利用阴离子交换色谱和疏水相互作用色谱从烟草疫霉菌培养液中分离出一种新的31ku激发子蛋白,选择了阴离子交换色谱流动相的的最佳pH值6.0,建立了疏水相互作用色谱硫酸铵Tris缓冲液-水洗脱模式,简化了纯化步骤和减少了活性损失的危险。  相似文献

选取了四种常用的弱阳离子交换(WCX)商品柱以研究标准蛋白在其上的色谱保留行为。发现在疏水色谱(HIC)模式下,蛋白在这四种WCX商品柱上也有不同程度的保留特征,且洗脱曲线呈现出保留值随盐浓度变化的U型。从分子力学角度定性解释了因疏水相互作用力的存在影响了蛋白在WCX色谱柱上洗脱顺序的改变。运用计量置换理论(SDT)中的两组线性方程进一步证实了WCX和HIC中蛋白与固定相间相互作用力的性质,在HIC中为非选择性作用力,而在离子交换色谱(IEC)中为选择性作用力。这四种色谱柱中的两种事实上可在WCX和HIC两种模式下,对标准蛋白进行分离且有较好的分离效果,有可能作为二维色谱柱来使用。  相似文献

用蛋白折叠液相色谱法(PFLC)对大肠杆菌表达的包涵体形式的重组人粒细胞集落刺激因子(rhG-CSF)进行了复性并同时纯化。用Cu2+-亚氨基二乙酸(IDA) Sepharose作为固定化金属离子亲和色谱的固定相。在低浓度脲存在下,以咪唑为洗脱剂,采用线性梯度洗脱rhG-CSF。该法仅通过一步PFLC分离,减少了复性过程中rhG-CSF的聚集,复性后的rhG-CSF的比活性为1.8×108 IU/mg,纯度为97%,质量回收率为32%。  相似文献

研究了用反相色谱模式, 在不同流动相流速条件下所得到的细胞色素C的肽谱图. 发现在低流速时, 保留弱的肽谱峰增多, 而在高流速时保留强的肽谱峰增多, 未发现溶质分子质量与溶质相对保留值(RRT)之间存在有特定的变化关系. 然而却发现了两种变化规律, 即溶质相对保留值(RRT)与流动相流速之间的双对数线性关系(1) log RRT=a+blog ν, 及所得该二线性参数, a与b之间的线性关系(2) a=c+db. 预计该结果可能会在液相色谱指纹图谱研究和蛋白质组学中分离低丰度蛋白时有潜在的应用价值.  相似文献

建立了大豆分离蛋白中三聚氰胺的液相色谱方法。大豆分离蛋白经过1%三氯乙酸和饱和乙酸铅溶液提取后,经过阳离子交换固相萃取柱净化处理,用5%氨化甲醇洗脱,洗脱液用高效液相色谱紫外检测器检测,外标法定量。三聚氢胺的质量浓度在0.4~40 mg/L范围内与色谱峰面积呈良好的线性关系,相关系数为0.9990。方法的检出限为0.6 mg/kg,定量限为2 mg/kg。2,4,20 mg/kg 3个浓度水平的加标回收率在78.82%~92.95%范围内。该方法分析成本低,测定结果准确,能满足常规检测及食品安全监控的需要。  相似文献

评价了麦科菲高聚物型(苯乙烯-二乙烯基苯共聚物,PS-DVB)反相色谱柱(MKF-RP色谱柱)的柱压,分离柱效,化学和机械稳定性,并研究了梯度条件、色谱柱长等因素对4种常见蛋白(胰岛素、溶菌酶、牛血清白蛋白、卵清蛋白)分离性能的影响.结果表明,在对4种常见蛋白分离时,每米理论塔板数均可达到105以上,且150 mm的柱长优于250 mm及50 mm.以牛血清白蛋白为例,在0.2~20 μg范围内,其进样量与峰面积的线性关系良好(r=0.9997);最大载样量为80 μg左右.经强酸强碱清洗不会改变色谱柱的柱效.  相似文献

对近两年来源于大肠杆菌(Escherichia coli,E.coli)的蛋白表达和用蛋白折叠液相色谱(protein folding liquid chromatography,PFLC)法对所形成的包涵体目标蛋白的复性并同时纯化的新近发展做了简要的介绍和评述.PFLC法用于包涵体蛋白分离、纯化很广,其特点是除了在色谱柱上将目标蛋白与其他组分分开,还同时要在色谱柱上进行包涵体蛋白折叠.可以说,现代生物技术中所用的大多数有价值蛋白产品的制备仍然有赖于不同机理的液相色谱(Lc)法.而用PFLC法对源于E.coli的蛋白的制备方法更具可塑性和容易达到规模化,其生成本可以成倍地降低.该文主要内容包括了E.coli蛋白的表达及样品前处理、PFLC的实用范围、PFLC的优化、PFLC中的新技术、新设备和新方法、PFLC的分子学机理、应用事例及对未来的展望.  相似文献

本文研究了液相色谱分离过程的吸附平衡常数、传质系数参数的估值模型,用色谱技术和以惰性物示踪的扰动应答实验技术测定甘露醇和山梨醇在钙型吸附剂上的吸附相平衡常数和总传质系数以及床层中的轴向扩展系数,结果表明:吸附平衡常数和吸附剂的选择性随温度的升高而下降;液固相间的总传质系数随温度的升高而增大;轴向扩散系数随流速的增大而增大,参数灵敏度分析的结果表明,相平衡关系,尤其是吸附选择性,对色谱分离的影响有较  相似文献

用高效疏水相互作用色谱(HPHIC)研究了将四氢糠醇加入盐酸胍(GuHCl)变性剂中或将四氢糠醇键合至硅胶基质时,对α-胰凝乳蛋白酶(α-Chy)复性效率的影响情况,并以α-Chy的生物活性回收率对结果进行了表征。用尺寸排阻色谱(SEG)与HPHIC的实验结果进行了比较,结果表明,四氢糠醇在变性剂中和键合至硅胶上时,均可提高α-Chy的复性效率,但提高的程度不同。四氢糠醇在溶液中或在HPHIC固定相表面时具有辅助α-Chy进行复性作用的机理可能是由于四氢糠醇与α-Chy形成了复合物的形式。实验中还对GuHCl中加入的四氢糠醇量进行了优化。  相似文献

Summary This paper further investigates the effects of silica base pore size and the molecular weight of polyethylene glycol (PEG) ligands on the coverage of PEG-bonded phases, as well as the resolution of protein separation in high-performance hydrophobic interaction chromatography (HPHIC). The results demonstrate that among the PEG-bonded phases examined in this study, the bonded phase coupled PEG-1500 on LiChrospher 500 silica exhibited the best resolution in protein separation.  相似文献

Protein refolding is a key step for the production of recombinant proteins, especially at large scales, and usually their yields are very low. Application of liquid chromatography to protein refolding is an exciting step forward for this field. In this work, recombinant human granulocyte colony-stimulating factor (rhG-CSF) expressed in Escherichia coli was renatured with simultaneous purification by ion exchange chromatography (IEC) with a Q Sepharose FF column. Several chromatographic parameters affecting the refolding yield of the denatured/reduced rhG-CSF, such as the urea concentration, pH value, concentration and ratio of reduced/oxidized glutathione in the mobile phase, as well as the flow rate of the mobile phase, were investigated in detail and indicated that the urea concentration and the pH value were of great importance. At the optimal conditions, the renatured and purified rhG-CSF was found to have a specific bioactivity of 3.0 x 10(8) IU/mg, a purity of 96%, and a mass recovery of 49%. Compared with the usual dilution method, the IEC method developed here is more effective for rhG-CSF refolding in terms of specific bioactivity and mass recovery.  相似文献

为使混合机理色谱(MMC)得到广泛地应用, 合成、表征和评价MMC固定相就成了首先要解决的问题. 依据离子交换色谱柱也具有疏水色谱(HIC)保留机理的特征, 选了4种弱阳离子交换(WCX)柱和一根二维[2D(WCX,HIC)]色谱柱, 研究了标准蛋白在这两类色谱柱上的保留行为. 这四种WCX色谱柱中的两种能在WCX和HIC两种分离模式下分离蛋白, 虽不如2D色谱柱效果好, 但有可能当成“准2D柱”来使用. 发现蛋白在这四种WCX柱上所显示的HIC分离特征各不相同, 且保留值随盐浓度变化呈现出的“U型”曲线也有大的差异. 实验结果显示, “U型”曲线的宽度和临界点分别与色谱动力学和热力学因素相关. 还对这两类色谱固定相的峰容量表征方法和命名提出了建议和说明.  相似文献

Interaction between proteins and stationary phase in hydrophobic interaction chromatography (HIC) is differentiated into two thermodynamic processes involving direct nonbonding/conformation interac- tion and surface hydrophobic effect of proteins, hence quantitatively giving rise to a binary linear rela- tion between HIC retention time (RT) at concentrated salting liquid and ligand-protein binding free en- ergy. Then, possible binding manners for 27 proteins of known crystal structures with hydrophobic ligands are simulated and analyzed via ICM flexible molecular docking and genetic algorithm, with re- sults greatly consistent with experimental values. By investigation, it is confirmed local hydrophobic effects of proteins and nonbinding/conformation interaction between ligand and protein both notably influence HIC chromatogram retention behaviors, mainly focusing on exposed portions on the protein surface.  相似文献

采用水包油型微乳液相色谱(MELC)分离了6种激素类药物(醋酸可的松、泼尼松龙、己烯雌酚、炔雌醇、醋酸氟轻松及黄体酮)。考察了微乳流动相的组成成分(包括表面活性剂的浓度、油相种类、有机添加剂种类)及固定相孔径等对分离的影响。实验得到的最佳分离条件: 色谱柱为Venusil ASB C18 (T)(粒径5 μm,孔径30 nm,250 mm×4.6 mm),微乳流动相为30 g/L十二烷基硫酸钠(SDS)-0.8%正辛烷-6.6%正丁醇,流速为0.8 mL/min,检测波长为254 nm,柱温为35 ℃。该方法可用于甾体药物及其制剂的分离鉴别以及快速测定。  相似文献

This paper discusses ultrahigh-speed, ultrahigh-resolution preparative protein separation using an in-house designed membrane chromatography device. The performance of the membrane chromatography device was systematically compared with an equivalent resin-packed preparative column. Experiments carried out using model proteins showed that membrane chromatography gave more than four times greater resolution than the preparative column, while at the same time being more than 19 times faster. Membrane chromatography was therefore a better option, not only in terms of higher productivity but also in terms of higher product purity. Membrane chromatography was also superior in terms of resolving and presenting tracer impurity peaks in the chromatogram. Experiments carried out using monoclonal antibody samples showed that membrane chromatography was suitable for ultrahigh speed, and ultrahigh resolution fractionation of charge variants. This paper highlights and explains the need for proper device design for enabling the use of membrane chromatography for the efficient purification of protein biopharmaceuticals.  相似文献

Summary The enantiomeric resolution of seven racemic sulphoxides on chiral stationary phases has been investigated by gas and liquid chromatography. In gas chromatography the separations were performed on octakis-(2,6-di-O-pentyl-3-O-butyryl)-γ-cyclodextrin (FS Lipodex-E) and heptakis-(2,6-di-O-methyl-3-O-pentyl)-β-cyclodextrin (DMP-β-CD). Both stationary phases were suitable for separation of the enantiomers of the sulphoxides. With one exception for each series all racemetes could be resolved on both stationary phases; FS Lipodex-E was more enantioselective than DMP-β-CD, whereas the latter seemed more generally applicable. Liquid chromatographic separations with Chiralcel-OB as stationary phase were significantly improved by optimization of mobile phase composition and temperature. Resolution factors up to Rs=6 were achieved indicating that the improved separations could now be easily used for preparative purposes.  相似文献

Summary A computer-assisted method is described for optimization of multi-component, mobile phase selection for separating enantiomers of four pesticides in normal-phase HPLC. The method is based on the triangle, solvent-selection concept using a statistical scanning method. The optimization of the separation over the experimental region is based on a special polynomial estimation from seven experimental runs, and resolution (Rs) is used as the selection criterion. The quinolones are derivatives of oxoquinolines and mostly known for their antibacterial and antiviral activities. Many quinolones are chiral compounds having asymmetric centers and important due to their enantioselective biological activities. In order to study the biological activities of quinolone enantiomers, to control the manufacturing of homochiral drugs and to prepare necessary quantities of pure enantiomers for preclinical or clinical trials, respective chiral separation methods are urgently needed. In this context, the present review discusses chromatographic and electrophoretic methods for the enantioseparation of chiral quinolones and provides some useful information on their physical and pharmaceutical properties. The drawbacks of currently used techniques are revealed and ways to overcome them are outlined. Moreover, recommendations for an optimal choice of a separation protocol are given.  相似文献
