Perinadine A, a novel tetracyclic alkaloid from marine-derived fungus Penicillium citrinum

[structure: see text] A novel tetracyclic alkaloid, perinadine A (1), was isolated from the cultured broth of the fungus Penicillium citrinum, which was separated from the gastrointestine of a marine fish, and the structure was elucidated on the basis of spectroscopic data including 2D NMR spectra. Biogenetically, perinadine A (1) may be derived from citrinin (4), a well-known mycotoxin, and a scalusamide A-type pyrrolidine alkaloid.     

Citrinin derivatives from the marine-derived fungus Penicillium citrinum

Three new citrinin derivatives, penicitrinols C, D, and E (1-3), along with two known compounds, citrinin (4) and decarboxydihydrocitrinone (5), were isolated from Penicillium citrinum. Their structures were determined by spectroscopic methods and X-ray diffraction analysis. Compounds 1 and 3 demonstrated weak cytotoxicity against the HL-60 cell line.     

Speradine A, a new pentacyclic oxindole alkaloid from a marine-derived fungus Aspergillus tamarii

A new pentacyclic oxindole alkaloid, speradine A (1), was isolated from the cultured broth of a fungus Aspergillus tamarii, which was separated from driftwood at a seashore in Okinawa. The structure and relative stereochemistry were determined by spectroscopic data and a single crystal X-ray diffraction analysis.     

Seragakinone A, a new pentacyclic metabolite from a marine-derived fungus

A new anthracycline-derived pentacyclic metabolite, seragakinone A (1), was isolated from the mycelium of an unidentified fungus, which was separated from the Okinawan marine rhodophyta Ceralodictyon spongiosum, and the structure was elucidated by spectroscopic data.     

Perpyrrospirone A,an unprecedented hirsutellone peroxide from the marine-derived Penicillium citrinum

Perpyrrospirone A (1) characterized an unprecedented 6/5/6/8/5/13/6 oxahexacyclic scaffold with a unique peroxide-bridged 8,9-dioxa-2-azaspiro[4.7]dodecane core from marine-derived Penicillium citrinum. Compounds 2 and 3 possessed rare oxatetracyclic (6/5/6/5) skeleton fused with a 13-menbered-ring macrocyclic moiety. Their structure and absolute configurations were determined by comprehensive spectroscopic analyses, ECD data coupled with TD-DFT calculations and X-ray diffraction experiments. In addition, 7 showed cytotoxicity and induced apoptosis of Hela cells in a dose-dependent manner after a 48 h treatment.     

Sorbiterrin A,a novel sorbicillin derivative with cholinesterase inhibition activity from the marine-derived fungus Penicillium terrestre

A novel sorbicillinoid named sorbiterrin A (1), possessing an unprecedented skeleton was isolated from a marine sediment derived fungus Penicillium terrestre. The structure including absolute stereochemistries was elucidated by analysis of NMR, MS data and TDDFT CD calculations. Sorbiterrin A showed moderate acetylcholinesterase (AChE) inhibitory effect with IC₅₀ 25 μg/mL.     

A new alkaloid from the marine-derived fungus Hypocrea virens

A new alkaloid, 2-methylimidazo[1,5-b]isoquinoline-1,3,5(2H)-trione, was isolated from the EtOAc extract of the marine-derived fungus Hypocrea virens. Its structure was confirmed by spectroscopic methods (1D and 2D NMR, MS).     

A new polyketide,penicillolide from the marine-derived fungus Penicillium sacculum

A new polyketide, penicillolide (1) was isolated from the fermentation broth of the marine-derived fungus Penicillium sacculum GT-308. Compound 1 is a polyketide with a unique carbon skeleton. The structure of this compound was established via extensive spectroscopic analyses including 1D-, 2D-NMR, and HRESI-MS.     

A novel antifouling alkaloid from halotolerant fungus Penicillium sp. OUCMDZ-776

Penispirolloid A (1), a novel alkaloid possessing a unique spiro imidazolidinyl skeleton, was isolated from a halotolerant fungal strain, Penicillium sp. OUCMDZ-776. Its structure was elucidated on the basis of spectroscopic methods and quantum chemical CD calculation. Compound 1 showed significant antifouling activity toward Bugula neritina larvae with EC₅₀ of 2.40 μg/ml. Plausible biogenesis of compound 1 was also proposed.     

Golmaenone, a new diketopiperazine alkaloid from the marine-derived fungus Aspergillus sp

A new diketopiperazine alkaloid, golmaenone (1). and related alkaloids, neoechinulin A (2). and L-alanyl-L-tryptophan anhydride (3). have been isolated from the culture broth of the marine-derived fungus Aspergillus sp. The structure and absolute stereochemistry of the new compound (1). was assigned by spectroscopic methods and the advanced Marfey's method. Compounds 1 and 2 exhibited a significant radical scavenging activity against 1,1-diphenyl-2-picrylhydrazyl (DPPH) with IC(50) values of 20 and 24 microM, respectively, which are similar to the positive control, ascorbic acid (IC(50), 20 microM). Compounds 1 and 2 also showed an ultraviolet-A (UV-A) (320--390 nm) protecting activity with ED(50) values of 90 and 170 microM, respectively, which are more active than oxybenzone (ED(50), 350 microM) currently being used as sunscreen.     

Cephalocyclidin A, a novel pentacyclic alkaloid from Cephalotaxus harringtonia var. nana

A novel alkaloid with an unprecedented fused-pentacyclic skeleton and six-consecutive asymmetric centers, cephalocyclidin A (1), has been isolated from the fruits of Cephalotaxus harringtonia var. nana, and the structure was elucidated on the basis of spectroscopic data. The relative and absolute stereochemistry of 1 was determined by a combination of NOESY correlations, X-ray crystallographic data, and the exciton chirality method.     

A new benzopyrans derivatives from a mangrove-derived fungus Penicillium citrinum from the South China Sea

One new benzopyran derivative (2R^*,4R^*)-3,4-dihydro-5-methoxy-2-methyl-2H-1-benzopyran-4-ol (1), together with five known compounds (2?6), were obtained from the EtOAc extract of the endophytic fungus Penicillium citrinum HL-5126 isolated from the mangrove Brguiera sexangula var. rhynchopetala collected in the South China Sea. Their structures were elucidated by the detailed analysis of comprehensive spectroscopic data. All compounds were evaluated for their antibacterial activities. Compound 6 exhibited potent inhibitory activity against Bacillus subtilis, Bacillus cereus and Micrococcus tetragenus with the same MIC values of 6.94 μM.     

Proangiogenic penibishexahydroxanthone A from the marine-derived fungus Penicillium sp. ZZ486A

Penibishexahydroxanthone A, a new hexahydroxanthone homodimer with a rare highly oxidized hexacyclic framework, was identified from the marine-derived Penicillium sp. ZZ486A. Its structure was determined by a combination of extensive NMR spectroscopical analyses, HRESIMS data, ECD calculation, and single crystal X-ray diffraction analysis. Penibishexahydroxanthone A showed in vivo activity in promoting angiogenesis in a dose-dependent manner by rescuing VRI-induced vascular insufficiency in zebrafish. This study is the first report of the proangiogenic activity of hexahydroxanthones.     

Phomactin H, a novel diterpene from an unidentified marine-derived fungus

Phomactin H (1), a diterpene has been isolated from cultures of an unidentified marine-derived fungus. The structure and relative stereochemistry of phomactin H (1) was determined by X-ray diffraction analysis. Phomactin H (1), having an oxepane moiety, is a new phomactin with a novel skeleton.     

Secondary metabolites from Antarctic marine-derived fungus Penicillium crustosum HDN153086

A new polyene compound (1) and a new diketopiperazine (2), as well as three known compounds (3–5), were isolated from the Antarctic marine-derived fungus Penicillium crustosum HDN153086. The structures of 1–5 were deduced based on MS, NMR and TD-DFT calculations of specific ECD spectra. These compounds were evaluated for their cytotoxic activities against K562 cell line and only compound 2 exhibited cytotoxicity against K562 cell, with IC₅₀ value of 12.7 μM.     

Bioactive polyketides from the sea fan-derived fungus Penicillium citrinum PSU-F51

Five new polyketides including two benzopyranones (1 and 2), one isochroman (3) and two anthraquinone-citrinin derivatives (4 and 5) were isolated from the sea fan-derived fungus Penicillium citrinum PSU-F51 together with thirteen known compounds. The structures were determined by spectroscopic methods. The anthraquinone-citrinin derivatives are rare natural products. Compound 4 displayed moderate antibacterial activity against both Staphylococcus aureus and methicillin-resistant S. aureus with equal MIC values of 16 μg/mL, while the known coniochaetone A displayed moderate antifungal activity against Candida albicans with MIC value of 16 μg/mL.     

Shimalactone A, a novel polyketide, from marine-derived fungus Emericella variecolor GF10

Shimalactone A (1), a novel polyketide having bicyclo[4.2.0]octadiene and oxabicyclo[2.2.1]heptane units, was isolated from a cultured marine fungus of Emericella variecolor GF10. The stereo structure of 1 was deduced from 2D NMR and X-ray crystallographic analyses of 1 and its derivatives. The absolute structure of 1 was determined by application of the CD allylic benzoate rule to the benzoate derivative of 1. Shimalactone A (1) induced neuritogenesis at 10 μg/mL against neuroblastoma Neuro 2A cells.     

Aspergilloxide, a novel sesterterpene epoxide from a marine-derived fungus of the genus Aspergillus

A new sesterterpene epoxide-diol, aspergilloxide (1), was isolated from the extract of a cultured marine-derived fungus (strain CNM-713) identified as an undescribed member of the genus Aspergillus. The structure of 1 was determined by interpretation of NMR data and by chemical methods. The absolute stereochemistry of aspergilloxide was assigned by application of the modified Mosher method. The carbon skeleton of 1 represents a new addition to the architectural diversity of the sesterterpenoid (C25) class of secondary metabolites.     

Two new benzophenones and one new natural amide alkaloid isolated from a mangrove-derived Fungus Penicillium citrinum

Two new compounds penibenzophenones A-B (1–2), and the synthetic α,β-unsaturated amide alkaloid (E)-tert-butyl(3-cinnamamidopropyl)carbamate (4), newly identified as a natural product, alone with three known ones (3, 5–6) were isolated from the EtOAc extract of the endophytic fungus Penicillium citrinum HL-5126 isolated from the mangrove Bruguiera sexangula var. rhynchopetala collected in the South China Sea. Compound 1 was a chlorinated benzophenone. The structures of 1–6 were elucidated by extensive NMR spectral interpretation, MS data and X-ray analysis. The new compound 2 displayed cytotoxic activity against human A549 cell lines with an IC₅₀ value of 15.7 μg/mL, and 1 showed antibacterial activity against Staphylococcus aureus with a MIC value of 20 μg/mL.
