The adsorptive and electrochemical behaviors of azithromycin were investigated on a glassy carbon electrode that was electrochemically treated by anodic oxidation at +1.8 V, following potential cycling in the potential range from -0.8 to +1.0 V. The resulting electrode showed good activity to improve the electrochemical response of the drug. An adsorptive stripping voltammetric method for the determination of azithromycin at an electrochemically activated glassy carbon electrode has been developed. Azithromycin was accumulated in phosphate buffer, pH 6, at a potential of +0.3 V (vs. Ag/AgCl electrode) for a certain time, and then determined by differential pulse voltammetry. The oxidative peak current at +0.82 V, at a scan rate of 20 mV s(-1), was a linear function of the concentration in the ranges of 0.25 - 2 microg mL(-1) and 1 - 10 microg mL(-1) using a 240 or 60 s(-1) preconcentration time, respectively. Application of the method to the determination of azithromycin in pharmaceuticals resulted in an acceptable deviation from the stated concentration. The preconcentration medium-exchange approach was utilized for the selective determination of the drug in spiked urine samples with satisfactory results. The peak current was linear with the drug concentration in the range of 0.5 - 3.5 microg per mL urine. The detection limit was 0.2 microg mL(-1) urine. The recovery levels of the method reached 96.3%.  相似文献

Rubbery ormosil films with immobilized aminofluorescein (AF) were investigated to develop an optochemical sensor for the determination of ammonia in water. The gel precursors with tetramethoxysilane (TMOS) and dimethyldimethoxysilane (DiMeDMOS) were deposited on glass supports, and characterized in terms of response to pH, and to dissolved ammonia at constant pH. After preconditioning the sensing film was stable for 6 months. The detection limit for ammonia in water was 0.2 μg mL–1 (S/N 2), the response being linearly dependent on concentration in the range of 0.5 to 80 μg mL–1 ammonia. The response time was less than 5 min. The effects of sodium chloride concentration, temperature, and coexisting metal ions and compounds were investigated. A spectrophotometric method is developed for the determination of traces of copper(II), based on the catalytic oxidative coupling reaction of 3-hydroxyacetanilide with 3-methyl-2-benzothiazolinone hydrazone in the presence of ammonia and hydrochloric acid. Beer's law is obeyed in the copper(II) concentration range of 0.008-0.16 microg mL(-1), and the molar absorptivity at 530 nm is 2.5x10(5) L mol(-1) cm(-1). The Sandell's sensitivity of the product is 0.000254 microg cm(-2). The optimum reaction conditions and other important analytical parameters have been investigated. The proposed method is applied to the analysis of water and soil samples and the results are compared with the literature method.  相似文献

Simple, accurate, reproducible, selective, sensitive and cost effective UV-spectrophotometric methods were developed and validated for the estimation of trigonelline in bulk and pharmaceutical formulations. Trigonelline was estimated at 265 nm in deionised water and at 264 nm in phosphate buffer (pH 4.5). Beer's law was obeyed in the concentration ranges of 1-20microg mL(-1) (r2=0.9999) in deionised water and 1-24 microg mL(-1) (r2=0.9999) in the phosphate buffer medium. The apparent molar absorptivity and Sandell's sensitivity coefficient were found to be 4.04 x 10(3)L mol(-1)cm(-1) and 0.0422 microg cm(-2)/0.001A in deionised water; and 3.05 x 10(3)L mol(-1)cm(-1) and 0.0567 microg cm(-2)/0.001A in phosphate buffer media, respectively. These methods were tested and validated for various parameters according to ICH guidelines. The detection and quantitation limits were found to be 0.12 and 0.37 microg mL(-1) in deionised water and 0.13 and 0.40 microg mL(-1) in phosphate buffer medium, respectively. The proposed methods were successfully applied for the determination of trigonelline in pharmaceutical formulations (vaginal tablets and bioadhesive vaginal gels). The results demonstrated that the procedure is accurate, precise, specific and reproducible (percent relative standard deviation <2%), while being simple and less time consuming and hence can be suitably applied for the estimation of trigonelline in different dosage forms and dissolution studies.  相似文献

A highly selective, sensitive and rapid method for the determination of trace amounts of inorganic mercury based on the reaction of Hg (II) with 6-mercaptopurine and the solid phase extraction of the complex on C18 membrane disks was developed. The 6-mercaptopurine selectively reacts with Hg (II) to form a complex in the pH range of 5-8. This complex was preconcentrated by solid phase extraction with C18 disks. An enrichment factor of 100 was achieved. The molar absorptivity of the complex is 0.26 x 10(-6) L. mol(-1) cm(-1) measured at 315 nm. The Beer's law is obeyed in the concentration range of 0.002-0.048 microg mL(-1). The relative standard deviation for eleven-replicated measurement of 0.04 microg mL(-1) is 1.5 %. The detection limit is 0.001 microg mL(-1) in the water samples. The advantage of the method is that the determination of Hg (II) is free from interference of almost all the cations and anions found in environment and wastewater samples. The determination of Hg (II) in water samples of different origins and marine sediment were carried out by the present method and cold vapor atomic absorption spectrometry (CVAAS). Also the method's accuracy was investigated by using SRM 2709. The obtained results by the present procedure were in good agreement with those of the CVAAS and certified value, so that the applicability of the proposed method was confirmed for the real samples.  相似文献

Preconcentration of chemical warfare agent degradation products (alkylphosphonic acids and alkyl alkylphosphonic acids) in low-conductivity matrices (purified water, tap water and local river water) by field-amplified sample stacking (FASS) was developed for capillary electrophoresis (CE) coupled to ion trap mass spectrometry. FASS was performed by adding a mixture of HCOONH(4) and NH(4)OH in appropriate concentrations to the sample. This allowed to control the conductivity and the pH of the sample in order to obtain FASS performances that are independent of analyte concentration. The influence of different parameters on FASS (sample to background electrolyte (BGE) conductivity ratio, injection volume and concentration of BGE) was studied to determine the optimal conditions and was rationalized by using the theoretical model developed by Burgi and Chien. A good correlation was obtained between the bulk electroosmotic velocity predicted by this model and the experimental value deduced from the migration time of the electroosmotic flow marker detected by mass spectrometry (MS). This newly developed method was successfully applied to the analysis of tap water and local river water fortified with the analytes and provided a 10-fold sensitivity enhancement in comparison to the signal obtained without preconcentration procedure. The quite satisfactory repeatability and linearity for peak areas obtained in the 0.5-5 microg mL(-1) concentration range allow quantitative analysis to be implemented. Limits of detection of 0.25-0.5 microg mL(-1) for the alkyl alkylphosphonic acids and of 0.35-5 microg mL(-1) for the alkylphosphonic acids were reached in tap water and river water.  相似文献

A simple and rapid capillary zone electrophoresis (CZE) method with UV detection has been developed for the determination of famotidine and its potential impurities in pharmaceutical formulations. The electrophoretic separation of these compounds was performed using a fused silica capillary and 37.5mmolL(-1) phosphate buffer pH 3.5 as the electrolyte. Under the optimised conditions, six impurities could be resolved from the famotidine peak in less than 7min. The calibration curves obtained for the seven compounds were linear over the concentration range investigated (from 1.5 to 78.5microg mL(-1)). The intra- and inter-day relative standard deviations for the migration times and corrected peak areas were less than 2% and 5%, respectively. The detection limits were found to be 0.09microg mL(-1) for famotidine, and from 0.1 to 0.62microg mL(-1) depending on the impurities. The method has been successfully applied to the determination of famotidine in commercial dosage forms.  相似文献

This work presents the development of an on-line pre-concentration system for simultaneous determination of Cd, Cu, Ni, V, Zn, Co and Pb in aqueous environmental samples and detection by inductively coupled plasma mass spectrometry. The system is based on cationic retention of the analytes onto a mini-column filled with silica gel modified with niobium(V) oxide. The effects of chemicals and flow variables have been investigated. The optimized operating conditions, selected as a compromise between sensitivity and analytical frequency were: sample pH 7.0, sample flow rate of 6.0mL min(-1), eluent flow rate of 2.0mL min(-1), and eluent (HNO(3)) concentration of 2.5mol L(-1). The relative standard deviation (n=7), enrichment factor and linear working range were 0.8-4.5%, 23.3-37.2 and 0.05-25.0microg L(-1), respectively. Limits of detection were between 0.01 and 0.03microg L(-1). The accuracy of the proposed method was checked with certified materials (NASS-4, NASS-5, CASS-4 and SRM 1643e). Values obtained were in accordance with those reported for the certified materials. Recovery was found to be in the range of 90-110% for a suit of water samples with variable matrices (seawater, tap water and ground water) collected in Florianopolis, Brazil.  相似文献

In this work, the development of two solid-phase extraction procedures (off-line and on-line formats) for the identification and quantification of several (fluoro)quinolones in hospital sewage water by HPLC-UV is described. Both procedures are based on the use of C18 and anion exchange (SAX) sorbents for the preconcentration and clean-up steps, respectively, and all variables influencing both steps were optimised. In the off-line format, after its pH was adjusted to 2.5, sample was preconcentrated on a C18 cartridge and eluted with 4 mL of methanol/ammonia (94/6). The methanolic extract must be diluted up to 10 mL with water to allow quantitative retention of the analytes on the SAX cartridge. In the on-line format, the addition of 2.5% of NH4Cl to the sewage water sample (pH = 2.5) was necessary to increase the breakthrough volumes of the analytes in the C18 precolumn. Quantitative transfer of the (fluoro)quinolones from the C18 precolumn to the SAX precolumn was accomplished by pumping 2 mL of a mixture methanol/water (40/60, pH = 9.2) at 2 mL min(-1). Elution of the analytes from the SAX precolumn by means of the chromatographic mobile phase required the inclusion of an additional isocratic step at the beginning of the gradient program. Both off-line and on-line solid phase extraction procedures coupled to HPLC-UV were applied to the analysis of a sewage water sample collected in the sewer system at the output of the St Dimphna Hospital (Geel, Belgium). The fluoroquinolone ciprofloxacin was found in this sample and quantified at 5.8 +/- 0.4 microg L(-1) (off-line method) and 5.6 +/- 0.5 microg L(-1) (on-line method). The analysis of spiked samples containing the seven (fluoro)quinolones studied provided quantitative recoveries in all cases with low RSD values (from 6 to 12%), and all the analytes could be identified by means of their UV spectra with match factors varying from 950 to 985 depending on the (fluoro)quinolone.  相似文献

A new fluorescent reagent 1,5-bis(4,6-dichloro-1,3,5-triazinylamino)naphthalene (DTAN) was synthesized. The optimum conditions of fluorescent reaction of this reagent with aniline (PA) were also investigated. Based on this reaction, a new spectrofluorimetric method was developed for the determination of aniline. The fluorescent intensity was directly proportional to the concentration of aniline in the ranges 0.05-2.0 microg mL(-1) and 2.0-50 microg mL(-1) with the detection limits of 34 ng mL(-1) and 90 ng mL(-1). This method is simple, practical and can afford good precision and accuracy and can be successfully applied to assess aniline in water samples. A possible mechanism of the change of fluorescence intensity introduced by putting the aniline into the system is also discussed.  相似文献

Even though monitoring of dissolved ammonia is acutely important for environmental studies, fish farms and for industrial surveillance, no system for the performance of online measurements at the concentrations needed exists so far. For many applications it is necessary to detect dissolved ammonia concentrations at sub mg/l-levels, because ammonia is reported to be toxic for aquatic organisms above 25 microg/l. We present new ammonia sensitive materials consisting of fluorescent pH indicators embedded into different cellulose esters. The low pK(a) value of the indicators and the high solubility of ammonia in the cellulose polymers lead to detection limits below 1 microg/l and a dynamic range between 5 and 1000 microg/l. Response times at these trace concentration levels are in the order of 20-30 min. The sensors are suitable for fresh and sea water monitoring by an additional silicon layer preventing the interference of protons and salinity. The fluorescent indicators Eosin ethylester and 2',7'-dichlorofluorescein methylester (DCF) were investigated to achieve sensors with a dynamic range matching the target concentrations. Sensors with improved performance were obtained by employing cellulose ester nanospheres with incorporated Eosin ethylester. The simple sensor design has a high potential to be applied in miniaturized optical measurement system for online ammonia detection.  相似文献

A new ammonia optical sensor was designed using bis(acetylacetoneethylendiamine)tributylphosphin cobalt(III) tetraphenylborate complex, coated on transparent triacetylcellulose film as membrane. The change in the absorbance of the optode at the maximum wavelength of 408 nm was related to ammonia concentration in aqueous samples. A buffer solution with a pH of 9 (sodium borate-HCl) was used. The optode was fully regenerated in pH 2. The linear dynamic range for determination of ammonia was 3.3 x 10(-4) to 6.9 x 10(-3) mol l(-1) with a detection limit of 5.0 x 10(-5) mol l(-1) and a response time range of 4 - 6 min. This membrane was successfully applied for determination of ammonia in drinking water.  相似文献

A novel method for the determination of proteins by using tetracarboxy manganese(II) phthalocyanine (MnC4Pc) as a resonance light scattering (RLS) probe has been developed. At pH 3.0 Britton-Robinson (B-R) buffer solution, the RLS intensity of MnC4Pc at 385 nm is greatly enhanced in the presence of proteins. The effects of pH, reaction time, concentration of MnC4Pc and interfering substances on the enhanced RLS intensity are investigated, respectively. Under optimal conditions, the linear ranges of the calibration curves are 0-2.00 microg mL(-1) for bovine serum albumin (BSA) and human serum albumin (HSA), 0.0-1.75 microg mL(-1) for human-IgG and ovalbumin, with a detection limit of 16.37 ng mL(-1) BSA, 17.62 ng mL(-1) HSA, 19.41 ng mL(-1) human-IgG and 20.72 ng mL(-1) ovalbumin. The method has been applied to the determination of total proteins in human serum samples collected from a hospital and the results are in good agreement with those reported by the hospital.  相似文献

This paper describes the enhanced separation of adenine (A), hypoxanthine (HX), 8-azaadenine (8-AA), thymine (T), cytosine (C), uracil (U) and guanine (G) by CZE dispersing carboxylic multiwalled carbon nanotubes (c-MWNTs) into the running buffer. The effect of important factors such as c-MWNT nanoparticle concentration, the acidity and concentration of running buffer, and separation voltage were investigated to acquire the optimum conditions. The seven purine and pyrimidine bases could be well separated within 16 min in a 35 cm effective length fused-silica capillary at a separation voltage of +8.0 kV in a 23 mM tetraborate buffer (pH 9.2) containing 8.0 x 10(-5) g/mL c-MWNTs. Under the optimal conditions, the linear ranges were of 2-250 microg/mL for A (R2 = 0.995), 3-200 microg/mL for U (R2 = 0.990) and G (R2 = 0.992), 3-250 microg/mL for T (R2 = 0.998), 2-200 microg/mL for C (R2 = 0.985) and 4-200 microg/mL for HX (R2 = 0.988) and 8-AA (R2 = 0.990). The detection limits were 0.9 microg/mL for A (S/N = 3), 2.4 microg/mL for U, 2.0 microg/mL for T, 1.5 microg/mL for C, 2.5 microg/mL for G and 3.0 microg/mL for HX and 8-AA. The proposed method was successfully applied for determining five purine and pyrimidine bases in yeast RNA.  相似文献

Two simple and accurate methods of analysis to determine pioglitazone hydrochloride (PIO) and mefformin hydrochloride (MET) in combined dosage forms were developed using second-derivative spectrophotometry and reversed-phase liquid chromatography (LC). PIO and MET in combined preparations (tablets) were quantified using the second-derivative responses at 227.55 nm for PIO and 257.25 nm for MET in spectra of their solutions in a mixture of methanol and acetonitrile (30 + 70). The calibration curves were linear [correlation coefficient (r) = 0.9984 for PIO and 0.9986 for MET] in the concentration range of 8-40 microg/mL for PIO and 4-12 microg/mL for MET. In the LC method, analysis was performed on a Hypersil ODS-C18 column with 5 microm particle size using the mobile phase acetonitrile-water-acetic acid (75 + 25 + 0.3), adjusted to pH 5.5 with liquor ammonia, at a flow rate of 0.5 mL/min. Measurement was made at a wavelength of 230 nm. Both the drugs were well resolved on the stationary phase, and the retention times were 8.5 min for PIO and 16.0 min for MET. The calibration curves were linear (r = 0.9933 for PIO and 0.9958 for MET) in the concentration range of 4-20 microg/mL for PIO and MET. Both methods were validated, and the results were compared statistically. They were found to be accurate, precise, and specific. The methods were successfully applied to the estimation of PIO and MET in combined tablet formulations.  相似文献

The analysis is described for separating seven beta-adrenergic blocking agents (atenolol, celiprolol, clorprenaline, fenoterol, metoprolol, propranolol, terbutaline) and clenbuterol (sympathomimetic beta-2 receptor stimulating agonist, decongestant and bronchodilator, illicit anabolic used in athletics) by CE with UV detection. In order to simultaneously separate all analytes, Tris-H3PO4 solution was applied containing titanium dioxide nanoparticles (TiO2 NPs) as BGEs. The effects of important factors, such as concentration of TiO2 NPs, optimum pH, run buffer concentration, and separation voltage, were investigated so as to achieve best CE separation. The eight analytes could be well separated applying a separation voltage of 15 kV in 75 mM Tris-H3PO4 buffer at a pH of 2.40, containing 6.0 x 10(-6) g/mL TiO2 NPs. Under these optimal conditions, the RSDs for peak areas and for migration times were less than 2.7 and 2.3%, respectively. The detection limits were 0.1 microg/mL for celiprolol, 0.1 microg/mL for propranolol, 0.2 microg/mL for fenoterol, 1.0 microg/mL for atenolol, 1.0 microg/mL for clenbuterol, 1.0 microg/mL for clorprenaline, 1.0 microg/mL for metoprolol, and 1.0 microg/mL for terbutaline. The proposed method was successfully applied for the rapid CE determination of the frequently applied antihypertensive beta-blocking compounds atenolol, metoprolol, terbutaline, and propranolol in pharmaceutical tablets.  相似文献

This work assesses the use of modified natural natrolite zeolite as an adsorptive material for the separation and preconcentration of trace amounts of ions. In this work we investigated the potential of modified natural natrolite zeolite for the simultaneous separation and preconcentration of trace amounts of copper and zinc ions. We have developed a simple, rapid, selective, sensitive and economical method for the simultaneous separation and preconcentration of trace amounts of copper and zinc in an aqueous medium using 2-(5-bromo-2-pyridylazo)-5-diethylaminophenol (5-Br-PADAP) as an analytical reagent. The sorption was quantitative in the pH range 7.5 - 9.5, whereas quantitative desorption occurred instantaneously with 5.0 mL of 2 mol L(-1) nitric acid. Linearity was maintained between 0.05 - 6.0 microg mL(-1) for copper and 0.02 - 1.5 microg mL(-1) for zinc in the final solution. Ten replicate determinations of 1.0 microg mL(-1) copper and 0.5 microg mL(-1) zinc in a mixture gave mean absorbances of 0.1687 and 0.2788 with relative standard deviations of +/-1.2% and +/-1.3%, respectively. The detection limits were 0.03 ng mL(-1) for Cu(II) and 0.006 ng mL(-1) for Zn(II) in the original solution (3 sigma(bl)/m). Different parameters, such as the effect of the pH, flow rate, breakthrough volume and interference of a large number of anions and cations, were studied and the proposed method was used for the determination of these metal ions in water as well as standard samples (e.g. Nippon Keikinzoku Kogyo (NKK) CRM, No. 916 and No. 920 aluminum alloy, National Institute for Environment Studies (NIES) No. 1 pepperbush and NIES No. 2 pond sediment). The determination of these metal ions in standard samples showed that the proposed method has good accuracy (recovery > 97%).  相似文献

A reliable and specific method for the determination of citalopram enantiomers was developed and validated. Chromatographic resolution of citalopram enantiomers was made on a Shim-pack (5 microm particle size) cyanopropyl column with beta-cyclodextrin (beta-CD) as an effective chiral mobile phase additive. The composition of the mobile phase was (90 + 10, v/v) aqueous 0.1% triethylammonium acetate buffer, pH 4.0 (adjusted with acetic acid), and acetonitrile, containing 12 mM beta-CD. The flow rate was 0.8 mL/min with ultraviolet detection at 240 nm. The effects of the mobile phase composition, concentration of beta-CD, and pH of the triethylammonium acetate buffer on peak shape and resolution of the enantiomers were investigated. The calibration graphs were linear (r = 0.9999, n = 8) in the range of 1-40 microg/mL for S(+) citalopram and R-(-) citalopram. The limit of detection values were 5.51 x 10(-3) and 4.35 x 10(-3) pg/mL, while the limit of quantification values were found to be 1.84 x 10(-2) and 1.45 x 10(-2) microg/mL for S-(+) citalopram and R-(-) citalopram, respectively.  相似文献

In this study, low concentrations of histamine2-receptor (H2-)antagonists were effected across a water plug, with separation taking place in a binary buffer comprising ethylene glycol and NaH2PO4 (pH 5.0), and detection at 214 nm. Liquid-liquid extraction with ethyl acetate- isopropanol is shown to provide extracts that are sufficiently clean. The calibration curves were linear over a concentration range of 0.1-2.00 microg/mL cimetidine, 0.2-5.0 microg/mL ranitidine-HCl, 0.3-5.0 microg/mL nizatidine, and 0.1-3.0 microg/mL famotidine. Mean recoveries were > 82%, while the intra- and interday relative standard deviations (RSDs) and relative errors (REs) were all < 13%. The method is sensitive with a detection limit of 3 ng/mL cimetidine, 30 ng/mL ranitidine HCl, 50 ng/mL nizatidine and 10 ng/mL famotidine (S/N = 3, electric-driven injection 90 s). This newly developed capillary electrophoresis (CE) method was applied for the determination of analytes extracted from plasma taken from a volunteer dosing a cimetidine, ranitidine, and nizatidine tablet simultaneously. These three H2-antagonists can be detected in real samples by this method, excluding the low dosing of famotidine tablet.  相似文献

An automated LPME device for a dynamic LPME method was manufactured and its extraction efficiency was tested using spiked urine samples. The developed home-made LPME device was a programmable automated syringe dispenser to overcome deteriorating precision and difficulties in manually manipulating the plunger repeatedly. To establish the optimum parameters for benzene ethylamines, the effects of sampling volume, solvent volume, pH, salt-effect, choice of solvents, plunger speed, and number of samplings were investigated. Good repeatabilities for the extraction of mephentermine, ephedrine, methoxyphenamine, selegiline, and bupropion were obtained and the RSD values were 2.4, 1.9, 1.3, 1.6 and 1.5% at a concentration of 3 microg mL(-1) in spiked urine samples, respectively. The limit of detection was below 0.05 microg mL(-1) for the investigated drugs. This developed device for LPME analysis gave good validation results and improved convenience.  相似文献
