Summary.  2-O-Ethyluracils were silylated with HMDS and condensed in the presence of TMS-triflate with β-D-glucose pentaacetate to give the corresponding β-nucleosides. Alternatively, these could be synthesized by nucleoside coupling of 2,3,4,6-tetra-O-acetyl-α-D-glucopyranosyl bromide with the sodium salts of 2-O-ethyluracils, which were deprotected with saturated ammonia in methanol. 6′-O-Tosylate nucleoside derivatives were prepared by treating of the latter with tosyl chloride in anhydrous pyridine. The compounds thus obtained were treated with sodium azide in anhydrous DMF to afford the corresponding 6′-azido nucleoside derivatives, which can also be prepared by treatment with sodium azide in the presence of carbon tetrabromide and triphenylphosphine in anhydrous DMF. Nucleophilic displacement of the 6′-tosyloxy group by morpholine gave 6′-deoxy-6′-morpholino nucleosides. The reduction of the azido group of the 6′-azido nucleosides using triphenylphosphine in pyridine afforded the 6′-amino analogues. Glucosylated 2-O-ethyluracils showed moderate activity against HBV. An efficient and high-yielding synthesis of N~2-arylaminopyrimidine-5-carbonitrile derivatives starting from arylamines and 2-methylthio-pyrimidine-5-carbonitriIe derivatives has been developed in the presence of cesium carbonate as basic reagent.This new protocol showed high chemical tolerance for a range of functional groups,and only the methylthio substituent on C2 of the pyrimidine ring was replaced with arylamine derivatives under the reaction conditions.  相似文献

The one-pot reaction between 2-aminobenzo[d]isothiazol-3-one and alkyl propiolates in presence of triphenylphosphine leads to the corresponding alkyl 4-amino-5-oxobenzo[f][1,4]thiazepine-3-carboxylates. A plausible mechanism of the reaction is proposed and unambiguous evidence for the structures is obtained from a detailed magnetic resonance spectral analysis. 1D and 2D NMR spectra such as COSY, 1H-13C and 1H-15N HSQC and HMBC heteronuclear correlations and an INADEQUATE experiment are reported and discussed.  相似文献

For the first time, amides and ureas based on both 5-nitroisoquinoline and 5-nitrosoisoquinoline were obtained by direct nucleophilic substitution of hydrogen in the 5-nitroisoquinoline molecule. In the case of urea and monosubstituted ureas, only 5-nitrosoisoquinoline-6-amine is formed under anhydrous conditions.  相似文献

Various methods of synthesizing functional derivatives of dithia-13(16)-crown-4(5) ethers are proposed. Complex-formation of the obtained compounds with Ag+ and Pb2+ ions has been studied using 1H NMR. Conditions for N-acylation of 2-amino-5-aryl-6H-1,3,4-thiadiazines with trifluoroacetic anhydride and halogen-substituted carboxylic acid halides with retention of the initial heterocyclic system were found. 5-Aryl-2-haloacylamino-6H-1,3,4-thiadiazines were obtained in preparative yields. The results of studies aimed at elaborating methods for synthesizing penta- and hexacoordinated organogermanium derivatives containing lactamo-N-methyl and related bidentate ligands and investigating their structure and reactivity are reviewed. Novel 5-oxazolone derivatives containing an N-phenyl-aza-15-crown-5 moiety were synthesized for the first time. The structures of the new derivatives were confirmed by 1H NMR, 13C NMR and FT-IR. In addition, evaluation of the visible absorption and emission properties of the structures were carried out in eight different solvents. The products show intense visible absorption maxima in the range 467-524 nm, and fluoresced strongly, with emission maxima from 496 to 689 nm in all the solvents tested.  相似文献

A series of 2-aryl-5-alkyl-7-methoxylbenzo[b]furan derivatives have been synthesized by utilizing the coupling of methyl 3-methoxy-4-hydroxy-5-bromocinnamate with cuprous phenylacetylide as the key step.The structures of the new compounds were confirmed by1H NMR,IR and MS.The structure of compound 14 was further confirmed by single crystal X-ray.Compound 17 showed cytotoxic activity against human lung carcinoma A549.  相似文献

Silylotropy in 4-substitutedN-trimethylsilypyrazoles is studied by dynamic1H,13C, and29Si NMR spectroscopy. The catalytic 1,2-migration of a trimethylsilyl group in 4-halo-N-trimethylsilylpyrazoles was detected. Synthetic procedures for new N2S4- and N2S5-donor macrocycles (2 and 4) were given. The ligands were prepared by the reaction of NaBH4 with the appropriate macrocyclic diamide in the presence of boron trifluoride ethyl etherate in dry tetrahydrofuran (THF). Solvent extraction method was used to evaluate metal-ion binding properties of the new ligands. The solvent extraction experiments suggested that the reduced macrocycles have Ag+ and Hg2+ selectivities compared to Pb2+, Co2+, Zn2+, Ni2+, Cu2+, Mn2+ and Cd2+ ions. The extraction constants (log K ex) and complex compositions were determined for Ag+ and Hg2+ complex of compound (4).  相似文献

Four 5-alkynyl-2′-deoxyuridines containing different bulky substituents and flexible linkers between the triple bond and the aromatic residue have been prepared and tested against HSV-1 in Vero cells. Two nucleosides containing carbonyl groups, 5-(4-benzoylphenoxypropyn-1-yl)-2′-deoxyuridine (19a) and 5-(estron-3-yloxypropyn-1-yl)-2′-deoxyuridine (19c), showed low cytotoxicity and moderate antiviral activity. The flexible linker appears not to be favorable for antiviral properties of 5-alkynyl-2′deoxyuridines: 5-[(perylen-3-yl)methoxypropyn-1-yl]-2′-deoxyuridine (19d) showed considerable cytotoxicity and no antiviral activity in contrast to the active and nontoxic 5-(perylen-3-ylethynyl)-2′-deoxyuridine (9), a nucleoside with a rigid triple-bond-connection of the aromatic system to the nucleobase.  相似文献

2-(1-Alkoxyimino-2,2,2-trifluoroethyl)-5-trimethylsilylfurans were synthesized by the condensation of 2-(trifluoroacetyl)-5-trimethylsilylfuran with alkoxyamines. According to 1H and 19F NMR spectroscopic data, the alkoxyimino group in the E-isomers descreens the H-3 and H-4 protons of the furan ring more strongly than in the Z-isomers, shifting their signals downfield. Summary 7-Chloroalkoxyisoflavones (10–26) have been prepared by chemoselective-in the case of 5,7-dihydroxyisoflavones also regioselective-alkylation of hydroxyisoflavones (3–9) with -bromo--chloroalkanes. Compounds10–26 were allowed to react either with 2,4-dihydroxy-3-n-propylacetophenone (1) or with 2-ethoxycarbonyl-7-hydroxy-8-n-propylchromone (2) to afford bridged isoflavone derivatives27–51 with methylene spacers of various length. Carboxylic acid ethyl esters43–51 have been saponified to obtain the carboxylic acids52–60.

The results of studies dealing with the synthesis of six- and ten-membered silicon-nitrogen-containing heterocyclic compounds are presented. The structures and reactivities of these compounds have been studied. High reactivities of 2,5-disilapiperazines and 1,6-dioxa-3,8-diaza-5,10-disilacyclodecane-2,7-dione derivatives have been noted. N6-methyladenosine(m6A) is the most abundant modification in mRNA. Studies on proteins that introduce and bind m6A require the efficient synthesis of oligonucleotides containing m6A. We report an improved five-step synthesis of the m6A phosphoramidite starting from inosine, utilising a 1-H-benzotriazol-1-yloxytris(dimethylamino)phosphoniumhexafluorophosphate (BOP)-mediated SNAr reaction in the key step. The route manifests a substantial increase in overall yield compared to reported routes, and is useful for the synthesis of phosphoramidites of other adenosine derivatives, such as ethanoadenosine, an RNA analogue of the DNA adduct formed by the important anticancer drug Carmustine.  相似文献
