyramidal geometry, in which the central copper ions are coordinated to N,N atoms of pzta and N,O atoms of amino acids. The interactions of the complexes with DNA were investigated using electronic absorption, competitive fluorescence titration, circular dichroism and viscosity measurements. These studies confirmed that the complexes bind to DNA through a groove binding mode with certain affinities (Kb = 4.71 × 103 and 1.98 × 103 M?1 for 1 and 2 , respectively). The human serum albumin (HSA) binding properties of the complexes were also evaluated using fluorescence and synchronous fluorescence spectroscopies, indicating that the complexes could quench the intrinsic fluorescence of HSA in a static quenching process. The relevant thermodynamic parameters revealed the involvement of van der Waals forces and hydrogen bonds in the formation of complex–HSA systems. Finally, molecular docking technology was also used to further verify the interactions of the complexes with DNA/HSA.  相似文献

Mononuclear and dinuclear copper(II) complexes with thiophenecarboxylic acid, [Cu(3‐TCA)2(2,2′‐bpy)] ( 1 ), [Cu(3‐Me‐2‐TCA)2(H2O)(2,2′‐bpy)] ( 2 ), [Cu(5‐Me‐2‐TCA)2(H2O)(2,2′‐bpy)] ( 3 ) and [Cu2(2,5‐TDCA)(DMF)2(H2O)2(2,2′‐bpy)2](ClO4)2 ( 4 ) (where 3‐TCA = 3‐thiophenecarboxylic acid; 3‐Me‐2‐TCA = 3‐methyl‐2‐thiophenecarboxylic acid; 5‐Me‐2‐TCA = 5‐methyl‐2‐thiophenecarboxylic acid; 2,5‐TDCA = thiophene‐2,5‐dicarboxylic acid; 2,2′‐bpy = 2,2′‐bipyridyl; DMF = N,N‐dimethylformamide), were synthesized. Compounds 1 – 4 were extensively characterized using both analytical and spectroscopic methods. Additionally, the solid‐state structures of 1 and 4 were unambiguously established from single‐crystal X‐ray diffraction studies. The hexacoordinated Cu(II) centre in 1 (CuO4N2) is a distorted octahedral geometry whereas the pentacoodinated 4 (CuO3N2) has distorted square pyramidal geometry. Compounds 1 and 4 exhibit intermolecular hydrogen bonding which leads to the formation of two‐ and three‐dimensional supramolecular architectures, respectively. Spectrophotometric and computational investigations suggest that these compounds bind with DNA in minor groove binding such that Kb = 4.9 × 105 M?1 and Ksv = 3.4 × 105 M?1, and binding score of ?5.26 kcal mol?1. The binding affinity of these complexes to calf thymus DNA is in the order 2 > 3 > 4 > 1 . Methyl‐substituted thiophene ring increases the DNA binding affinity whereas unsubstituted thiophene ring DNA binding rate is reduced. The methyl group on the thiophene ring would sterically hinder π–π stacking of the ring with DNA base pairs, and subsequently they are involved in hydrophobic interaction with the DNA surface rather than partial intercalative interaction. Compounds 1 – 4 show pronounced activity against B16 mouse melanoma skin cancer cell lines as measured by MTT assay yielding IC50 values in the micromolar concentration range. The compounds could prove to be efficient anti‐cancer agents, since at a concentration as low as 2.1 μg ml?1 they exerted a significant cytotoxic effect in cancer cells whereas cell viability was not affected in normal cells.  相似文献

The cobalt(II) complexes [Co(Cl)2(met)(o‐phen)] ( 1 ), [Co(Cl)2(en)(met)] ( 2 ) and [Co(Cl)2(met)(opda)] ( 3 ) (met = metformin, o‐phen = ortho‐phenanthroline, en = ethylenediamine, opda = ortho‐phenylenediamine) were synthesized and characterized using liquid chromatography–mass spectrometry, elemental analysis, molar conductance measurements, thermal analysis, infrared spectroscopy, magnetic moment measurements, electronic spectroscopy and X‐ray diffraction. The metal centre was found to be in an octahedral geometry. UV–visible absorption, fluorescence and viscosity measurements were conducted to assess the interaction of the complexes with calf thymus DNA. The complexes showed absorption hyperchromism in UV–visible spectra with DNA. The binding constants from UV–visible absorption studies were 1.38 × 105, 2.1 × 105 and 3.1 × 105 M?1 for 1 , 2 and 3 , respectively, and Stern–Volmer quenching constants from fluorescence studies were 0.146, 0.176 and 0.475, respectively. Viscosity measurements revealed that the binding of the complexes with DNA could be surface binding, mainly due to groove binding. The activities of the complexes in DNA cleavage decrease in the order 3 > 2 > 1 . The complexes were docked into DNA topoisomerase II using Discovery Studio 2.1 software.  相似文献

Two new novel complexes, [Cu4(Endc)4(phen)4]⋅7(H2O)⋅2(O) and [Mn2(Endc)2(phen)2(H2O)2]⋅(H2O) (phen =1,10‐phenanthroline, H2Endc = endo ‐norbornene‐cis ‐5,6‐dicarboxylic acid), were synthesized and structurally characterized using IR and 1H NMR spectroscopies, elemental analysis and single‐crystal X‐ray diffractometry. Their reactivity with calf thymus DNA and HeLa cell DNA was measured using UV absorption and fluorescence spectroscopies. The results indicated that these complexes can bind to DNA with different binding affinity. Gel electrophoresis assay demonstrated the ability of the complexes to cleave pBR322 plasmid DNA. Apoptotic study showed that the complexes exhibit significant cancer cell inhibitory rates. Eventually, the complexes can suitably dock with a special DNA (PDB ID: 1AIO).  相似文献

Six novel organometallic half sandwich complexes [(η5‐C5Me5)M(L1–3)Cl]Cl.2H2O were synthesized using [{(η5‐C5Me5)M(μ‐Cl)Cl2], where M = Ir (III)/Rh (III) and L1–3 = three pyridyl pyrimidine based ligands; and characterized by NMR, Infra‐red spectroscopy, conductance, elemental and thermal analysis. The complex‐DNA binding mode and/or strength evaluated using absorption titration, electrochemical studies and hydrodynamic measurement proposed intercalative binding mode, which was also confirmed by molecular docking study. Differential pulse voltammetry and cyclic voltammetry studies indicated an alteration in oxidation and reduction potentials of complexes (M+4/M+3) in presence of CT‐DNA. The metal complexes can cleave plasmid DNA as proposed in gel electrophoretic analysis. The LC50 values of complexes evaluated on brine shrimp suggested their potent cytotoxic nature.  相似文献

The antitumor activity shown by many platinum complexes has produced a strong interest in research of new organometallic compounds. Among the metal compounds synthesized and tested, copper compounds have received considerable attention because of their cytotoxic activity against solid tumors. A novel wheel‐shaped octanuclear copper(II) complex with a 1,2,4‐triazole derivative ligand formulated as [Cu8L4](ClO4)8?11H2O ( 1 ) (L = 3,5‐bis((bis(2‐hydroxylethyl)amino)methyl)‐4 H‐1,2,4,‐triazole‐4‐amine) has been synthesized and structurally characterized. In 1 , eight Cu atoms are linked through 1,2,4‐triazole units and alkoxide bridges to form a centrosymmetric octanuclear Cu(II) metallomacrocycle. The interaction of complex 1 with calf thymus DNA has been studied using UV absorption, fluorescence and circular dichroism spectroscopies, viscosity measurements and cyclic voltammetry. The apparent binding constant (kapp) value for 1 is 1.59 × 105 M?1. Furthermore, complex 1 displays efficient oxidative cleavage of supercoiled DNA in the presence of external agents, the rate constant for the conversion of supercoiled to nicked DNA being 2.67 × 10?5 s?1. Interestingly, cytotoxicity studies on the MCF‐7 human breast cancer cell line show that the IC50 value of 1 is less than that of cisplatin for the same cell line, revealing that it has the potential to act as an effective metal‐based anticancer drug. Three novel compounds, (η6‐p‐cymene)RuCl2(2‐fluoro‐5‐aminopyridine) (compound 1), (η6‐p‐cymene)RuCl2(5‐amino‐2‐chlorpyridine) (compound 2) and (η6‐p‐cymene)RuCl2(2‐bromo‐ 5‐aminopyridine) (compound 3), were synthesized and characterized. The compound 1 and 3 were determined by X‐ray diffraction, showing a distorted piano‐stool type of geometry with similar bond lengths and angles around the ruthenium. Compound 2 exhibited moderate in vitro activity against A549 and MCF‐7 human cancer cells, the other two lower activities. The UV–vis and fluorescent absorption titrations showed that three compounds binded with CT‐DNA in a minor groove. The intrinsic binding constants (Kb) were calculated to be 2.13(±0.03) × 105 M?1, 2.89(±0.03) × 105 M?1 and 2.45(±0.03) × 105 M?1 for compound 1, 2 and 3, respectively, by using UV–vis absorption titrations data. Among the three compound, the highest value of intrinsic binding constant of compound 2 was consistent with its highest cytoxicity against A549 and MCF‐7 human cancer cells in vitro.  相似文献

Two novel chiral ruthenium(II) complexes, Δ‐[Ru(bpy)2(dmppd)]2+ and Λ‐[Ru(bpy)2(dmppd)]2+ (dmppd = 10,12‐dimethylpteridino[6,7‐f] [1,10]phenanthroline‐11,13(10H,12H)‐dione, bpy = 2,2′‐bipyridine), were synthesized and characterized by elemental analysis, 1H‐NMR and ES‐MS. The DNA‐binding behaviors of both complexes were studied by UV/VIS absorption titration, competitive binding experiments, viscosity measurements, thermal DNA denaturation, and circular‐dichroism spectra. The results indicate that both chiral complexes bind to calf‐thymus DNA in an intercalative mode, and the Δ enantiomer shows larger DNA affinity than the Λ enantiomer does. Theoretical‐calculation studies for the DNA‐binding behaviors of these complexes were carried out by the density‐functional‐theory method. The mechanism involved in the regulating and controlling of the DNA‐binding abilities of the complexes was further explored by the comparative studies of [Ru(bpy)2(dmppd)]2+ and of its parent complex [Ru(bpy)2(ppd)]2+ (ppd = pteridino[6,7‐f] [1,10]phenanthroline‐11,13 (10H,12H)‐dione).  相似文献

Two complexes [MnL2 (H2O)2]·2ClO4 (complex 1) and [CuL(H2O)3]·2NO3 (complex 2) (where L = 3,5‐bis(1‐imidazoly) pyridine) were designed and synthesized. The structures of the complexes were characterized by X‐ray crystallography, elemental analyses, and infrared spectrum. The interaction capacity of the complexes with calf thymus DNA has been investigated by UV and fluorescence spectroscopy. Gel electrophoresis assay demonstrated the ability of the complexes to cleave the pBR322 plasmid DNA. Efficient binding properties of DNA were established by UV–vis, fluorescence, and gel electrophoresis. The intrinsic binding constants (Kb) were calculated to be 0.1524, 0.1041 for complexes 1–2, respectively. The cytotoxic activity of the two complexes exhibited a higher cytotoxicity against HeLa cell lines and lower cytotoxicity toward the normal cell lines. Flow cytometry demonstrated the cancer cell inhibitory rate of two complexes. Furthermore, computer‐aided molecular docking studies were performed to visualize the binding mode of the drug candidate at the molecular level. Interestingly, complex 1 exhibited a significant cancer cell inhibitory rate than cisplatin and other complexes.  相似文献

A series of novel cytotoxic compounds, [Mn(cpt)2], [Zn(tpt)(H2O)2]?DMA?2(H2O) and [Cu(tpt)]?DMA (cpt = 4′‐(4‐carboxyphenyl)‐2,2′:6′,2″‐terpyridine, tpt = 4‐(2,4,6‐tricarboxylphenyl)‐2,2′:6′,2″‐terpyridine, DMA = (CH3)2NH), were isolated and characterized. The structures of these complexes were characterized using single‐crystal X‐ray diffraction. The mode and extent of binding between fish sperm DNA and the complexes were investigated using fluorescence spectroscopy and molecular docking. These results indicate the ability of the complexes to bind to DNA with different binding affinities. The binding of the Zn(II) complex with DNA is stronger than that of the corresponding Cu(II) analogue, which is expected due to the z* effect and geometry. The ability of these complexes to cleave pBR322 plasmid DNA was demonstrated using gel electrophoresis assay, showing that the complexes have effective DNA cleavage activity. In addition, the cytotoxic effects of these complexes were examined on HeLa cells (human cervix epithelia carcinoma cells) in vitro. The three complexes exhibit different cytotoxic effects and decent cancer cell inhibitory rate. This means that the structures and type of metal have a great influence on the activity of these novel complexes.  相似文献

The Schiff base N,N′‐bis(salicylidene)‐1,5‐diamino‐3‐oxapentane (H2L) and its lanthanide(III) complexes, PrL(NO3)(DMF)(H2O) ( 1 ) and Ho2L2(NO3)2 · 2H2O ( 2 ), were synthesized and characterized by physicochemical and spectroscopic methods. Single crystal X‐ray structure analysis revealed that complex 1 is a discrete mononuclear species. The PrIII ion is nine‐coordinate, forming a distorted capped square antiprismatic arrangement. Complex 2 is a centrosymmetric dinuclear neutral entity in which the HoIII ion is eight‐coordinate with distorted square antiprismatic arrangement. The DNA‐binding properties of H2L and its LnIII complexes were investigated by spectrophotometric methods and viscosity measurements. The results suggest that the ligand H2L and its LnIII complexes both connect to DNA in a groove binding mode; the complexes bind more strongly to DNA than the ligand. Moreover, the antioxidant activities of the LnIII complexes were in vitro determined by superoxide and hydroxyl radical scavenging methods, which indicate that complexes 1 and 2 have OH · and O2– · radical scavenging activity.  相似文献

Three water‐soluble complexes, [Cu2L2Cl2] ( 1 ), [CoL2(im)2] ( 2 ) and [ZnLClH2O] ( 3 ) (HL = 8‐hydroxyquinoline‐5‐sulphonic acid; im = N ‐methylimidazole), were prepared and characterized using various spectral techniques. The DNA binding behaviour of complexes 1 – 3 was studied using UV–visible and circular dichroism (CD) spectra and cyclic voltammetry. All three complexes exhibit hypochromism but complexes 1 and 3 alone give a red shift of 4 nm with a significant binding constant of K b = 2.1 × 104 and 1.0 × 104 M−1, respectively, but complex 2 shows no red shift with lower K b of 4.1 × 103 M−1. The voltammetric E 1/2 of complex 1 on interaction with herring sperm DNA shifts to a more positive potential, as expected, than complex 2 due to higher DNA affinity. Additionally, analysis of electrochemical data yields a value of K +/K 2+ greater than one suggesting that complex 1 binds to DNA through intercalation in the M(I) state. Evidently in CD spectral analysis, complex 1 exhibits a decrease in molar ellipticity with a red shift of 10 nm and a significant decrease in intensity compared to complexes 2 and 3 . This clearly indicates that complex 1 induces the B → A transition to a greater extent than 2 and 3 . Oxidative cleavage using circular plasmid pUC18 DNA with complex 1 was investigated using gel electrophoresis. Interestingly, complex 1 displays a strong DNA binding affinity and is efficient in cleaving DNA in the presence of H2O2 at pH = 8.0 at 37 °C.  相似文献

Two transition metal complexes, [Cu(FH)3]⋅2Cl⋅2H2O and [Ni(FH)3]⋅2Cl⋅2H2O, were synthesized from the reactions of furan‐2‐carboxylic acid hydrazide with CuCl2⋅2H2O and NiCl2⋅6H2O. The synthesized complexes were characterized using analytical and various spectral techniques. The structures of the complexes were determined using single‐crystal X‐ray diffraction. The interactions of the complexes with calf thymus DNA (CT‐DNA) were studied using absorption, fluorescence, cyclic voltammetric and viscosity measurements. The experimental results showed that the complexes could interact with CT‐DNA through intercalation. A gel electrophoresis assay demonstrated the ability of the complexes to cleave pBR322 DNA. The binding interaction of the complexes with bovine serum albumin was investigated using a fluorescence spectroscopic method. The radical scavenging ability, assessed using a series of antioxidant assays involving 2,2‐diphenyl‐2‐picrylhydrazyl radical, hydroxyl radical and nitric oxide radical, showed that the complexes possess significant radical scavenging properties. Further, the in vitro cytotoxic effect of the complexes examined on cancerous cell lines, such as human cervical cancer cells (HeLa) and human breast cancer cell line (MCF‐7), showed that the complexes exhibit significant anticancer activity.  相似文献

Hydrothermal reactions of tridentate rigid 2,4,6‐tris‐(benzimidazolyl‐2‐yl)pyridine (pytbzim) ligand and Zn(II)/Cd(II) salts generate binuclear complexes {[Cd2Cl2(pytbzim)2(H2O)2]·2NO3}n ( 1 ) and two isomorphs {[M2Cl2(pytbzim)2(H2O)2]Cl2·2H2O}n [M=Cd ( 2 ), Zn ( 3 )]. All complexes include [M2Cl2(pytbzim)2(H2O)2] dimers, which are further connected into a three‐dimensional supramolecular networks through ?‐? stacking interaction and hydrogen bonds. The solid state photoluminescent studies reveal good fluorescent properties of the pytbzim ligand and complexes 1 – 2 at room temperature.  相似文献

[MCl(H2L)(OH2)]·1.5H2O (M = Pd(II) ( 1 ) and Pt(II) ( 2 )) and [Ru(H2L)2(OH2)2]·3H2O ( 3 ) (H3L: N‐phenyl, N`‐(3‐triazolyl)thiourea) were synthesized, characterized and tested for their antibacterial activities against Staphylococcus aureus and Escherichia coli bacteria. The thiourea derivative is coordinated to Mn+ ions as a mono‐negatively N,S‐bidentate ligand via the enolization of C = S group and triazole N center. The density functional theory calculations reveal that presence of a water molecule in a trans position to triazole ring increased the stability of d8 metal ions complexes via the formation of strong Cl…NH intramolecular H‐bond. The cis‐Ru(II)‐isomer with two isoenergetically H2L? molecules are more stable than the trans‐analog. Coordination of H3L to Ru(II) ion did not alter the toxicity of the free ligand, while the interaction with the d8 metal ions gave rise to inactive compounds.  相似文献

The dinuclear Cu(II) complexes [Cu2(L1)2(mb)]?ClO4 ( 1 ) and [Cu2(L2)2(mb)]?ClO4 ( 2 ) (HL1 = 2‐[(2‐diethylaminoethylimino)methyl]phenol; HL2 = 2‐[1‐(2‐diethylaminoethylimino)propyl]phenol; mb = 4‐methylbenzoate) were synthesized and characterized using X‐ray crystal structure analysis and spectroscopic methods. Complexes 1 and 2 are dinuclear with distorted square pyramidal Cu (II) geometries, where Schiff base coordinates with tridentate (N,N,O) chelating mode and mb bridges two metal centres. Optimized structures and photophysical properties of ligands and complexes were calculated using density functional theory and time‐dependent density functional theory methods using B3LYP functional with 6‐31G (d,p) and LanL2MB basis sets. Interactions of the complexes with bovine serum albumin (BSA) and human serum albumin (HSA) were studied using UV–visible absorption and fluorescence spectroscopies and the calculated values of association constants (M?1) are 1.7 × 105 ( 1 –BSA), 5.7 × 105 ( 2 –BSA), 1.6 × 105 ( 1 –HSA) and 6.9 × 105 ( 2 –HSA). Interactions of the complexes with calf thymus DNA were also investigated and the binding affinities are 1.4 × 105 and 1.6 × 105 M?1 for 1 and 2 , respectively. Both complexes catalytically oxidize 3,5‐di‐tert‐butylcatechol to 3,5‐di‐tert‐butylbenzoquinone in the presence of molecular oxygen.  相似文献

The title complexes, hexaaquacobalt(II) bis(μ‐pyridine‐2,6‐dicarboxylato)bis[(pyridine‐2,6‐dicarboxylato)bismuthate(III)] dihydrate, [Co(H2O)6][Bi2(C7H4NO4)4]·2H2O, (I), and hexaaquanickel(II) bis(μ‐pyridine‐2,6‐dicarboxylato)bis[(pyridine‐2,6‐dicarboxylato)bismuthate(III)] dihydrate, [Ni(H2O)6][Bi2(C7H4NO4)4]·2H2O, (II), are isomorphous and crystallize in the triclinic space group P. The transition metal ions are located on the inversion centre and adopt slightly distorted MO6 (M = Co or Ni) octahedral geometries. Two [Bi(pydc)2] units (pydc is pyridine‐2,6‐dicarboxylate) are linked via bridging carboxylate groups into centrosymmetric [Bi2(pydc)4]2− dianions. The crystal packing reveals that the [M(H2O)6]2+ cations, [Bi2(pydc)4]2− anions and solvent water molecules form multiple hydrogen bonds to generate a supramolecular three‐dimensional network. The formation of secondary Bi...O bonds between adjacent [Bi2(pydc)4]2− dimers provides an additional supramolecular synthon that directs and facilitates the crystal packing of both (I) and (II).  相似文献

Three 1H‐benzimidazole‐5‐carboxylate (Hbic)‐based coordination polymers, {[Ni(H2O)(Hbic)2] · 2H2O}n ( 1 ), {[Ni(H2O)2(Hbic)2] · 3H2O}n ( 2 ), and {[Co2(H2O)4(Hbic)4] · 4DMF · 3H2O}n ( 3 ) were obtained by reactions of the ligand H2bic and NiII or CoII salts in the presence of different structure directing molecules. They were structurally characterized by single‐crystal X‐ray diffraction, IR spectra, elemental analysis, thermal stability, luminescent, and magnetic measurements. Structural analysis suggests that the three polymers exhibit a 2D (4, 4) layer for 1 and 1D linear double chains for both 2 and 3 due to the variable binding modes and the specific spatial orientation of the Hbic ligand towards the different paramagnetic metal ions, which were further aggregated into different 3D supramolecular architectures by popular hydrogen‐bonding interactions. Weak and comparable antiferromagnetic couplings mediating by Hbic bridge are observed between the neighboring spin carriers for 2 and 3 , respectively.
