A revised structure for hermonionic acid

Hermonionic acid and its decarboxylated product have been isolated from $\text{Garcinia}$$\text{quaesita}$. ¹³C NMR spectral and chemical evidence indicate that hermonionic acid is 2-0-[2-(3-methylbut-2-enyl)-3-methoxy- 4-hydroxy-5-(3,7-dimethylocta-2,6-dienyl]-4-methoxy-5-(3-methylbut-2-enyl-6-hydroxybenzoic acid. The previously assigned dienone structure for this acid is incorrect.     

Calozeylanic acid,a new bark acid from three calophyllum species (guttiferae)

Calozeylanic acid has been isolated from the bark of three $\text{calophyllum}$ species: $\text{C.lankaensis}$ (= $\text{C.zeylanicum}$), $\text{C.thwaitesii}$ and $\text{C.walkeri}$. Its structure has been established as 2(R),3(R)-2,3-Dimetyl- 5-hydroxy-6-(3-methylbutanyl)-6-(3,7-dimethylocta-3,6-dienyl)-7-oxo-8- (2-carboxy-1-phenylethyl)-2,3,6,7-tetrahydrobenzo-4-pyrone.     

Synthesis and revised structure of the o-succinylbenzoic acid coenzyme a ester,an intermediate in menaquinone biosynthesis

Synthesis of the two isomers $\text{2}$, $\text{3}$ of the mono coenzyme A ester of o-succinylbenzoic acid ($\text{1}$, OSB, i.e. 4-(2-carboxyphenyl)-4-oxobutanoic acid) and enzymic conversion of $\text{3}$, to 1,4-dihydroxy-2-naphthoic acid $\text{7}$ shows that as opposed to previous assumptions the “aliphatic” rather than the “aromatic” carboxyl group in o-succinylbenzoic acid $\text{1}$ is activated during vitamin K₂ biosynthesis in Escherichia coli and Mycobacterium phlei.     

Synthesis of 4′-amino-4′-deoxy analog of pantothenic acid

The synthesis of $\text{D}$- and $\text{L}$-N-(4-amino-3, 3-dimethyl-2-hydroxybutyryl)- β-alanine ($\text{7}$) is described. Compound $\text{7}$ is an analog of pantothenic acid in which the 4′-hyroxy group is replaced by amino group. The synthetic sequence leading to $\text{7}$ involved the synthesis $\text{D}$L-4-amino-3,3-dimethyl-2-hydroxybutyric acid and its resolution. Coupling of N-benzyloxycarbonyl N-hydroxysuccinimide ester ($\text{5}$) with β-alanine and followed by removal of the protecting group gave $\text{7}$.     

A practical process for large-scale synthesis of (S)-5-hydroxy-6-trans-8,11,14-cis-eicosatetraenoic acid (5-HETE)

($\text{S}$)-5-Hydroxy-6-$\text{trans}$-8,11,14-$\text{cis}$-eicosatetraenoic acid (5-HETE) (1) and its enantiomer are readily available by a chemical synthesis from arachidonic acid which includes a chromatographic separation of diastereomeric urethanes (3) made from (±)-5-HETE methyl ester and the isocyanate 4 derived from dehydroabietylamine.     

Enantioselective synthesis of β-hydroxyisobutyric acid: a useful synthon in the synthesis of polypropionate-type natural products

The aldol reaction of formaldehyde with the dicyclopentylborinyl enolate derived from $\text{S}$-(or $\text{R}$-) 1-tert-butyldimethylsilyloxy-l-cyclohexylbutan-2-one, followed by desilylation and sodium meta-periodate oxidation, provides $\text{R}$-(or $\text{S}$-) β-hydroxyisobutyric acid.     

Efficient asymmetric synthesis of α-amino acids through hydrogenation of α,β-dehydroamino acid residue in cyclic dipeptides

A series of cyclo (Δaminoacyl-L-Ala) ($\text{4}$) (Δ=α,β-dehydro) were prepared from cyclo(Gly-L-Ala) and corresponding aldehyde, and hydrogenated with Pd black in MeOH. Chiral inductions producing cyclo (L-aminoacyl-L-Ala) ($\text{5}$) from $\text{4}$ were 96–99% in the case of L-Aba (2-aminobutanoic acid), L-Val, L-Leu, and L-App (2-amino-5-phenylpentanoic acid) as an L-aminoacyl moiety in $\text{5}$. Pure L-Leu, L-Aba, and L-App were synthesized in preparative scale from corresponding $\text{4}$ through asymmetric hydrogenation and acid-hydrolysis.     

Structure elucidation and synthesis of (2s)-4-oxo-1-azabicyclo[3.3.0]octa-5,7-diehe-2-carboxylic acid,a new metabolite isolated ercm stheptomyces oliyacbjs

The structure of a new metabolite isolated from $\text{Streptomyces}$$\text{olivaceus}$ has been shown to be (2S)-4-oxo-1-azabicyclo[3.3.0]octa-5,7-diene-2-carboxylic acid by means of its spectroscopic properties and total synthesis.     

A Wittig reaction on mucochloric and mucobromic acids. Preparation of ethyl (2E, 4Z -4,5-dihalo-2,4-pentadienoates and alkyl 2E -5-bromo-2-penten-4-ynoates

Reaction of mucochloric and mucobromic acid with (ethoxycarbonylmethylene) triphenyl-phosphorane affords the ethyl ester of (2$\text{E}$, 4$\text{Z}$ -4,5-dichloro- and (2$\text{E}$, 4$\text{Z}$) -4, 5-dibromo-2,4-pentadienoic acid respectively; in the latter case ethyl (2$\text{E}$ -5-bromo-2-penten-4-ynoate is formed concomittantly.     

Methylthiomethyl esters from acid chlorides reexamination of a reported β-ketosulfoxide synthesis

Reexamination of the reported (1) synthesis of β-ketosulfoxides from acid chlorides and reagent $\text{2}$ revealed that the products formed were actually methylthiomethyl esters ($\text{4}$).     

An efficient and stereocontrolled synthesis of platelet activating factor from (s)-(−)-malic acid

A stereocontrolled synthesis of C₁₆-PAF ($\text{11}$) from (S)-(?)-malic acid ($\text{1}$), employing regioselective hydrogenolytic cleavage of benzylidene acetal derivatives of (S)-1,2,4-butanetriol ($\text{2}$) with borane-tetrahydrofran complex, is described.     

Eugenol and safrole as starting materials for the synthesis of 3-alkyl muconic acid mono and dimethyl esters and 4,4-dialkyl butenolides

Compounds $\text{1b-e}$ and $\text{2b-d}$ prepared from eugenol ($\text{1a}$) and safrole ($\text{2a}$) respectively were submitted to ozonation in methanol at 0°C. The 3-alkyl muconic acid dimethyl esters $\text{4b-e}$ were produced from $\text{1b-e}$ (～ 45%), while starting from $\text{2b-d}$ the 3-alkyl muconic acid monomethyl esters $\text{6b-d}$ were obtained regiospecifically (～ 75%). These latter compounds were transformed (～ 85%) into the 4,4-dialkyl butenolides $\text{7b-d}$.

Condensation of 2-acetamido-6-$\text{O}$-acetyl-2-deoxy-D-mannose with pyruvic acid in the presence of immobilized acylneuraminate pyruvate lyase (2.8 units) gave 5-acetamido-9-$\text{O}$-acetyl-3,5-dideoxy-D-$\text{glycero}$-D-$\text{galacto}$-2-nonulopyranos-1-onic acid ($\text{N}$-acetyl-9-$\text{O}$-acetylneuraminic acid) on the 0.3 mmole scale.

(+)-Thalictrifoline ($\text{3}$) has been synthesized from an optically resolved (+)-8-oxo-13-carboxytetrahydroprotoberberine ($\text{13}$) whose absolute configuration was established by correlation with (+)-$\text{18}$. This determines the absolute configurations of (+)-thalictrifoline ($\text{3}$) as 13R, 14R aria (+)-corydalic acid methyl ester ($\text{9}$) as 3R, 4R.     

Fluorocarbon derivatives of nitrogen. Part 11. Synthesis of some 2-(trifluoromethyl)imidazo[1,2-a]pyridines from trifluoroacetonitrile

3-(t-Butoxycarbonyl)-2-(trifluoromethyl)imidazo[1,2-$\text{a}$]-pyridine, prepared from trifluoroacetonitrile and pyridinium t-butoxycarbonylmethylide, reacts smoothly with trifluoroacetic acid to provide 2-(trifluoromethyl)imidazo[1,2-$\text{a}$]pyridine-3-carboxylic acid, which gives 2-(trifluoromethyl)imidazo[1,2-$\text{a}$]-pyridine when heated. 3-Cyano-2-(trifluoromethyl)imidazo[1,2-$\text{a}$]pyridine can be obtained $\text{via}$ treatment of trifluoroacetonitrile with pyridinium cyanomethylide, which is sufficiently reactive to effect nucleophilic displacement of fluorine from pentafluoropyridine under mild conditions [→pyridinium cyano(tetrafluoro-4-pyridyl)methylide].     

Synthesis of enantiomerically enriched atrolactic acid and other α-hydroxy acids

The α-anions of 2-substituted 1,3-dioxolan-4-ones derived from chiral mandelic and lactic acid ($\text{2a}$, $\text{2b}$; $\text{3a}$; $\text{10a}$, $\text{10b}$) were alkylated with high stereoselectivity. The products formed were hydrolysed to α-hydroxy acids with 65–85% e.e. ((S)(+)-$\text{5}$,$\text{7}$,$\text{9}$, (R)(–)-$\text{13}$).     

Novel (4 + 1) fragment combination approach to chiral cyclopentanoids from tartaric acid

A $\text{chiral}$ cyclopentanoid building block $\text{29}$ has been synthesized in “one pot” cyclization process from epoxides $\text{22}$ - $\text{25}$ (which are readily accessible from (R,R)-(+) tartaric acid) with the carbanion derived from phenylthioacetonitrile (PhS-CH₂-CN).     

Conversion of clavulanic acid into thiadeoxa nuclear analogues

1,4-Addition of sulphur nucleophiles to the diene ($\text{12}$) derived $\text{via}$ the pen-2-em ($\text{5}$) from clavulanic acid provides the thiadeoxa analogues ($\text{14}$–$\text{15}$). X-ray analysis of the ester ($\text{14}$) shows the thermodynamically stable isomers to have the same relative stereochemistry as clavulanic acid.     

Heavenly blue anthocyanin II trans-4-0-(6-0-(trans-3-0-(β -d-glucopyranosyl)caffeyl)-β-d-glucopyranosyl)- caffeic acid,a novel component in heavenly blue anthocyanin

Structure of an alkaline hydrolysis product of heavenly blue anthocyanin was determined to be $\text{trans}$-4-0-(6-0-($\text{trans}$-3-0-(β-D-glucopyranosyl)caffeyl)-β-D- glucopyranosyl)caffeic acid ($\text{1}$).     

Amino acids derivatives that stabilize secondary structures of polypeptides-- : I. Synthesis of LL-3-amino-2-piperidone-6-carboxylic acid (LL-Acp), a novel beta-turn-forming amino acid

A six-step synthesis is reported of $\text{LL}$-3-amino-2-piperidone-6-carboxylic acid from $\text{L}$-homoserine lactone and the $\text{mono}$-$\text{t}$-butyl ester on $\text{N}$-benzyloxycarbonylaminomalonic acid. Evidence of chiral integrity is discussed.
