Human skin is repeatedly exposed to UVR that influences the function and survival of many cell types and is regarded as the main causative factor in the induction of skin cancer. It has been traditionally believed that skin pigmentation is the most important photoprotective factor, as melanin, besides functioning as a broadband UV absorbent, has antioxidant and radical scavenging properties. Besides, many epidemiological studies have shown a lower incidence for skin cancer in individuals with darker skin compared to those with fair skin. Skin pigmentation is of great cultural and cosmetic importance, yet the role of melanin in photoprotection is still controversial. This article outlines the major acute and chronic effects of UVR on human skin, the properties of melanin, the regulation of pigmentation and its effect on skin cancer prevention.  相似文献

Melanin pigments are important regulators for the evolution of essential functions of human skin. The concentration of melanin, as well as its depth distribution, is strongly affected by ultraviolet radiation. In un-tanned skin, melanin pigments are found only in the basal layer of the epidermis, while in tanned skin it is distributed throughout the epidermis. So far, mainly the amount of melanin, and not its distribution, has been considered in view of skin photobiology. With an advanced radiative transfer model we investigate, for the first time, how the depth distribution of melanin influences the amount of ultraviolet radiation that reaches living cells in the epidermis, and thus can damage the DNA in the cells. The simulations are performed for average pigmented skins (type III-IV). A surprisingly large factor, up to 12, is found between the ultraviolet protection of skin with melanin distributed throughout the epidermis, and skin with melanin only in the basal layer of the epidermis. We also show that the synthesis of previtamin D3, in skin, can vary with more than 100% if the depth distribution of melanin is changed, while the degradation of folate in dermal blood is almost un-affected by variations in the melanin depth distribution.  相似文献

Throughout the body, melanin is a homogenous biological polymer containing a population of intrinsic, semiquinone-like radicals. Additional extrinsic free radicals are reversibly photo-generated by UV and visible light. Melanin photochemistry, particularly the formation and decay of extrinsic radicals, has been the subject of numerous electron spin resonance (ESR) spectroscopy studies. Several melanin monomers exist, and the predominant monomer in a melanin polymer depends on its location within an organism. In skin and hair, melanin differs in content of eumelanin or pheomelanin. Its bioradical character and its susceptibility to UV irradiation makes melanin an excellent indicator for UV-related processes in both skin and hair. The existence of melanin in skin is strongly correlated with the prevention against free radicals/ROS generated by UV radiation. Especially in the skin melanin (mainly eumelanin) ensures the only natural UV protection by eliminating the generated free radicals/ROS. Melanin in hair can be used as a free radical detector for evaluating the efficacy of hair care products. The aim of this study was to investigate the suitability of melanin as protector of skin against UV generated free radicals and as free radical indicator in hair.  相似文献

From mutants of the fungusVerticillium dahliae Kleb. having different genetic blocks in the biosynthesis of melanin we have isolated and identified scytalone, flaviolin, 4-hydroxyscytalone, and 2-hydroxyjuglone and have detected 3,4,8-trihydroxytetralone and 4,8-dihydroxytetralone, which, in combination with the results of complementation analysis, have confirmed the scheme of melaninogenesis inV. dahliae put forward previously. Summary The resistance of melanin to acid attack is used in a new gravimetric procedure for the determination of melanin in animal tissues. Lipids are removed by solvent extraction after acid hydrolysis of the samples.
Zusammenfassung Eine neue gravimetrische Methode zur Bestimmung von Melanin in tierischen Geweben wurde entwickelt, die auf dessen Widerstandsfähigkeit in saurem Milieu beruht. Die vorgeschlagene Methode ist einfach; ihre Resultate werden in absoluten Werten ohne Kalibrierung gewonnen. Sie läßt sich für verschiedene Pigmentgewebe anwenden. Das beschriebene Verfahren scheint vorteilhafter als visuelle Schätzungsmethoden und solche Verfahren zu sein, die eine Melaninauflösung vorsehen. Die Melaninkonzentration in einigen Pigmentgeweben wurde angegeben.

Abstract— A method for estimating the amount of pigment in normal human skin in uivo is presented. This method is based on remittance spectroscopy. The spectrum of normal skin is compared to amelanotic skin and the logarithm of the ratio is fitted with a straight line in the range 620-720 nm. The parameters obtained are strongly correlated for all the volunteers in this study. therefore each spectrum determines one parameter for each individual. When similar analysis was performed on DOPA-melanin we obtained the same strong correlation among different concentrations. We are thus able to determine a relation between the coefficient obtained from the remittance spectrum from normal skin and an equivalent concentration of DOPA-melanin in aqueous solution. We can thus estimate. to a first approximation, the total melanin mass in human skin non-invasively, and can determine a parameter that is uniquely correlated to the amount of melanin pigment in the skin.  相似文献

Melanins perform their biological activity (photoprotection and light enhanced chemical reactivity) under the form of porous aggregates on which ions and neutral molecules can be adsorbed. For this reason, the photochemistry of natural and synthetic melanins must be investigated in the framework of the physico-chemical theory of the heterogeneous reactions and a detailed knowledge of the surface properties, is therefore, necessary. In this work, some surface characteristics of melanin particles have been investigated taking advantage of the photophysical behaviour of pyrene, a dye widely used in studies of the interface properties of micelles and colloidal semiconductors. Our fluorescence study has allowed to obtain valuable informations regarding the micro-environmental polarity of the melanin surface (that influences the vibronic structure of the emission spectra), the excimer formation, the lifetimes of the emissions and the kinetics of quenching by Cu2+.  相似文献

The mammalian eye consists of several layers of pigmented tissues that contain melanin. The eye is a unique organ for pigment cell research because one can isolate and compare melanosomes from different tissues and embryonic origins. Retinal, iris and ciliary pigment epithelial cells are derived from the neural ectoderm, more specifically from the extremity of the embryonic optical cup, which is also the origin of the retina. In contrast, the pigment-generating cells in the choroid and in the stroma of the iris and ciliary body, uveal melanocytes, are developed from the neural crest, the same origin as the melanocytes in skin and hair. This review examines the potential functions of ocular melanin in the human eye. Following a discussion of the role of melanins in the pigment epithelium and uveal melanocytes, three specific topics are explored in detail-photo-screening protective effects, biophysical and biochemical protective effects, and the biologic and photobiologic effects of the two main classes of melanins (generally found as mixtures in ocular melanosomes)--eumelanin and pheomelanin.  相似文献

A new approach is proposed to model a collective ion channel dynamics. We have assumed that ion channels create a two-component spatio-temporal interaction field. Every channel at its current spatial location in membrane contributes permanently to this field with its state (open or closed) and coupling strength to other channels. This field is described by a reaction-diffusion equation, the transition of ion channel from closed to open state (and vice versa) is described by a master equation, and migration of channels in membrane is described by a set of Langevin equations coupled by the interaction field. Within this model, we have investigated critical conditions for spatial distribution of ion channel activity.  相似文献

Antioxidant properties of melanin in retinal pigment epithelial cells   总被引:7,自引:0,他引:7  
The retinal pigment epithelium (RPE) is a monolayer of highly pigmented cells lining the inner aspect of Bruch's membrane. This pigmentation is due to eumelanin and a possible antioxidant role of melanin is reported here. The photo-oxidation of A2E, a constituent of RPE lipofuscin, leads to the sequential addition of up to nine oxygen atoms and/or the addition or loss of two hydrogen atoms. These photo-oxidations were investigated in the presence and absence of either calf or human RPE melanin in A2E-laden RPE cells. It was found that calf melanin was protective against the photo-oxidation of A2E, with an inhibition of oxidation of up to 50% in the case of the addition of two oxygen atoms. Calf melanin was also protective against blue light-induced damage to RPE cells. In addition this ability appears to decrease in humans as they grow older. With aging, a melanin-lipofuscin complex called melanolipofuscin forms. It is suggested that the oxidation or photo-oxidation of A2E in vivo may contribute to the age-related deterioration of the anti-oxidant role of RPE melanin and lead to various retinal disorders, such as age-related macular degeneration.  相似文献

An electrochemical method for self-assembling melanin films on the Au(111) surface from melanin aggregates in alkaline media is reported. Electrochemical data combined with scanning tunneling microscopy (STM), atomic force microscopy, and Auger electron spectoscopy show that the amount and structure of the deposited melanin film depend on the potential (E) applied to the electrochemical interface and deposition time. Film formation takes place at a noticeable rate at E = -1.0 V (vs SCE). High-resolution STM images at the early stages of growth show small particles, 5-8 nm in size and 0.3-0.4 nm in height, forming ordered arrays that follow closely the Au(111) topography. The size of the melanin particles increases as the film thickness increases, reaching 150 nm for deposits grown for 16 h. The deposited films are electrochemically active, showing well-defined redox couples preceding the hydrogen evolution reaction.  相似文献

A comparison between the effects of aluminum and cupric ions on the dopachrome (DC) conversion and the cooperation effect of the both ions in the DOPA oxidation to melanin pathway has been studied by UV-Vis spectrophotometric method. Both aluminum and cupric ions catalyze the DC conversion reaction, which is an important step in the melanin synthesis pathway. However, cupric ions catalyze the conversion of DC to yield 5,6-dihydroxyindole-2-carboxylic acid (DHICA) but the product of DC conversion catalyzed by aluminum is 5,6-dihydroxyindole (DHI). DOPA oxidation catalyzed by aluminum and cupric ions is studied in the presence of hydrogen peroxide. The results from our experiments provide evidence that aluminum can markedly increase the oxidative stress of copper-mediated the melanin formation and influence the properties of the melanin by means of changing the ratio of DHICA/DHI in the acidic environment (pH 5.5).  相似文献

This study reports depigmenting potency of selenium-containing carbohydrates, which would be based upon the finding of direct inhibition to mushroom tyrosinase. Two selenoglycosiede, SG-3 (bis(2,3,4-tri-O-acetyl-beta-D-arabinopyranosyl) selenide) and SG-8 (4'-methylbenzoyl 2,3,4,6-tetra-O-acetyl-D-selenomanopyranoside) among eleven selenium-containing compounds examined, were discovered to be effective depigmenting compounds on a mushroom tyrosinase inhibitory assay. SG-3 exhibited a competitive inhibition effect that was similar to kojic acid, well-known tyrosinase inhibitor. At 100 microM and 150 microM, SG-8 had an uncompetitive inhibitory effect that was higher than kojic acid. A study of a melan-a cell originated-tyrosinase inhibition assay showed that SG-8 had a lower inhibitory effect than kojic acid. SG-3 showed a similar inhibition effect to kojic acid on the melan-a cell-originated tyrosinase inhibitory assay. SG-8 showed dose-dependently cytotoxicity in a study of inhibition melanin synthesis by melan-a cells. Most melan-a cells did not survive after being treated with 20 microM of SG-8. At 10 microM, SG-3 inhibited melanin synthesis in the melan-a cells, and the effect was similar to phenylthiourea, which is a well-known inhibitor of melanin synthesis. Therefore, SG-3 is a new candidate for depigmenting reagents.  相似文献

In this work the calculated nuclear quadrupole coupling constants (NQCC; chi) of 17O in artemisinin and some of its derivatives and the effects of charge density due to the nature of ligands on NQCC of 17O were investigated. All calculations were performed at the HF/3-21G level using the Gaussian 98 program. The results show that the O-O linkage has a characteristic role in the antimalarial activity of artemisinin. In addition, various substitutions on C4 change the charge density on these oxygens and consequently change the pharmaceutical effect of artemisinin. Our results suggest that due to a larger charge density on O1, the heme iron approaches the endoperoxide moiety at the O1 position with preference to the O2 position.  相似文献

The oxidation of tyrosine by monophenol monooxygenase (tyrosinase: EC 1.10.3.1) to melanin has been studied by a combination of ultraviolet, circular dichroism, and nuclear magnetic resonance techniques. Abstract Induction and fate of ultraviolet radiation-induced pyrimidine dimers in DNA have been measured in the epidermis of the marsupial, Monodelphis domestica, using damage-specific endonucleases from Micrococcus luteus. Approximately 90% of the dimers are lost when irradiated animals are subjected to photoreactivating light for 180 min. No loss of dimers was detected when the animals were held for a similar period of time in the dark. The capacity of these epithelial cells to photorepair pyrimidine dimers may provide a useful whole animal system in which to determine the role of pyrimidine dimers in photobiological responses of the skin.  相似文献
