Self‐assembled thermo‐ and pH‐responsive poly(acrylic acid)‐b‐poly(N‐isopropylacrylamide) (PAA‐b‐PNIPAM) micelles for entrapment and release of doxorubicin (DOX) was described. Block copolymer PAA‐b‐PNIPAM associated into core‐shell micelles in aqueous solution with collapsed PNIPAM block or protonated PAA block as the core on changing temperature or pH. Complexation of DOX with PAA‐b‐PNIPAM triggered by the electrostatic interaction and release of DOX from the complexes due to the changing of pH or temperature were studied. Complex micelles incorporated with DOX exhibited pH‐responsive and thermoresponsive drug release profile. The release of DOX from micelles was suppressed at pH 7.2 and accelerated at pH 4.0 due to the protonation of carboxyl groups. Furthermore, the cumulative release of DOX from complex micelles was enhanced around LCST ascribed to the structure deformation of the micelles. © 2008 Wiley Periodicals, Inc. Layer‐by‐layer (LbL) assembly was conducted on CaCO3 microparticles pre‐doped with polystyrene‐block‐poly(acrylic acid) (PS‐b‐PAA) micelles, and resulted in micelles encapsulation in the microcapsules after core removal. Distribution of the micelles in the templates and capsules was characterized by transmission electron microscopy and confocal laser scanning microscopy. The micelles inside the capsules connected with each other to form a chain and network‐like structure with a higher density near the capsule walls. The hydrophobic PS cores were then able to load small uncharged hydrophobic drugs while the negatively charged PAA corona could induce spontaneous deposition of water‐soluble positively charged drugs such as doxorubicin.

In acidic solution, complex micelles were formed by diblock copolymers of poly (ethylene glycol)-b-poly (ε-caprolactone) (PEG-b-PCL) and folate-poly (2-(dimethylamino) ethyl methylacrylate)-b-poly (ε-caprolactone) (Fol-PDMAEMA-b-PCL) with a PCL core, a mixed PEG/Fol-PDMAEMA shell. The surface charge of the complex micelles was positive at acidic surroundings for the protonated PDMAEMA. With increasing pH value to 7.4 (above pK a of PDMAEMA), these micelles could convert into a core-shell-corona (CSC) structure composing a hydrophobic PCL core, a collapsed PDMAEMA shell, and a soluble PEG corona. Compared to core-shell micelles formed by PEG-b-PCL, micelles with CSC structure can prolong degradation by enzyme. Doxorubicin was physically loaded into the PCL core. The drug release rate was pH-dependent. At pH 5.5, complex micelles with core-shell structure showed faster drug release rate, while at pH 7.4, complex micelles gained CSC structure which control the drug release at a lower rate. The multifunctional complex micelles were prepared for enhanced tumor therapy.  相似文献

A series of amphiphilic triblock copolymers, poly[oligo(ethylene glycol) methacrylate]xblock‐poly(ε‐caprolactone)‐block‐poly[oligo(ethylene glycol) methacrylate]x, POEGMACo(x), were synthesized. Formation of hydrophobic domains as cores of the micelles was studied by fluorescence spectroscopy. The critical micelle concentrations in aqueous solution were found to be in the range of circa 10?6 M. A novel methodology by modulated temperature differential scanning calorimetry was developed to determine critical micelle temperature. A significant concentration dependence of cmt was found. Dynamic light scattering measurements showed a bidispersed size distribution. The micelles showed reversible dispersion/aggregation in response to temperature cycles with lower critical solution temperature between 75 and 85 °C. The interplay of the two hydrophobic and one thermoresponsive macromolecular chains offers the chance to more complex morphologies. © 2010 Wiley Periodicals, Inc. Triblock copolymers (MPEG‐b‐PCEMA‐b‐PHQHEMA) bearing cinnamoyl and 8‐hydroxyquinoline side groups with different block length are synthesized by a two‐step reversible addition fragmentation chain transfer polymerization of cinnamoyl ethyl methacrylate (CEMA) and 2‐((8‐hydroxyquinolin‐5‐yl)methoxy)ethyl methacrylate (HQHEMA), respectively. The self‐assembly of MPEG‐b‐PCEMA‐b‐PHQHEMA in mixture of THF and ethanol is investigated by varying the ratio of THF and ethanol. Spheric micelles with diameter of 63.7 nm and polydispersity of 0.128 are obtained for MPEG113b‐PCEMA15b‐PHQHEMA17 in THF/ethanol with a volume ratio (v/v) of 5/5. The PCEMA inner shell of the resulted micelles is photo‐crosslinked under UV radiation to give stabilized micelles. The complex reaction of the stabilized micelles with Zn(II) is investigated under different conditions to give zinc(II)‐bis(8‐hydroxyquinoline)(Znq2)‐containing micelles. When the complex reaction is carried out in THF/ethanol (v/v = 5/5) or THF/toluene (v/v = 6/4) with zinc acetate, fluorescent Znq2‐containing micelles are obtained without obvious change in diameters and morphologies. A dissipative particle-dynamics method was used to simulate the self-aggregation behavior of 15 alkylimidazoline surfactants of different structures. The effects of concentration and structure of hydrophilic and lipophilic groups of the alkylimidazolines on the configuration and aggregation number (Nagg) of the micelles were also explored. Results show that the concentration of the alkylimidazoline has a significant influence on the configuration of the micelles generated. More specifically, alkylimidazolines of different concentrations are found to generate different structures (spherical, rod-shaped, layered, and interlaced) in aqueous solution. At low (1–10?wt%) and medium (40?wt%) concentrations, the micelles generated in aqueous solution are spherical and rod-shaped, respectively. At higher concentrations (80?wt%), the micelles generated present interlaced shapes with rod-shaped and layered micelles when the length of the lipophilic chain is greater than C13 or the hydrophilic group is Hc. Nagg is strongly dependent on alkylimidazoline concentration and increases as concentration increases. Nagg is also greatly dependent on the structure of the hydrophilic and lipophilic groups present: it increases as the chain length of the lipophilic group increases but decreases as that of the hydrophilic group increases.  相似文献

The conformation of various basic poly (-amino acid)s was investigated by CD measurements in aqueous solutions containing bis (2-ethylhexyl)sodium sulfosuccinate (AOT) as well as in the AOT reversed micelles. The addition of AOT into an aqueous solution of poly(L-lysine) induces the conformational transition from coil to ordered structure, followed by aggregation. On the other hand, poly(L-lysine) assumes-structure in the reversed micelles at low wovalue (wo=[H2O]/[AOT]). Similarly to poly(L-lysine), poly(L-ornithine) takes an ordered structure in the aqueous solution containing AOT and-structure in the reversed micelles. In this case, however, these ordered structures are not so stable, compared with that of poly(L-lysine). Poly(L-arginine) undergoes the conformational transition from coil to helix by addition of AOT into the aqueous solution. Further addition of AOT allows transformation into-structure. Copoly(L-lysyl-L-leucine) with 63% leucine residue was shown to take a stable helical conformation even in pure water. In the reversed micelles, however, this ordered structure is significantly changed probably because the hydrophobic interaction among the leucyl residues is lowered in the reversed micelles.  相似文献

A new atom transfer radical polymerization (ATRP) initiator, namely, 2‐(1‐(2‐azidoethoxy)ethoxy)ethyl 2‐bromo‐2‐methylpropanoate containing both “cleavable” acetal linkage and “clickable” azido group was synthesized. Well‐defined azido‐terminated poly(N‐isopropylacrylamide)s (PNIPAAm‐N3)s with molecular weights and dispersity in the range 11,000–19,000 g mol?1 and 1.20–1.28, respectively, were synthesized employing the initiator by ATRP. Acetal containing PCL‐b‐PNIPAAm block copolymer was obtained by alkyne–azide click reaction of azido‐terminated PNIPAAm‐N3 with propargyl‐terminated PCL. Critical aggregation concentration (CAC) of PCL‐b‐PNIPAAm copolymer in aqueous solution was found to be 8.99 × 10?6 M. Lower critical solution temperature (LCST) of PCL‐b‐PNIPAAm copolymer was found to be 32 °C which was lower than that of the precursor PNIPAAm‐N3 (36.4 °C). The effect of dual stimuli viz . temperature and pH on size and morphology of the assemblies of PCL‐b‐PNIPAAm block copolymer revealed that the copolymer below LCST assembled in spherical micelles which subsequently transformed to unstable vesicles above the LCST. Heating these assemblies above 40 °C led to the precipitation of PCL‐b‐PNIPAAm block copolymer. Whereas, at decreased pH, micelles of PCL‐b‐PNIPAAm copolymer disintegrate due to the cleavage of acetal linkage and precipitation of hydrophobic hydroxyl‐terminated PCL. The encapsulated pyrene release kinetics from the micelles of synthesized PCL‐b‐PNIPAAm copolymer was found to be faster at higher temperature and at lower pH. © 2016 Wiley Periodicals, Inc. J. Micelles prepared from amphiphilic block copolymers in which a poly(styrene) segment is connected to a poly(ethylene oxide) block via a bis‐(2,2′:6′,2″‐terpyridine‐ruthenium) complex have been intensely studied. In most cases, the micelle populations were found to be strongly heterogeneous in size because of massive micelle/micelle aggregation. In the study reported in this article we tried to improve the homogeneity of the micelle population. The variant preparation procedure developed, which is described here, was used to prepare two “protomer”‐type micelles: PS20‐[Ru]‐PEO70 and PS20‐[Ru]‐PEO375. The dropwise addition of water to a solution of the compounds in dimethylformamide was replaced by the controlled addition of water by a syringe pump. The resulting micelles were characterized by sedimentation velocity and sedimentation equilibrium analyses in an analytical ultracentrifuge and by transmission electron microscopy of negatively stained samples. Sedimentation analysis showed virtually unimodal size distributions, in contrast to the findings on micelles prepared previously. PS20‐[Ru]‐PEO70 micelles were found to have an average molar mass of 318,000 g/mol (corresponding to 53 protomers per micelle, which is distinctly less than after micelle preparation by the standard method) and an average hydrodynamic diameter (dh) of 18 nm. For PS20‐[Ru]‐PEO375 micelles, the corresponding values were M = 603,000 g/mol (31 protomers per micelle) and dh = 34 nm. The latter particles were found to be identical to the “equilibrium” micelles prepared in pure water. Both micelle types had a very narrow molar mass distribution but a much broader distribution of s values and thus of hydrodynamic diameters. This indicates a conformational heterogeneity that is stable on the time scale of sedimentation velocity analysis. The findings from electron microscopy were in disagreement with those from the sedimentation analysis both in average micelle diameter and in the width of the distributions, apparently because of imperfections in the staining procedure. Both laser light scattering (LLS) and small-angle X-ray scattering (SAXS) were used to study the water-induced formation and structure of micelles and supramolecules of Pluronic P103 [(EO)17(PO)60(EO)17] in o-xylene, a selective solvent for the long middle block. In pure o-xylene, P103 molecules exist as unimer coils with an equivalent hard-sphere radius of 1.6 nm even at fairly high concentrations. Micelles with a PEO/water core and a PPO dominated corona were formed in the presence of water when the P103 concentration ≥0.046 g/mL. The size and structure of micelles have been studied as a function of solubilized water content Z (the molar ratio of water to EO units) in micelles. The micelles change from a somewhat open structure with some EO units either dangling out of the micellar core or being incorporated into neighboring micellar cores at low Z values to a flower-like structure with relatively sharp interface at high Z values. At low Z values (< about 2.9), micelles tend to have a structure with part of the poorly solvated PEO blocks present in the corona. With more water added to the core, the PEO blocks in the corona gradually entered into the core, and the PPO blocks backfolded to form loops. With increasing Z, the micellar core radius, Rc, and the hard-sphere volume fraction, ϕ, of micelles increased; the aggregation number, N, kept nearly a constant; but the hydrodynamic radius, 〈Rh0, and the corona thickness, Rs, decreased. At high Z values (> about 2.9), micelles have a flower-like structure with the two end PEO blocks belonging to the small micellar core. With increasing Z, the values of Rc, ϕ, and N increased, while Rs kept nearly a constant. In the concentrated regime (C > 0.30 g/mL), a stiff polymer network at a critical ϕ value of 0.49 was formed. The supramolecular structures with a face-center cubic packing, and a possible hexagonal packing at higher polymer concentrations (i.e. > 0.55 g/mL), were observed, respectively. © 1998 John Wiley & Sons, Inc. The effect of cationic micelles of cetyltrimethylammonium bromide (CTAB) on the kinetics of interaction of copper dipeptide complex [Cu(II)‐Gly‐Gly]+ with ninhydrin has been studied spectrophotometrically at 70°C and pH 5.0. The reaction follows first‐ and fractional‐order kinetics, respectively, in complex and ninhydrin. The reaction is catalyzed by CTAB micelles, and the maximum rate enhancement is about twofold. The results obtained in the micellar medium are treated quantitatively in terms of the kinetic pseudophase and Piszkiewicz models. The rate constants (kobs or kΨ), micellar‐binding constants (kS for [Cu(II)‐Gly‐Gly]+, kN for ninhydrin), and index of cooperativity (n) have been evaluated. A mechanism is proposed in accordance with the experimental results. The influence of different inorganic (NaCl, NaBr, Na2SO4) and organic (NaBenz, NaSal) salts on the reaction rate has also been seen, and it is found that tightly bound/incorporated counterions are the most effective. © 2007 Wiley Periodicals, Inc. Supramolecular capsules were assembled by neutral halogen bonding (XB) and studied in the solid state, in solution, and in the gas phase. The geometry of the highly organized capsules is shown by an X‐ray crystal structure which features the assembly of two XB hemispheres, geometrically rigidified by H‐bonding to eight MeOH molecules and encapsulation of two benzene guests. To enhance capsular association strength, tuning the XB donor is more efficient than tuning the XB acceptor, due to desolvation penalties in protic solvents, as shown for a tetraquinuclidine XB acceptor hemisphere. With a tetra(iodoethynyl) XB donor and a tetralutidine XB acceptor, the association in deuterated benzene/acetone/methanol 70:30:1 at 283 K reaches K a=(2.11±0.39)×105 m −1G =−6.9±0.1 kcal mol−1). The stability of the XB capsules in the gas phase was confirmed by electrospray ionization mass spectrometry (ESI‐MS). A new guest binding site was uncovered within the elongated iodoethynyl capsule.  相似文献

In this article, a mononuclear Mn(Niten)2H2O complex was prepared to catalyze hydrolysis of p-nitrophenyl acetate (PNPA) in presence of Cetyltrimethylammonium bromide (CTAB) micellar system in different pH range from 6.5–10 at 25°C. These results obtained indicate that the complex exhibits good catalytic function. It also appears the complex accelerates the hydrolytic cleavage of PNPA in cationic CTAB micellar solution which may be due to coordinating ability of substrate to complex, electrostatic interaction between micelles and complex and due to electrostatic interaction between micelles with reactant.  相似文献

Amphiphilic AB‐type diblock copolymers composed of hydrophobic poly(L ‐lactide) (PLA) segments and hydrophilic poly(glycolic acid lysine) [poly(Glc‐Lys)] segments with amino side‐chain groups self‐associated to form PLA‐based polymeric micelles with amino surfaces in an aqueous solution. The average diameter of the loose core–shell polymeric micelles for poly(Glc‐Lys) [number‐average molecular weight (Mn) = 1240]‐b‐PLA (Mn = 7000) obtained by a dimethyl sulfoxide/water dialysis method was estimated to be about 50 nm in water by dynamic light scattering measurements. The size and shape of the obtained polymeric micelles were further observed with transmission electron microscopy and atomic force microscopy. To investigate the possibility of applying the obtained PLA‐based polymeric micelles as bioabsorbable vehicles for hydrophobic drugs, we tested the entrapment of drugs in poly(Glc‐Lys) (Mn = 1240)‐b‐PLA (Mn = 7000) micelles and their release with doxorubicin as a hydrophobic drug. © 2002 Wiley Periodicals, Inc. The effects of Brij 35 micelles, CTABr micelles, and mixed Brij 35–CTABr micelles on the acid–base behavior of phenyl salicylate (PST) have been studied in aqueous solution containing 2% v/v acetonitrile. The apparent pKb (pKappb) of PST is decreased by 1.5 pK units with the increase in [Brij 35] from 0 to 0.02 M which is attributed to micellar medium effect. The values of pKappb remain almost independent of [CTABr] within its range 0.01–0.03 M. The increase in [CTABr] from 0 to 0.03 M in aqueous solution containing 0.02 M Brij 35 has not resulted in a change in pKappb. This shows that the characeristic structural features of nonionic Brij 35 micelles remain essentially unchanged on addition of CTABr under the present experimental conditions.  相似文献

An approach for the preparation of block copolymer vesicles through ultrasonic treatment of polystyrene‐block‐poly(2‐vinyl pyridine) (PS‐b‐P2VP) micelles under alkaline conditions is reported. PS‐b‐P2VP block copolymers in toluene, a selective solvent for PS, form spherical micelles. If a small amount of NaOH solution is added to the micelles solution during ultrasonic treatment, organic‐inorganic Janus‐like particles composed of the PS‐b‐P2VP block copolymers and NaOH are generated. After removal of NaOH, block copolymer vesicles are obtained. A possible mechanism for the morphological transition from spherical micelles to vesicles or Janus‐like particles is discussed. If the block copolymer micelles contain inorganic precursors, such as FeCl3, hybrid vesicles are formed, which may be useful as biological and chemical sensors or nanostructured templates. © 2014 Wiley Periodicals, Inc. Biodegradable chitosan-graft-poly(D,L-lactide) (CHI-PLA) have been prepared by in situ ring-opening polymerization of DL-lactide (LA) grafting from chitosan. The chemical structure and physical properties of these chitosan derivatives were characterized by FTIR, 1H-NMR, TGA and XRD. Formation and characteristics of polymeric micelles based on these chitosan derivatives were studied by fluorescence spectroscopy and dynamic light scattering (DLS). The critical micelle concentration (CMC) in water varied from 0.048 to 0.021 mg/mL as PLA grafting percentage (GP) increased from 92% to 132%. The mean diameters of CHI-PLA micelles have been readily tuned by controlling GP and the pH of micelles solution. The quasi-spherical micelles were observed by transmission electron microscopy (TEM). In particular, complexes formed by CHI-PLA micelles and plasmid DNA were investigated by agarose gel electrophoresis. Compared to chitosan, it could exhibit enhanced transfection efficiency in some cases and lower cell toxicity, as confirmed by in vitro transfection and cytotoxicity assay in L929 cell lines. These results indicated that CHI-PLA may be used as a new nonviral gene delivery vector for future gene therapy applications.  相似文献

The hydrolysis of bis(p‐nitrophenyl)phosphate (BNPP) catalyzed by N‐methyldiethanolamine‐Ce(III) complex in the presence and absence of cetyltrimethylammonium bromide (CTAB) and Brij35 surfactants at pH 7.20 and 303 K has been studied. The experimental results indicate that N‐methyldiethanolamine‐Ce(III) complex remarkably accelerates the hydrolysis of BNPP. The observed first‐order rate constant of the hydrolysis of BNPP catalyzed by N‐methyldiethanolamine‐Ce(III) complex at pH 7.20 and 303 K is 1.22 × 10?2 s?1, which is 1.09 × 109 times of that of spontaneous hydrolysis of BNPP at pH 7. It is close to the activity of natural enzyme. A general quantitative treatment of the catalytic reaction involved a ternary complex as MmLlS has also been proposed in this paper. Applying this method to the catalytic hydrolysis of BNPP, we have obtained its thermodynamic and kinetic parameters. CTAB and Brij35 surfactant micelles obviously influence the rate constants of the catalytic hydrolysis of BNPP. Functional nanostructures of self‐assembled block copolymers (BCPs) incorporated with various inorganic nanomaterials have received considerable attention on account of their many potential applications. Here we demonstrate the two‐dimensional self‐assembly of anisotropic titanium dioxide (TiO2) nanocrystals (NCs) and metal nanoparticles (NPs) directed by monolayered poly(styrene)‐block‐poly(4‐vinylpyridine) (PS‐b‐P4VP) copolymer inverse micelles. The independent position‐selective assembly of TiO2 NCs and silver nanoparticles (AgNPs) preferentially in the intermicelle corona regions and the core of micelles, respectively, for instance, was accomplished by spin‐coating a mixture solution of PS‐b‐P4VP and ex situ synthesized TiO2 NCs, followed by the reduction of Ag salts coordinated in the cores of micelles into AgNPs. Hydrophobic TiO2 NCs with a diameter and length of approximately 3 nm and 20–30 nm, respectively, were preferentially sequestered in the intermicelle nonpolar PS corona regions energetically favorable with the minimum entropic packing penalty. Subsequent high‐temperature annealing at 550 °C not only effectively removed the block copolymer but also transformed the TiO2 NCs into connected nanoparticles, thus leading to a two‐dimensionally ordered TiO2 network in which AgNPs were also self‐organized. The enhanced photocatalytic activity of the AgNP‐decorated TiO2 networks by approximately 27 and 44 % over that of Ag‐free TiO2 networks and randomly deposited TiO2 nanoparticles, respectively, was confirmed by the UV degradation property of methylene blue.  相似文献

Sugar-lipid hybrids of the type CnCm were prepared by coupling an alkane chain (Cn) with a maltooligosaccharide (Gm) over an amide linkage. Coupling was performed with maltobionolactone (G2) and n-alkylamine chains Cn withn=8,10,12,14,16, i.e. variation of the hydrophobic part of the molecule, and with hexadecylamine (C16) and different maltooligosaccharides (Gm, m=2,3,4,6). The solution properties of the various products were studied by means of static and dynamic light scattering (LS) and by electron-microscopy (EM).The results may be summarized as follows: If the alkane chain is shorter thann=14, small spherical micelles with a radius of about 3 nm are observed. In time these micelles aggregate further to form increasingly larger spherical clusters which eventually precipitate. Long rod-like micelles form whenn 14. Contour length and chain stiffness were determined by applying theories of semiflexible chains. A qualitative confirmation of the light scattering results, i.e., micelle size and shape, was obtained from electron microscopy.  相似文献
