[structure: see text] A novel tetracyclic alkaloid, perinadine A (1), was isolated from the cultured broth of the fungus Penicillium citrinum, which was separated from the gastrointestine of a marine fish, and the structure was elucidated on the basis of spectroscopic data including 2D NMR spectra. Biogenetically, perinadine A (1) may be derived from citrinin (4), a well-known mycotoxin, and a scalusamide A-type pyrrolidine alkaloid.  相似文献

Three new citrinin derivatives, penicitrinols C, D, and E (1-3), along with two known compounds, citrinin (4) and decarboxydihydrocitrinone (5), were isolated from Penicillium citrinum. Perpyrrospirone A (1) characterized an unprecedented 6/5/6/8/5/13/6 oxahexacyclic scaffold with a unique peroxide-bridged 8,9-dioxa-2-azaspiro[4.7]dodecane core from marine-derived Penicillium citrinum. Compounds 2 and 3 possessed rare oxatetracyclic (6/5/6/5) skeleton fused with a 13-menbered-ring macrocyclic moiety. Their structure and absolute configurations were determined by comprehensive spectroscopic analyses, ECD data coupled with TD-DFT calculations and X-ray diffraction experiments. In addition, 7 showed cytotoxicity and induced apoptosis of Hela cells in a dose-dependent manner after a 48 h treatment.  相似文献

A novel sorbicillinoid named sorbiterrin A (1), possessing an unprecedented skeleton was isolated from a marine sediment derived fungus Penicillium terrestre. The structure including absolute stereochemistries was elucidated by analysis of NMR, MS data and TDDFT CD calculations. Penispirolloid A (1), a novel alkaloid possessing a unique spiro imidazolidinyl skeleton, was isolated from a halotolerant fungal strain, Penicillium sp. OUCMDZ-776. Its structure was elucidated on the basis of spectroscopic methods and quantum chemical CD calculation. Compound 1 showed significant antifouling activity toward Bugula neritina larvae with EC50 of 2.40 μg/ml. Plausible biogenesis of compound 1 was also proposed.  相似文献

A new diketopiperazine alkaloid, golmaenone (1). and related alkaloids, neoechinulin A (2). and L-alanyl-L-tryptophan anhydride (3). have been isolated from the culture broth of the marine-derived fungus Aspergillus sp. The structure and absolute stereochemistry of the new compound (1). was assigned by spectroscopic methods and the advanced Marfey's method. Compounds 1 and 2 exhibited a significant radical scavenging activity against 1,1-diphenyl-2-picrylhydrazyl (DPPH) with IC(50) values of 20 and 24 microM, respectively, which are similar to the positive control, ascorbic acid (IC(50), 20 microM). Compounds 1 and 2 also showed an ultraviolet-A (UV-A) (320--390 nm) protecting activity with ED(50) values of 90 and 170 microM, respectively, which are more active than oxybenzone (ED(50), 350 microM) currently being used as sunscreen.  相似文献

One new benzopyran derivative (2R*,4R*)-3,4-dihydro-5-methoxy-2-methyl-2H-1-benzopyran-4-ol (1), together with five known compounds (2?6), were obtained from the EtOAc extract of the endophytic fungus Penicillium citrinum HL-5126 isolated from the mangrove Brguiera sexangula var. rhynchopetala collected in the South China Sea. Their structures were elucidated by the detailed analysis of comprehensive spectroscopic data. All compounds were evaluated for their antibacterial activities. Compound 6 exhibited potent inhibitory activity against Bacillus subtilis, Bacillus cereus and Micrococcus tetragenus with the same MIC values of 6.94 μM.  相似文献

Penibishexahydroxanthone A, a new hexahydroxanthone homodimer with a rare highly oxidized hexacyclic framework, was identified from the marine-derived Penicillium sp. ZZ486A. Its structure was determined by a combination of extensive NMR spectroscopical analyses, HRESIMS data, ECD calculation, and single crystal X-ray diffraction analysis. Penibishexahydroxanthone A showed in vivo activity in promoting angiogenesis in a dose-dependent manner by rescuing VRI-induced vascular insufficiency in zebrafish. A new polyene compound (1) and a new diketopiperazine (2), as well as three known compounds (35), were isolated from the Antarctic marine-derived fungus Penicillium crustosum HDN153086. The structures of 15 were deduced based on MS, NMR and TD-DFT calculations of specific ECD spectra. These compounds were evaluated for their cytotoxic activities against K562 cell line and only compound 2 exhibited cytotoxicity against K562 cell, with IC50 value of 12.7 μM.  相似文献

Five new polyketides including two benzopyranones (1 and 2), one isochroman (3) and two anthraquinone-citrinin derivatives (4 and 5) were isolated from the sea fan-derived fungus Penicillium citrinum PSU-F51 together with thirteen known compounds. The structures were determined by spectroscopic methods. The anthraquinone-citrinin derivatives are rare natural products. Compound 4 displayed moderate antibacterial activity against both Staphylococcus aureus and methicillin-resistant S. aureus with equal MIC values of 16 μg/mL, while the known coniochaetone A displayed moderate antifungal activity against Candida albicans with MIC value of 16 μg/mL.  相似文献

Shimalactone A (1), a novel polyketide having bicyclo[4.2.0]octadiene and oxabicyclo[2.2.1]heptane units, was isolated from a cultured marine fungus of Emericella variecolor GF10. The stereo structure of 1 was deduced from 2D NMR and X-ray crystallographic analyses of 1 and its derivatives. The absolute structure of 1 was determined by application of the CD allylic benzoate rule to the benzoate derivative of 1. Shimalactone A (1) induced neuritogenesis at 10 μg/mL against neuroblastoma Neuro 2A cells.  相似文献

A new sesterterpene epoxide-diol, aspergilloxide (1), was isolated from the extract of a cultured marine-derived fungus (strain CNM-713) identified as an undescribed member of the genus Aspergillus. The structure of 1 was determined by interpretation of NMR data and by chemical methods. The absolute stereochemistry of aspergilloxide was assigned by application of the modified Mosher method. The carbon skeleton of 1 represents a new addition to the architectural diversity of the sesterterpenoid (C25) class of secondary metabolites.  相似文献

Two new compounds penibenzophenones A-B (1–2), and the synthetic α,β-unsaturated amide alkaloid (E)-tert-butyl(3-cinnamamidopropyl)carbamate (4), newly identified as a natural product, alone with three known ones (3, 5–6) were isolated from the EtOAc extract of the endophytic fungus Penicillium citrinum HL-5126 isolated from the mangrove Bruguiera sexangula var. rhynchopetala collected in the South China Sea. Compound 1 was a chlorinated benzophenone. The structures of 1–6 were elucidated by extensive NMR spectral interpretation, MS data and X-ray analysis. The new compound 2 displayed cytotoxic activity against human A549 cell lines with an IC50 value of 15.7 μg/mL, and 1 showed antibacterial activity against Staphylococcus aureus with a MIC value of 20 μg/mL.  相似文献
