Tertiary aliphatic β- and γ-nitro nitriles and ketones deuterated in (several) selected positions had been synthesized. The deuterated nitro compounds served as a starting material for the corresponding deuterium labeled nitrones or hydroxylamines (reducing with aluminum amalgam). Further oxidation of the last two groups of compounds with sodium periodate or m-CPBA afforded the relevant deuterated tertiary C-nitroso compounds.  相似文献

The structure of a constrained bicyclic chiral derivatizing agent (CDA), 1,2,3,4-tetrahydro-1,4-epoxynaphthalene-1-carboxylic acid, THENA 1, was modified by replacing both exo-methylene protons with deuterium atoms. The modified CDA, THENA-d22, could be used to assign the absolute configuration of chiral secondary alcohols with good reliability. Compared with THENA, the multiplicity of the methylene proton signals in the 1H NMR spectra of THENA-d2 derivatives is less complicated and the new CDA thus offers simpler NMR spectra for data interpretation.  相似文献

Detailed dynamic nuclear polarization and electron spin resonance studies were carried out for 3‐carbamoyl‐2,2,5,5‐tetramethyl‐pyrrolidine‐1‐oxyl, 3‐carboxy‐2,2,5,5‐tetramethyl‐pyrrolidine‐1‐oxyl,3‐methoxycarbonyl‐2,2,5,5‐tetramethy pyrolidine‐1‐oxyl nitroxyl radicals and their corresponding deuterated nitroxyl radicals, used in Overhauser‐enhanced magnetic resonance imaging for the first time. The dynamic nuclear polarization parameters such as dynamic nuclear polarization (DNP) factor, longitudinal relaxivity, saturation parameter, leakage factor and coupling factor were estimated for deuterated nitroxyl radicals. DNP enhancement increases with agent concentration up to 3 mm and decreases above 3 mm . The proton spin–lattice relaxation time and the longitudinal relaxivity parameters were estimated. The leakage factor increases with increasing agent concentration up to 3 mm and reaches plateau in the region 3–5 mm . The coupling parameter shows the interaction between the electron and nuclear spins to be mainly dipolar in origin. DNP spectrum exhibits that the full width at half maximum values are higher for undeuterated nitroxyl radicals compared with deuterated nitroxyl radicals, which leads to the increase in DNP enhancement. The ESR parameters such as, the line width, line shape, signal intensity ratio, rotational correlation time, hyperfine coupling constant and g‐factor were calculated. The narrow line width was observed for deuterated nitroxyl radicals compared with undeuterated nitroxyl radicals, which leads to the higher saturation parameter value and DNP enhancement. The novelty of the work permits clear understanding of the DNP parameters determining the higher DNP enhancement compared with the undeuterated nitroxyl radicals. Summary Deuterated compounds have been used as mobile phases for microcolumn high-performance liquid chromatography (HPLC) employing flow-cell Fourier transform infrared spectroscopy for detection. Separations were carried out on adsorption, reversed-phase, non-aqueous size-exclusion and aqueous size-exclusion chromatographic columns. Due to the IR transparency of deuterated compounds in a C–H stretching region they represent nearly ideal eluents in terms of universal detection. In addition, due to the shift in the absorption wavenumber following deuteration, deuterated solvents allow FTIR detection of solutes in other regions, where otherwise it would be prohibited, or sensitivity sacrified by interfering solvent absorption.  相似文献

Infrared monitoring was accomplished in reversed phase HPLC on using deuterated solvents. The end of a micro HPLC column was flattend and used as a flow cell for on-column IR detection. Novel di-, tetra- and octadeuterated derivatives of mono-N-alkylated diaminopropanes, spermidines, spermines, symmetrically bis-N-alkylated spermines and unsymmetrically bis-N-alkylated spermines were synthesized. A highly deuterated novolac‐type phenolic resin was prepared by polycondensation of deuterated phenol and formaldehyde using oxalic acid as an acid catalyst. The polycondensation of deuterated monomers and the formation of the highly deuterated phenolic resin were confirmed by the gel permeation chromatography, IR, and 1H NMR analyses. With the exception of hydroxyl groups, the degree of deuteration was estimated to be more than 98%. The polymer conformation in THF solution was evaluated by the scaling exponent of the Mark–Houwink–Sakurada equation. The exponent of the deuterated phenolic resin is 0.26 in THF at 40 °C and is close to that of a nondeuterated phenolic resin, which suggests that phenolic resins behave like a compact sphere irrespective of deuteration. The curing behavior of the deuterated phenolic resin with hexamethylenetetramine was confirmed by differential scanning calorimetry analysis. The cured highly deuterated phenolic resin exhibits a lower incoherent neutron scattering background than that of the nondeuterated phenolic resin, which suggests that the former is suitable for matrix resins with low incoherent backgrounds for small‐angle neutron scattering studies of thermosetting resins. © 2011 Wiley Periodicals, Inc. Chimeric oligonucleotides with the novel morpholino modification and the phosphoramidate linkers have been synthesized and characterized. These oligonucleotides showed moderate thermal stability with complementary RNA and DNA, and enhanced resistance toward the nuclease (t1/2 > 10 h). The phosphoramidate linker made the synthesis of such oligonucleotides applicable on a DNA synthesizer. Under the acidic condition (pH 3.0), the phosphoramidate linkers were readily cleaved, and such property might be useful for the DNA-sequence determination.  相似文献

The use of calcium carbide (in the presence of H2O) as a source of acetylene in the reaction with methyl 2-amino-4,6-O-benzylidene-2-deoxy-β-d-glucopyranoside under superbasic conditions (KF, KOH, DMSO, 130 °C, 3 h) led to the corresponding vinyl ether in 78% yield. The vinylation does not affect other reaction centers. Replacement of water with deuterium oxide in the reaction mixture gave a stable deuterated analog of this vinyl ether in 72% yield. The one-step isotopic enrichment was practically quantitative (isotopic purity >96%). Introduced deuterium atoms can be used as a convenient label in further transformations.

This paper deals with the synthesis of m32,2,7G5′pppAmpUmpApd(CpTpTpApCpCpT), a 5′-TMG-capped RNA-DNA chimeric oligonucleotide, which is expected to be conveyed to the nucleus by snurportin 1, a nuclear-transport protein. This 5′-TMG-capped RNA-DNA chimeric molecule was synthesized by the enzymatic condensation of m32,2,7G5′pppAmpUmpA with A5′ppd(CpTpTpApCpCpT) in the presence of RNA ligase. An inherent serious side reaction was disclosed in the 5′-adenylation of d(CpTpTpApCpCpT) on the solid support, but the use of an active ester intermediate as the pA donor gave an improved result.  相似文献
