[structure: see text] Two new diterpenes, lagaspholones A (1) and B (2), have been isolated from the methanolic extract of Euphorbia lagascae, along with the known compounds (+)-dehydrovomifoliol, scopoletin, dehydrodiconiferyl diacetate, 3-indolcarbaldehyde, and 4-hydroxy-3,5-dimethoxybenzaldehyde. Their structures were elucidated by spectroscopic methods. Compounds 1 and 2 contain the rare jatropholane-type skeleton, characterized by a 5:6:7:3 fused ring system. A possible biosynthetic pathway for lagaspholones is proposed. 相似文献

Jatrophalactone (1) and jatrophalone (2), together with jatrophadiketone (3), a 6/6/6 tricyclic diterpene transformed from rhamnofolane skeleton, have been isolated from the methanolic extract of Jatropha curcas. The structures were established on the basis of extensive spectroscopic methods and further confirmed by X-ray crystallographic analysis. The cytotoxicity against selected cell lines was evaluated. Among them, compound 1 displayed cytotoxicity against HL-60, SMMC-7721, A-549, MCF-7, and SW480 cell lines with IC50 values of 8.5, 20.6, 19.7, 20.1, and 19.2 μM, respectively. The possible biosynthetic pathways for 1 and 3 are proposed. 相似文献

Birhodomolleins A-C (1-3), three novel diterpenoids dimerized from two grayanane diterpenes through an oxygen bridge, were isolated from the flowers of Rhododendron molle. Their structures were elucidated by interpretation of their 1D and 2D NMR and other spectroscopic data. Birhodomollein A (1) contains a rare chloro-substitution on one of the grayanane moieties. Acetone:chloroform (1:2) extract of the aerial parts of Euphorbia connata Boiss. (Euphorbiaceae) was investigated for its diterpenoids. This led to the isolation of one known and two new diterpenes, belonging to the pentahydroxy-13(17)-epoxy-8,10(18)-myrsinadiene and tetrahydroxy-5,6-epoxy-14-oxo-jatropha-11(E)-ene classes. The structures were elucidated based on 13C and 1H NMR as well as 2D NMR, IR and MS spectra and the cytotoxicity for compounds 1–3 were evaluated by using MTT assay against two human breast cancer cell lines. Myrsinane-type compounds – 3,7,14,15-tetraacetyl-5-propanoyl-13(17)-epoxy-8,10(18)-myrsinadiene (1) and 3,7,10,14,15-pentaacetyl-5-butanoyl-13,17-epoxy-8-myrsinene (2) – exhibited moderate inhibitory effects, with IC50 values of 24.53 ± 3.39 and 26.67 ± 1.41 μM against the MDA-MB cell line, and 37.73 ± 3.41 and 34.57 ± 2.12 μM against the MCF-7 cell line, respectively. Jatrophane-type diterpene – 5,6-epoxy-8,9,15-triacetyl-3-benzoyl-14-oxo-jatropha-11(E)-ene (3) – showed weak cytotoxicity, with IC50 values of 55.67 ± 7.09 μM against MDA-MB, and moderate cytotoxicity with IC50 values of 24.33 ± 3.21 μM against MCF-7 cell line. 相似文献

The aerial parts of Euphorbia cornigera Boiss., on extraction with MeOH, yielded new bio-active constituents (1, 2) and known compounds (3 and 4) after MTT cytotoxicity assay-guided fractionation and chromatographic separation were conducted. From the aerial parts of E. cornigera Boiss., new bio-active constituents were extracted in methanol. The extract was partitioned in different organic solvents and the ethylacetate-soluble portion was subjected to Craig's distribution. The MTT cytotoxicity assay-guided chromatographic separation yielded four (1-4), out of which two (1, 2) were new and two known (3, 4) bio-active compounds, and they are reported for the first time from this source. Their structure and relative stereochemistry were established by analysing spectroscopic and mass measurement data. The isolates were named as: 13-O-[(2Z ,4 E ,6 Z)]-deca-2,4,6-trienoylingenol (1), 13-O-( 2 Z ,4 E ,6 Z)-deca-2,4,6-trienoyl-20-O-angeloylingenol (2), 13-O-dodecanoyl-20-O-hexanoylingenol (3) and 3-O-(2,3-dimethylbutanoyl)-13-O-dodecanoyl-20-O-hexadecanoylingenol (4). Literature revealed that compounds 1 and 2 are new metabolites, while 3 and 4 are known, and are reported for the first time from this source. Cytotoxicities of isolates were evaluated in terms of IC(50) against RAW and HT-29 cell lines through MTT assay using ambrucin hydrochloride as a control. Three new diterpenes, namely jasonin-a (1), jasonin-b (2), and jasonin-c (3) were isolated from the aerial parts of Jasonia montana (Asteraceae). Their structures were elucidated on the basis of spectral data as [(1E)-2-((2S)-1,2,5-trimethylbicyclo[3.2.l]octan-8-yl)vinyl] benzene-3-carboxylic acid (1), [3-((2S, 5S)-1,2, 5-trimethylcycloheptanyl)propyl]benzene-3-carboxylic acid (2), and [(1E)-3-((7R)-1,7-dimethy-4-methylenecycloheptanyl)prop-1-enyl] benzene-3-carboxylic acid (3). In addition, the previously reported 5,7,3'-trihydroxy-3,6,4'-trimethoxy flavone designated as centaureidin (4), was also isolated and characterized from this source. The different extracts of the plant were also screened for hypoglycemic, antidiabetic, and antimicrobial activities, wherein the petroleum ether and ethanolic extracts exhibited hypoglycemic and antidiabetic activity, and the petroleum ether and chloroform extracts showed antimicrobial activity. 相似文献

Fractionation of a cytotoxic extract obtained from a southern Australian marine sponge, Phorbas sp., yielded the known diterpenes phorbasins B-F () together with five new members of the phorbasin family, phorbasins G-K (). Structures were assigned to the new phorbasins based on detailed spectroscopic analysis. Four fractions were prepared from the crude extract of Caesalpinia minax Hance and the inhibitory activity of nitric oxide (NO) production release of RAW 264.7 cells stimulated by lipopolysaccharide (LPS) was evaluated. The ethyl acetate (EtOAc) fraction showed obvious inhibitory effect. Bioassay-guided fractionation led to the isolation of three new cassane diterpenes, caesalmin X (1), caesalmin Y (2) and caesalmin Z (3), together with 19 known cassane diterpenoids (4–22). Their structures were mainly characterised on the basis of extensive spectroscopic analyses and comparison with reported data. Moreover, three compounds (20–22) which possessed furanditerpenoid 7,17-lactone structures, displayed moderate activities, with IC50 value of 29.85, 27.38 and 25.40 μM, respectively. 相似文献

A new tricyclic diterpenoid, named pre-segetanin (1), and two new diterpenes, named segetanin A and B (2 and 3), the latter based on the rare segetane skeleton, have been identified from the whole plant of sea spurge along with four known segetane diterpenes (4-7). Among them, pre-segetanin (1) has an unprecedented carbon skeleton, whose isolation provides a first insight into the biosynthesis of diterpenoids with a segetane skeleton. Six new tigliane diterpenoids, euphkanoids A ? F (1–6), along with 12 known diterpenoids (7–18) were isolated from the roots of Euphorbia kansuensis, a traditional Chinese medicinal plant. Their structures were elucidated by spectroscopic analysis, ECD calculation and chemical methods. Compounds 1–3 represent the unusual examples of tigliane diterpenoids with a conjugated olefinic aldehyde moiety. 1–18 and two hydrolyzed analogues (1c and 4a) were screened for the inhibitory effects on nitric oxide (NO) production induced by lipopolysaccharide (LPS) in RAW264.7 cells. The results showed that 1–3 and 6 markedly inhibited NO production with IC50 values in the range of 4.8–11.3 μM, which were more active than the positive control quercetin (IC50 = 12.3 μM). 相似文献

Chemical investigation of an Okinawan marine sponge resulted in the isolation of two new spongian-class diterpenes 1 and 2 together with two known compounds, chromodorolide B (3) and chromodorolide C (4). Compound 1, named chromodorolide D, is an example of a diterpenoid with a highly rearranged chromodorane carbon skeleton, while compound 2 retains the open side chains. The structures of the new compounds were elucidated on the basis of extensive spectroscopic analysis and chemical conversion. Five new brominated diterpenes, along with two known, have been isolated from the organic extract of the red alga Laurencia obtusa, collected from the coastal rocks of Preveza in the Ionean Sea, Greece. The novel metabolites prevezols B-E possess two new carbon skeletons, to the best of our knowledge, unprecedented in the literature. The structures and the relative stereochemistry of the new natural products were established by means of spectral data analyses. Three new abietane diterpenes, sugikurojins D (1), E (2), and F (3), and two new abietanes which incorporate cadinane, sugikurojins G (4) and H (5) were isolated from the bark of Cryptomeria japonica. These structures were elucidated primarily by extensive NMR experiments. The structure of sugikurojin D (1) was deduced to be 6alpha-acetoxy-7beta,11-dihydroxy-12-methoxy-8,11,13-abietatriene. Sugikurojin E (2) was deduced to be 6alpha-acetoxy-7beta,12-dihydroxy-8,11,13-abietatriene. Sugikurojin F (3) was 7alpha-methoxy-8,13-abietadien-11,12-dione. Sugikurojins G (4) and H (5) had a unique skeleton incorporating an alpha-cadinol or a 1alpha-hydroxy-T-cadinol in ferruginol, respectively. Also obtained in this investigation were the known diterpenes (6-14). An antibacterial activity of ten among these against Staphylococcus aureus and Escherichia coli was inactive at the (MIC: 125 microg/ml) level. Meanwhile, in the cytotoxic activity against HL-60, compounds 4, 8, and 11 showed moderate (IC50: 4, 35.4; 8, 28.0; 11, 52.4 microM) though weak (IC50: 4, 100; 8, 80.8; 11, 100 microM) activity against HCT-15. 相似文献

Chromatographic fractionation of a hexane extract from stem-bark of Premna latifolia yielded three new icetexane diterpenes (1-3). Their structures were elucidated on the basis of spectroscopic data, mainly NMR and MS, data. Compounds 1-3 were also evaluated for the cytotoxicity against the cancer cell lines (HT-29, A-431, MCF-7, Hep-G2, PC-3, A-549, B-16 F10 and ACHN). Compounds 1 and 2 displayed potent cytotoxic activity against HT-29 and Hep-G2 cell lines with IC50 values of 0.04 and 0.18 μg/mL, respectively. 相似文献

Two new and one known cyclomyrsinane-type diterpenes (1–3) were isolated from the methanol extract of the whole, undried plant of Euphorbia falcata, native to Hungary. The structures and relative configuration of the compounds were established on the basis of extensive spectroscopic analysis, including HRESIMS and one- and two-dimensional NMR techniques. These compounds are hexa- and heptaesters of diterpene polyols, esterified with acetic, propanoic, isobutanoic, 2-methylbutanoic, and benzoic acids. This type of diterpenes was previously detected only in three Euphorbia species (Euphorbia teheranica, Euphorbia seguieriana, and Euphorbia prolifera). The compounds, together with premyrsinanes isolated previously from this plant were evaluated for their antiproliferative activity against HeLa, Ishikawa, and MCF7 mouse lymphoma cells, and for their capacity to modulate resistance to doxorubicin in L5178 mouse lymphoma cells that over-express the MDR1 efflux pump. Compounds did not show substantial antiproliferative activity against any of the tested cell lines. However all compounds act in a mild to a very strong synergism with doxorubicin against the MDR mouse lymphoma cell line. 相似文献
