The mechanism of long chain free fatty acid (FFA) transport across cell membranes is under active investigation. Here we describe the use of multi imaging mass spectrometry (MIMS) to monitor intracellular concentrations of FFA and provide new insight into FFA transport in cultured adipocytes. Cells were incubated with 13C-oleate:BSA and either dried directly or dried after washing with a medium deprived of 13C-oleate:BSA. Cells were analyzed with MIMS using a scanning primary Cs+ ion beam and 12C-, 13C-, 12C14N-, 13C14N-) (or 12C 15N-) were imaged simultaneously. From these quantitative images the values of the 13C/ 12C ratios were determined in the intracellular lipid droplets, in the cytoplasm and outside the 3T3F442A adipocytes. The results indicate that after incubation with 13C-oleate:BSA the droplet 13C/ 12C ratio was 15 +/- 6%. This value is about 14-fold higher than the 13C/ 12C terrestrial ratio (1.12%). After washing the 13C-oleate:BSA, the droplet 13C/ 12C ratios decreased to 1.6 +/- 0.1%, about 40% greater than the natural abundance. Results for washed cells indicate that relatively little FFA was esterified. The unwashed cell results, together with the value of the lipid water partition coefficient, reveal that intracellular unbound FFA (FFAu) concentrations were on average about 4.5-fold greater than the extracellular FFAu concentrations. These results are consistent with the possibility that FFA may be pumped into adipocytes against their electro-chemical potential. This work demonstrates that MIMS can be used to image and quantitate stable isotope labeled fatty acid in intracellular lipid droplets.  相似文献

The imidazol side group of histidine has two nitrogen atoms capable of being protonated or participating in metal binding. Hence, histidine can take on various metal-bound and protonated forms in proteins. Because of its variable structural state, histidine often functions as a key amino acid residue in enzymatic reactions. Ab initio (HF and MP2) calculations were done in modeling the cation (H+, Li+, Na+, K+, Mg2+, Ca2+) interaction with side chain of histidine. The region selectivity of metal ion complexation is controlled by the affinity of the side of attack. In the imidazol unite of histidine the ring nitrogen has much higher metal ion (as well as proton) affinity. The complexation energies with the model systems decrease in the following order: Mg2+ > Ca2+ > Li+ > Na+ > K+. The variation of the bond lengths and the extent of charge transfer upon complexation correlate well with the computed interaction energies.  相似文献

A new insoluble solid functionalized ligand system bearing chelating ligand group of the general formula P-(CH2)3-N[CH2CONH(C6H4)NH2]2, where P represents [Si–O] n polysiloxane network, was prepared by the reaction of the immobilized diethyliminodiacetate polysiloxane ligand system, P-(CH2)3N(CH2CO2Et)2 with 1,2-diaminobenzene in toluene. 13C CP-MAS NMR, XPS and FTIR results showed that most ethylacetate groups (–COOEt) were converted into the amide groups (–N–C=O). The new functionalized ligand system exhibits high capacity for extraction and removal of the metal ions (Fe3+, Co2+, Ni2+, Cu2+ and Zn2+) with efficiency of 95–97% after recovery from its primary metal complexes. This functionalized ligand system formed 1:1 metal to ligand complexes.  相似文献

The binding of cations Li+, Na+, K+, Cs+, Ag+, Zn2+, Ni2+, Co2+, NH4 + (group I), H+, Mg2+, Al3+, Ga3+ (group II), and Ca2+, Pb2+ (group III) by 21,31-diphenyl-l 2,42-dioxo- 7,10,13-trioxa-l,4(3,1)-diquinoxalina-2(2,3),3(3,2)-diindolizinacyclopentadecaphane (1), which contains two indolizine and two quinoxaline fragments and 3,6,9-trioxaundecanes spacer, and by its acyclic analog (2) was studied using cyclic voltammetry in MeCN/0.1 M Bu4NBF4. It was concluded that the ions of group I are not bound by these compounds, the ions of group II exhibit the reversible redox-switched binding by the carbamoyl groups of the quinoxaline fragments, whereas the ions of group III are bound not only by the initial compounds and radical cations 1 and 2, but also by dication 1. This binding of the Ca2+ and Pb2+ ions stabilizes dication 1.  相似文献

By making modifications to our previously established measurement setup, we increased our abundance sensitivity for 233U by three orders of magnitude and can now measure 233U/238U ratios as low as 10−13. Because 233U has separate production pathways than 236U, it can provide valuable information on the particular source of anthropogenic uranium in a sample. We demonstrated the utility of our improved capability by using 233U to distinguish separate sources of anthropogenic uranium in a set of samples collected from a contaminated site. In the future, we plan to apply our new capability to characterizing 233U in a wide range of uranium materials.  相似文献

Quantification of 241Am in urine at low levels is important for assessment of individuals’ or populations’ accidental, environmental, or terrorism-related internal contamination, but no convenient, precise method has been established to rapidly determine these low levels. Here we report a new analytical method to measure 241Am as developed and validated at the Centers for Disease Control and Prevention (CDC) by means of the selective retention of Am from urine directly on DGA resin, followed by SF-ICP-MS detection. The method provides rapid results with a limit of detection (LOD) of 0.22 pg/L (0.028 Bq/L), which is lower than 1/3 of the C/P CDG for 241Am at 5 days post-exposure. The results obtained by this method closely agree with CDC values as measured by liquid scintillation counting, and with National Institute of Standards Technology (NIST) Certified Reference Materials (CRM) target values.  相似文献

The migration of 237Np in an undisturbed Chinese loess column was investigated by direct γ-ray method. The column was taken from a field test site and installed in a laboratory simulation hall. Radionuclide 237Np in the form of neptunium nitrate, mixed with quartz, was introduced into the column and covered with loess. Artificial rainfall was applied to the column for about 3 years and, the counting rates of 237Np in the column from 56 to 616 days at different vertical positions were detected with a γ-ray detection system. Based on the counting rates of 237Np in the simulation column at different vertical positions and the distance from the source layer, the relationship of the mass center of 237Np in the column at different experimental periods to the experimental time was established, C m = 0.36 log(t)-2.75. Here C m is the mass center of 237Np in the column, cm, and t is the experimental time in days. Based on this relationship, the mass center of 237Np for the 1,073-day experiment was predicted and compared to that obtained with the final destructive method. The good agreement between the prediction and the experimental values indicates that the direct γ-ray method could be used to predict the migration of strongly adsorbed radionuclides such as 237Np in environmental media with the help of laboratory simulation columns.  相似文献

Intra-hepatic administration of radioactive glass microspheres is a treatment for patients with primary liver cancer and hepatic metastases. The purpose of this study was radionuclide purity assessment of new glass particles containing two radionuclide, 90Y as a therapeutic source and also 177Lu as a source of diagnostic gamma. For the mixed source, activity measurement using a dose calibrator cannot be used and we need new calibration methods. YAS (Yb) and YAS compositions were sol–gel derived glass particles and production of 90Y (177Lu) and 90Y particles was performed using the Tehran Research Reactor. The radionuclide purity was carried out using γ-spectrometry with HPGe detector. A non-destructive spectroscopic assay was employed due to a newly updated low uncertainty positron branching ratio of 90Y that emit 511 keV annihilation radiations. In another method, a new calibration of 90Y using a non-destructive spectroscopic assay of 88Y were investigated. Potential radionuclide impurity include: 88Y, 152Eu, 60Co with activity 100, 50 and 5 Bq per 1 mg of that are not harmful for patients due to delivering radioactive particles about 20–50 mg in 90Y(177Lu) glass microspheres. Among of radionuclide impurity, 152Er with a half life of 13.54 years and 88Y with a half life of 106.65 days was important in the residual delivery device. For calibration of 90Y with monitoring of 511 keV, errors were12.2–21%. In calibration of 90Y using gamma spectroscopic assay of 88Y, there was an error less than 14%. Spectroscopic assay of 88Y can be performed easily and has more repeat for our purpose.  相似文献

Summary This paper reports the radiochemical study of the ion-exchange of Cs+, Na+, Sr2+ and Eu3+ ions with H+ by chromium hexacyanoferrate(II) which was prepared in a granular form using a gel method. The slow steps which determine the rate of exchange of these ions are directly proportional to the particle diameter and this is confirmed from the linearity test of Bt vs. t plots at different particle diameters. Boyed’s equation and Reichenberg’s tables were used for evaluating all the kinetic parameters. The results reveal that the effective particle radii are unchanged for both chromium hexacyanoferrate(II) dried at 60 and 120 °C. The obtained data were analyzed using McKay plots and Arrhenius equation and the kinetic and thermodynamic parameters, e.g., effective diffusion coefficient, activation energies and entropies of activation have been evaluated. The mobility of these ions inside the particles of chromium hexacyanoferrate(II) decrease in the order of Eu3+>Sr2+>Na+⊃Cs+.  相似文献

Radiolabeled fatty acids as myocardial metabolic agent are used for detecting ischemic heart disease, however, no 99mTc-labeled fatty acids have potential use in clinical diagnosis. In this work, five fatty acid analogues labeled with 99mTc were firstly synthesized and characterized, their biological behaviors were investigated. All Radiotracers had good stability when incubated in rat serum for 3 h at 37 °C. 99mTc -CpT-12-ODPPA (8b) showed higher initial myocardial uptake (8.17% ID/g at 1 min postinjection) and good heart/blood ratio (2.58 at 30 min postinjection). 99mTc-11-dpa-OUFA (2b) as positively charged lipophilic compounds had reasonable heart uptake (5.59% ID/g at 1 min postinjection) and good retention (1.89% ID/g at 60 min postinjection). Unfortunately, no great improvement was obtained by the five 99mTc-labeled fatty acid analogues for the short myocardial retention and poor heart/liver ratios.  相似文献

Low-lying structures of water cationic clusters and the compounds with the OH radical have become a hot topic in recent years. We here investigate the cluster \( {\left({\mathrm{H}}_2\mathrm{O}\right)}_{10}^{+} \) and calculate its ideal structures by the quantum chemical calculation together with the particle swarm optimization method. We analyzed the properties of the obtained lower-energy isomers of \( {\left({\mathrm{H}}_2\mathrm{O}\right)}_{10}^{+} \). Their energies are further re-optimized and demonstrated at three different methods with two basis sets. Based on our numerical calculations, a new cage-like structure of \( {\left({\mathrm{H}}_2\mathrm{O}\right)}_{10}^{+} \) with the lowest energy is obtained at MP2/aug-cc-pVDZ level. Our results showed the comparison of energy order at different conditions and demonstrated the influence of temperature on the relative Gibbs energy and IR spectra. Moreover, we also contained the molecule orbitals to discuss the stability of these representative isomers.  相似文献

The production cross-section of the medical isotope, 99Mo from the enriched 100Mo(n,2n) reaction with the average neutron energies of 21.9 and 26.5 MeV has been determined for the first time by using an off-line γ-ray spectrometric technique. The average neutron energies were generated by using the 9Be(p,n) reaction with the proton energies of 35 and 45 MeV from the MC50 cyclotron of the Korea Institute of Radiological and Medical Sciences (KIRAMS) at Seoul, South Korea. The 100Mo(n,2n) reaction cross-section as a function of neutron energy was also calculated theoretically by using the computer code TALYS-1.8 and EMPIRE-3.2 Malta. The experimental results are in close agreement with the theoretical values from TALYS-1.8. However, the present data at the neutron energy of 21.9 MeV is slightly lower and at 26.5 MeV is higher than the values from EMPIRE-3.2 Malta.  相似文献

The dehydrogenation reaction mechanism of cyclohexane catalyzed by dimer transition metal cluster V2+ has been investigated at the B3LYP/6-31G (d, p) level of density functional theory. Density of states (DOS) graph is used to understand more deeply the roles of the front molecular orbital of the initial complexes. After the first molecular dehydrogenation, the reaction mainly consists of two competition mechanisms. First, the C-H bonds of cyclohexane can be effectively activated by the V2+ cation, yielding the same-face dehydrogenation products. Second, the C-C bonds are activated, forming the different-face dehydrogenation products. Our calculations indicate that the reaction takes place more easily along the low-spin potential energy surface on the same-face and is a low-barrier or even barrier-free transformation. Carbon-carbon single bonds are nonpolar and generally far less reactive. A comparison of the reaction mechanism of V2+ and congener Ti2+ with cyclohexane has been presented. The bond dissociation energies (BDEs) of V2+ are greater than that of Ti2+, leading to difficulties in forming sandwich complexes in the different-face dehydrogenation of cyclohexane, and the same-face dehydrogenation is an important reaction channel.  相似文献

The purpose of this work was to indirect label IgG with fac-[188Re(CO)3(H2O)3]+ and to check the radiochemical behavior of the labeled product. The compound of (bis(2-pyridylmethyl)-amino)-acetic acid (L2H) was synthesized and labeled with fac-[188Re(CO)3(H2O)3]+. The labeling yield of 188Re(CO)3–L2H was more than 90%. The effects of protein concentration, reaction time, pH and reaction temperature of labeling of IgG with 188Re(CO)3–L2H were investigated. The conjugation conditions were optimized. The labeled product was analyzed by size exclusion HPLC and TLC. The stability of 188Re(CO)3–L2H–IgG in vitro was high. The results of this study may be useful for [188Re(CO)3(H2O)3]+ labeling of protein for radioimmunotherapy.  相似文献

Two new ruthenium(II) polypyridyl complexes (1) (dtmi = 3-(pyrazin-2-yl)-as-triazino[5,6-f]-5-methoxylisatin) and (2) (dtni = 3-(pyrazin-2-yl)-as-triazino[5,6-f]-5-nitroisatin) have been synthesized and characterized by elemental analysis, FAB-MS, ES-MS and 1H-n.m.r. The DNA-binding patterns of complexes were investigated by spectroscopic titration, viscosity measurements and thermal denaturation. The results indicate that the complexes (1) and (2) interact with calf thymus DNA (CT-DNA) by intercalative mode. Due to the withdrawing electronic substitutent in the intercalative ligand, ptni, the DNA-binding affinity of the complexes (2) is larger than that complex (1) does.  相似文献

The influence of Ca2+-, Cu2+- and Fe3+-exchanged montmorillonite (MMT) on the type of interaction with aniline in the interlayer space of MMT has been studied by means of X-ray powder diffraction and infrared spectra. Results of X-ray diffraction showed that aniline was successfully intercalated into the interlayer space of MMT. Based on IR spectra evaluation, aniline was indirectly coordinated through a water-bridge in Ca2+- and Fe3+-MMT and it was indirectly coordinated through a water-bridge as well as protonated in Cu2+-MMT (the spectrum of protonated aniline showed deformation and changes in the NH 3 + absorption at approximately 1521 cm?1). It is important to point out that Cu2+-MMT indirect coordination and protonation occur simultaneously.  相似文献

Cationic metal ion-coordinated N-diisopropyloxyphosphoryl dipeptides (DIPP-dipeptides) were analyzed by electrospray ionization multistage tandem mass spectrometry (ESI-MS n ). Two novel rearrangement reactions with hydroxyl oxygen or carbonyl oxygen migrations were observed in ESI-MS/MS of the metallic adducts of DIPP-dipeptides, but not for the corresponding protonated DIPP-dipeptides. The possible oxygen migration mechanisms were elucidated through a combination of MS/MS experiments, isotope (18O, 15N, and 2H) labeling, accurate mass measurements, and density functional theory (DFT) calculations at the B3LYP/6-31 G(d) level. It was found that lithium and sodium cations catalyze the carbonyl oxygen migration more efficiently than does potassium and participation through a cyclic phosphoryl intermediate. In addition, dipeptides having a C-terminal hydroxyl or aromatic amino acid residue show a more favorable rearrangement through carbonyl oxygen migration, which may be due to metal cation stabilization by the donation of lone pair of the hydroxyl oxygen or aromatic π-electrons of the C-terminal amino acid residue, respectively. It was further shown that the metal ions, namely lithium, sodium, and potassium cations, could play a novel directing role for the migration of hydroxyl or carbonyl oxygen in the gas phase. This discovery suggests that interactions between phosphorylated biomolecules and proteins might involve the assistance of metal ions to coordinate the phosphoryl oxygen and protein side chains to achieve molecular recognition.  相似文献
