A one-pot approach based upon the Povarov reaction has been efficiently employed with a number of exo-glycals and para-substituted benzanilines to synthesize novel open-ring, carbohydrate-derived quinolines. The mechanism of this reaction was studied and an explanation for the observed stereoselectivity is proposed. Treatment of the compounds with the Lewis acid, boron trichloride, successfully removes the benzyl ether protecting groups in good yields. Several of the prepared compounds have been screened in the National Cancer Institute’s (NCI’s) 60 cell line model. Moderate activity was observed for several leukemia cell lines.  相似文献

The synthesis is reported of novel P-stereogenic binaphthyl substituted monophosphines via a short five-step synthesis using a nickel coupling reaction with separation of the borane-protected diastereomeric products. Extensive coordination studies of these ligands with a number of well-known metal precursors were performed to more effectively understand their behaviour during catalysis.These ligands and some previously reported P-stereogenic ligands were tested in the rhodium catalysed asymmetric addition of phenyl boronic acid to napthaldehyde. These studies in asymmetric catalysis were used to compare the chiral induction of ligands that combine both axial and central chirality with ligands lacking P-stereogenicity.  相似文献

The bioluminescent system of the univalve shell Latia neritoides exhibits a luciferin-luciferase reaction. We study the enol formate structure of Latia luciferin, which is expected to be important for luminescent activity. The Latia luciferin analogues with an enol substituted benzoate moiety were synthesized and their bioluminescent activity was measured. The Latia luciferin benzoate analogues delay emission for natural luciferin in bioluminescence, indicating that the Latia bioluminescent activity can be controlled by the design of the enol ester.  相似文献

A rare silylative hydroxyalkylation of amide compounds with chiral aldehydes has been developed utilizing a Lewis acid-Lewis base promoter system consisting of an equimolecular mixture of tert-butyldimethylsilyl trifluoromethanesulfonate and N-diisopropylethylamine. This approach culminated in the synthesis of several enantiopure acyclic nucleoside representatives comprising thymidine analogues 6, 7, 9, 10, 12 and 13, uridine analogues 15 and 16, and 6-chloropurine derivatives 18 and 19.  相似文献

Treatment of 4-nitrophenyl chloroformate with alkylammonium hydrochloride salts and solid anhydrous Na2CO3 in either CH2Cl2 or CH3CN gave 4-nitrophenyl N-methylcarbamate and other N-alkylcarbamate analogues in excellent yields. Of particular interest is the observation that 4-nitrophenyl N-methylcarbamate, a safer alternative to the highly toxic methyl isocyanate, is obtained in quantitative yield (?95% pure as determined by 1H NMR) after simple filtration and solvent evaporation.  相似文献

Efficient methods of preparation of disubstituted exo-glycals by palladium cross-coupling reaction on the readily available dibromo exo-glycal and methoxycarbonyl exo-glycal have been developed. Hydrogenation of these new monosubstituted and disubstituted exo-glycals proceeded with a high stereocontrol and led to original C-glycosyl compounds.  相似文献

A new strategy has been designed for the preparation of gem-difluoro-bisarylic derivatives. It starts from easily accessible and reactive gem-difluoro-propargylic intermediates and elaborates the aromatic rings by a Diels-Alder-aromatization sequence. Heterocyclic systems can be also obtained by 1,3 dipolar cycloadditions, affording mixed aromatic/heteroaromatic derivatives with CF2 as a linker. Since this motif is a bioisostere of O and CO, corresponding bisarylic scaffolds could be of use to prepare chemical libraries of fluorinated analogues of bioactive natural products and/or drugs.  相似文献

A general method is presented for the preparation of tert-butyl esters by the gentle warming of the carboxylic acid in the presence of excess of tert-butyl acetoacetate and a catalytic amount of acid. Long-chain-branched poly(p-dioxanone)s (LCB-PPDOs) with different branch densities were prepared via the chain-extending reaction of hydroxyl group terminated linear bi-functional PPDO (2a-PPDO) and star-like tri-functional PPDO (3a-PPDO) prepolymers, which were synthesized by the ring-opening polymerization of p-dioxanone (PDO) using 1,4-butanediol (BD) and trimethylolpropane (TMP) as multi-functional initiators, respectively. The undesirable gelation was successfully depressed by adjusting the chain length and feed ratio of prepolymers. The average molecular weight between branch points (Mb) and the average number of branch per 100,000 g/mol (Bn) of LCB-PPDOs were calculated from the 1H NMR spectra. The average number of branch ranged from 0 to 6.72 branch points per 100,000 g/mol, and the number-average molecular weights between branch points ranged from 6900 to 20,500 g/mol. The results of differential scanning calorimetry (DSC) showed that the crystallization behavior of LCB-PPDOs was changed evidently with the branch density. Small-amplitude dynamic oscillatory rheometer was used to investigate the rheological properties of the melts of LCB-PPDO including zero-shear viscosity, storage modulus, relaxation times and loss angle, which largely depended on the branch density and length of LCB-PPDOs. Therefore, the rheological behaviors of PPDO can be well-controlled via synthesizing LCB-PPDOs with the desired architectures.  相似文献

An efficient synthesis of microtubule-stabilizing agent ceratamine A analogues is described. The key step is the application of either reductive Heck reaction or Heck reaction to construct the imidazo[4,5-d]azepine core. Thirteen novel analogues of ceratamine A were synthesized and evaluated for their in vitro cytotoxicity against five human cancer cell lines (HCT-116, HepG2, BGC-823, A549 and A2780). It was the first report about the systematic structural modification and SARs study of ceratamine A. The results demonstrated that bulky substituents at C-14 and C-16 could enhance the cytotoxicy and modification at N-7 was crucial for high potency. Especially compound 1f bearing methyl group at C-14 and C-16, and compound 1k bearing benzyl group on N-7, showed better cytotoxicy than ceratamine A against A549.  相似文献

The preparation of methyl (R)-o-chloromandelate via Ru-catalyzed asymmetric hydrogenation and transfer hydrogenation was investigated. With Ru-(R,R)-2,4,6-triisopropyl C6H2SO2-DPEN as the catalyst and HCOOH-Et3N azeotrope as the hydrogen donor, up to 92% ee was obtained in an optional condition. A detailed study, involving the synthesis of a single-source precursor containing two metal ions sharing the same crystallographic site, has been undertaken to elucidate the use of such a single-source precursor in a CVD process for growing thin films of oxides comprising these two metals, ensuring a uniform composition and distribution of metal ions. The substituted complexes Cr1−xAlx(acac)3, where acac = acetylacetonate, have been prepared by a co-synthesis method, and characterized using UV–Vis spectroscopy, TGA/DTA measurements, and single crystal X-ray diffraction at low temperature. All the studied compositions crystallize in the monoclinic space group P21/c with Z = 4 in the unit cell. It was observed that the ratio (Al:Cr) of the site occupancy for the metal ions, obtained from single crystal refinement, is in agreement with the results obtained from complexometric titrations. All the solid state structures have the metal in an octahedral environment forming six-membered chelate rings. M–O acac bond lengths and disorder in the terminal carbon have been studied in detail for these substituted metal–organic complexes. One composition among these was chosen to evaluate their suitability as a single-source precursor in a LPMOCVD process (low-pressure metal–organic chemical vapour deposition) for the deposition of a substituted binary metal oxide thin film. The resulting thin films were characterized by X-ray diffraction, scanning electron microscopy, and infrared spectroscopy.  相似文献

The enantioselective synthesis of the diterpenic moiety in the abeo-breviano skeleton is reported. The synthesis is carried out starting from 2-methyl-1,3-cyclohexanedione and EVK in eleven steps following a ring-expansion strategy once the tricyclic perhydrophenantrene skeleton has been obtained. A new Tiffeneau-Demjanov rearrangement under dark conditions is reported in which the insertion of the new methylene group is directed towards the most hindered side of the carbonyl group. This result is new and opposite to those reported in the literature under light (filtered or un-filtered wavelengths) conditions where insertion in the less hindered side is usually preferred.  相似文献

Hydroxylamine-O-sulfonic acid (HOSA) was used as an efficient nucleophilic amination reagent for 2-chloropyrimidines, 2-chloroquinolines, and 1-chloroisoquinoline. The newly obtained heteroaromatic hydroxylamine-O-sulfonates subjected to the reaction with acyl isothiocyanates underwent tandem nucleophilic addition-electrophilic 5-endo-trig cyclization. The mechanism of the cyclization was investigated with use of the long-range corrected hybrid density functional ωB97X-D/6-31+G and SM8 (DMF) solvation model. The structures of the heteroaromatic hydroxylamine-O-sulfonates and N-(5-methoxy-2H-[1,2,4]thiadiazolo[2,3-a]pyrimidin-2-ylidene)benzamide were confirmed by single crystal X-ray analysis. N-(2H-[1,2,4]thiadiazolo[3,2-a]isoquinolin-2-ylidene)benzamide exhibited a pronounced in vitro cytostatic activity against human tumor cell lines SISO, LCLC, A-427, DAN-G, and RT-4 (IC50 in the range 1.47-2.97 μM).  相似文献

β-CF3-α,β-diphenylvinyl sulfide 3a was prepared stereoselectively in 77% yield from the reaction of 2 with phenyllithium at room temperature for 5 h. Oxidation of 3a with MCPBA afforded the corresponding vinyl sulfone 4a, in which (E)-4a can be crystallized in a mixture of CH2Cl2 and hexane. The addition-elimination reaction of (E)-4a with phenyllithium having substituents on the benzene ring provided 5a-j in 51-82% yields stereospecifically. Similarly, the treatment of (E)-4a with p-chloroethoxyphenyllithium in the presence of 12-crown-4 (20 mol %) at −10 °C, followed by slowly warming to room temperature, resulted in the formation of the corresponding panomifene precursor 6 in 82% yield.  相似文献

The combination of the 5-N-tert-butoxycarbonyl (Boc) group of laurylthio sialoside and cyclopentyl methyl ether (CPME) as a solvent enhanced the reactivity and α-selectivity of the sialyl donor during sialylation. Selective deprotection of the N-Boc group of sialoside, including an acid-sensitive isopropylidene function, was successfully achieved by Yb(OTf)3-SiO2. Transformation of N,N-Ac,Boc into an N-acetylglycolyl group of sialoglycoside was easily performed via selective N-deacylation of the mixed Ac-N-Boc carbamate, subsequent Boc group removal, and acylation.  相似文献

This article presents a systematic analysis of the numerous intramolecular reactions of electronically excited alkenes categorized according to substance class and reaction type. The rearrangements and fragmentations observed in simple alkenes are usually rediscovered in non-conjugated and conjugated dienes and polyenes, where they are accompanied by reaction types such as rearrangements involving several double bonds, cyclizations, and intramolecular cycloadditions. Geometrical factors largely determine which reaction of a series of energetically feasible competing processes actually takes place. The industrial syntheses of vitamin D utilize many of the reaction types to be discussed.  相似文献

Three new sesquiterpenoids, a new unusual dimeric eudesmanolide, bialantolactone, a new nor-eudesmanolide, trinoralantolactone, and a new seco-guaiene, 7S,1(10)Z-4,5-seco-guaia-1(10),11-diene-4,5-dioxo, together with 13 known sesquiterpenoids, were isolated from the roots of Inula helenium. Their structures were elucidated by comprehensive spectroscopic analyses. The absolute configurations of bialantolactone and 7S,1(10)Z-4,5-seco-guaia-1(10),11-diene-4,5-dioxo were defined via the experimental and computational optical rotation and CD data. The plausible biosynthetic pathways to bialantolactone and 7S,1(10)Z-4,5-seco-guaia-1(10),11-diene-4,5-dioxo are discussed. 16 compounds were evaluated for their anti-bacterial activities against six bacteria.  相似文献

A new method was developed to obtain preparatively important quantities of β-pseudouridine, which was used as an intermediate.
