Eleven novel 5‐methyl‐2‐[(un)substituted phenyl]‐4‐{4,5‐dihydro‐3‐[(un)substituted phenyl]‐5‐(1,2,3,4‐tetrahydroisoquinoline‐2‐yl)pyrazol‐1‐yl}‐oxazole derivatives were synthesized and characterized by elemental analysis, ESI‐MS, 1H NMR and 13C NMR. All of the compounds have been screened for their antiproliferative activities against PC‐3 cell (human prostate cancer) and A431 cell (human epidermoid carcinoma cancer) lines in vitro. The results revealed that compounds 4g , 4j and 4k exhibited the strong inhibitory activities against the PC‐3 cell lines (with IC50 values of 2.8±0.11, 3.1±0.10 and 3.0±0.06 μg/mL, respectively).  相似文献

N‐(trialkoxysilylalkyl) derivatives of 1,2,3,4‐tetrahydroquinoline, 1,2,3,4‐tetrahydroisoquinoline and 4,4‐dimethyl‐4‐sila‐1,2,3,4‐tetrahydroisoquinoline were prepared and characterized by elemental analysis, 1H, 13C and 29Si NMR spectroscopy. In vivo psychotropic properties and in vitro cytotoxic effects of 3‐[N‐(1,2,3,4‐tetrahydroisoquinolyl)]propyltriethoxysilane methiodide and 3‐[N‐(1,2,3,4‐tetrahydroisoquinolyl)]propylsilatrane are reported. Comparative study of 29Si shifts in newly synthesized compounds suggested donor–acceptor interaction between nitrogen and silicon atom, which increased electron density at Si nuclei, revealing a stronger increment of N → Si transannular bond in comparison with N → Si α‐effect. The molecular structure of 3‐[N‐(1,2,3,4‐tetrahydroisoquinolyl)]propylsilatrane features a penta‐coordinate silicon atom having CSiO3 pattern and Si…N intramolecular interaction. Superparamagnetic iron oxide/oleic acid nanoparticles bearing lipid‐like organosilicon N‐(2‐hydroxyethyl)‐1,2,3,4‐tetrahydroisoquinoline derivatives have been synthesized with the aim of their potential biomedical application. X‐ray diffraction analysis, Dynamic light‐scattering measurements, method of magnetogranulometry and some others have been employed to investigate the morphology and properties of the nanoparticles synthesized. The magnetic core diameter of mixed covered nanoparticles ranged between 4.8 and 9.6 nm. The magnetization analyses showed that the particles are superparamagnetic at room temperature. In vitro cell cytotoxicity and intracellular NO generation caused by the water magnetic solution of nanoparticles possessing cytotoxic organosilicon heterocyclic choline analogue, namely N‐(2‐dimethyl‐n‐hexadecylsiloxyethyl)‐N‐methyl‐1,2,3,4‐tetrahydroisoquinolinium iodide, was examined in relation to monolayer human fibrosarcoma (HT‐1080) and mouse hepatoma (MG‐22A) tumour cell lines and normal mouse fibroblasts (NIH 3T3). The biological studies have revealed its selective cytotoxicity in tumour cells and strong effect on MG‐22A cell morphology. Incorporation of the synthesized nanoparticles into cells was observed. Some new N-alkoxycarbonyl-N″-benzoyl-benzamidrazones (p-toluamidrazones) 3a-3d, and 1,3,5-trisubstituted 1,2,4-triazole 4a-4h derivatives by starting from N-benzoylbenzimidates or N-benzoyl-p-toluimidates. The structures of compounds 3 and 4 were established on the basis of elemental analyses, IR, ^1H NMR, ^13C NMR and UV data. Antimicrobial experiments of the compounds performed by using agar-well diffusion and broth microdilution methods revealed that only compounds 3a-3d, 4a and 4b showed inhibitory effect only on Candida albicans ATCC 60193. However, compound 4b had also specific antibacterial activity against Staphylococcus aureus ATCC 25923. The other compounds showed neither antifungal nor antibacterial activities. Compounds 3a, 4a and 4b have been screened on three human tumor cell lines, breast cancer (MCF7), non small cell lung cancer (NCI-H460), and CNS cancer (SF-268) at the National Cancer Institute (NCI), USA, which were found to exhibit low antiproliferative activity.  相似文献

The diorganotin(Ⅳ) complexes of N-(3,5-dibromosalicylidene)-α-amino acid, R2Sn(2-O-3,5-Br2C6H2CH= NCHRCOO)(where R=H, Me, i-Pr, Bz; R'=n-Bu, Cy), were synthesized by the reactions of diorganotin dichlorides with in situ formed potassium salt of N-(3,5-dibromosalicylidene)-α-amino acid and characterized by elemental analysis, IR and NMR (^1H, ^13C and ^119Sn) spectra. The crystal structures of n-Bu2Sn(2-O-3,5-Br2C6H2CH= NCHRCOO)(R=i-Pr, Bz) and Cy2Sn(2-O-3,5-Br2C6H2CH=NCHRCOO)(R=Me, Bz) were determined by X-ray single crystal diffraction and showed that the tin atoms are in a distorted trigonal bipyramidal geometry to form five- and six-membered chelate rings with the tridentate ligand. Bioassay results indicated that the compounds possess better in vitro antitumour activity against three human tumour cell lines, HeLa, CoLo205 and MCF-7, than cis-platin and moderate anti-bacterial activity against two bacteria, E. coli and S. aureus.  相似文献

New hydroselenites of the different silylalkylthio‐substituted N‐heterocycles have been prepared by the reaction of selenium dioxide with N‐heterocycles in an aqueous medium. Their structure was confirmed by 1H, 13C, and 77Se NMR data. Most of these silylalkylthio‐substituted N‐heterocycles and their hydroselenites have an expressed cytotoxic activity on the MG‐22A (mouse hepatoma), HT‐1080 (human fibrosarcoma), B16 (mouse melanoma), and Neuro 2A (mouse neuroblastoma) cell lines. Some of the hydroselenites exhibit free‐radical protection simultaneously with a high cytotoxic effect. The substances studied were also active in vivoagainst sarcoma S‐180. The reaction of homophthalic anhydride and N‐(1‐methyl‐1H‐pyrrol‐2‐yl‐methylidene)‐benzylamine in boiling benzene afforded as a main product the expected substituted trans‐1,2,3,4‐tetrahydroisoquinoline‐4‐carboxylic acid 5 . The carboxylic group of 5 was transformed in four steps into cyclic amino‐methyl groups yielding numerous new tetrahydroisoquinolinones 11a‐j incorporating a given fragment of pharmacological interest. Natural and synthetic isoquinoline alkaloids display a wide variety of potent biological activities. The title 1‐aryl‐2‐hydroxyethyl‐1,2,3,4‐tetrahydroisoquinoline, C19H23NO4, crystallizes with two molecules in the asymmetric unit related by pseudo‐translation but differing only slightly in conformation. The pseudosymmetry is also reflected in the diffraction pattern. The subset of reflections corresponding to the smaller cell and average structure are on average twice as intense as those subtending the larger cell. Tentative refinement in the subcell leads to a disordered structural model with satisfactory agreement factors and, after appropriate use of restraints, acceptable molecular geometry but significantly larger and more anisotropic displacement parameters. In the correct unit cell, the independent molecules differ with respect to the orientation of the hydroxyethyl group. Intramolecular hydrogen bonding occurs between the hydroxyphenyl group and the N atom.  相似文献

Twenty nine novel N‐4‐methyl‐1,2,3‐thiadiazole‐5‐carbonyl‐N′‐phenyl ureas were designed and synthesized, and their structures were confirmed by proton nuclear magnetic resonance (1H NMR), infra red spectroscopy (IR) and high‐resolution mass spectroscopy (HRMS). Compounds V‐9 , V‐11 , V‐12 , V‐15 , V‐19 , V‐21 , V‐22 and V‐24 exhibit excellent activity against Culex pipiens pallens. Compounds V‐12 and V‐22 present good insecticidal activity against Plutella xylostella L. Their median lethal concentrations (LC50) are 164.15 and 89.69 mg·L?1, respectively. Compound V‐11 also has potential wide spectrum of fungicide activity. Its median effective concentrations (EC50) detected from 3.82 µg·mL?1 against Physalospora piricola to 31.60 µg·mL?1 against Cercospora arachidicola. Compounds V‐15 and V‐24 show outstanding induction activities as same as positive controls TDL and ningnanmycin, furthermore V‐24 has the highest induction activity of 41.85%±4.43%. To elucidate the structure activity relationship in these compounds, a 3D‐QSAR model has been built. The established model showed a reliable predicting ability with q2 values of 0.643 and r2 values of 0.982.  相似文献

Fourteen new diorganotin(IV) complexes of N‐(5‐halosalicylidene)‐α‐amino acid, R′2Sn(5‐X‐2‐OC6H3CH?NCHRCOO) (where X = Cl, Br; R = H, Me, i‐Pr; R′ = n‐Bu, Ph, Cy), were synthesized by the reactions of diorganotin halides with potassium salt of N‐(5‐halosalicylidene)‐α‐amino acid and characterized by elemental analysis, IR and NMR (1H, 13C and 119Sn) spectra. The crystal structures of Bu2Sn(5‐Cl‐2‐OC6H3CH?NCH(i‐Pr)COO) and Ph2Sn(5‐Br‐2‐OC6H3CH?NCH(i‐Pr)COO) were determined by X‐ray single‐crystal diffraction and showed that the tin atoms are in a distorted trigonal bipyramidal geometry and form five‐ and six‐membered chelate rings with the tridentate ligand. Bioassay results of a few compounds indicated that the compounds have strong cytotoxic activity against three human tumour cell lines, i.e. HeLa, CoLo205 and MCF‐7, and the activity decreased in the order Cy>n‐Bu>Ph for the R′ group bound to tin. Novel N‐aryl (and N‐alkyl) γ‐ and δ‐imino esters 2a–g ( 3a–g ) and N‐aryl (and N‐alkyl) ketimines 2h–j ( 3h–j ) were synthesized in high yields (80–99%) from their corresponding γ‐ and δ‐keto esters and ketones in this study. The structures of the synthesized compounds were clarified by Fourier transform infrared (FT‐IR), NMR (1H and 13C), mass spectrometry, and elemental analyses. Isomerizations [E/Z] were also determined by their 1H NMR spectra. The free‐radical scavenging activity of imines was evaluated using the 1,1‐diphenyl‐2‐picryl‐hydrazyl (DPPH) method. The relationships between the structure and antioxidant activity of these compounds are discussed. Among these compounds, 2a–c (at the concentration 1000 μg/mL) exhibit high antioxidant activity similar to those of the standards (butylated hydroxyanisole [ BHA], butylated hydroxytoluene [ BHT], and ascorbic acid).  相似文献

Two new dinuclear phenyltin(IV) complexes derived from N,N′‐bis(2‐hydroxybenzyl)‐1,2‐ethanebis(dithiocarbamate) ligand, [2‐HOC6H4CH2N(CS2SnPh3)CH2]2 ( 1 ) and [2‐HOC6H4CH2N(CS2SnClPh2)CH2]2 ( 2 ) have been synthesized and characterized by elemental analysis, IR and NMR (1H, 13C and 119Sn) spectra. The crystal structures of complexes 1 and 2 were determined by X‐ray single crystal diffraction and show that the dithiocarbamate ligand is coordinated to the tin atom in the anisobidentate manner and the tin atom is five‐coordinated. The coordination geometry of tin atom is best described as an intermediate between trigonal bipyramidal and square pyramidal with τ‐values of 0.63 and 0.53, respectively. Intermolecular hydrogen bonds (O H···S and O H···Cl) in 1 and 2 connect neighboring molecules into a one‐dimensional supramolecular chain with the centrosymmetric cyclic motifs. Complex 1 has potent in vitro cytotoxic activity against two human tumor cell lines, CoLo205 and Bcap37, while complex 2 displays weak cytotoxic activity. Salts of meta‐xylyl‐linked N‐ethyl/n‐butyl/benzyl‐substituted bis‐benzimidazolium having hexafluorophosphate counterions have been synthesized. The corresponding binuclear Ag(I)‐N‐heterocyclic carbene complexes were prepared by the reaction of Ag2O. The N‐heterocyclic carbene (NHC) ligand precursor 7 and Ag(I)–NHC complexes 10 and 11 have been structurally characterized by single‐crystal X‐ray diffraction technique. All of the reported compounds have been tested for their anticancer activity using human colorectal (HCT 116) cancer cell lines. Sterically varied benzimidazolium salts displayed significant activity against HCT 116 cell line, yielding IC50 values in the range 0.1–19.4 µ m , while Ag(I)–carbene complexes showed exceptionally good activity (0.2–1.3 µ m ) against tested cancer cell lines. A series of novel N‐aryl‐4‐(tert‐butyl)‐5‐(1H‐1,2,4‐triazol‐1‐yl)thiazol‐2‐amines synthesized in a green way. H2O2‐NaBr Brominating circulatory system was used in the synthesis of the key intermediate in a mild condition. All of the target compounds were confirmed by 1H NMR and elemental analysis and tested for their cytotoxicity against two different human cancer cell lines. The cytotoxicity assay revealed that some of the title compounds showed moderate to strong cytotoxic activities. Compound 2i was the most potent compound with the IC50 values of 9 μM against Hela cells and 15 μM against Bel–7402 cells, respectively.  相似文献

On the basis of the principle of combination of active groups, a series of novel N‐(4‐([2,2′:5′,2′′‐terthiophen]‐5‐yl)‐2‐methylbut‐3‐yn‐2‐yl) benzamide derivatives were designed, synthesized and systematically evaluated for their antiviral activity against tobacco mosaic virus (TMV). The bioassay results showed that most of these compounds displayed good anti‐TMV activity, and some of them exhibited higher antiviral activity than commercial Ningnanmycin. Especially, compound 8e with excellent anti‐TMV activity (inactivation activity, 92.3%/500 µg·mL?1; curative activity, 85.7%/500 µg·mL?1 and protection activity, 64.7%/500 µg·mL?1) emerged as a potential inhibitor of plant virus TMV. Quantitative structure‐activity relationship studies proved that the van der Waals volume (V) and electronic parameter (∑(∑σo+σp) and ∑σm) for the substituent R1 were very important for antiviral activities in this class of compounds.  相似文献

A series of diorganotin complexes with Schiff base ligands, (E)‐N′‐(5‐bromo‐2‐hydroxybenzylidene)‐3‐hydroxy‐2‐naphthohydrazide, H2L1, and (E)‐N′‐(5‐chloro‐2‐hydroxybenzylidene)‐3‐hydroxy‐2‐naphthohydrazide, H2L2, were synthesized and characterized by elemental analysis, IR, 1H, 13C and 119Sn NMR spectroscopy. The molecular structures of the complexes, [(5‐bromo‐2‐oxidobenzylidene)‐3‐hydroxy‐2‐naphthohydrazidato]di(o‐chlorobenzyl)tin(IV) 6 and [(5‐chloro‐2‐oxidobenzylidene)‐3‐hydroxy‐2‐naphthohydrazidato]dibutyltin(IV) 9, were determined through single‐crystal X‐ray diffraction and revealed a distorted trigonal‐bipyramidal configuration. The in vitro cytotoxic activity of the Schiff bases and their diorganotin complexes was also evaluated against several human carcinoma cell lines, namely HT29 (human colon carcinoma cell line), SKOV‐3 (human ovarian cancer cell line), MCF7 (hormone‐dependent breast carcinoma cell line) and MRC5 (non‐cancer human fibroblast cell line). [(5‐Bromo‐2‐oxidobenzylidene)‐3‐hydroxy‐2‐naphthohydrazidato]dibutyltin(IV) 2 and [(5‐bromo‐2‐oxidobenzylidene)‐3‐hydroxy‐2‐naphthohydrazidato]dibenzyltin(IV) 5 were the most active diorganotin complexes of H2L1 ligand. Among the diorganotin complexes of H2L2 ligand, [(5‐chloro‐2‐oxidobenzylidene)‐3‐hydroxy‐2‐naphthohydrazidato]dicyclohexyltin(IV) 11 showed good cytotoxic activity against all the tested cell lines. As such, the above compounds can be considered agents with potential anticancer activities, and can therefore be investigated further in in vitro or in vivo anticancer studies. 本文首次合成标题化合物N-(4-甲基苯甲酰氨基)-N’-[5-(2-三氟甲基苯基)-2-呋喃甲酰硫脲。化合物(C21H16F3N3O3S, Mr = 447.43)单晶经测定为单斜晶体,空间群为P -1。在晶体中,存在一些分子内和分子间的相互作用,分子间还有C—H···π 的相互作用,这可能导致晶体更稳定的原因。目标产物的结构经IR, H NMR和元素分析测定确证。初步生物活性测试表明,部分化合物对棉花枯萎病、黄瓜灰霉病、苹果轮纹病和棉花炭疽病有较好的选择性杀菌活性;部分目标化合物有较好的除草活性。  相似文献
