To study the photoreactions between thymidine and psoralen and to devise a synthetic route to prepare thymidine-5-methoxypsoralen adducts, we have synthesized a compound in which thymidine is linked to a 5-methoxypsoralen derivative by a short flexible chain located to allow only the formation of syn intramolecular [2+2] cycloadducts. Irradiation at 365 nm carried out at usual concentrations (10?2-10?3M) resulted in a regioselective dimerization involving the 3,4 double bonds. Extreme dilution (c 5 × 106M) was necessary to observe the intramolecular photoaddition, which produces exclusively the 3,4-trans-syn adduct. These results are compared to those obtained previously with similar heterodimers that lead exclusively to the 3,4-cis-anti thymine or thymidine-psoralen adduct on exposure to near UV light.  相似文献

UVA‐activated psoralens are used to treat hyperproliferative skin conditions due to their ability to form DNA photoadducts, which impair cellular processes and may lead to cell death. Although UVA (320–400 nm) is more commonly used clinically, studies have shown that UVB (280–320 nm) activation of psoralen can also be effective. However, there has been no characterization of UVB‐induced adduct formation in DNA alone. As psoralen derivatives have a greater extinction coefficient in the UVB region (11 800 cm?1 M?1 at 300 nm) compared with the UVA region (2016 cm?1 M?1 at 365 nm), a greater extent of adduct formation is expected. SELDI‐TOF, a proteomic technique that combines chromatography with mass spectrometry, was used to detect photoadduct formation in an alternating A–T oligonucleotide. 8‐Methoxypsoralen (8‐MOP) and DNA solutions were irradiated with either UVA or UVB. An adduct peak was obtained with SELDI‐TOF. For UVB‐activated 8‐MOP, the extent of adducts was three times greater than for UVA. HPLC ESI‐MS analysis showed that UVB irradiation yielded high levels of 3,4‐monoadducts (78% of total adducts). UVA was more effective than UVB at conversion of 4′,5′‐monoadducts to crosslinks (17% vs 4%, respectively). This report presents a method for comparing DNA binding efficiencies of interstrand crosslink inducing agents.  相似文献

The reaction of 3-(2-oxocycloalkylidene)indol-2-one 1 with thiourea and urea derivatives has been investigated. Reaction of 1 with thiourea and urea in ethanolic potassium hydroxide media leads to the formation of spiro-2-indolinones 2a-f in 40–50% yield and a novel tetracyclic ring system 4,5-cycloalkyl-1,3-diazepino-[4,5-b]indole-2-thione/one 3a-f in 30–35% yield. 3-(2-Oxocyclopentylidene)indol-2-one afforded 5′,6′-cyclopenta-2′-thioxo/ oxospiro[3H-indole-3,4′(3′H)pyrimidin]-2(1H)-ones 2a,b and 3-(2-oxocyclohexylidene)indol-2-one gave 2′,4′a,5′,6′,7′,8′- hexahydro-2′-thioxo/oxospiro[3H-indole-3,4′ (3′H)-quinazolin]-2(1H)-ones 2c-f . Under exactly similar conditions, reaction of 1 with fluorinated phenylthiourea/cyclohexylthiourea/phenylurea gave exclusively spiro products 2g-1 in 60–75% yield. The products have been characterized by elemental analyses, ir pmr. 19F nmr and mass spectral studies.  相似文献

Abstract. The UVA-mediated photoreaction of the monofunctional 7-methylpyrido[3,4-c]psoralen (MePyPs) with 2′-deoxy-adenosine was investigated in the dry state. Two main 7-methylpyrido[3,4-c]psoralen monoadducts to 2′-deoxy-adenosine were isolated by HPLC and characterized by soft ionization mass spectrometry (fast atom bombardment) and extensive 1H NMR analyses including correlation spectroscopy, total correlation spectroscopy and nuclear Overhauser effect measurements. The two unusual photoadducts exhibit a covalent bond between the C-4′ of the furan moiety of the psoralen on one hand and either the C-l' or the C-4′ of the furanose ring of the purine nucleoside on the other hand. The furan-side monoadducts appear to be specific for both 2′-deoxy-adenosine and MePyPs. They are likely to result from the recombination of the 4′,5′-dihydrofuran-4′-yl radical of MePyPs with 2′-deoxyadenosyl carbon-centered sugar radicals at either the 1′ or the 5′ position.  相似文献

Bromination of dithieno[3,4-b:3′,4′-d]pyridine ( 1 ) and dithieno[2,3-b:3′,2′-d]pyridine ( 2 ) has been studied. Disubstitution occurred at both positions of the C ring. The substitution pattern is found to be similar to that of the nitration reaction. The structures of bromo derivatives were established by 1H and 13C nmr spectroscopy.  相似文献

The synthesis and selected reactions of the versatile heterocycle 2-amino-3-cyanomethylsulfonyl-4,5-dimethylfuran is reported. In particular, cyclization reaction of the aminofuran yielded a number of novel furo[3,2-b]thiazine 1,1-dioxides. Additionally, a novel tetracyclic system, namely a pyrrolo[2′,3′:5,6]-[1,4]thiazino[3,2-b]quinoline 4,4-dioxide, is prepared via an intramolecular triple-cyclization in which the furan ring is opened and reclosed as a pyrrole.  相似文献

Irradiation at 365 nm carried out at usual concentrations (10–3 M ) leads exclusively to one diastereoisomer of an intramolecular 3,4 cis-anti [2+2] adduct. No photodimerization is observed in these conditions. Aminocyanopyrazole derivatives and pyrazolo[2,3-a]quinazolones were obtained in good yields from hydrazidoyl halides and malononitrile. Pyrazolo[3,4-d]pyridazine and pyridazo[4′,5′: 1,2]pyrazolo[1,5-a]quinazoline derivatives were synthesized in quantitative yields by reaction of hydrazine hydrate with 2 and 16 , respectively. A novel ring system, a 3-substituted tetrahydro derivative of 7-oxo-6H,8H-pyridazo[3′,4′,5′-c'd']-pyrazolo[3,4-d]pyrimidine was prepared by reaction of 6 with dimethyl carbonate. Pyrazolo[3,4-d]pyrimidine-4,6-dithiones were obtained in good yields by reaction of 2 with carbon disulfide. The structures of the products were assigned and confirmed on the basis of their elemental analyses, spectral data, and alternate synthesis wherever possible. The structures of the parent fused heterocyclic systems discussed in this work are summarized in Scheme 1 .  相似文献

A series of 7,12-dihydropyrimido[1′2′:1,2]pyrido[3,4-b]mdole-4(6H)-ones was prepared by Fischer indolization of 9-arylhydrazono-6,7,8,9-tetrahydro-4H-pyrido[1,2-a]pyrirmdin-4-ones. Quantum chemical calculations (ab initio and AM1) indicate that position 3 of 7,12-dihydropyrimido[1′,2′:1,2]pyrido-[3,4-b]indole-4(6H)-one can be involved in electrophilic substitutions, while position 2 is sensitive towards nucleophilic attack. Bromination of 6-methyl-7,12-tetrahydropyrimido[1′,2′:1,2]pyrido-[3,4-b]indol-4(6H)-one 16 with bromine afforded 3-bromo derivative 25 , which was reacted with cyclic amines to give 2-ammo-7,12-dihydropyrirmdo[1′2′:1,2]pyrido[3,4-b]indol-4(6H)-ones 26–30 in an addition-elimination reaction. Vielsmeier-Haack formylation of compound 16 gave 12-formyl 31 and 3,12-diformyl 32 derivatives (an N-formyl-1-deaza derivative of nauclefidine alkaloid 34 ) at 60° and 100°, respectively. 3,12-Diformyl compound 32 was oxidized to 3-carboxyl derivative 33 with potassium permanganate. The quaternary salt 35 , obtained from compound 16 with dimethyl sulfate, suffered a ring opening on the action of aqueous sodium hydroxide. The new compounds have been characterized by elemental analyses uv, 1H nmr and in some cases by 13C ruler, CD spectra and X-ray investigations.  相似文献

The polycyclic heterocyclic compound with a novel ring system, 15-chloro[1]benzothieno[2″,3″:3′,4′]-naphtho[1′,2′:4,5]thieno[2,3-c]quinoline was synthesized via photocyclization of 3-chloro-N-phenyl[1]-benzothieno[2′,3′:3,4]naphtho[2,1-b]fhiophene-2-carboxamide followed by chlorination with phosphorus oxychloride. Treatment of easily prepared l-substituted-4-benzylidene-2,3-dioxopyrrolidines with ammonium formate produces 1,2,4,6,7,8-hexahydro-2,6-disubstituted-8-aryldipyrrolo [3,4-b:3′,4′-e]pyridine-3,5-diones (II), usually in yields of 50 to 60%. Aromatization of the dihydropyridine ring of the hexahydroderivatives II yields corresponding 1,2,6,7-tetrahydro-2,6-disubstituted-8-aryldipyrrolo[3,4-b:3′,4′-e]pyridine-3,5-diones (III). These compounds appear to be the first to incorporate the dipyrrolo[3,4-b:3′,4′-e]pyridine ring system.  相似文献

The involvement of the 5′-hydroxyl group on β-D-arabinofurano[1′,2′:4,5]oxazolo-s-triazin-4-one-6-thione ( 1b ), to form an intramolecular covalent adduct at C6, is postulated to explain the formation of almost equal amounts of 5-N-alkyl-β-D-arabinofurano[1′,2′:4,5]oxazolo-s-triazin-4-one-6-thione and 5-N-alkyl-β-D-arabinofurano[1′,2′:4,5]oxazolo-s-triazine-4,6-dione during alkylation of 1b . An X-ray crystallographic study was conducted on 5-N-methyl-β-D-arabinofurano[1′,2′:4,5]oxazolo-s-triazine-4,6-dione ( 2a ) and its solid state structure was established. This was compared to the energy minimized structure of the same compound that was generated by the molecular modeling program, MACROMODEL. Force field calculations (Allinger's MM2) on this structure and other intermediates lend support to the concept of formation of the intramolecular covalent adduct.  相似文献

As a continuation of recent study on the synthesis of a bis[1,4]oxathiinodipyridine ring system, we would now like to report the preparation of 7-chlorobenzo[1″,2″:5,6:3″,4″:5′,6′]-bis[1,4]oxathiino[3,2-b: 3′,2′-b]dipyridine. Although a potentially complex reaction with several products possible, the title compound was formed exclusively, suggesting considerable mechanistic selectivity. The characterization of the product by FT-1H-nmr as well as its mass spectral fragmentation pathways are also reported.  相似文献

Abstract— In order to investigate the interactions and the photoreactions in solution between the thymine (thy) and the psoralen (Pso) rings, we have prepared model compounds Thy-(CH2)n-Pso in which two aromatic chromophores Thy and Pso are linked by flexible polymethylene chains of varying length (CH2)n. Two series of compounds were examined and compared as models for the two important drugs 5-methoxypsoralen and 8-methoxypsoralen. Results concerning the 5-alkoxypsoralen series are reported here. In water, these model molecules exhibit intramolecular ring-ring stacking interactions as indicated by hypochromism in the UV and by shielding of the protons in 1H NMR spectroscopy. These interactions disappear in organic solvents. The photochemical properties of the models were examined in relation with their ground state interaction properties. Irradiation at 365 nm carried out at the usual concentrations (10-2-10-3 M) leads exclusively to a stereoselective dimerization involving the psoralen moieties of the models at the 3,4 double bonds. However, when operating at exceedingly low concentrations (2 × 10-5 M ), the psoralen photodimerization is avoided and a highly regio and stereo-selective psoralen thymine photoaddition is observed involving the 3,4 double bond of psoralen leading to the cis adduct. The same reaction occurs for all models under study being independent of the length of the (CH2)n polymethylene linking chain, n = 2 to 6, 12 and of the solvent used. This is unambiguous proof for the highest intrinsic photoreactivity of the 3,4 vs the 4',5' double bond in 5-alkoxy psoralen.  相似文献

A new series of pyrazolopyrazinoselenolotriazolopyrimidines was synthesized by a facile method based on condensation of 5-amino-3-methyl-1-phenyl-1H-pyrazolo[3,4-b]selenolo[3,2-e]pyrazine-6-carbonitrile ( 3 ) with triethyl orthoformate followed by intramolecular cyclization with hydrazine to afford 7-amino-8-imino-3-methyl-1-phenyl-1,8-dihydro-7H-pyrazolo[3″,4″:5′,6′]pyrazino[2′,3′:4,5] selenolo[3,2-d]pyrimidine ( 5 ). The latter compound was utilized as a multipurpose precursor for the construction of other new triazoles fused to the pyrazolopyrazino- selenolopyrimidine moiety. Alternatively, acetylation and chloro-acetylation of compound 3 using acetic anhydride and chloroacetyl chloride yielded the acetyl amino 11 and chloroacetamido 12 derivatives, respectively. Compound 12 underwent nucleophilic substitution upon reaction with morpholine to provide the morpholinyl acetamide 13 . Furthermore, the pyrazolopyridoselenolopyrazine ring system 14 was synthesized by the reaction of the o-amino-carbonitrile 3 with malononitrile. Assignment of the chemical structures for the new compounds was confirmed depending on elemental and spectral techniques. On the other hand, most of the synthesized compounds revealed promising results against various bacterial and fungal strains.  相似文献

The synthesis of several dipyridazinothiazines have been accomplished by: (a) cyclization in concentrated hydrochloric acid solution of the appropriate intermediates; and (b) via the Smiles rearrangement in either basic or glacial acetic acid solution of the appropriate intermediates. The following ring systems have been prepared and characterized: 10H-dipyridazino-[4,3-b:4′,5′-e]-1,4-thiazine, 5H-dipyridazino[3,4-b:4′,5′-e]-1,4-thiazine, 10H-dipyridazino[4,5-b:-4′,5′-e]-1,4-thiazine, 5Hdipyridazino[5,4-b:4′,3′-e]-1,4-thiazine, and 10H-dipyridazino[3,4-b:-3′,4′-e]-1,4-thiazine.  相似文献

From an analysis of nmr spectral data, 1,6,7,12b-tetrahydro-2H,4H-[1,3 ]oxazino[3′, 4′ :1,2]-pyrido[ 3,4-b ]indole is shown to exist in solution at room temperature almost entirely in the cis-fused ring conformation with the nitrogen lone pair bisecting the C4 methylene group whereas under the same conditions 1,2,3,6,7,12b-hexahydro-3-methyl-4H-pyrimido[3′,4′:1,2] pyrido-[3,4-b ]indole exists as an approximately 50:50 equilibrium mixture of the cis and trans-fused ring conformations.  相似文献
