In a quantitative single particle analysis, named the low-Z particle EPMA, number concentration data for chemical species encountered in aerosol sample are provided. However, it will be more useful if mass concentration data can be obtained from single particle analysis; i.e., the single particle analysis data for weight fractions of chemical species can be complementarily used in combination with the bulk analysis data, for more clearly understanding the behavior of airborne aerosols. In order to investigate how reliably mass concentration data can be obtained from the low-Z particle EPMA technique, a potassium feldspar powdered standard reference material (SRM), of which elemental weight fractions are well defined by various bulk analytical techniques, was analyzed using the low-Z particle EPMA technique. In this work, it is demonstrated that weight fractions of major elements in the powdered SRM sample obtained by the low-Z particle EPMA are within 8% to the certified values obtained by bulk analytical techniques, although the single particle and bulk analyses employ different approaches. Further, it is shown that the quantitative single particle analysis, i.e., low-Z particle EPMA, can provide molecular mass concentration data for chemical species, which is not easy to obtain using bulk analysis.  相似文献

Using a chiral base from a norephedrine-derived diamine, the enantioselective rearrangement of a meso-cyclohexene oxide can be performed in 94% yield and with 94% enantioselectivity. The enantioselectivity is lower (86% ee) with the diastereoisomeric chiral base. Reaction of phenol with sulfuryl chloride in the presence of a catalytic amount of the heterogeneous amine catalyst, t-butylaminomethyl polystyrene, in a nonpolar solvent, proceeds with high conversion (∼98%) and with high selectivity (∼89%) to ortho-chlorophenol. The catalyst is stable under the chlorination conditions, and can easily be regenerated by filtration and reused. This method could be applicable to other phenols.  相似文献

The effects of two types of biochar on corn production in the Mediterranean climate during the growing season were analyzed. The two types of biochar were obtained from pyrolysis of Pinus pinaster. B1 was fully pyrolyzed with 55.90% organic carbon, and B2 was medium pyrolyzed with 23.50% organic carbon. B1 and B2 were supplemented in the soil of 20 plots (1 m2) at a dose of 4 kg/m2. C1 and C2 (10 plots each) served as control plots. The plots were automatically irrigated and fertilizer was not applied. The B1-supplemented plots exhibited a significant 84.58% increase in dry corn production per square meter and a 93.16% increase in corn wet weight (p << 0.001). Corn production was no different between B2-supplemented, C1, and C2 plots (p > 0.01). The weight of cobs from B1-supplemented plots was 62.3%, which was significantly higher than that of cobs from C1 and C2 plots (p < 0.01). The grain weight increased significantly by 23% in B1-supplemented plots (p < 0.01) and there were no differences between B2-supplemented, C1, and C2 plots. At the end of the treatment, the soil of the B1-supplemented plots exhibited increased levels of sulfate, nitrate, magnesium, conductivity, and saturation percentage. Based on these results, the economic sustainability of this application in agriculture was studied at a standard price of €190 per ton of biochar. Amortization of this investment can be achieved in 5.52 years according to this cost. Considering the fertilizer cost savings of 50% and the water cost savings of 25%, the amortization can be achieved in 4.15 years. If the price of biochar could be reduced through the CO2 emission market at €30 per ton of non-emitted CO2, the amortization can be achieved in 2.80 years. Biochar markedly improves corn production in the Mediterranean climate. However, the amortization time must be further reduced, and enhanced production must be guaranteed over the years with long term field trials so that the product is marketable or other high value-added crops must be identified.  相似文献

The substrate range of a novel recombinant (S)-alcohol dehydrogenase from Rhodococcus erythropolis is described. In addition, an enzyme-compatible biphasic reaction medium for the asymmetric biocatalytic reduction of ketones with in situ-cofactor regeneration has been developed. Thus, reductions of poorly water soluble ketones in the presence of the alcohol dehydrogenase from R. erythropolis and a formate dehydrogenase from Candida boidinii can be carried out at higher substrate concentrations of 10-200 mM. The resulting (S)-alcohols were formed with moderate to good conversion rates, and with up to >99% ee.  相似文献

A broad range of arylaliphatic ketones is efficiently reduced to the corresponding optically active (R)-alcohols by a recombinant alcohol dehydrogenase from Pseudomonas fluorescens (PFADH) produced by overexpression in Escherichia coli. PFADH shows high activity and stereoselectivity in the reduction of acetophenone and various derivatives (45–99% e.e.), as well as in the reduction of 3-oxobutyric acid methyl ester (>99% e.e.). The highest activity was observed between 10 and 20°C. The cofactor NADH can be efficiently recycled by the addition of 10–20% (v/v) of iso-propanol.  相似文献

Lipophilic trifluoroacetophenone derivatives incorporated in plasticized PVC membranes are able to selectively extract water and alcohols from the sample solution into the organic membrane phase, reversibly forming hydrates and hemi-acetals, respectively. Since this is accompanied by a change in the absorption spectrum of the acetophenone isologue, the chemical recognition process can directly be translated into an optical signal. With N-acetyl-N-dodecyl-4-trifluoroacetylaniline (ETH 6022) as the electrically neutral, lipophilized carrier ethanol can be determined from 0.5 to 35% (v/v) in aqueous solutions. The calibration curve for different ethanol-water mixtures shows a good correlation with the mathematically derived formalism and thus confirms the theoretically expected behavior. Besides high reproducibility of the optical signals, very short response times of less than 30 s were realized. The optode membrane presented exhibits a preference for ethanol compared to water by a factor greater than 11. The selectivities for several primary alcohols, such as methanol, ethanol, 1-propanol and 1-n-butanol, are comparable, but isopropanol and tert.-butanol are rejected by a factor of about 10. The alcohol concentration in different beverages was determined to evaluate the reliability of the system. The values obtained for wine, beer and different spirits show an excellent correlation with those obtained by a conventional approach involving distillation and density measurements. A residual standard deviation of ± 0.27% (v/v) over the 0.7–40% (v/v) range was found.  相似文献

Chicken stomachs can be processed into collagen hydrolysate usable in cosmetic products. The aim of the study was to verify the effects of a carbopol gel formulation enriched with 1.0% (w/w) chicken hydrolysate on the properties of the skin in the periorbital area after regular application twice a day for eight weeks in volunteers ageed 50 ± 9 years. Skin hydration, transepidermal water loss (TEWL), skin elasticity and skin relief were evaluated. Overall, skin hydration increased by 11.82% and 9.45%, TEWL decreased by 25.70% and 17.80% (always reported for the right and left area). Generally, there was an increase in skin elasticity, a decrease in skin roughness, as the resonance times decreased by 85%. The average reduction of wrinkles was 35.40% on the right and 41.20% on the left. For all results, it can be seen that the longer the cosmetic gel formulation is applied, the better the results. Due to the positive effect on the quality and functionality of the skin, it is possible to apply the cosmetic gel formulation in the periorbital area. The advantage of the product with chicken collagen hydrolysate is also the biocompatibility with the skin and the biodegradability of the formulation.  相似文献

A tandem reaction for the synthesis of chroman framework via a one-pot reaction of phenol, paraformaldehyde, and 5-benzylimidazolidin-4-one mediated by p-nitrobenzoic acid is described. Current techniques being used for sunscreen analysis are incapable of direct determination of the active ingredients in sunscreen formulations. Therefore, the development of methodologies for rapid in situ analysis of sunscreens is desirable. This paper describes the application of ultraviolet resonance Raman spectroscopy (UVRRS) to the direct in situ analysis of sunscreen formulations. High-quality UV resonance Raman spectra were obtained for five sunscreen active ingredients (AIs), mixtures of the AIs and real sunscreen formulation samples. The spectra from the sunscreen formulations gave distinct spectral signatures indicative of the sunscreen AIs in each sample, with essentially no interference from the complex sunscreen matrix. Also, despite the fact that many of the AIs are fluorescent, no fluorescence interferences in the resonance Raman spectra were observed. Excitation wavelength-dependent studies throughout the 244-275 nm region demonstrate that the best discrimination of the AIs was achieved at an excitation wavelength of 244 nm. Thus, by tuning the excitation wavelength within the absorption bands of the AIs, complete identification of these analytes can be achieved in situ without any sample pretreatment or separation. The limit of detection found for a common AI in situ with this technique is 0.23% (w/w), the limit of quantitation is 0.78% (w/w), while the dynamic range is between 0.8% and 50% (w/w). The technique is fast, robust, lacks any major interference, and can be adapted for routine online quality control.  相似文献

Toward the standardization of common evening primrose (Oenothera biennis) sprout extract (OBS-E), we aimed to obtain indicator compounds and use a validated method. HPLC-UVD allowed simultaneous quantification of the indicator compounds quercetin-3-O-glucuronide and ellagic acid. The method was validated in terms of specificity, linearity, precision, accuracy, and limit of detection/limit of quantification (LOD/LOQ). High specificity and linearity was demonstrated, with correlation coefficients of 1.0000 for quercetin-3-O-glucuronide and 0.9998 for ellagic acid. The LOD/LOQ values were 0.486/1.472 μg/mL for quercetin-3-O-glucuronide and 1.003/3.039 μg/mL for ellagic acid. Intra-day and inter-day variability tests produced relative standard deviation for each compound of <2%, a generally accepted precision criterion. High recovery rate were also obtained, indicating accuracy validation. The OBS-E prepared using various concentrations of ethanol were then analyzed. The 50% ethanol extract had highest content of quercetin-3-O-glucuronide, whereas the 70% ethanol extract possessed the lowest. However, the ellagic acid content was highest in the 70% ethanol extract and lowest in the 90% ethanol extract. Thus, quercetin-3-O-glucuronide and ellagic acid can be used industrially as indicator compounds for O. biennis sprout products, and our validated method can be used to establish indicator compounds for other natural products.  相似文献

Three-phase partitioning (TPP) was used to purify and recover catalase from potato crude extract. The method consists of ammonium sulfate saturation, t-butanol addition, and adjustment of pH, respectively. The best catalase recovery (262 %) and 14.1-fold purification were seen in the interfacial phase in the presence of 40 % (w/v) ammonium sulfate saturation with 1.0:1.0 crude extract/t-butanol ratio (v/v) at pH 7 in a single step. The sodium dodecyl sulfate–polyacrylamide gel electrophoresis (SDS-PAGE) analysis of the enzyme showed comparatively purification and protein molecular weight was nearly found to be 56 kDa. This study shows that TPP is a simple, economical, and quick method for the recovering of catalase and can be used for the purification process.  相似文献

The prevalence of cephalosporine-resistant (3GC-R) strains among United States community-related research samples ranged from 5.6 to 10.8%, while, in the European countries, it was 1.2% to 10.1%. Several studies suggest that meat of animal origin could be one of the reservoirs of 3GC-R bacteria. Here, 86 raw meat samples (turkey, pork, chicken and beef) were collected randomly and verified for the presence of 3GC-R bacteria. The 3GC-R bacteria were isolated, identified and characterized phenotypically (antibiotic resistance, motility and biofilm) and genotypically (repetitive-sequence-based rep-PCR) to elucidate any correlations with principal component analysis (PCA). From 28 3GC-R positive samples, 41 strains were isolated, from which the majority belonged to Serratia fonticola (39%), followed by Escherichia coli (19.5%), Enterobacter cloacae (17.1%) and Klebsiella pneumoniae (14.6%). The isolates of E. coli and S. fonticola presented diverse profiles in rep-PCR. Generally, 3GC-R strains were more resistant to antibiotics used in veterinary medicine than in human medicine. PCA derived from antibiotic resistance, motility and biofilm formation of S. fonticola and E. coli strains showed that resistance to beta-lactams was separated from the resistance to other antibiotic classes. Moreover, for the S. fonticola, E. coli and En. cloacae, the type of meat can create a specific tendency towards antibiotic resistance and phenotypic characteristics for S. fonticola, while these relationships were not found for other tested species.  相似文献

Amplification of enantiomeric enrichment is a key feature for the chemical evolution of biological homochirality from the origin of chirality. The aggregations of the enantiomers by diastereomeric interactions enable the modification of their enantiomeric excess during some chemical processes. Fluorine-containing chiral compounds possess large amplification effect via distillation, sublimation and achiral chromatography by self-disproportionation. Asymmetric amplifications in enantioselective catalysis occur by the differential formation and reactivity between homochiral and heterochiral aggregate in solution.We described the amplification of ee in asymmetric autocatalysis of 5-pyrimidyl alkanol in the reaction between diisopropylzinc and pyrimidine-5-carbaldehdye. During the reactions extremely low ee (ca. 0.00005% ee) can be amplified to achieve more than 99.5% ee. Since the proposed origins of chirality such as CPL, quartz, chiral organic crystals of achiral compounds and statistical fluctuation of ee can initiate the asymmetric autocatalysis with amplification of ee, the proposed origin of chirality can be linked with enantiopure organic compound in conjunction with amplification of ee by asymmetric autocatalysis. In addition, we described that the carbon isotopically chiral compound triggers the asymmetric autocatalysis of 5-pyrimiodyl alkanol to afford the enantioenriched product with the absolute configuration correlated with that of carbon isotope chirality, that is, isotope chirality including hydrogen isotopes can control the enantioselectivity of asymmetric addition of alkyl metal reagent to aldehyde.  相似文献

Butanol tolerance is a critical factor affecting the ability of microorganisms to generate economically viable quantities of butanol. Current Clostridium strains are unable to tolerate greater than 2% 1-butanol thus membrane or gas stripping technologies to actively remove butanol during fermentation are advantageous. To evaluate the potential of alternative hosts for butanol production, we screened 24 different microorganisms for their tolerance to butanol. We found that in general, a barrier to growth exists between 1% and 2% butanol and few microorganisms can tolerate 2% butanol. Strains of Escherichia coli, Zymomonas mobilis, and non-Saccharomyces yeasts were unable to surmount the 2% butanol growth barrier. Several strains of Saccharomyces cerevisiae exhibit limited growth in 2% butanol, while two strains of Lactobacillus were able to tolerate and grow in up to 3% butanol.  相似文献

Biofouling presents a significant obstacle to the long-term use of electrochemical sensors in complex media. Drinking water biofilms reduce performance of sensors by insulating electrode surfaces by inter alia inhibiting mass transport. Boron-doped diamond (BDD) electrodes are relatively resistant to biofouling and inert at high potentials. These qualities can be exploited to create a drinking water quality sensor that resists biofouling to meet performance criteria for longer, and to enable electrochemical cleaning of the sensor surface in situ using high potentials without disconnecting or disassembling the sensor.A purpose-built BDD wall-jet sensor was compared with a glassy carbon (GC) sensor in ability to determine free chlorine, detect biofilm and remove biofilm in situ. It was found that the BDD produced accurate and reliable readings with a 4.86% standard error and a LOD of 0.18 ppm. The BDD could be electrochemically cleaned in situ whereas this was less successful with the GC electrode. The BDD electrode could also detect electroactive pyocyanin, secreted in the biofilm of the drinking water biofilm indicator organism Pseudomonas aeruginosa, potentially enabling biofouling and non-biological fouling such as scaling to be distinguished. Observed changes in flow sensitivity and current-voltage curves that correspond to fouling provide multiple fouling detection methods, resulting in an accurate, sensitive, water quality sensor that can be cleaned without disassembly or replacement of parts and can identify when cleaning is required.  相似文献

Microscale regenerable biosensors are described and utilized to measure the natural fluorophor benzo[a]pyrene tetraol (BPT). The sensors combine laser-excited/fiber-optic remote sensing principles with a unique capillary tube delivery system to make repetitive, heterogeneous fluoroimmunoassay measurements. Two sensor configurations and modes of operation are described. Concentrations of BPT in the nanomolar range are easily measured with a reproducibility of 10% or better, depending on the sensor design, selective measurements can be made in ca. 20 min, then the sensor can be regenerated by delivering new reagents to the sensing chamber, without removing the sensor from the sample.  相似文献

Rice weevil, Sitophilus oryzae L. (Coleoptera: Curculionidae), is one of the most destructive stored-product pests that is resistant to a wide range of chemical insecticides. In the present study, we investigated whether a lectin extracted from Polygonum persicaria L. (PPA) can be used as a biorational agent to control such insect pests. Along with the lethal digestive assay, the sub-lethal insecticidal activities of PPA, including the effects on digestive, detoxifying, and antioxidant enzyme activities, were evaluated against S. oryzae adults. The effect of feeding a diet containing PPA and carob extract as a food attractant on the mortality of S. oryzae adults was also investigated. Feeding on the diet containing PPA resulted in a significant mortality of S. oryzae adults with a LC50 (Lethal Concentration to kill 50% of insects) of 3.68% (w/w). The activity of digestive enzymes, including α-amylase, α-glucosidase, TAG-lipase, trypsin, chymotrypsin, elastase, and carboxy- and aminopeptidase, were decreased by the sub-lethal concentration of PPA. Detoxifying and antioxidant enzymes, including esterase, superoxide dismutase, catalase, glutathione-S-transferase, ascorbate peroxidase, glucose 6-phosphate dehydrogenase, and malondialdehyde, were activated in adults affected by PPA. These findings indicated that PPA, in addition to causing digestive disorders, leads to oxidative stress in S. oryzae. The presence of carob extract had no effect on the PPA-induced mortality of the insect. According to the results of the present study, PPA has promising insecticidal efficiency against S. oryzae. In addition, the usage of PPA with a food attractant carob extract in bait traps can be recommended as a new biorational formulation in S. oryzae management.  相似文献

An assay based on optical biosensor technology has been developed to detect a broad range of nitroimidazole drug residues and their metabolites (dimetridazole (DMZ), metronidazole (MNZ), ronidazole (RNZ), hydroxymetronidazole (HO-MNZ) and hydroxydimetridazole (HO-DMZ)) in chicken muscle. The detection limit for the procedure was determined as 0.5 ppb for DMZ and detection capabilities (CCβs) ranged from <1 ppb for DMZ, MNZ and RNZ to <2 ppb for HO-MNZ and HO-DMZ. Intra-assay variation (n = 6) was calculated as 11.6% at a concentration of 1 ppb DMZ and 4.7% at a concentration of 2 ppb DMZ. Inter-assay variation (n = 3) was determined to be 14.2% at a concentration of 1 ppb DMZ and 3.5% at a concentration of 2 ppb DMZ.A prototype kit based on this assay was produced and a multinational study was undertaken to independently evaluate its performance. The resulting data showed that the kit can be implemented with little difficulty in laboratories of varying expertise and is sensitive enough to meet the standards required by international law. Feedback from this study led to the incorporation of some minor improvements to the kit. The commercial partner in the project, XenoSense Ltd., was consulted with regards to producing a commercial test kit based on the prototype assay. As feedback from the collaborative study had been positive with respect to speed, ease of use and performance of the kit, the decision to commercialise the kit was taken. In conclusion, the prototype nitroimidazole kit was shown to offer numerous advantages over existing analytical techniques.  相似文献

In this study, we present a powerful stirred tank reactor system that can efficiently hydrolyse lignocellulosic material at high solid content to produce hydrolysates with glucose concentration > 100 g/kg. As lignocellulosic substrates alkaline-pretreated wheat straw and organosolv-pretreated beech wood were used. The developed vertical reactor was equipped with a segmented helical stirrer, which was specially designed for high biomass hydrolysis. The stirrer was characterised according to mixing behaviour and power input. To minimise the cellulase dosage, a response surface plan was used. With the empirical relationship between glucose yield, cellulase loading and solid content, the minimal cellulase dosage was calculated to reach at least 70 % yield at high glucose and high substrate concentrations within 48 h. The optimisation resulted in a minimal enzyme dosage of 30 FPU/g dry matter (DM) for the hydrolysis of wheat straw and 20 FPU/g DM for the hydrolysis of beech wood. By transferring the hydrolysis reaction from shaking flasks to the stirred tank reactor, the glucose yields could be increased. Using the developed stirred tank reactor system, alkaline-pretreated wheat straw could be converted to 110 g/kg glucose (76 %) at a solid content of 20 % (w/w) after 48 h. Organosolv-pretreated beech wood could be efficiently hydrolysed even at 30 % (w/w) DM, giving 150 g/kg glucose (72 %).  相似文献
