By means of the two-compartment system. PTL-channels, the first lectin channels formed on planar lipid bilayers by Pinellia ternata lectin (PTL) have been studied. The results show (i) PTL-channels are voltage-independent and have apparent subunits; (ii) in 50 mmol/L KC1 and 25 mmol/L BaCl2 solutions, a single channel has unit conductance of 21 pS and 42 pS, respectively; (iii) the channel exhibits a slightly higher permeability to divalent than monovalent cation (PBa/PK=4.1), and (iv) the selectivity among divalent or monovalent cations is poor. The cation selectivity sequence for the channel will follow PBa(7. 0)-Psr (6. 4)~PMg(6. 4)>PK(1. 7)>PN.(1. 0)~PLi(l. 0).Moreover, these data also give explanation to the facilitatory action of PTL on the release of acetyl-choline from motor nerve terminals.  相似文献

Further experiments were conducted to examine the effect of scoparone (6,7-dimethoxycoumarin), a coumarin derivative found in the Chinese crude drug "Capillaris Flos," on calcium mobilization. Scoparone does not affect Ca2+ influx through the voltage-dependent channel due to membrane depolarization. Its inhibitory action may be dependent not only on the inhibition of Ca2+ release from sarcoplasmic reticulum but also on the inhibition of extracellular Ca2+ influx through the receptor-operated channel.  相似文献

The membrane potential (Um), the main factor of the excitation-contraction coupling, of human allantochorial placental vascular smooth muscle cells (VSMCs) has been previously shown to depend on voltage-sensitive K+ channels. These channels were blocked by high external K+. To characterize other channels which regulated Um, various constrictor or/and vasodilators and channel blockers were used. Serotonin depolarized VSMCs, in normal medium, but induced a more marked depolarization in VSMCs predepolarized by high external K+. This depolarization was inhibited by nifedipine, a blocker of voltage-gated Ca2+ channels. Acetylcholine, sodium nitroprusside (without effect on Um in normal medium), hyperpolarized the predepolarized-high K+ medium VSMCs. This hyperpolarization was inhibited after addition of charybotoxin (a blocker of Ca2+-activated K+ channels) or/and glibenclamide (a blocker of ATP-sensitive K+ channels). A similar effect was obtained with isoproterenol. These results indicated that membrane potential of human placental allantochorial VSMCs was regulated by voltage-gated, Ca2+- and ATP-sensitive K+ channels and by voltage-dependent Ca2+ channels.  相似文献

Excessive release of glutamate induces excitotoxicity and causes neuronal damage in several neurodegenerative diseases. Natural products have emerged as potential neuroprotective agents for preventing and treating neurological disorders. Dehydrocorydaline (DHC), an active alkaloid compound isolated from Corydalis yanhusuo, possesses neuroprotective capacity. The present study investigated the effect of DHC on glutamate release using a rat brain cortical synaptosome model. Our results indicate that DHC inhibited 4-aminopyridine (4-AP)-evoked glutamate release and elevated intrasynaptosomal calcium levels. The inhibitory effect of DHC on 4-AP-evoked glutamate release was prevented in the presence of the vesicular transporter inhibitor bafilomycin A1 and the N- and P/Q-type Ca2+ channel blocker ω-conotoxin MVIIC but not the intracellular inhibitor of Ca2+ release dantrolene or the mitochondrial Na+/Ca2+ exchanger inhibitor CGP37157. Moreover, the inhibitory effect of DHC on evoked glutamate release was prevented by the mitogen-activated protein kinase (MAPK)/extracellular signal-regulated kinase (ERK) inhibitor PD98059. Western blotting data in synaptosomes also showed that DHC significantly decreased the level of ERK1/2 phosphorylation and synaptic vesicle-associated protein synapsin I, the main presynaptic target of ERK. Together, these results suggest that DHC inhibits presynaptic glutamate release from cerebrocortical synaptosomes by suppressing presynaptic voltage-dependent Ca2+ entry and the MAPK/ERK/synapsin I signaling pathway.  相似文献

Excess synaptic glutamate release has pathological consequences, and the inhibition of glutamate release is crucial for neuroprotection. Kaempferol 3-rhamnoside (KR) is a flavonoid isolated from Schima superba with neuroprotective properties, and its effecton the release of glutamate from rat cerebrocortical nerve terminals was investigated. KR produced a concentration-dependent inhibition of 4-aminopyridine (4-AP)-evoked glutamate release with half-maximal inhibitory concentration value of 17 µM. The inhibition of glutamate release by KR was completely abolished by the omission of external Ca2+ or the depletion of glutamate in synaptic vesicles, and it was unaffected by blocking carrier-mediated release. In addition, KR reduced the 4-AP-evoked increase in Ca2+ concentration, while it did not affect 4-AP-evoked membrane potential depolarization. The application of selective antagonists of voltage-dependent Ca2+ channels revealed that the KR-mediated inhibition of glutamate release involved the suppression of P/Q-type Ca2+ channel activity. Furthermore, the inhibition of release was abolished by the calmodulin antagonist, W7, and Ca2+/calmodulin-dependent protein kinase II (CaMKII) inhibitor, KN62, but not by the protein kinase A (PKA) inhibitor, H89, or the protein kinase C (PKC) inhibitor, GF109203X. We also found that KR reduced the 4-AP-induced increase in phosphorylation of CaMKII and its substrate synapsin I. Thus, the effect of KR on evoked glutamate release is likely linked to a decrease in P/Q-type Ca2+ channel activity, as well as to the consequent reduction in the CaMKII/synapsin I pathway.  相似文献

用膜片钳全液泡记录方式研究了CuCl2对萝卜SV通道的影响.研究表明,SV通道为阳离子选择性通道,细胞质钙可激活此通道,且其表现出钙依赖性.细胞质钙达2mmol/L时,SV通道电流基本上达到饱和.当在外液中加入不同浓度的CuCl2时,SV通道电流有明显的变化.高浓度的铜离子对通道电流有抑制作用,而低浓度(<0.1mmol/L)则增强通道电流.电极内液中的CuCl2对SV通道电流也有抑制作用.这为进一步研究铜离子对植物生理活动的影响以及铜肥对农作物的作用在通道水平上提供重要的依据.  相似文献

[structure: see text] Chlorahololides A(1) and B(2), two highly complex sesquiterpenoid dimers, were isolated from Chloranthus holostegius. Their structures and absolute configurations were established by NMR spectroscopy, X-ray crystallography, and CD. Chlorahololides A (1) and B (2) exhibited potent and selective inhibition on the delayed rectifier (IK) K+ current, with an IC50 of 10.9 and 18.6 microM, respectively.  相似文献

目的:建立不同产地半夏无机元素的分析方法和溯源体系,为半夏药材的质量控制和道地性评价提供技术支持。方法:采用电感耦合等离子体质谱法(ICP-MS)和电感耦合等离子体原子发射光谱法(ICP-AES)对我国6个主产区72份半夏样品中K、Ca、Na、Mg、Al等37种无机元素的含量进行测定,并采用方差分析、主成分分析、因子分析等计量学方法进行统计与评价。通过对比BP-神经网络算法、K-最近邻算法、最小二乘支持向量机等多种模式识别方法,探索适合半夏产地溯源的最佳模型。结果:不同产地半夏中无机元素的构成各具特征,各无机元素含量在产地间差异显著(P<0.05),其中La、Pb、As、Na、Bi、Hg、Sn、Cd、Ag 9种元素在不同产区间的差异最为明显;3D-plots图显示不同产地半夏样品分布相对集中,具备产地分类的可行性;KNN分类模型(曼哈顿距离)是半夏产地溯源的最佳方法,测试集的正确率达到100%。结论:无机元素分析技术结合适当的计量学模型可以实现半夏的产地溯源。  相似文献

Transmembrane ion transport by arachidonic acid (AA) through bilayer lipid membranes (BLMs) was investigated by means of electrochemical measurements to provide a basis for designing a sensor membrane. We found that AA induces a channel-type current, in addition to a carrier-type current, across planar BLMs. A linear relation between the logarithmic value of the AA concentration and the current responses (given as integrated currents) was observed for a carrier-type current, while a sigmoid relation was found for a channel-type current. Although AA transports Na+, Ca2+ and Mg2+ and exhibits ion selectivity between Na+ and Mg2+ for the carrier-type current, ion transport for the channel-type current was non-selective. It was found that ion transport via the channel mechanism occurs frequently for AA, while channel-type currents were only occasionally observed for y-linolenic acid and prostaglandin D2. No channel-type currents were induced by other fatty acids (oleic, linoleic, stearic, myristic, eicosapentanoic and docosahexanoic acids) and metabolites of AA (12-HETE and 5-HETE). The carrier-type ion transport occurs selectively to these compounds if the concentration is below 1.0 microM. These results suggest that AA selectively facilitates an ion flux through the BLMs, generating channel-type and/or carrier-type currents, which can be used as a measure of the AA concentration.  相似文献

Suspension of intact human red cells in media with low chloride and sodium concentrations (isotonic sucrose substitution) results in strongly inside positive membrane potentials, which activate the voltage-dependent non-selective cation (NSVDC) channel. By systematic variation of the initial Nernst potentials for chloride (degree of ion substitution) as well as the chloride conductance (block by NS1652), and by exploiting the interplay between the Ca(2+)-permeable NSVDC channel, the Ca(2+)-activated K+ channel (the Gárdos channel) and the Ca(2+)-pump, a graded activation of the NSVDC channel was achieved. Under these conditions, it was shown that the NSVDC channels exist in two states of activation depending on the initial conditions for the activation. The hysteretic behaviour, which in patch clamp experiments has been found for the individual channel unit, is thus retained at the cellular level and can be demonstrated with red cells in suspension.  相似文献

Nanoparticles (NPs) are considered as an important environmental risk factor for cognitive impairments and neurodegenerative disorders. Recent evidences have reported a glutamatergic system response against air-borne NPs. Zinc and copper oxides (ZnO and CuO) NPs are the most common metal oxide NPs in industries. The effects of these NPs on hippocampal voltage-dependent ion channels and spatial cognition have been previously studied. However, there is a lack of evidence regarding the effects of ZnO and CuO NPs on glutamatergic synapse neurotransmission in central nervous system. In the present study, the effects of ZnO and CuO NPs on glutamate (Glu) release and uptake have been investigated in isolated nerve terminals (synaptosomes). Our findings have shown that, even in high doses of ZnO and CuO NPs, no significant effect on Glu release is observed. However, a decrease has been observed in uptake of Glu. Hence, ZnO and CuO NPs can be considered as hazardous agents inducing neurodegenerative disorders through Glu excitotoxicity.  相似文献

A series of gas-phase reactions involving molecular Ca-containing ions was studied by the pulsed laser ablation of a calcite target to produce Ca(+) in a fast flow of He, followed by the addition of reagents downstream and detection of ions by quadrupole mass spectrometry. Most of the reactions that were studied are important for describing the chemistry of meteor-ablated calcium in the earth's upper atmosphere. The following rate coefficients were measured: k(CaO(+) + O --> Ca(+) + O(2)) = (4.2 +/- 2.8) x 10(-11) at 197 K and (6.3 +/- 3.0) x 10(-11) at 294 K; k(CaO(+) + CO --> Ca(+) + CO(2), 294 K) = (2.8 +/- 1.5) x 10(-10); k(Ca(+).CO(2) + O(2) --> CaO(2)(+) + CO(2), 294 K) = (1.2 +/- 0.5) x10(-10); k(Ca(+).CO(2) + H(2)O --> Ca(+).H(2)O + CO(2)) = (13.0 +/- 4.0) x 10(-10); and k(Ca(+).H(2)O + O(2) --> CaO(2)(+) + H(2)O, 294 K) = (4.0 +/- 2.5) x 10(-10) cm(3) molecule(-1) s(-1). The quoted uncertainties are a combination of the 1sigma standard errors in the kinetic data and the systematic errors in the models used to extract the rate coefficients. Rate coefficients were also obtained for the following recombination (also termed association) reactions in He bath gas: k(Ca(+).CO(2) + CO(2) --> Ca(+).(CO(2))(2), 294 K) = (2.6 +/- 1.0) x 10(-29); k(Ca(+).H(2)O + H(2)O --> Ca(+).(H(2)O)(2)) = (1.6 +/- 1.1) x 10(-27); and k(CaO(2)(+) + O(2) --> CaO(2)(+).O(2)) < 1 x 10(-31) cm(6) molecule(-2) s(-1). These recombination rate coefficients, as well as those for the ligand-switching reactions listed above, were then interpreted using a combination of high level quantum chemistry calculations and RRKM theory using an inverse Laplace transform solution of the master equation. The surprisingly slow reaction between CaO(+) and O was explained using quantum chemistry calculations on the lowest (2)A', (2)A' and (4)A' potential energy surfaces. These calculations indicate that reaction mostly occurs on the (2)A' surface, leading to production of Ca(+)((2)S) + O(2)((1)Delta(g)). The importance of this reaction for controlling the lifetime of Ca(+) in the upper mesosphere and lower thermosphere is then discussed.  相似文献

The effect of the calcium-binding protein regucalcin on the Ca2+ transport system in the liver microsomes from fed rats was investigated. Ca2+ transport was assayed by the method of Millipore filtration to estimate microsomal 45Ca2+ accumulation following addition of 10 mM adenosine triphosphate (ATP). 45Ca2+ uptake was retarded by the presence of regucalcin (1.0-4.0 microM). This retardation was remarkable at 1 min after regucalcin addition, while appreciable retardation was no longer seen at 5 min. Regucalcin (2.0 microM)-induced retardation of 45Ca2+ uptake was prevented by the presence of calmodulin (5 micrograms/ml). Calmodulin alone (1 and 5 micrograms/ml) caused a significant increase in 45Ca2+ uptake at 3 min after the start of incubation. Also, regucalcin (2.0 microM)-induced retardation of 45Ca2+ uptake was completely blocked by the presence of a Ca2(+)-trapping agent, oxalate (3 mM). On the other hand, 45Ca2+, which accumulated in microsomes during 5 min after ATP addition, was markedly released by the addition of regucalcin. This release was dose-dependent (0.5-4.0 microM). Guanosine triphosphate (GTP; 10-100 microM) caused a significant release of 45Ca2+ from the microsomes. The presence of regucalcin (2.0 microM) further enhanced the GTP effect. Regucalcin (2.0 microM)-induced release of 45Ca2+ was not blocked by the presence of the protein thiol-protecting agent dithiothreitol (0.1 mM). The presence of oxalate (3 mM) completely blocked the effect of regucalcin on 45Ca2+ release from the microsomes. These results indicate that regucalcin stimulates Ca2+ release from liver microsomes, and that the protein retards the microsomal Ca2+ uptake. The present study suggests that regucalcin can regulate the Ca2+ transport system in rat liver microsomes.  相似文献

利用全细胞膜片钳技术, 在急性分离的大鼠海马锥形细胞上研究了La3+对快速激活与失活的瞬时外向钾电流IA和平台电流IK的影响. 结果发现, 随着细胞外液中La 3+浓度的增加, La 3+对IA的抑制率逐渐增大, 而且具有电压依赖性; 10-5 mol/L La 3+可改变IA激活曲线和失活曲线的半数激活电压Vh值, 使激活曲线和失活曲线左移, 但不改变其斜率因子k值. 细胞外液中 La 3+对IK及其激活曲线都没有明显作用. 这可能是镧导致海马神经元异常的作用机制之一.  相似文献

The effect of heparin on the calcium-binding protein regucalcin-stimulated Ca2+ release from rat liver microsomes was investigated. Ca2+ release was assayed by the method of Millipore filtration to estimate microsomal 45Ca2+ accumulation following the addition of 10 mM adenosine triphosphate. The addition of regucalcin (1.0 microM) or inositol 1,4,5-trisphosphate [Ins(1,4,5)P3; 1.0 microM] stimulated 45Ca2+ release from rat liver microsomes. These effects were completely inhibited by the presence of heparin (10.0 micrograms/ml). Regucalcin did not enhance the effect of Ins(1,4,5)P3. These results suggest that regucalcin affects 45Ca2+ release involved in Ins(1,4,5)P3 action in rat liver microsomes.  相似文献

Guanidine has been used with some success to treat myasthenia gravis and myasthenic syndrome because it increases acetylcholine release at nerve terminals through K?, Na? and Ca2? channels-involving mechanisms. Currently, guanidine derivatives have been proposed for treatment of several diseases. Studies aimed at providing new insights to the drug are relevant. Experimentally, guanidine (10 mM) induces on mouse phrenic nerve-diaphragm (PND) preparations neurotransmission facilitation followed by blockade and a greatest secondary facilitation after its removal from bath. Herein, we hypothesized that this peculiar triphasic response may differ in muscles with distinct twitch/metabolic characteristics. Morphological alterations and contractile response of PND, extensor digitorum longus (EDL) and soleus (SOL) preparations incubated with guanidine (10 mM) for 15, 30, 60 min were analyzed. Guanidine concentrations of 5 mM (for PND and EDL) and 1 mM (for EDL) were also tested. Guanidine triphasic effect was only observed on PND regardless the concentration. The morphological alterations in muscle tissue varied along time but did not impede the PND post-wash facilitation. Higher doses (20-25 mM) did not increase EDL or SOL neurotransmission. The data suggest a complex mechanism likely dependent on the metabolic/contractile muscle phenotype; muscle fiber types and density/type of ion channels, sarcoplasmic reticulum and mitochondria organization may have profound impact on the levels and isoform expression pattern of Ca2? regulatory membrane proteins so reflecting regulation of calcium handling and contractile response in different types of muscle.  相似文献

聚丙烯酰胺(HPAM)是油田常用的驱油聚合物,用油田污水配制HPAM溶液易导致其黏度明显降低,影响驱油效果。依据埕东油田污水实测的各种金属阳离子含量来配制HPAM溶液,测得各金属阳离子对其黏度影响由大到小的顺序为:Na+>Fe2+>Ca2+>K+>Mg2+;通过红外光谱和扫描电镜分析金属阳离子导致HPAM溶液降黏的机理,Na+、K+、Ca2+、Mg2+主要是通过与HPAM链上的羧酸根阴离子静电引力相互作用,降低HPAM分子表面原有的电荷密度,造成分子链卷曲,同时减弱了极性基团的溶剂化能力,释放大量的"束缚水",从而使黏度显著降低;Fe2+离子主要是与水中溶解氧共同作用,引发自由基反应,导致HPAM骨架水解断裂,致使黏度显著降低。  相似文献
