The separation of hydroxyaromatic compounds in vegetable oils, including synthetic antioxidants (3-tert-butyl-4-hydroxyanisol and 2,6-di-tert-butyl-4-hydroxytoluene), E-vitamers and other natural oil components, by nonaqueous capillary electrophoresis in an oil-miscible background electrolyte (BGE) was investigated. The BGE contained 40 mM KOH in a methanol/1-propanol (PrOH) mixture (15:85 v/v). The oil samples were 1:1 diluted with PrOH and directly injected in the capillary. Under negative polarity (cathode at the injection end), the anionic solutes moved faster than the electroosmotic flow, being well-resolved among them and from the triacylglycerols. Using virgin palm, extra virgin olive, wheat germ, virgin soybean and other oils, the capability of the procedure to quickly yield a characteristic profile of the biophenols present in the sample was demonstrated. The alpha-, (beta + gamma)- (as unresolved pair) and delta-tocopherols of a soybean oil sample were quantified.  相似文献

A nonaqueous capillary electrophoresis (NACE) method, 30 mM ammonium acetate in methanol as background electrolyte (BGE), was developed for separation of the organomercury species without complexing reagents. The effects of different solutes and solvents in BGE were studied. Three species of organomercury, methylmercury, ethylmercury and phenylmercury, were separated well and all the number of theoretical plates were over 106. The present NACE method was also coupled with sample stacking and electrokinetic injection techniques to enhance the detection sensitivity. Under the optimum conditions, the limit of detection (S/N = 3) is 18 ng mL−1 and the linear relation range from 40 to 750 ng mL−1 were obtained for methylmercury.  相似文献

In this article, optimization of BGE for simultaneous separation of inorganic ions, organic acids, and glutathione using dual C4D‐LIF detection in capillary electrophoresis is presented. The optimized BGE consisted of 30 mM 2‐[4‐(2‐hydroxyethyl)piperazin‐1‐yl]ethanesulfonic acid, 15 mM 2‐amino‐2‐hydroxymethyl‐propane‐1,3‐diol, and 2 mM 18‐crown‐6 at pH 7.2 and allowed simultaneous separation of ten inorganic anions and cations, three organic acids and glutathione in 20 min. The samples were injected hydrodynamically from both capillary ends using the double‐opposite end injection principle. Sensitive detection of anions, cations, and organic acids with micromolar LODs using C4D and simultaneously glutathione with nanomolar LODs using LIF was achieved in a single run. The developed BGE may be useful in analyses of biological samples containing analytes with differing concentrations of several orders of magnitude that is not possible with single detection mode.  相似文献

Capillary ion electrophoresis–capacitively coupled contactless conductivity detection (CIE-C4D) with a polyvinyl alcohol chemically coated capillary (PVA capillary) was used to analyze inorganic cations (Na+, K+, NH4+, Mg2+, and Ca2+) commonly found in human saliva. The PVA capillary, which was made by our laboratory, minimized electro-osmotic flow in the wide pH range of the background electrolyte (BGE), and the PVA layer adsorbed to capillary wall did not affect the conductimetric background level. In this study, we determined an optimized BGE of 30 mM lactic acid/histidine plus 3 mM 18-crown-6 for the CIE-C4D system using the PVA capillary, which could simultaneously improve the separation of Mg2+ and Ca2+ from Na+ and that of K+ from NH4+. This system obtained highly reproducible separation of cations in human saliva samples within 8 min at 20 kV without deprotonation. The quantifiability of cations in human saliva samples on the CIE-C4D system was demonstrated through identification by ion chromatography with satisfactory results.  相似文献

In-line solid-phase extraction–capillary electrophoresis coupled with mass spectrometric detection (SPE–CE–MS) has been used for determination of 2-ethylidene-1,5-dimethyl-3,3-diphenylpyrrolidine (EDDP), codeine (COD), hydrocodeine (HCOD), and 6-acetylmorphine (6AM) in urine. The preconcentration system consists of a small capillary filled with Oasis HLB sorbent and inserted into the inlet section of the electrophoresis capillary. The SPE–CE–MS experimental conditions were optimized as follows: the sample (adjusted to pH 6.0) was loaded at 930 mbar for 60 min, elution was performed with methanol at 50 mbar for 35 s, 60 mmol L−1 ammonium acetate at pH 3.8 was used as running buffer, the separation voltage was 30 kV, and the sheath liquid at a flow rate of 5.0 μL min−1 was isopropanol–water 50:50 (v/v) containing 0.5% acetic acid. Analysis of urine samples spiked with the four drugs and diluted 1:1 (v/v) was studied in the linear range 0.08–10 ng mL−1. Detection limits (LODs) (S/N = 3) were between 0.013 and 0.210 ng mL−1. Repeatability (expressed as relative standard deviation) was below 7.2%. The method developed enables simple and effective determination of these drugs of abuse in urine samples at the levels encountered in toxicology and doping.  相似文献

Organic acids in cerebrospinal fluid (CSF) are potential diagnostic markers for neurological diseases and metabolic disorders. A capillary electrophoretic (CE) method for the direct analysis, i.e., without any sample preparation, of six organic acids in CSF was developed. A capillary coating consisting of a triple layer of charged polymers (polybrene-dextran sulfate-polybrene) was used in combination with a negative separation voltage, providing fast and efficient analysis of acidic compounds. Separation conditions, such as background electrolyte (BGE) concentration and pH were optimized, and the influence of albumin and sodium chloride was systematically studied using a set of test compounds. With injection volumes of ca. 44 nL, plate numbers of up to ca. 150,000 were obtained with a BGE of 200 mM sodium phosphate (pH 6.0). It appeared that high sodium chloride concentrations in the sample hardly affected the peak width and shape of the organic acids, most probably due to transient isotachophoresis effects occurring in the sample zone. Adverse effects of CSF proteins, which frequently compromise the CE performance, could be effectively minimized by the triple layer coating in combination with rinses of 0.1 M hydrochloric acid. Overall, the developed CE system allowed direct injections of CSF samples, yielding good separation efficiencies and stable migration times (RSDs < 2%) for organic acids. Validation of the method with artificial and real CSF samples showed good linear responses (r > 0.99), and LODs for the organic acids were in the range of 2–8 μg/mL when applying UV detection. RSDs for migration times and peak areas were <2% and <7%, respectively. The applicability of the CE system is shown for the determination of organic acids in CSF samples.  相似文献

Summary The potential of a non-aqueous, capillary electrophoresis (NACE) system for separating oxytetracycline from three of its impurities—tetracycline, 4-epioxytetracycline and 4-epitetracycline—using UV detection has been studied. The running buffer was: 25 mM sodium acetate, 1 mM EDTA, methanesulfonic acid, pH 4, dissolved in MeOH-ACN (50∶50,v/v). The method was also used to determine these compounds in pig plasma. A solid-phase extraction (SPE) procedure as a clean-up step has also developed. For this we tested Sep Pak C18, LiChrolut EN and OASIS cartridges. OASIS cartridges were best. Recoveries were 90–100% for all compounds except EOTC which had a recovery of 74%.  相似文献

Summary A fluorimetric-detection RP-HPLC method is developed for the separation and quantification of bisphenol A diglycidyl ether (BADGE), bisphenol F diglycidyl ether (BFDGE), its hydrolysis and chlorohydroxy derivatives, and NOGE and BADGE oligomers. This method was applied to test an epoxy resin used as a coating for food contact packaging materials with two different heat treatments in the curing process. Migration of these compounds was evaluated in three food simulants: 3% (w/v) acetic acid, 10% (v/v) ethanol, rectified olive oil and also in acetonitrile. A simple extraction procedure was also applied to obtain olive oil extracts. Quantification was performed by using external calibration, and correlation coefficients were greater than 0,996 and the limit of detection (LOD) was 0.02 mg L–1 for all substances. This contribution describes use of a separation method based on on-line coupling of a multisyringe flow system with a chromatographic monolithic column for simultaneous determination of hydrochlorothiazide and losartan potassium in tablets. The system comprised a multisyringe module, three low-pressure solenoid valves, a monolithic C18 column (25 mm × 4.6 mm i.d.), and a diode-array detector. The mobile phase was 10 mmol L−1 potassium dihydrogen phosphate (pH 3.1)-acetonitrile-methanol (65:33:2 v/v/v) at a flow rate 0.8 mL min−1. UV detection was carried out at 226 nm. The multi-syringe chromatographic (MSC) method with UV spectrophotometric detection was optimized and validated. Results from validation were very good. The analysis time was about 400 s. The method was found to be applicable to routine analysis of both compounds in tablets. The coupling of the monolithic columns with a multi-syringe flow-injection analysis manifold provides an excellent and inexpensive tool to solve the separation problems without use of HPLC instrumentation.  相似文献

In this work, a capillary electrophoresis (CE) method for the determination of a group of eleven triazine compounds by micellar electrokinetic capillary chromatography (MEKC) with diode array detection was developed. The eleven herbicides studied were: desethylatrazin-2-hydroxy (DEA), simazine, prometon, atrazine, simetryn, ametryn, propazine, prometryn, trietazine, terbutylazine, and terbutryn The separation of these compounds was optimised as a function of buffer concentration and pH, concentration of sodium dodecyl sulphate (SDS) and voltage applied. To increase the selectivity of the separation and the resolution of the solutes, different organic solvents were tested as buffer additives, obtaining the best results when 1-propanol was used. The optimised buffer (24 mM of sodium borate, 18 mM of disodium hydrogen phosphate, 25 mM of SDS, pH 9.5, and 5% of 1-propanol) provides the best separation in terms of resolution and migration time. This method allowed the determination of these compounds at concentrations of 0.05 μg l−1 in ground water samples pretreated using solid-phase extraction (SPE).  相似文献

Summary The chiral separation of two newly synthesized arylpropionic acids of pharmaceutical interest, namely 2-[(5′-benzoil-2′-hydroxy)phenyl]-propionic acid (DF-1738y) and 2-[(4′-benzoiloxy-2′-hydroxy)phenyl]-propionic acid (DF-1770y), was performed by Capillary Zone Electrophoresis (CZE) using either cyclodextrins or antibiotics as chiral selectors in coated capillary. In order to optimize the separation, the effect on the migration time and resolution of type and concentration of the chiral selector, the buffer pH and the capillary temperature were studied. Several cyclodextrins, namely the charged 6A-monomethylamino-β-cyclodextrin (MeNH-β-CD) and the neutral methyl-β-cyclodextrins (M-β-CD) and heptakis-2,3,6-tri-O-methyl-β-cyclodextrin (TM-β-CD), were tested for the enantiomeric separation of aryl propionic acids (APAs) compounds. Of these TM-β-CD provided the highest enantiomeric resolution at pH 5, however only DF-1738y optical isomers were baseline resolved. Using background electrolytes (BGEs) at higher pHs (pH=6–7) supported with the above listed CDs, an enantioresolution increase was recognized only for compound DF-1738y. In contrast DF-1770y exhibited the highest resolution at the lowest pH value studied (pH 4). The macrocyclic antibiotic vancomycin was therefore added to the BGE and tested as chiral selector using the partial filling counter current mode in order to obtain a sensitive analysis, high resolution and reduced antibiotic adsorption on the capillary wall. 5 mM vancomycin dissolved in the BGE at pH 5 and 25°C provided relatively high enantiomeric resolution (R DF-1738y=3.4,R DF-1770y=2.22) of both compounds.  相似文献

Summary A capillary zone electrophoretic method for the analysis of phenolic acids in soil and plant extracts was developed with direct UV detection using a phosphate electrolyte solution. The electrophoretic separation required the phenolic acids to be charged at a pH above their pKa in order to achieve their migration towards the anode. Electroosmotic flow (EOF) was reversed in direction by adding tetradecyltrimethylammonium bromide (TTAB). Factors affecting the separation selectivity, including the buffer pH and EOF modifiers, were investigated systematically. Eight phenolic acids were separated and detected in 10 min using an electrolyte containing 25 mM phosphate, 0.5 mM TTAB and 15% acetonitrile (v/v) at pH of 7.20. Linear plots for the test phenolic acids were obtained in a concentration range of 0.01–1 mM with detection limits in the range of 1.0–7.0 μM. The recoveries ranged from 92.8 to 102.3% in soil and plant tissues samples spiked at 100 μM and the relative standard deviation based on the peak area were ranged 2.0 to 4.5%. The proposed method was used for the determination of phenolic acids in plant tissue and soil extracts with direct injection.  相似文献

Summary MECC separation of 11 coumarins has been achieved by use of a running electrolyte at pH 10.4 prepared from 50 mM boric acid, 10 mM sodium tetraborate and 100 mM sodium hydroxide. The buffer solution contained 50 mM SDS and, as organic modifier 1%n-propanol. The applied voltage was 25 kV and the temperature of the capillary was kept constant at 20°C. HPLC baseline separation of the coumarin mixture was obtained by use of a reversed-phase column and an acetonitrile-water solvent gradient. UV detection was performed at 205 nm. Peak assignment and purity control were achieved by HPLC-mass spectrometry with either an electrospray interface or an atmospheric-pressure chemical-ionization interface. Compounds were detected in either negative- or positive-ion modes. These MECC and HPLC-MS methods are suitable for ‘fingerprint’ analysis of a number of coumarin-containing plants, e.g. Fr.Ammi visnagae, Rd.Scopoliae and Rd.Imperatoriae.  相似文献

A simple, rapid method using CE and microchip electrophoresis with C4D has been developed for the separation of four nonsteroidal anti-inflammatory drugs (NSAIDs) in the environmental sample. The investigated compounds were ibuprofen (IB), ketoprofen (KET), acetylsalicylic acid (ASA), and diclofenac sodium (DIC). In the present study, we applied for the first time microchip electrophoresis with C4D detection to the separation and detection of ASA, IB, DIC, and KET in the wastewater matrix. Under optimum conditions, the four NSAIDs compounds could be well separated in less than 1 min in a BGE composed of 20 mM His/15 mM Tris, pH 8.6, 2 mM hydroxypropyl-beta-cyclodextrin, and 10% methanol (v/v) at a separation voltage of 1000–1200 V. The proposed method showed excellent repeatability, good sensitivity (LODs ranging between 0.156 and 0.6 mg/L), low cost, high sample throughputs, portable instrumentation for mobile deployment, and extremely lower reagent and sample consumption. The developed method was applied to the analysis of pharmaceuticals in wastewater samples with satisfactory recoveries ranging from 62.5% to 118%.  相似文献

A simple, rapid, and sensitive non-aqueous capillary electrophoresis procedure for the quantitative determination of matrine and oxymatrine is established. Optimum separation conditions were obtained when the sample was injected under pressure for 3 s at 50 mbar and separated with the buffer containing 70 mM ammonium acetate, 7.0% (v/v) acetic acid, and 10% (v/v) acetonitrile in methanol medium at 25 kV applied voltage. The analytes were detected at 205 nm. The two alkaloids can be separated within 12 min and quantified with high sensitivity. The method was validated in terms of reproducibility, linearity, and accuracy when applied to the analysis of matrine and oxymatrine in Sophora flavescens and its medicinal preparations.  相似文献

A new hydrophilic interaction liquid chromatographic (HILIC) method for the simultaneous determination of isoascorbic (IAA) and ascorbic acid (AA) was developed. The separation of IAA and AA was studied in various HILIC stationary phases and the influence of the composition of the mobile phase, the ionic strength and the column temperature to the chromatographic characteristics is presented. The final method used an aminopropyl column under isocratic elution with acetonitrile–100 mM ammonium acetate solution (90:10, v/v) at a flow rate of 0.4 mL/min and a detection wavelength of 240 nm. This method was validated and the calibration curves were found to be linear in the range of 1.0–65 μg/mL for both IAA and AA. The method limit of detection for IAA determination in fish tissue was 2.3 μg/g. Inter-day precision (as %RSDR) was ranged between 0.56% and 8.3% at three concentration levels, whereas the respected recoveries ranged between 82% and 98%. This method was applied to the determination of IAA (as additive E315) in frozen redfish samples. The hyphenation of the HILIC separation with a tandem mass spectrometer was also studied and the problems encountered with negative electrospray ionization under HILIC separation conditions are discussed.  相似文献

Summary Capillary electrophoresis (CE) has been successfully applied to the separation of the enantiomers of venlafaxine (Vx) and its main active metabolite,O-desmethylvenlafaxine (ODV), by use of a mixture of two cyclodextrins (CDs) added to the background electrolyte (BGE). The use of carboxymethylated β-cyclodextrin (CMB) enabled the enantioseparation of both Vx and ODV, although separation of all four enantiomers was not achieved. Addition of a second cyclodextrin (α-CD) to the BGE resulted in the simultaneous resolution of the enantiomers of both compounds. In this system, α-CD enabled discrimination between Vx and ODV whereas CMB enabled the enantioseparation. Resolution was strongly influenced by the ratio of the concentrations of the two complexing agents. Baseline resolution of Vx and ODV was achieved in an uncoated capillary. Addition of organic modifiers to the BGE had a substantial effect on the interaction of the analytes with the α-CD.  相似文献

A practical chiral CE method, using sulfated‐β‐CD as chiral selector, was developed for the enantioseparation of glycopyrrolate containing two chiral centers. Several parameters affecting the separation were studied, including the nature and concentration of the chiral selectors, BGE pH, buffer type and concentration, separation voltage, and temperature. The separation was carried out in an uncoated fused‐silica capillary of (effective length 40 cm) × 50 μm id with a separation voltage of 20 kV using 30 mM sodium phosphate buffer (pH 7.0, adjusted with 1 M sodium hydroxide) containing 2.0% w/v sulfated‐β‐CD at 25°C. Finally, the method for determining the enantiomeric impurities of RS‐glycopyrrolate was proposed. The method was further validated with respect to its specificity, linearity range, accuracy and precision, LODs, and quantification in the expected range of occurrence for the isomeric impurities (0.1%).  相似文献

A nonaqueous micellar electrokinetic capillary chromatography method with indirect LIF was developed for the determination of strobilurin fungicide residues in fruits and vegetables. Hydrophobic CdTe quantum dots (QDs) synthesized in aqueous phase were used as background fluorescent substance. The BGE solution, QD concentration, and separation voltage were optimized to obtain the best separation efficiency and the highest signal intensity. The optimal BGE solution consists of 40 mM phosphate, 120 mM sodium dodecyl sulfate, 15% v/v water and 15% v/v hydrophobic CdTe QDs in formamide, of which apparent pH is 9.5. The optimized separation voltage is controlled as 25 kV. The resultant detection limits of azoxystrobin, kresoxim‐methyl, and pyraclostrobin are all 0.001 mg/kg, their linear dynamic ranges are 0.005–2.5 mg/kg, and the recoveries of the spiked samples are 81.7–96.1%, 86.5–95.7%, and 87.3–97.4%, respectively. This method has been proved to be sensitive enough to detect the aforementioned fungicides in fruits and vegetables at the maximum residue limits.  相似文献

A fast and accurate micellar electrokinetic capillary chromatography (MEKC) method was developed for monitoring N-methyl-2-pyrrolidone (NMP) exposure. Baseline separation of NMP and its main metabolites: 5-hydroxy-N-methyl-2-pyrrolidone (5HNMP), N-methylsuccinimide (MSI), 2-hydroxy-N-methylsuccinimide (2HMSI), and 2-pyrrolidone (2P) was obtained within 6 min in an uncoated fused silica capillary using 5 mM phosphate buffer and 140 mM sodium dodecyl sulfate (pH 7.1) as background electrolyte (BGE). On-line UV-detection was performed at 200 nm and the applied electric field was 400 V cm−1. Possible interference of BGE-induced system peaks on separation was investigated by computer simulation and no such interference was observed. The developed MEKC method combined with solid phase extraction for sample preparation was successfully applied to the analysis of urine of rats exposed to NMP. The urinary excretion was determined in 0–6 h and 6–24 h specimens collected after an intragastic administration of 308 mg NMP / kg rat body weight. The results of NMP disposition kinetics in rat urine are reported for NMP and metabolites.   相似文献
