Comprehensive two-dimensional liquid chromatography (LC?×?LC) has received much attention because it offers much higher peak capacities than separation in a single dimension. The advantageous peak capacity makes it attractive for the separation of complex samples. Various gradient methods have been used in LC?×?LC systems. The use of continuous shift gradient is advantageous because it combines the peak compression effect of full gradient mode and the tailed gradient program in parallel gradient mode. Here, a comparison of LC?×?LC analysis of Chinese herbal medicine with full gradient mode and shift gradient mode in the second dimension was performed. A correlation between the first and second dimensions was found in full gradient mode, and this was significantly reduced with shift gradient mode. The orthogonality increased by 43.7 %. The effective peak distribution area increased significantly, which produced better separation.  相似文献

In the present study, we investigated the possibilities and limitations of computer-assisted method development (CAMD) for the HILIC separation optimization of a mixture of 13 isomeric hydroxy- and aminobenzoic acids on a ZIC-HILIC column. The isocratically obtained Neue and Kuss retention parameters enabled the accurate gradient retention modeling for peaks eluting well within the gradient (mean error of 2.7 %). The prediction errors for peaks eluting at the end of the gradient could be reduced from 8.8 to 6.1 % by implementing the isocratic regime after the gradient into the expression for the gradient retention factor. The prediction of the corresponding peak widths improved significantly for certain compounds and gradient profiles using individual gradient N values for each compound compared to employing a single N value for all compounds and gradient profiles. Two gradient optimization strategies (constructing the R s map based on individual retention modeling and predictive elution stretching and shifting, PEWS2) resulted in a reasonable separation of the challenging mixture of 13 isomeric hydroxy- and aminobenzoic acids on the ZIC-HILIC column. Overall, the optimization was limited by the steep decrease in N (dropping to the isocratic N value) and corresponding increase in peak width when increasing the gradient time. The discrimination factors d 0 were used to assess the resolution between peaks varying widely in height. The best separation was found to be obtained via the PEWS2 approach. Both the individual retention modeling and PEWS2 strategies corresponded to a total instrument time less than 12 h (including equilibration). Finally, it was found that the salt concentration had a significant effect on both the retention and the peak shape of the compounds, resulting in a small “solution domain” at 10 mM. Coupled columns with higher efficiencies are suggested to improve the resolution and robustness of the separation.  相似文献

The gradient elution behaviour of five synthetic decapeptide analogues has been investigated using an octadecylsilica stationary phase and trifluoroacetic acid-water-acetonitrile mobile phases. The influence of gradient time and flow-rate on the relative retentions and bandwidths of these peptides was assessed using quantitative expressions derived from linear solvent strength theory and general plate height theory. Linear relationships between logarithmic median capacity factors, log k, and the mole fraction of organic solvent modifier, phi, were observed over the experimental range of conditions used. The slopes of these plots were different for all peptides, which indicates that divergences will occur in the prediction of peptide retention times due to conformation dependent changes in hydrophobic contact area occupancy at the stationary phase surface. However, the differences in S values (tangent to the curve obtained in a plot of log k versus phi) for these peptides were not substantial enough to seriously affect the prediction of peptide retention times at one gradient slope from those observed at another. In addition, significant differences existed between experimental and theoretical peak capacity data of these peptide analogues of similar molecular weight and overall polarity, particularly at lower flow-rates or longer residence times. These results once again demonstrate that additional diffusional and interactive processes occur during the reversed-phase separation of peptides and proteins which are not yet adequately formalized by current chromatographic theory.  相似文献

Summary The effects of pH on both the solute retention and the peak shape of ionogenic compounds are studied in order to propose accurate models for pH optimization purposes. Several mathematical models (theoretical and empirical) for describing the variation of the retention factor versus pH are compared within different pH ranges. Limits of such models used for optimizing the pH by requiring only 3 preliminary experimental runs, are discussed in terms of deviations (≤±5%) of predicted retention times from experimental retention times. An original procedure is developed for selecting the most convenient retention model, from a given set of three retention data. This set is also applied to modeling the variation of both peak width and peak asymmetry with mobile phase pH conditions. Such a procedure is demonstrated as helpful for the separation of ionogenic solutes by considering mobile phase pH as an additional variable that can be useful during optimization procedures.  相似文献

In the present work it is shown that the linear elution strength (LES) model which was adapted from temperature-programming gas chromatography (GC) can also be employed to predict retention times for segmented-temperature gradients based on temperature-gradient input data in liquid chromatography (LC) with high accuracy. The LES model assumes that retention times for isothermal separations can be predicted based on two temperature gradients and is employed to calculate the retention factor of an analyte when changing the start temperature of the temperature gradient. In this study it was investigated whether this approach can also be employed in LC. It was shown that this approximation cannot be transferred to temperature-programmed LC where a temperature range from 60°C up to 180°C is investigated. Major relative errors up to 169.6% were observed for isothermal retention factor predictions. In order to predict retention times for temperature gradients with different start temperatures in LC, another relationship is required to describe the influence of temperature on retention. Therefore, retention times for isothermal separations based on isothermal input runs were predicted using a plot of the natural logarithm of the retention factor vs. the inverse temperature and a plot of the natural logarithm of the retention factor vs. temperature. It could be shown that a plot of lnk vs. T yields more reliable isothermal/isocratic retention time predictions than a plot of lnk vs. 1/T which is usually employed. Hence, in order to predict retention times for temperature-gradients with different start temperatures in LC, two temperature gradient and two isothermal measurements have been employed. In this case, retention times can be predicted with a maximal relative error of 5.5% (average relative error: 2.9%). In comparison, if the start temperature of the simulated temperature gradient is equal to the start temperature of the input data, only two temperature-gradient measurements are required. Under these conditions, retention times can be predicted with a maximal relative error of 4.3% (average relative error: 2.2%). As an example, the systematic method development for an isothermal as well as a temperature gradient separation of selected sulfonamides by means of the adapted LES model is demonstrated using a pure water mobile phase. Both methods are compared and it is shown that the temperature-gradient separation provides some advantages over the isothermal separation in terms of limits of detection and analysis time.  相似文献

A strategy for rapid optimization of liquid chromatography column temperature and gradient shape is presented. The optimization as such is based on the well established retention and peak width models implemented in software like e.g. DryLab and LC simulator. The novel part of the strategy is a highly automated processing algorithm for detection and tracking of chromatographic peaks in noisy liquid chromatography-mass spectrometry (LC-MS) data. The strategy is presented and visualized by the optimization of the separation of two degradants present in ultraviolet (UV) exposed fluocinolone acetonide. It should be stressed, however, that it can be utilized for LC-MS analysis of any sample and application where several runs are conducted on the same sample. In the application presented, 30 components that were difficult or impossible to detect in the UV data could be automatically detected and tracked in the MS data by using the proposed strategy. The number of correctly tracked components was above 95%. Using the parameters from the reconstructed data sets to the model gave good agreement between predicted and observed retention times at optimal conditions. The area of the smallest tracked component was estimated to 0.08% compared to the main component, a level relevant for the characterization of impurities in the pharmaceutical industry.  相似文献

The overall peak capacity in comprehensive two-dimensional liquid chromatographic (LC x LC) separation can be considerably increased using efficient columns and carefully optimized mobile phases providing large differences in the retention mechanisms and separation selectivity between the first and the second dimension. Gradient-elution operation and fraction-transfer modulation by matching the retention and the elution strength of the mobile phases in the two dimensions are useful means to suppress the band broadening in the second dimension and to increase the number of sample compounds separated in LC x LC. Matching parallel gradients in the first and second dimension eliminate the necessity of second-dimension column re-equilibration after the independent gradient runs for each fraction, increase the use of the available second-dimension separation time and can significantly improve the regularity of the coverage of the available retention space in LC x LC separations, especially with the first- and second-dimension systems showing partial selectivity correlations. Systematic development of an LC x LC method with parallel two-dimensional gradients was applied for separation of phenolic acids and flavone compounds. Several types of bonded C18, amide, phenyl, pentafluorophenyl and poly(ethylene glycol) columns were compared using the linear free energy relationship method to find suitable column combination with low correlation of retention of representative standards. The phase systems were optimized step-by-step to find the mobile phases and gradients providing best separation selectivity for phenolic compounds. The optimization of simultaneous parallel gradients in the first and second dimension resulted in significant improvement in the utilization of the available two-dimensional retention space.  相似文献

The linear solvent strength model was used to predict coverage in online comprehensive two‐dimensional reversed‐phase liquid chromatography. The prediction model uses a parallelogram to describe the separation space covered with peaks in a system with limited orthogonality. The corners of the parallelogram are assumed to behave like chromatographic peaks and the position of these pseudo‐compounds was predicted. A mix of 25 polycyclic aromatic compounds were used as a test. The precision of the prediction, span 0–25, was tested by varying input parameters, and was found to be acceptable with root mean square errors of 3. The accuracy of the prediction was assessed by comparing with the experimental coverages. Less than half of experimental coverages were outside prediction ± 1 × root mean square error and none outside prediction ± 2 × root mean square error. Accuracy was lower when retention factors were low, or when gradient conditions affected parameters not included in the model, e.g. second dimension gradient time affects the second dimension equilibration time. The concept shows promise as a tool for gradient optimization in online comprehensive two‐dimensional liquid chromatography, as it mitigates the tedious registration and modeling of all sample constituents, a circumstance that is particularly appealing when dealing with complex samples.  相似文献

In this work we develop a practical approach to optimization in comprehensive two dimensional liquid chromatography (LC x LC) which incorporates the important under-sampling correction and is based on the previously developed gradient implementation of the Poppe approach to optimizing peak capacity. The Poppe method allows the determination of the column length, flow rate as well as initial and final eluent compositions that maximize the peak capacity at a given gradient time. It was assumed that gradient elution is applied in both dimensions and that various practical constraints are imposed on both the initial and final mobile phase composition in the first dimension separation. It was convenient to consider four different classes of solute sets differing in their retention properties. The major finding of this study is that the under-sampling effect is very important and causes some unexpected results including the important counter-intuitive observation that under certain conditions the optimum effective LC x LC peak capacity is obtained when the first dimension is deliberately run under sub-optimal conditions. In addition, we found that the optimum sampling rate in this study is rather slower than reported in previous studies and that it increases with longer first dimension gradient times.  相似文献

The development and application of new separation mechanisms such as hydrophilic interaction chromatography (HILIC) is of high importance for the simultaneous analysis of polar molecules such as primary metabolites. However the retention mechanism in HILIC is not fully understood and as a result retention prediction tools are not at hand for this chromatographic approach. In the present report we study the utility of a simple algorithm, based on a simple linear and/or a simple logarithmic retention model, for retention prediction in HILIC gradient separation of a mixture of 23 selected compounds including (poly)amines, amino acids, saccharides, and other molecules. Utilizing two types of gradient elution programs with or without an isocratic part, retention data were collected in order to build prediction models. Starting from at least three gradient runs the prediction of analyte retention was very satisfactory for all gradient programs tested, providing useful evidence of the value of such retention time prediction methodologies.  相似文献

Summary After a brief introduction of the subject, the paper focusses on the first step in any optimization procedure: the delineation of the parameter space, wherein the global optimum is to be found. For organic modifier optimization in reversed-phase liquid chromatography it is shown that the necessary information can be derived from a single water-methanol gradient. It first yields an estimate of the total number of solutes in the sample, which is vital to define the peak capacity needed to achieve separation at a certain confidence level. Next, the gradient allows the prediction of suitable isocratic methanol binary solvents, and transfer rules formulate the iso-eluotropic composition of the common binary solvents (tetrahydrofuran and acetonitrile). Because all predictions are based on a statistical analysis of a limited data base, attention is given to the practical situation where an actual sample deviates from the average solute behaviour. Such deviations are revealed in the first isocratic run and can be used to arrive at a better estimate of solute retention.  相似文献

Previous studies demonstrated that quantitative structure-retention relationships (QSRR) combined with the linear solvent strength (LSS) model allow for prediction of gradient reversed-phase liquid chromatography retention time for any analyte of a known molecular structure under defined LC conditions. A QSRR model derived at the selected gradient time and at the same gradient time was tested. The aim the present study was to evaluate the accuracy of QSRR predictions used during the predictions of LC gradient retention times with variable gradient times. For this purpose, predictions of retention times at two gradient times were used to find the optimal, different gradient times. In the first step, experimental retention data for the model set of analytes were used to derive appropriate QSRR models at two gradient times. These QSRR models were further used to predict gradient retention times for another set of testing analytes at the two selected above gradient times. Then, applying linear solvent-strength (LSS) theory, the predicted retention times for test analytes were used to find other optimal gradient times for those analytes. Satisfactory predictions of gradient retention times for test analytes were obtained at gradient times different from those applied for model analytes.  相似文献

采用微管液相色谱法同时测定白术中的白术内酯Ⅲ、白术内酯Ⅰ和苍术酮。采用美国Micro-tech Ultra plusⅡTM微管柱通用型二元梯度高效液相色谱仪和Microsil C18微管色谱柱(150 mm×1.0 mm),以甲醇-乙腈-水(A相体积比为90∶5∶5;B相体积比为55∶5∶40)为流动相,梯度洗脱,流速50 μL/min,在220 nm波长下检测。在上述色谱条件下,3种组分分离良好,在相应的进样质量范围内具有良好的线性关系。白术内酯Ⅲ、Ⅰ和苍术酮的加标回收率分别为96.86%,97.13%和98.06%;相对标准偏差分别为1.63%,1.31%和0.39%。该法简便、快捷,能够用于白术的质量控制。  相似文献

A novel comprehensive two-dimensional liquid chromatographic (LC×LC) system was developed for the quantification of antioxidant phenolic compounds in wine and juice. The system allows parts of the sample that are well separated in the first column to pass directly to the detector after the first column, while the rest of the sample proceeds to the second column. The optimised LC×LC system employed a combination of two C18 columns, the latter column with an ion-pair reagent (tetrapentylammonium bromide). The relative standard deviations (RSD) for the retention times were better than 0.01% in the first dimension and on average 6.3% in the second. The RSD values of the peak volumes varied from 3% (protocatechuic acid) to 13% (caffeic acid, n = 3, 10 μg/ml).  相似文献

The mixed-mode separation of a selection of anionic and cationic pharmaceutically related compounds is studied using ion-exchange columns and eluents consisting of ionic salts (potassium hydroxide or methanesulfonic acid) and an organic modifier (methanol). All separations were performed using commercially available ion-exchange columns and an ion chromatography instrument modified to allow introduction of methanol into the eluent without introducing compatibility problems with the eluent generation system. Isocratic retention prediction was undertaken over the two-dimensional space defined by the concentration of the competing ion and the percentage of organic modifier in the eluent. Various empirical models describing the observed relationships between analyte retention and both the competing ion concentration and the percentage of methanol were evaluated, with the resultant model being capable of describing the separation, including peak width, over the entire experimental space based on six initial experiments. Average errors in retention time and peak width were less than 6% and 27%, respectively, for runs taken from both inside and outside of the experimental space. Separations performed under methanol gradient conditions (while holding the competing ion concentration constant) were also modelled. The observed effect on retention of varying the methanol composition differed between analytes with several analytes exhibiting increased retention with increased percentage methanol in the eluent. An empirical model was derived based on integration of the observed tR vs. %methanol plot for each analyte. A combination of the isocratic and gradient models allowed for the prediction of retention time using multi-step methanol gradient profiles with average errors in predicted retention times being less than 4% over 30 different 2- and 3-step gradient profiles for anions and less than 6% over 14 different 2- and 3-step gradient profiles for cations. A modified peak compression model was used to estimate peak widths under these conditions. This provided adequate width prediction with the average error between observed and predicted peak widths being less than 15% for 40 1-, 2- and 3-step gradients for anions and less than 13% over 14 1-, 2- and 3-step gradients for cations.  相似文献

The development of a rapid, reliable and reproducible LC method for the determination and quantification of 13 polyphenols (gallic acid, protocatechuic aldehyde, gentisic acid, catechin, vanillinic acid, caffeic acid, vanillin, epicatechin, syringaldehyde, p-coumaric acid, ferulic acid, sinapic acid and resveratrol) in grapes and derived products is reported. The polyphenols were separated in less than 8 min. Employed was an RP-18e (100 mm × 4.6 mm) monolithic type column. A gradient method with the following solvents was utilised for the chromatographic separation: A: 90% water, 2% acetic acid in methanol, and B: 90% methanol, 2% acetic acid in water. Two detectors in series were employed: a UV–Vis detector and a fluorescence excitation/emission detector. Influence of temperature (15–40 °C) and solvent flow rate (2–5 mL min?1) on the separation were studied, and 25 °C and 2.5 mL min?1 were found to be the optimum conditions. The relative standard deviations of the resulting peak areas, for both intra- and inter- experiments, were less than 2.4 and 2.6%, respectively. Finally, the developed method has been utilised for the quantification of the polyphenols in real samples.  相似文献

Computer models have been widely used to predict the chromatographic behaviour of liquid chromatography systems. With the introduction of mass spectrometric detection and the use of lower mobile phase flow rates with conventional LC equipment, the influence of the dwell volume on the shape of the gradient curve becomes an issue in predicting the retention times. A new straight forward algorithm is proposed for the modelling of retention times in reversed-phase LC, taking the effect of the dwell volume on the gradient shape into account. The results show that the dwell volume has a large effect at lower flow rates and on the retention times of the analytes eluting at the end of fast gradient curves. The proposed model is able to make reliable predictions and can be helpful in LC-MS method development.  相似文献

Previous studies of peptide separation by normal-phase liquid chromatography have shown a linear relationship between the logarithm of the capacity factor and the logarithm of the volume fraction of modifier in the mobile phase. This permitted the use of a model to predict isocratic and gradient retention times based on data obtained by two initial gradient runs. In the present study, chromatographic behavior of 25 peptides in normal-phase liquid chromatography with isocratic elution have been studied and a linear relationship between the slope (S) and intercept [log k(0)] was obtained. This relationship was combined with the algorithm of prediction reported in the previous paper. The prediction of peptide retention times with only a single experimental gradient retention data was investigated.  相似文献
