A new p-tert-butyl-calix[8]arene-bonded silica gel stationary phase (CABS) was used for the separation of some aromatic carboxylic acids by HPLC. The chromatographic behaviour of the solutes on CABS was studied in comparison with conventional ODS. The results show that the calix[8]arene-bonded phase exhibits a stronger retention and better selectivity than ODS for the aromatic carboxylic acids. The different elution order of the analytes was also observed on both packings, which show the existence of distinct retention mechanisms. According to chromatographic data, it can be concluded that the high selectivity of CABS for aromatic carboxylic acids ascribes to various interactions between CABS and the analytes, such as hydrophobic interaction, hydrogen bonding interaction, π-π interaction and inclusion interaction.  相似文献

Summary For the first time calix[6]arene has been chemically combined with silica gel via a longer spacer to prepare calix[6]arene-bonded silica gel stationary phase for high-performance liquid chromatography (HPLC). The separation of positional isomers and polycyclic aromatic hydrocarbons on the calix[6]arene-bonded phase was achieved with methanol-water as mobile phase. Some nucleosides were also separated on the bonded phase. The reversed-phase chromatographic performance of the bonded phase was studied. The results showed that the calix[6]arene-bonded phase was highly hydrophobic. A possible separation mechanism has been proposed.  相似文献

In this paper, the chromatographic behavior of some water-soluble vitamins was studied on a new p-tert-butyl-calix[8]arene-bonded silica gel stationary phase (CABS, 5 μm particle size, the bonded amount 0.071 mmol g−1) by using vitamin standards as probes for HPLC. The comparative study of the separation of these compounds was done by using CABS and ODS as stationary phases under the same chromatographic conditions. The better separation of six vitamins including: B1, B2, B6, B12, C, and nicotinic acid (B5), on CABS can be achieved by using isocratic mode with methanol-phosphate buffer (25:75, (v/v)) as mobile phase within 20 min. The results show that the calix[8]arene-bonded phase exhibits high selectivity for water-soluble vitamins. We found that the elution order of B2 (12.08 min) and B12 (16.42 min) on CABS was very different from that of B12 (7.76 min) and B2 (18.47 min) on ODS, which indicate that different retention mechanisms exist in the chromatographic processes of the two stationary phases. According to the chromatographic data, it can be concluded that various chromatographic retention mechanisms are responsible for the separation of above compounds on CABS, such as hydrophobic interaction, hydrogen bonding interaction, and π-π interaction. The new packing has two advantages over ODS. On one hand, the polar and ionized analytes, such as C and B5, exhibited stronger affinities to CABS because of hydrogen bonding interaction. On the other hand, the retention of B2 and B12 became shorter on CABS with weaker hydrophobicity in comparison with ODS. The new material exhibits the promising application in the separation of water-soluble vitamins.  相似文献

The chromatographic behavior of some nucleosides, pyrimidines and purines on a new p-tert-butyl-calix[8]arene-bonded silica gel stationary phase (CABS) were studied by high performance liquid chromatography. Their retention behavior on CABS were compared with those on ODS. The influence of mobile phase variables, such as methanol content, pH and ionic strength on the retention behavior were studied. Some nucleosides, pyrimidines and purines on CABS were successfully separated. The results show that the calix[8]arene-bonded phase exhibits high selectivities for the above analytes in high aqueous mobile phases. According to the chromatographic data, it is indicated that hydrophobic interaction, hydrogen-bonding interaction, and dipole-dipole interaction are mainly responsible for the retention behavior. In addition, in some extent, the vertical stacking action of the analytes on CABS can also change the retention behavior. CABS was superior to ODS in the routine fast separation of nucleosides and bases.  相似文献

In this paper, perhydroxyl-cucurbit[6]uril ((HO)12CB[6]) was, for the first time, grafted to silica gel as a hydrophilic-interaction chromatographic stationary phase. Several alkaloids were used as probes to investigate the retention mechanism on the new stationary phase. The effect of mobile phase variables such as acetonitrile content, ionic strength and pH on their chromatographic behavior was investigated. The results indicate that the stationary phase behaves as a hydrophilic-interaction chromatographic packing. Finally, several alkaloids were separated on the stationary phase.  相似文献

A new 1,3-alternate 25,27-dibenzoiloxy-26,28-bis-[3-propyloxy]-calix[4]arene-bonded silica gel stationary phase (1,3-Alt CalixBn) has been prepared and used for the separation of aromatic positional isomers by high-performance liquid chromatography (HPLC). The effect of organic modifier content and pH of the mobile phase on retention and selectivity of these compounds were studied. We have synthesized the 1,3-alternate 25,27-dioctyloxy-26,28-bis-[3-aminopropyloxy]-calix[4]arene and then immobilized onto γ-chloropropylsilica gel (CPS). The high-performance liquid chromatographic behavior of some aromatic hydrocarbons, phenolic compounds, aromatic amines and drug compounds was studied on this 1,3-alternate-calix[4]arene-bonded silica gel stationary phase (CIMS). The effect of organic modifier content and pH of the mobile phase on retention and selectivity of these compounds were investigated. 采用固液相表面连续反应法,先将偶联剂γ-[(2,3)-环氧丙氧]丙基三甲氧基硅烷(KH-560)键合到球形硅胶表面,然后再将植物有效成分丁香酚与硅胶上的KH-560活性基团反应,合成了丁香酚键合硅胶液相色谱固定相(EGSP)。采用元素分析、热重分析和红外光谱对该固定相的结构进行了表征。以萘作为溶质探针,乙腈-水(35:65, v/v)为流动相,流速为0.8 mL/min,测得EGSP柱的柱效。以一系列的中性、碱性和酸性化合物为溶质探针,C18柱和苯基柱作参比,对该固定相的色谱性能及保留机理进行了研究。结果表明,硅胶表面成功键合上了丁香酚配体,键合量为0.28 mmol/g, EGSP柱理论塔板数约为24707 N/m。该固定相不仅具有良好的反相色谱性能,同时由于配体结构中含有芳环、烯基和甲氧基,还能与溶质发生π-π电荷转移、偶极-偶极和氢键作用。与传统的反相C18柱和苯基柱相比,EGSP在极性芳香族化合物的快速、简便分离中占优势。  相似文献

A new p-tert-butyl-calix[8]arene-bonded silica gel stationary phase (CABS) was prepared via 3-glycidoxypropyltrimethoxysilane as a coupling reagent for HPLC. Its structure was characterized by diffuse reflectance infrared Fourier transform spectroscopy (DRIFT), elemental analysis and thermal analysis. The chromatographic performance of new packing was evaluated by using basic, acidic and neutral aromatic compounds as probes compared with conventional ODS. The results show that the new stationary phase has an excellent reversed-phase property and high selectivities for substituted aromatics compared with ODS, because CABS can provide various sites for the analytes, such as hydrogen-bonding interactions, pi-pi interactions, and inclusion complex, besides hydrophobic interactions.  相似文献

Summary After summarizing published results on silica gel and alkyl-bonded silica gel solubilities, the experimental solubility of unmodified silica and copper silicate gel (used in ligand exchange chromatography) in watermethanol-ammonia and water-acetonitrile-ammonia mixtures are given. These results demonstrate that silica gel solubility, measured by a static method, varies greatly with the water volume fraction of a ternary mixture. Curiously, no influence from the organic nature of the solvent was demonstrated. This observation is inconsistent with the fact that the copper silicate gel used in ligand-exchange chromatography with water-methanol-ammonia as mobile phase is attacked more rapidly than with water-acetonitrile-ammonia.An explanation based upon the variations of acid-base properties of silicic acid versus mobile phase dielectric constant values in conjunction with the methanol protophilic properties is proposed. Finally, it is shown that chromatographic columns filled with copper silicate gel have lifetimes of several months, if they are used with a procolumn (of copper silicate gel) and with acetonitrile as organic solvent with a volume fraction greater than 0.5 in the mobile phase.  相似文献

The sulfonamides are stable chemotherapeutics used against the bacterial disease affecting bees, known as American foulbrood (Bacillus larvae), so their residues could appear in the honey of treated bees. Their presence at a concentration above the limit value is a potential hazard to human health. Brazilian authorities have included in the National regulatory monitoring program, the control of the three most widely used sulfonamides in honey production, i.e., sulfathiazole, sulfamethazine and sulfadimethoxine. A method for the determination of residual sulfonamides in honey, using sulfapyridine as an internal standard has been developed, optimized and validated. Some changes were implemented on current available methodologies for the analysis of sulfonamides in honey in order to adopt such procedures to Brazilian honey samples. Sulfonamides were extracted from honey with dichloromethane after dissolution with 30% sodium chloride, and cleaned up with solid phase extraction on Florisil columns. The eluate was analyzed by high-performance liquid chromatography with ultraviolet detection. The limit of detection was determined at 3 μg kg−1, 4 μg kg−1 and 5 μg kg−1 for sulfathiazole, sulfamethazine and sulfadimethoxine, respectively with average recoveries of 61.0% for sulfathiazole; 94.5% for sulfamethazine and 86.0% for sulfadimethoxine at the 100 μg kg−1 level. As the final step of validation procedure, the analysts were submitted to a blind spiked sample prepared by the quality assurance officer which results were successfully obtained regarding recovery and deviations.  相似文献

Summary A new stationary phase was prepared by immobilizing the chitosan, a natural hydrophilic polymer, on microparticolate silica gel. The effect of the polarity, pH and ionic strength of the mobile phase has been studied in order to find optimal conditions for the separation of nucleotides and aminoacids. The influence of the properties of the mobile phase on the retention was examined, allowing to employ the chitosan-coated silica gel both to anion exchange and adsorption, depending on the pH and the polarity of the eluent used.  相似文献

High-performance liquid chromatography (HPLC) separation of drugs at elevated pressure with 1.7 microm hybrid C18 stationary phase columns was investigated. This technique, which uses instrumentation engineered to handle the narrow peaks and high back pressures generated by 1.7 microm particle columns, provided significantly better resolution and/or faster analysis than conventional HPLC and capillary electrophoresis (CE). The use of 2mm internal diameter (i.d.) columns of 3-10 cm length has been evaluated for the separation of basic and neutral drugs, drug profiling, and general screening (including acidic drugs). For these applications, compared to conventional HPLC and CE, it provided up to 12x and 3x faster analyses, respectively. Precision was excellent for both isocratic and gradient analyses. For retention time and peak area, RSDs of < or =0.1% were obtainable. Fifteen anabolic steroids and esters were well separated in a 2.5 min gradient. For drug profiling, compared to HPLC and CE, approximately twice as many peaks were resolved. HPLC at elevated pressure is also well suited as a general screening technique. Twenty-four solutes of varying drug classes including narcotic analgesics, stimulants, depressants, hallucinogens, and anabolic steroids were fully separated in a 13.5 min gradient.  相似文献

将新制备的厚朴酚键合硅胶固定相(MSP)用于嘌呤、嘧啶、蝶呤及黄酮类化合物的液相色谱分离分析。选取了4种嘌呤、8种嘧啶、4种蝶呤及5种黄酮类药物作为极性化合物的代表,以商品反相碳十八烷基键合硅胶柱(ODS)作参照,研究了新固定相对碱性化合物的选择性和相关分离机理。实验发现,在简单流动相条件下,厚朴酚键合硅胶固定相对上述药物表现出较高的选择性及分离效果。尽管MSP没有进行封尾处理,但含氮类极性化合物(嘌呤、嘧啶、蝶呤)仍表现出基本对称的色谱峰形。多数药物在两柱上的洗脱顺序大致相同,说明疏水作用始终存在,这说明新固定相具有反相色谱性能。比较研究还发现,MSP在分离上述极性药物时能够提供除疏水性作用之外的其他作用位点。例如,在分离嘌呤、嘧啶及蝶呤时,氢键和偶极作用明显存在;同时MSP与溶质结构中的芳环(硫唑嘌呤、紫花牡荆素)之间有较强的π-π电子相互作用等,使得含氮类极性化合物和黄酮的保留一般比ODS强,分离度也有一定的改善。多种作用可以合理地解释MSP柱对极性溶质有更强的分离能力,厚朴酚键合硅胶固定相可在一定程度上弥补ODS单一疏水作用的不足,有利于分类碱性化合物。  相似文献

基于四氮杂杯[2]芳烃[2]三嗪键合硅胶吸附剂(NC-Si),构建了固相萃取-高效液相色谱法同时测定河水中3种硝基苯酚和己烯雌酚的新方法。考察并获得了固相萃取和液相色谱分离的优化条件:将样品溶液pH调至5,以5 mL/min上样,经自制固相萃取柱净化,2 mL氨水-甲醇(2:98,v/v)洗脱;在C8柱上以甲醇-0.1%磷酸溶液为流动相进行梯度洗脱。4种目标分析物的检出限(LOD,S/N=3)为0.03~0.3 μg/L,定量限(LOQ,S/N=10)为0.1~1.0 μg/L;加标回收率为75.5%~104.2%,相对标准偏差(RSD,n=5)小于6.3%。该方法准确、可靠,可用于河水中硝基苯酚及己烯雌酚的灵敏检测。  相似文献

By using ionic liquid as membrane liquid and tri-n-octylphosphine oxide (TOPO) as additive, hollow fiber supported liquid phase microextraction (HF-LPME) was developed for the determination of five sulfonamides in environmental water samples by high-performance liquid chromatography with ultraviolet detection The extraction solvent and the parameters affecting the extraction enrichment factor such as the type and amount of carrier, pH and volume ratio of donor phase and acceptor phase, extraction time, salt-out effect and matrix effect were optimized. Under the optimal extraction conditions (organic liquid membrane phase: [C8MIM][PF6] with 14% TOPO (w/v); donor phase: 4 mL, pH 4.5 KH2PO4 with 2 M Na2SO4; acceptor phase: 25 μL, pH 13 NaOH; extraction time: 8 h), low detection limits (0.1–0.4 μg/L, RSD ≤ 5%) and good linear range (1–2000 ng/mL, R2 ≥ 0.999) were obtained for all the analytes. The presence of humic acid (0–25 mg/L dissolved organic carbon) and bovine serum albumin (0–100 μg/mL) had no significant effect on the extraction efficiency. Good spike recoveries over the range of 82.2–103.2% were obtained when applying the proposed method on five real environmental water samples. These results indicated that this present method was very sensitive and reliable with good repeatabilities and excellent clean-up in water samples. The proposed method confirmed hollow fiber supported ionic liquid membrane based LPME to be robust to monitoring trace levels of sulfadiazine, sulfamerazine, sulfamethazine, sulfadimethoxine and sulfamethoxazole in aqueous samples.  相似文献

Summary Polypyrrole chloride and polypyrrole dodecylsulfate coated-silica packing materials have been synthesized. Reverse-phase chromtographuy was employed to characterise both packings. A series of test compounds with known properties was used as molecular probes. They included benzene and derivatives, basic drugs, and polyaromatic hydrocarbons. This work describes the separation performance of a perhydroxylcucurbit[6]uril stationary phase for capillary gas chromatography. The perhydroxylcucurbit[6]uril stationary phase exhibits higher resolving capabilities for critical analytes with better peak shapes than cucurbit[6]uril and conventional stationary phases. The advantageous separation feature of the perhydroxylcucurbit[6]uril stationary phase may originate from its unique structure and favorably balanced interactions with the analytes. Also, the perhydroxylcucurbit[6]uril column shows good separation repeatability with relative standard deviations in the range of 0.01–0.13% for intraday, 0.37–0.82% for interday, and 1.0–4.7% for column‐to‐column repeatability.  相似文献

采用固液相连续反应法,以γ-环氧丙氧基)丙基三甲氧基硅烷(KH560)为偶联剂,制备了一种喹啉醚基键合硅胶固定相(QBS),采用元素分析、漫反射红外光谱和热分析表征了固定相的结构。多种溶质为探针(包括非极性的烷基苯和多环芳烃、芳香族化合物位置异构体及极性的核苷和碱基等),较系统地研究了该新固定相的色谱性能。研究表明,新固定相与ODS相比,除具有弱的疏水性外,还能与溶质发生多种作用,如:氢键和π-π作用等。在分离非极性的多环芳烃时主要基于疏水作用;在分离极性的核苷和碱基时,氢键和络合作用较重要;在分离芳香族化合物位置异构体时,溶质极性取代基与喹啉醚基键合相的氢键作用。溶质苯环与喹啉基配体之间的π-π作用,两协同作用提高了QBS对位置异构体的分离选择性。  相似文献
