2-Hydroxy-4H-benzo[4,5]thiazolo[3,2-a]pyrimidin-4-one 2a and 7-hydroxy-5H-thiazolo[3,2-a]pyrimidin-5-one 2b, were obtained in high yields under mild conditions from the cyclization reactions of bis-(2,4,6-trichlorophenyl) malonate and 2-aminobenzothiazole or 2-aminothiazole, respectively. A new class of compounds, 2,3-dihydro-4H-benzo[4,5]thiazolo[3,2-a]furo[2,3-d]pyrimidin-4-ones and 6,7-dihydro-5H-furo[2,3-d]thiazolo[3,2-a]pyrimidin-5-ones, were synthesized via the microwave assisted radical addition of compounds 2a and 2b to various alkenes using manganese(III) acetate. A preliminary acetylcholine esterase (AchE) inhibition test of compound 4e showed excellent (92%) inhibitory potential, comparable with the standard drug Donapezil®.  相似文献

A novel series of conjugates of benzosuberone and 1,2,3-triazole i.e. 3-(4-phenyl-1H-1,2,3-triazol-1-yl)propyl-9-chloro-2,3-dimethyl-6,7-dihydro-5H-benzo[7]annulene-8-carboxylic acids (8a-j) were synthesized in good to excellent yields catalysed by CuSO4 under milder reaction conditions and evaluated for their anti-proliferative activity. The structural elucidation of the prepared compounds was carried out using IR, 1H NMR, 13C NMR and Mass spectral analysis. The newly synthesized derivatives (8a-j) were evaluated for their anti-proliferative activity against four human cell lines and the novel derivatives showed moderate to excellent activity. The obtained results suggest that these compounds can be considered as new hits for anti-proliferative drug development programme and further SAR studies can help obtain better anticancer agents.  相似文献

A cotarnine alkaloid-based synthesis was developed for new heptacyclic condensed diisoquinolines via the double intramolecular pseudosalt bis[1,3]dioxolo[4,5-g;4′,5′-g′][1,3,4]oxadiazolo[2,3-a;5,4-a′]diisoquinoline 6. Substitution of the central O atom in 6 by C, S, or N nucleophiles afforded the first representatives of the new ring systems bis[1,3]dioxolo[4,5-g:4,5-g′]pyrazolo[3,2-a:5,1-a′]diisoquinoline (7a-d), bis[1,3]dioxolo[4,5-g:4,5-g′][1,3,4]thiadiazolo[2,3-a:5,4-a′]diisoquinoline (8), and bis[1,3]dioxolo[4,5-g:4,5-g′][1,2,4]triazolo[3,2-a:5,1-a′]diisoquinoline (9a-d) under simple reaction conditions.  相似文献

Hydroxylamine-O-sulfonic acid (HOSA) was used as an efficient nucleophilic amination reagent for 2-chloropyrimidines, 2-chloroquinolines, and 1-chloroisoquinoline. The newly obtained heteroaromatic hydroxylamine-O-sulfonates subjected to the reaction with acyl isothiocyanates underwent tandem nucleophilic addition-electrophilic 5-endo-trig cyclization. The mechanism of the cyclization was investigated with use of the long-range corrected hybrid density functional ωB97X-D/6-31+G and SM8 (DMF) solvation model. The structures of the heteroaromatic hydroxylamine-O-sulfonates and N-(5-methoxy-2H-[1,2,4]thiadiazolo[2,3-a]pyrimidin-2-ylidene)benzamide were confirmed by single crystal X-ray analysis. N-(2H-[1,2,4]thiadiazolo[3,2-a]isoquinolin-2-ylidene)benzamide exhibited a pronounced in vitro cytostatic activity against human tumor cell lines SISO, LCLC, A-427, DAN-G, and RT-4 (IC50 in the range 1.47-2.97 μM).  相似文献

A series of tetrachloride platinum(IV) compounds of the general formulae PtCl4L2, where L = 1,2,4-triazolo[1,5-a]pyrimidine (tp) (1), 5,7-dimethyl-1,2,4-triazolo[1,5-a]pyrimidine (dmtp) (2), 5,7-ditertbutyl-1,2,4-triazolo[1,5-a]pyrimidine (dbtp) (3) and 5-methyl-1,2,4-triazolo[1,5-a]pyrimidin-7(4H)-one (HmtpO) (4) have been prepared and characterized by thermal analysis, 1H, 13C, 15N, 195Pt NMR and IR spectroscopy. Spectral data suggest that the triazolopyrimidines act as a monodentate ligand via the nitrogen atom N(3). The preliminary assessments of antitumor properties of the four complexes were evaluated as in vitro antiproliferative activity against three cell lines: HL-60 human acute promyelocytic leukemia, SW707 rectal adenocarcinoma and HCV29T bladder cancer. PtCl4(dbtp)2 exhibits high cytotoxic activity against all human cell lines, whereas the other complexes are only moderately active.  相似文献

An efficient one-pot synthetic method for the highly substituted 5H-[1,3,4]thiadiazolo[3,2-a]pyrimidine-6-carboxylate derivatives has been accomplished via a microwave irradiation of MCRs of various aldehydes, 2-aminothiadiazole, and acetoacetate without any catalyst in acetic acid. This approach provides a convenient one pot method for the synthesis of 5H-[1,3,4]thiadiazolo[3,2-a]pyrimidine-6-carboxylate derivatives.  相似文献

2-Substituted-4a-hydroxy-9H-cycloalkyl[1,2-e]oxazolo[3,2-a]pyrimidin-9-ones 2a-c were synthesized by an one-step cyclocondensation from the 5-substituted-2-amino-2-oxazolines 1a-c with ethyl 2-oxocyclohexanecarboxylate in ethanol at room temperature, and easily dehydrated to provide 2-substituted-9H-cycloalkyl[1,2-e]oxazolo[3,2-a]pyrimidin-9-ones 3. In refluxing xylene, the reaction conducted with various ethyl 2-oxocycloalkanecarboxylates led to the two isomeric 2-substituted-8/9H-cycloalkyl[1,2-e]oxazolo[3,2-a]pyrimidin-8/9-ones 3 and 2-substituted-5H-cycloalkyl[1,2-d]oxazolo[3,2-a]pyrimidin-5-ones 4. The structure of some compounds was unambiguously established using X-ray crystallography. According to results from the DSC analysis of compound 2a, formation of the thermodynamically stable pyrimidinones 4 could be related to an intramolecular rearrangement of kinetically controlled pyrimidinones 3.  相似文献

A series of pyrrolo[3′,2′:4,5]thiopyrano[3,2-b]pyridin-2-ones 4 was prepared in good yields by reacting enaminoketones with cyanomethylene active compounds such as phenylsulfonylacetonitrile, benzoylacetonitrile, and malononitrile. Derivatives of the title ring system were tested by the National Cancer Institute of Bethesda against a panel of about 60 human tumor cell lines, and one of them showed inhibitory activity against all cancer cell lines reaching on 48% of them GI50 values at submicromolar level and on the majority of the remaining ones in the low micromolar concentration.  相似文献

The compounds (3R,5S)-(+)-5-methyl-3-phenyl-2,3,5,6,7,8-hexahydro-oxazolo[3,2-a]pyridin-4-ylium iodide 4 and (3R,5S)-(+)-5-n-propyl-3-phenyl-2,3,5,6,7,8-hexahydro-oxazolo[3,2-a]pyridin-4-ylium iodide 5 were synthesized in two steps starting from the bicyclic thiolactam trans (3R,2aS)-(−)-5-thio-3-phenyl-2,3,6,7,8,2a-hexahydro-oxazolo[3,2-a]pyridine 1. A novel series of 6-alkyl-4-bromopyrano[3,2-c]quinoline-2,5-diones ( 2a–c ), 6-alkyl-3,4-dibromopyrano[3,2-c]quinoline-2,5-diones ( 4a–c ), and 6-alkyl-3-amino-bromopyrano[3,2-c]quinoline-2,5-diones ( 6a–c ) were synthesized via appropriate conventional methods and in good yields. The structures of target compounds were approved by elemental analysis, IR, 1H NMR, 13C NMR, and mass spectrometry. The antitumor inhibitory activities of the new compounds were evaluated on several cancer cell lines, A-549 (human lung cancer), HCT-116 (human colon cancer), MCF-7 (breast cancer), and HePG-2 (human liver cancer). Moreover, 50% inhibitory concentrations, IC50, were established. 5-Fluorouracil was used as a positive control in the viability assay. The screening results showed that most of the examined compounds exposed powerful inhibition activity toward various cell lines. Particularly, Compound 4c exhibited higher cytotoxic activity against four tumor cell lines than the reference drug, 5-fluorouracil, with significantly lower IC50 values. Accordingly, most of the synthesized compounds would be a better prospective growth in the anticancer drug discovery.  相似文献

Novel spiro(3-hydroxy-5,5-dimethyl)-1,5’-(2’-substituated[1,3,4]-oxadiazole/thiadiazolo[3,2-c] thiazolines and 3-hydroxy-5,5-dimethyl-spiro[cyclohex-2-ene-thiazolo/thiazino[3,2-b]-s-tetrazine were prepared from previously reported hydrazone, semicarbazone, carbazone, and spiro-s-tetrazine intermediates derived from dimedone precursor. All the newly synthesized heterocycles have been characterized by analytical and spectral (1H NMR and hC NMR) data and screened their in vitro antibacterial activity against gram negative bacteria Escherichia coli and gram positive bacteria Staphylococcus aureus. Some of the compounds showed moderate to good activities towards the bacteria chosen.  相似文献

A convenient method for the preparation of benzofuro[3,2-c]isoquinoline derivatives is described. The condensation reaction of methyl 2-(chloromethyl)-benzoate with substituted salicylonitriles 7a-c and intramolecular cyclization of the resulting substituted methyl 2-[(2-cyanobenzyl)oxy]benzoates 10a-c using potassium tert-butoxide results in the substituted benzofuro[3,2-c]isoquinolin-5(6H)-ones 1a-c. The same sequence of reactions starting from 2-(chloromethyl)benzonitrile and compounds 7a-c gave substituted 5-aminobenzofuro[3,2-c]isoquinolines 13a-c. In addition, this method is useful for the synthesis of other heterocycles. For example, using 1-cyano-2-naphthol 16, instead of the salicylonitriles 7a-c, gives naphtho[1′,2′:4,5]furo[3,2-c]isoquinolines.  相似文献

A new series of pyridothienopyrimidine derivatives was designed and evaluated as antimicrobial and anticancer agents. The target compounds were synthesized starting with 3-aminothieno[2,3-b]pyridine-2-carboxamide derivative 1 which underwent cyclocondensation reaction with aromatic aldehydes to give the key intermediates 2a,b. By further treatment of 2a,b with various reagents, the target 2,4-disubstituted-pyrido[3′,2′:4,5]thieno[2,3-d]pyrimidines 3a,b11a,b were obtained. To evaluate the antimicrobial activity of the new compounds, they were tested against five bacterial and five fungal strains. Compounds 6c, 8b, 9a and 9b revealed the most significant antimicrobial activity against the tested microorganisms with MIC values range (4–16 μg/mL). Also, compounds 2a,b11a,b were screened for their in vitro cytotoxic activity against HepG-2 and MCF-7 cancer cell lines compared with doxorubicin and cisplatin as references drugs. Moreover, compounds (2b, 4a, 6a, 7b, 7c and 9a) which exhibited the most potent anticancer activity, were further subjected to EGFRWT enzyme inhibition assay utilizing erlotinib as a standard drug. The compounds 6a, 7b, 7c and 9a which showed the most promising suppression effects were also evaluated as inhibitors against the mutant forms EGFRL858R and EGFRT790M. The 4-aminopyrazolone analogue 9a showed superior anticancer activity against both HepG-2 and MCF-7 cell lines (IC50 = 1.27, 10.80 μM, respectively) and more potent enzymatic inhibition activity against EGFRWT and its mutant forms EGFRL858R and EGFRT790M than that obtained by erlotinib (IC50 = 0.021, 0.053, 0.081 µM, respectively, IC50erlotinib; 0.027, 0.069, 0.550 µM, respectively). Finally, the molecular docking study showed good binding patterns of the most active compounds with the prospective target EGFRWT.  相似文献

3-Amino-4-hydroxy-6-phenylpyrano[3,2-c]quinoline-2,5(6H)-dione was produced by a smooth reduction of its nitro precursor. Reaction of this 3-amino-4-hydroxypyranoquinoline derivative with different electrophiles, leading to a variety of oxazolo and oxazinopyrano[3,2-c]quinoline derivatives, was described. The structure of the new products was elucidated via elemental analysis, IR, 1H NMR, 13C NMR, and mass spectra. Also, screening the antitumor activity of new compounds against two human cell lines (HepG-2 and HCT-116) was carried out. Some new products showed significant antitumor activities.  相似文献

A simple and convenient route was performed for the synthesis of some new heterocyclic compounds based on thieno[3,2-c]pyrazole derivative for antimicrobial evaluation. The key intermediate, 5-amino-3-methyl-1-phenyl-1H-thieno[3,2-c]pyrazole-6-carbohydrazide 3, was prepared by Gewald’s synthesis of Ethyl 5-amino-3-methyl-1-phenyl-1H-thieno[3,2-c]pyrazole-6-carboxylate 2. This intermediate reacted with various reagents to afford different fused and polyfunctional substituted. The structures of these compounds were confirmed by elemental analysis, IR, 1H NMR, 13C NMR and mass spectral data. All the new synthesized compounds were screened for various microorganisms such as Aspergillus fumigatus; Geotrichum candidum; Syncephalastrum racemosum (Fungus); Candida albicans (Yeast fungus); Staphylococcus aureus; Bacillus subtilis (as Gram-positive bacteria); Pseudomonas aeruginosa and Escherichia coli (as Gram-negative bacteria) by the disc diffusion method. In general, the novel synthesized compounds possessed moderate to high antimicrobial activity against the previously mentioned microorganisms.  相似文献

A series of 3-diphenylmethyl-6-substituted-1,2,4-triazolo[3,4-b]-1,3,4-thiadiazole derivatives (4aj and 5ad) were synthesized by condensation of 4-amino-5-diphenylmethyl-4H-1,2,4-triazole-3-thiol with various substituted aromatic acids and aryl/alkyl-isothiocyanates. The structures of synthesized compounds were characterized by elemental analysis, IR, 1H NMR, 13C NMR and mass spectroscopic studies. These compounds were tested in vivo for their anti-inflammatory activity. The compounds which showed activity comparable to the standard drug ibuprofen were screened for their analgesic, ulcerogenic, lipid peroxidation and hepatotoxic effects. Compounds 6-(4-chlorophenyl)-3-diphenylmethyl-1,2,4-triazolo[3,4-b]-1,3,4-thiadiazole (4a) and 6-(2,4-dichlorophenyl)-3-diphenylmethyl-1,2,4-triazolo[3,4-b]-1,3,4-thiadiazole(4c) emerged as the most active compounds of the series and were moderately more potent than the standard drug ibuprofen.  相似文献

Reaction of 11-aminoacronycine, 10-aminobenzo[a]acronycine, and 10-aminobenzo[b]acronycine with paraformaldehyde gave the corresponding Tröger’s bases 11, 14, and 16, respectively. The cytotoxic activity of those three new compounds was determined against L-1210 leukemia and KB-3-1 solid tumor cell lines, in comparison with their parent compounds, acronycine, benzo[a]acronycine, and benzo[b]acronycine.  相似文献

Several derivatives of two novel mesoionic ring systems, i.e., isoxazolo[2,3-a]pyrimidinedione and 1,3,4-oxadiazolo[3,2-a]pyrimidinedione, were prepared for evaluation as adenosine antagonists. Whereas both ring systems are relatively stable when the 6-position (i.e., that position corresponding to the purine 1-position) is substituted by an alkyl group, neither ring system is stable when this position is unsubstituted. An example of a 6-unsubstituted non-mesoionic isoxazolopyrimidinedione exhibited similar behavior. As adenosine antagonists, the mesoionic compounds were found to be less potent than their previously evaluated mesoionic thiadiazolo[3,2-a]pyrimidinedione counterparts.  相似文献

According to the biological screening reports, a thiadiazoloquinazoline scaffold constitutes a promising source of inspiration for the synthesis of novel bioactive molecules. The current review article summarizes the most recent synthetic routes toward thiadiazoloquinazoline derivatives. The present paper is devoted to the search for drug-like molecules with anticancer properties using the thiazolo[3,2-b][1,2,4]triazole-6-one scaffold. A series of 24 novel thiazolo-[3,2-b][1,2,4]triazole-6-ones with 5-aryl(heteryl)idene- and 5-aminomethylidene-moieties has been synthesized employing three-component and three-stage synthetic protocols. A mixture of Z/E-isomers was obtained in solution for the synthesized 5-aminomethylidene-thiazolo[3,2-b]-[1,2,4]triazole-6-ones. The compounds have been studied for their antitumor activity in the NCI 60 lines screen. Some compounds present excellent anticancer properties at 10 μM. Derivatives 2h and 2i were the most active against cancer cell lines without causing toxicity to normal somatic (HEK293) cells. A preliminary SAR study had been performed for the synthesized compounds.  相似文献
