Previously, we reported an interesting reaction by which esculeogenin A [(5alpha,22S,23S,25S)-3beta,23,27-trihydroxyspirosolane], a sapogenol of tomato-saponin, esculeoside A, was easily converted into a pregnane derivative, 5alpha-pregn-16-en-3beta-ol-20-one, merely by refluxing with pyridine and water. Its chemical mechanism including air oxidation is here described.  相似文献

The stems of the Chinese traditional medicine Stephanotis mucronata were screened for immunologically active pregnane glycosides using high-performance liquid chromatography (HPLC) coupled with electrospray ionization tandem mass spectrometry. In the mass spectra of pregnane glycosides, predominant [M+Na]+ ions were observed and used to determine the molecular masses, while fragmentation reactions of the [M+Na]+ ions were recorded to provide information on the primary sequences of oligosaccharide chains in terms of classes of monosaccharide. Fragment ions from the side-chain cleavage of aglycone portions can provide mass information about side-chain substitutions. To further confirm the fragment ion structures, Fourier transform ion cyclotron resonance tandem mass spectrometry (MSn) with low-energy collision-induced dissociation was performed using samples collected from HPLC fractions, which provided accurate elemental compositions of fragment ions. Based on fragmentation patterns and comparison with standards, ten pregnane glycosides were identified as stemucronatosides C, D, F, and G, mucronatosides A, B, and C, stephanoside E, and two glycosides that are identified in the S. mucronata extracts for the first time. The latter two pregnane glycosides are 12-O-cinnamoyldeacetylmetaplexigenin-3-O-6-deoxy-3-O-methyl-beta-D-allopyranosyl-(1 --> 4)-beta-D-cymaropyranosyl-(1 --> 4)-beta-D-cymaropyranoside and 12-O-cinnamoyl-20-O-acetyl (20S)-pregn-6-ene-3beta,5alpha,8beta,12beta,14beta,17beta,20-heptaol 3-O-beta-D-thevetopyranosyl-(1 --> 4)-beta-D-cymaropyranosyl-(1 --> 4)-beta-D-cymaropyranoside.  相似文献

A spirosolane derivative possessing a hydroxyl group at C-23, esculeogenin A, a sapogenol of tomato saponin, was found to be easily converted into the corresponding pregnane derivative by refluxing with aqueous pyridine. Therefore, introduction of a hydroxyl group into the C-23 of diosgenin (as representative of spirostane derivatives) and solasodine (as representative of spirosolane derivatives) was attempted by the reaction of NaNO(2)-BF(3)?·?Et(2)O. In diosgenin, the objective compound was obtained by the reaction in AcOH. However, in solasodine, we obtained a 23-nitroso derivative by the reaction in AcOH and 23,24-bisnorcholanic acid 22-16 lactone, or vespertilin, in AcOH and CHCl(3).  相似文献

Moderate acetylation of tomatidine with anhydrous acetic acid and pyridine for 20 h at r.t., followed by pseudomerization in ice-water, gave a delta(20(22))-pseudo compound, which was then subjected to ozonolysis to provide a pregnane derivative in a total 54% yield showing neuritogenic and NGF-enhancing activities.  相似文献

Two novel reactions with regard to steroidal alkaloid sapogenols obtained from ripe tomato have been found. These reactions have been attained by simple facile method in only refluxing with pyridine and water. One is isomerization of a spirosolane derivative, isoesculeogenin A (2), into a rare solanocapsine derivative, esculeogenin B (3), disclosing a chemical correlation between isoesculeogenin A and esculeogenin B. Another is conversion of a 23-hydroxyspirosolane derivative, esculeogenin A (1), into a pregnane derivative (4), presenting an epoch-making method for derivation of steroidal hormone material.  相似文献

The isolevuglandin 17-isoLGE4 (10-acetyl-11-formyl-14-hydroxynonadeca-4(Z),7(Z),12(E),16(Z)-tetr aenoic acid) is a levulinaldehyde derivative that is expected to be generated during the free radical-induced oxidation of docosahexaenoic acid. A total synthesis was executed to facilitate detection and identification of 17-isoLGE4 in biological samples. Conjugate addition of a higher order vinyl cyanocuprate to a gamma-alkoxy enone achieved the final carbon-carbon bond formation to complete a convergent elaboration of the 17-isoLGE4 carbon skeleton. Attempted construction of the requisite vinyl nucleophile synthon using hydrostannylation of an alkyne was foiled by tri-n-butylstannyl radical-promoted isomerization of a cis to a trans double bond. Derivatization of 17-isoLGE4 with methoxylamine under anhydrous or wet conditions generated bismethoximes of 17-isoLGE4 or the isomerized delta11-17-isoLGE4 respectively. Analysis of the mass spectrum of a bismethoxime-pentafluorobenzyl ester-trimethylsilyl ether derivative of 17-isoLGE4 provided presumptive evidence that an incorrect structure was proposed earlier for C22-PGF4alpha, the only F4-isoprostane which is produced enzymatically. We conclude that the 22-carbon analogue of PGF2alpha, produced from docosahexaenoic acid by a cyclooxygenase from trout gill, does not have the same side chains as 17-isoLGE4. Furthermore, we now propose that mass spectral data reported for "C22-PGF4alpha" support a 14-PGF4alpha structure rather than the 17-PGF4alpha structure suggested previously.  相似文献

The roots of Trachycalymma fimbriatum (WEIMARCK ) BULLOCK contain both cardenolide and pregnaneglycosides. Elimination of 2-deoxysugars by mild acid hydrolysis gave a mixture from which some anhydroderivatives and the following compounds could be isolated: uzarigenin ( l ), ascleposide = 3-O-(6-deoxy-β-D -allopyranosyl)-uzarigenin ( 4 ), coroglaucigenin ( 6 ) and two pregnane derivatives (H and J). Compound H could be identified as 3β,14β-dihydroxy5α, 17α-pregnan-20-one ( 10 ). Compound J is probably a new substance, for which we tentatively assign structure 18 , i.e. 3β8β,14β-trihydroxy-5α,17α-pregnan-20-one. We suspect H and J to be artefacts produced from the corresponding 17b-derivatives during acid hydrolysis. 17-iso-H is probably a precursor in the biosynthesis of uzarigenin. The cardenolides of Trachycalymma fimbriatum are the same as found in Asclepias glaucophylla, a closely related species, while the pregnane derivatives of the latter are distinctly higher hydroxylated.  相似文献

Reaction of K(10)[alpha(2)-P(2)W(17)O(61)] or K(10)[alpha(1)-P(2)W(17)O(61)] or [Bu(4)N][OsCl(4)N] in a water/methanol mixture, and subsequent precipitation with (Bu(4)N)Br provided [alpha(2)-P(2)W(17)O(61){Os(VI)N}](7-) and [alpha(1)-P(2)W(17)O(61){Os(VI)N}](7-) Dawson structures as tetrabutylammonium salts. Reactions of [(Bu(4)N)(4)][alpha-H(3)PW(11)O(39)] with either [ReCl(3)(N(2)Ph(2))(PPh(3))(2)] or [Bu(4)N][ReCl(4)N] are alternatives to the synthesis of [(Bu(4)N)(4)][alpha-PW(11)O(39){Re(VI)N}]. It has been shown that under mild conditions the reactions of 17α-hydroxy 3,20-diones of the pregnane series with acid hydrazides lead to the formation of 3-monohydrazones, while under severe conditions 3,20-dihydrazones are formed regardless of whether a C4–C5 ethylenic bond or a chain of conjugation of C4–C5 and C6–C7 ethylenic bonds is present in the steroid molecule. The reaction of 17α-acetoxy 3,20-diketones of the pregnane series with acid hydrazides takes place only with the formation of 3-monohydrazones. An investigation of the gestagenic action of some of the compounds synthesized has shown that the presence of a 3-keto group in the steroids of the pregnane series is not necessary for the retention of this effect. The replacement of the keto group at carbon atom 3 by an azomethine group does not abolish the gestagenic action.  相似文献

New 1-acyl-sialosyl-glycerol derivatives (1a--d alpha, 1a--d beta, 2 alpha, 2 beta, which mimic the structure of the capsular polysaccharide of group C meningococcal were synthesized by the use of a chiral glycerol derivative, and were found to have phospholipases A2 and C inhibitory activities. Furthermore, synthesis of 2-palmitoyl-sialosyl-glycerol derivative (4 alpha, 4 beta, 5 alpha, 5 beta), galactosyl-glycerol derivative (6), and sialosyl-galactosyl-glycerol derivative (7) were carried out to examine the difference between these activities. Among these sialosyl derivatives, 3-palmitoyl-sialosyl-glycerol derivatives (1--3 alpha, 1--3 beta) demonstrated the most potent inhibitory activities.  相似文献

In the field of functionalized polyoxometalates, organosilyl derivatives of polyoxotungstate constitute a special class of hybrid organic-inorganic system. The first organosilyl derivative of the monovacant Dawson heteropolyoxotungstate [alpha2-P2W17O61]10- was obtained by three different methods. The use of two organosilanes as reagents enabled the preparation of the functionalized polyoxometalate [alpha2-P2W17O61(RSi)2O]6- in good yield. Electrospray (ESI-MS) and matrix-assisted laser desorption ionization time-of-flight (MALDI-TOF) mass spectrometry, and 183W, 31P, and 29Si NMR spectroscopy were used to characterize the composite systems. In several cases, ESI-MS analyses generated reduction processes which were compared to those related to [PMo11VO40]4-, the highly reducible Keggin polyoxometalate.  相似文献

[reaction: see text]. A method was found for the novel synthesis of ortho ester derivatives that are potentially useful as selective epsilon opioid receptor ligands. An unexpected 17-(cyclopropylmethyl)-4,5alpha-epoxy-6alpha-hydroxy-3,7,7-trimethoxy-8-oxa-6,14-endoethanomorphinan was produced when 17-(cyclopropylmethyl)-4,5alpha-epoxy-3-methoxy-6alpha,14-dihydroxy-6beta-(1,3-dithia-2-yl)-morphinan was treated in methanol with trimethyl orthoformate and CuO/CuCl2. This ortho ester derivative was then converted to an ester with acid. The structure of the ortho ester was determined by 2D NMR (HMBC) and mass spectra.  相似文献

Stemmosides C and D, two novel pregnane glycosides characterized by an unusual C-17 side chain were isolated from the pericarps of Solenostemma argel. In addition, stemmoside D displays an uncommon 14β proton configuration, apparently being the first pregnane isolated from plants known to have a 15 keto, cis CD ring junction. Their structures have been established by ESIMS and NMR experiments. The relative configuration of the molecules was determined using a strategy based on the simulation of 1H, 13C, and J coupling NMR parameters. DFT calculations of 1H and 13C chemical shifts, and of the 1H homonuclear spin–spin coupling constants were performed with the mPW1PW91 functional using the 6-31G(d,p) basis set on the fully optimized geometries of all the possible stereoisomers.  相似文献

Four novel withanolide-type steroids named cilistols p, pm, p1 and u (1-4, respectively), were isolated from the leaves of Solanum cilistum. The respective structures were characterized by spectroscopic means as follows: cilistol p (1) was (22R,24R,25R,26S)-1-oxo-22,26-epoxy-3alpha,5alpha-cycloergostane-6beta,17alpha, 24,25,26-pentaol 26-O-beta-D-glucopyranoside, cilistol pm (2) corresponded to the 6-O-methyl ether derivative of 1; cilistol p1 (3) was represented as the 24-O-methyl ether of 1, and cilistol u (4) was shown to be the epoxide between C-24 and -25, presumably bearing cilistols p, pm and p1 by ring-opening.  相似文献

Treatment of an alpha,beta-unsaturated ketimine with an alpha,beta-unsaturated carbonyl compound in the presence of a rhenium complex, Re2(CO)10, gave a cyclopentadienyl-rhenium complex. This reaction proceeds via rhenium-catalyzed C-H bond activation of an olefinic C-H bond, insertion of an alpha,beta-unsaturated carbonyl compound into a Re-C bond of the alkenylrhenium intermediate, intramolecular nucleophilic cyclization, reductive elimination, elimination of aniline to give a cyclopentadiene derivative, followed by the formation of a cyclopentadienyl-rhenium complex from the cyclopentadiene derivative and the rhenium complex.  相似文献

A "spin diverse" S = 3/2 ground-state complex, NN-SQCuTpCum,Me, has been prepared. The three S = 1/2 spin carriers are nitronyl nitroxide (NN), o-semiquinone (o-SQ), and cupric ion. The solid-state structure of the ZnII derivative, NN-SQZnTpCum,Me (C56H69BN8O4Zn), was determined: monoclinic, P2(1)/c, a = 12.5781(12) A, b = 17.7408(17) A, c = 24.440(2) A, alpha = 90.00 degrees, beta = 98.240(2) degrees, gamma = 90.00 degrees, Z = 4. The results of X-ray structural characterization of the ZnII derivative suggest substantial interaction between the two spin carriers NN and o-SQ. Indeed, strong intramolecular exchange coupling has been determined by variable-temperature magnetic susceptibility studies. Intraligand ferromagnetic exchange is considerably greater than kT, such that only the triplet state is populated at 300 K, and CuII-ligand exchange is ferromagnetic, with J = +75 cm-1.  相似文献

The gas chromatographic (GC) and GC-mass spectrometric properties of the diethylhydrogensilyl-cyclic diethylsilylene (DEHS-DES) derivatives of prostaglandin (PG) F1 alpha methyl ester, PGF2 alpha methyl ester, 6-keto-PGF1 alpha methyl ester-alkyloxime and thromboxane (TX) B2 methyl ester-alkyloxime and the DES derivative of 13,14-dihydro-15-keto-PGF2 alpha methyl ester-alkyloxime were studied. When the ketonic PGs and TXB2 were converted into their methyloxime derivatives, the methylene unit values of these five prostanoid derivatives were slightly greater than those of the corresponding dimethylethylsilyl ether derivatives. When the ketonic PGs were converted into their corresponding ethyloxime derivatives, baseline separation was achieved in 20 min by use of a methylsilicone cross-linked fused-silica capillary column. The mass spectra of these derivatives were characterized by the ion at m/z 157 for F alpha prostaglandins and m/z 269 for TXB2. The major fragmentations were directed by the DES group, and other fragmentations common to the prostanoid derivatives were losses of an ethyl radical at the silicon atom, C5H11 hydrocarbon fragment, diethylhydrogensilanol and C15-C20 hydrocarbon fragment. The mass fragmentations of these prostanoid derivatives are briefly discussed. GC with high-resolution selected-ion monitoring was carried out for the TXB2 derivative at a resolution of 8000 by monitoring the ion at m/z 269.1573. A 25-pg amount of this derivative showed a well shaped doublet with a signal-to-noise ratio of more than 300:1.  相似文献
